Vitamin D Receptor Polymorphisms and Cancer.

Increasing scientific evidence supports the link between vitamin D and cancer risk. The active metabolite 1,25(OH)2D exerts its activity by binding to the vitamin D receptor (VDR), an intracellular receptor that mediates transcriptional activation and repression of target genes. The binding of 1,25(OH)2D to VDR is able to regulate hundreds of different genes. VDR is active in virtually all tissues including the colon, breast, lung, ovary, bone, kidney, parathyroid gland, pancreatic b-cells, monocytes, T lymphocytes, melanocytes, keratinocytes, and also cancer cells. The relevance of VDR gene re

The genetics of hepatitis C virus underlie its ability to escape humoral immunity.

Hepatitis C virus (HCV) is a leading cause of chronic liver disease, and efforts to develop therapeutic vaccine strategies have been limited by immune escape due to HCV variants that are resistant to current vaccines or HCV variants that rapidly acquire new resistance-conferring mutations. Recently, the crystal structure of the viral envelope protein E2 region was resolved as well as how E2 docks to the host CD81 protein; therefore, antibodies that block this interaction should prevent viral entry into host cells. In this issue of the JCI, Bailey and colleagues show that immune escape of HCV c

Phylogenetic reconstruction of the Legionella pneumophila Philadelphia-1 laboratory strains through comparative genomics.

Over 20 years ago, two groups independently domesticated Legionella pneumophila from a clinical isolate of bacteria collected during the first recognized outbreak of Legionnaires' disease (at the 1976 American Legion's convention in Philadelphia). These two laboratory strains, JR32 and Lp01, along with their derivatives, have been disseminated to a number of laboratories around the world and form the cornerstone of much of the research conducted on this important pathogen to date. Nevertheless, no exhaustive examination of the genetic distance between these strains and their clinical progenito

IL1A-889 C/T gene polymorphism in irritant contact dermatitis.

BACKGROUND: Upon skin contact to irritants, interleukin-1 alpha (IL-1α) is released in the stratum corneum as a primary step of skin inflammation. Variations in the IL-1A gene have been shown to alter the expression of IL-1α. This may influence the susceptibility to skin inflammation and the development of irritant contact dermatitis (ICD). OBJECTIVE: To determine effects of an IL1A-889 C/T polymorphism in view of susceptibility to develop irritant contact dermatitis. METHODS: In a case-control study, 478 Caucasian patients with occupational ICD of the hands were genotyped for an IL1A-889 C/

The contribution of different prion protein types and host polymorphisms to clinicopathological variations in Creutzfeldt-Jakob disease.

Creutzfeldt-Jakob disease is a fatal neurodegenerative disease that primarily affects the central nervous system. In this respect, it can be considered alongside the more frequently occurring neurodegenerative diseases, such as Alzheimer's disease. Creutzfeldt-Jakob disease is perhaps the paradigmatic protein misfolding disorder, so comparisons between the mechanisms involved in Creutzfeldt-Jakob disease and other neurodegenerative diseases associated with protein misfolding (such as the tauopathies and synucleinopathies) may also be informative. Like many of these diseases, Creutzfeldt-Jakob

Beneficial laggards: multilevel selection, cooperative polymorphism and division of labour in threshold public good games.

BACKGROUND: The origin and stability of cooperation is a hot topic in social and behavioural sciences. A complicated conundrum exists as defectors have an advantage over cooperators, whenever cooperation is costly so consequently, not cooperating pays off. In addition, the discovery that humans and some animal populations, such as lions, are polymorphic, where cooperators and defectors stably live together--while defectors are not being punished--, is even more puzzling. Here we offer a novel explanation based on a Threshold Public Good Game (PGG) that includes the interaction of individual an

[Genetic basis of dyslipidemia].

Genetic disorders play an important role in the ethiopathogenesis of disease processes in humans. In the present paper we have described documented genes polymorphisms associated with dyslipidemia. Some of them have been known for years, but some have been discovered quite recently. Their characteristics an impact on lipid profile are presented. Discussed are their direct relationship with atherosclerosis and cardiovascular diseases.

Polymorphism of the angiotensin-converting enzyme gene and cardiovascular disease.

The angiotensin-converting enzyme (ACE) is an integral part of two enzymatic cascades, one leading to the generation of angiotensin II and the other to the degradation of bradykinin. The great variety of cardiovascular effects mediated by these vasoactive peptides and the efficacy of ACE inhibitors in the treatment of hypertension and heart failure emphasize the prominent role of ACE in the cardiovascular system. Early in this decade convincing experimental evidence demonstrated the induction of this enzyme in several pathophysiological conditions, including myocardial infarction and left vent

A novel polymorphism in the coding region of the vasopressin type 2 receptor gene.

Nephrogenic diabetes insipidus (NDI) is a rare disease characterized by renal inability to respond properly to arginine vasopressin due to mutations in the vasopressin type 2 receptor (V2(R)) gene in affected kindreds. In most kindreds thus far reported, the mode of inheritance follows an X chromosome-linked recessive pattern although autosomal-dominant and autosomal-recessive modes of inheritance have also been described. Studies demonstrating mutations in the V2(R) gene in affected kindreds that modify the receptor structure, resulting in a dys- or nonfunctional receptor have been described,

Investigation on dimorphism of Blastomyces dermatitidis by agar-implantation method.

By the agar-implantation developed by the authors the process of conversion on Blastomyces dermatitidis from mycelial phase to yeast phase was observed. First of all slide cultures of the fungus were prepared at room temperature. Upon confirmation of good hyphal growth, a cover glass was removed and a part of medium was cut out in a square of about 3 mm a side. After mice were laparotomied, each agar block cut out was implanted in the peritoneal cavity of mouse. The mice implanted with the agar blocks were killed, two each, every day for 14 days, and thereafter at intervals of a week for 2 mon

[Association polymorphic variants of GRIN2B gene with paranoid schizophrenia and response to common neuroleptics in Russians and Tatars from Bashkortostan Republic].

An analysis of the association of paranoid schizophrenia seeking with polymorphic variants of GRIN2B gene was performed in order to identify genetic risk factors of disease development and genetic markers of the response to therapy by neuroleptics in Russian and Tatar patients from Bashkortostan Republic (BB). In the course of the analysis, we revealed the following: 1) genetic markers of increased risk of developing paranoid schizophrenia in various ethnic groups, including, in Tatars, the GRIN2B* T/*Tgenotype (p = 0.003; OR = 2.33) and GRIN2B*T allele (p = 0.001; OR = 2.36), rs1805247; in Ru

Cytokine Gene Polymorphisms Modulate Isohemagglutinin Titers and Classes: Another Aspect Towards the Link Between ABO Groups and Human Pathologies?

Anti-A and anti-B antibodies are essential for monitoring adverse reactions in organ transplants and transfusions. However, their importance is also growing due to their involvement in the pathophysiology of various human diseases, such as infections, although this is currently the subject of heated debate. A characteristic heterogeneity in the titers and classes of anti-A and anti-B antibodies is observed among individuals. Several factors appear to be responsible, such as everyone's specific immune profile, age, sex, microbiota composition, lifestyle, and health status. The immune profile, t
