Coronary heart disease (CHD) is closely associated with lipid metabolic disorders, inflammatory activation, and myocardial energy imbalance. Heart-type fatty acid-binding protein (H-FABP), a major intracellular fatty acid transporter in cardiomyocytes, plays a critical role in fatty acid trafficking, mitochondrial β-oxidation, and lipid signaling. Under pathological conditions such as hypoxia, oxidative stress, and inflammation, H-FABP dysfunction disrupts lipid homeostasis, leading to lipotoxicity, mitochondrial impairment, endothelial injury, and microcirculatory dysfunction. These pathologi

Cytoplasmic Dynein-2 / IFT-dynein is the only known retrograde motor for intraflagellar transport. Dysfunction of the two intermediate dynein chains WDR34 and WDR60 causes Short Rib Thoracic Dystrophy (SRTD), a human skeletal chondrodysplasias with high lethality. However, individual protein functions are incompletely understood as complete loss of function of WDR34 or WDR60 is lethal in vertebrates and individuals with SRTD carry at least one putative hypomorphic missense allele. Gene knockout is therefore not suitable to study the effect of these human missense disease alleles. Therefore, us

Microfold cells (M cells) in the gut serve as a key gateway to enhance the potency of oral vaccines. Although natural polysaccharides show promise as carriers, their development is limited by inherent drawbacks such as scarce availability and lack of intrinsic targeting capacity. Therefore, it is essential to identify suitable materials from well-defined, commercially available polysaccharide standards. In this study, a library of well-defined, high-purity commercial polysaccharides was screened, and identified Xylan as a lead candidate with superior M cell transport efficiency in an in vitro

Mitochondrial dysfunction contributes to numerous human diseases. Voltage‑dependent anion channel 2 (VDAC2) is an essential outer mitochondrial membrane porin with distinct structural and functional properties that are non-redundant with those of VDAC1 and VDAC3. VDAC2 precisely controls ATP, ADP, NADPH and Ca2+ transport, thereby acting as a key hub for energy metabolism, calcium homeostasis, redox balance and cell fate. It directly binds BAX/BAK to differentially regulate apoptosis and is also involved in ferroptosis, necroptosis, Parkin‑dependent mitophagy, and lipid transport through prote

The advent of nanotechnology in biotechnology and medicine has accelerated the development of nanomaterials as innovative tools for diagnostics, therapy, and drug delivery. Despite their versatility, many nanoparticle systems fail in vivo due to their limited ability to traverse biological barriers, resulting in off-target distribution, rapid clearance, or undesired bioaccumulation. Structural parameters-including size and surface chemistry-critically shape nanoparticle biodistribution and fate. Albumin, the most abundant serum protein, naturally acts as a long-circulating carrier capable of t

Water exchange between cerebrospinal fluid (CSF) and blood is fundamental to brain fluid homeostasis, yet ventricular CSF-to-blood water transport has never been quantified as a physiological parameter in adult humans. We investigated ventricular water absorption kinetics in 40 adults undergoing diagnostic evaluation for hydrocephalus. During ventricular infusion testing, 15O-labelled water was administered intraventricularly via an external ventricular drain, with continuous jugular bulb blood sampling and simultaneous intracranial pressure (ICP) monitoring. Ventricular water absorption was e

High-density lipoprotein cholesterol (HDL-C) has long been inversely associated with atherosclerotic cardiovascular disease risk, but pharmacologic efforts to raise HDL-C have consistently failed to reduce cardiovascular events. This has shifted focus from HDL quantity to quality, emphasizing functional properties such as cholesterol efflux, antioxidative capacity, and anti-inflammatory activity. Dysfunctional HDL, often modified by oxidative and inflammatory processes mediated by myeloperoxidase, loses its ability to promote reverse cholesterol transport, support endothelial function, and sup

Despite the known benefits of exercise, an in-depth comparison of the effects of regular versus irregular exercise on brain-body interactions and molecular mechanisms remains lacking. This study demonstrates that regular exercise robustly enhances fear memory extinction in mice, whereas irregular exercise has only mild effects. This divergence arises from regular rather than irregular exercise strongly inhibiting axonal mRNA transport and local translation of choline acetyltransferase (ChAT) in septal cholinergic neurons projecting to the hippocampus, thereby reducing hippocampal acetylcholine

Angiotensin-converting enzyme inhibitory peptides (ACEIPs) from natural food sources are effective alternatives for treating hypertension. This study aims to screen highly active ACEIPs from yellow tuna peptides, and to elucidate their inhibitory mechanism, stability, transport mechanism and potential antihypertensive mechanism by a multiple approach. Two novel ACEIPs STHPHF (IC50 = 30.71 μg/mL) and LTFSY (IC50 = 20.72 μg/mL) were screened by bio-activity guided approach and de novo sequencing. Inhibition kinetics and molecular docking indicated that STHPHF demonstrated a mixed inhibitor, wher

Regulating the microscale carrier transport in semiconductor materials to enhance the macroscopic photoelectrochemical properties would drive technological advancement and industrial upgrading. In this work, we integrated the maturity of microfluidic technology control methods with current photoelectrochemical analysis sensing techniques to establish a microfluidic photoelectrochemical induction system by modulating the photon-to-current efficiency under zero bias. Traditionally, target recognition has been achieved by affecting the transport of carriers through the space resistance equivalent

Persistent synovitis is a pivotal pathological feature of rheumatoid arthritis (RA). However, the current rheumatoid drugs are accompanied by severe side effects and have limited anti-inflammatory capabilities. In this work, we designed a bioactive material-folic acid modified layered double hydroxides (FA-LDH), aiming at targeting M1 macrophages and modulating macrophage repolarization. The in vitro experiment showed that FA-LDH mitigated the release of proinflammatory cytokines and promoted the expression of M2 macrophage markers. In terms of the action mechanism, FA-LDH modulated the nucleo

BACKGROUND: The neuromuscular junction (NMJ) is the synapse between motor neurons and skeletal muscle and controlls movement. Impaired synaptic transmission and NMJ degeneration has been observed during healthy ageing and is also implicated in several neuromuscular diseases. On account of the high energy demands of being distally located and large sized, NMJs are enriched with mitochondria. This enrichment is dependent on transport of mitochondria across the axon to the NMJ. METHODS: We first established a human 3D neuromuscular assembloid model to study in-vitro NMJs, by fusing human stem cel

The ATP6V0A4 gene encodes the a4 subunit of vacuolar H+-ATPase (V-ATPase), which mediates hydrogen ion transport across the membrane. Previous studies have suggested that mutations in ATP6V0A4 consistently result in a loss of function, impairing the hydrogen ion transport efficacy of V-ATPase and leading to distal renal tubular acidosis and sensorineural hearing loss. Here, we identified a 32-year-old male patient and his father, both of whom harbored a heterozygous ATP6V0A4 p.V512L mutation and exhibited hypochloremic metabolic alkalosis, acidic urine, and hypokalemia. Through a series of pro

Cystinosis is a rare, autosomal recessive, lysosomal storage disease caused by mutations in the gene CTNS, leading to cystine accumulation in the lysosomes. While cysteamine lowers the cystine levels, it does not cure the disease, suggesting that CTNS exerts additional functions besides cystine transport. This study investigated the impact of infantile and juvenile CTNS mutations with discrepant genotype/phenotype correlations on CTNS expression, and subcellular localisation and function in clinically relevant cystinosis cell models to better understand the link between genotype and CTNS funct

PURPOSE: FLVCR1 encodes a solute carrier protein implicated in heme, choline, and ethanolamine transport. Although Flvcr1-/- mice exhibit skeletal malformations and defective erythropoiesis reminiscent of Diamond-Blackfan anemia (DBA), biallelic FLVCR1 variants in humans have previously only been linked to childhood or adult-onset ataxia, sensory neuropathy, and retinitis pigmentosa.
METHODS: We identified individuals with undiagnosed neurodevelopmental disorders and biallelic FLVCR1 variants through international data sharing and characterized the functional consequences of their FLVCR1 vari

Fatty acid-binding protein 7 (FABP7) is vital for uptake and trafficking of fatty acids in the nervous system. To investigate the involvement of FABP7 in noise-induced hearing loss (NIHL) pathogenesis, we used Fabp7 knockout (KO) mice generated via CRISPR/Cas9 in the C57BL/6 background. Initial auditory brainstem response (ABR) measurements were conducted at 9 weeks, followed by noise exposure at 10 weeks. Subsequent ABRs were performed 24 h later, with final measurements at 12 weeks. Inner ears were harvested 24 h after noise exposure for RNA sequencing and metabolic analyses. We found no sig

Diffusion Tensor Imaging (DTI) and Manganese Enhanced MRI (MEMRI) are noninvasive tools to characterize neural fiber microstructure and axonal transport. A combination of both may provide novel insights into the progress of neurodegeneration. To investigate the relationship of DTI and MEMRI in white matter of tauopathy, twelve optic nerves of 11-month-old p301L tau mice were imaged and finished with postmortem immunohistochemistry. MEMRI was used to quantify Mn2+ accumulation rates in the optic nerve (ON, termed ONAR) and the Superior Colliculus (SC, termed SCAR), the primary terminal site of

Rising prevalence of type 2 diabetes mellitus (T2DM) in sub-Saharan Africa has necessitated surveys of antidiabetic medicinal plants. This study assessed the antidiabetic mechanism of Terminalia catappa aqueous leaf extract (TCA) in high fat/low dose streptozotocin-induced type 2 diabetic rats. T2DM was induced by a combination of high-fat diet and low dose STZ (30 mg/kg bw) and the animals were administered with TCA (400 and 800 mg/kg bw) orally daily for 28 days. Biochemical parameters and indices for diabetes including renal function tests and pancreatic histology were evaluated. Relative e
