Caveolin-1 stabilizes ATP7A, a copper transporter for extracellular SOD, in vascular tissue to maintain endothelial function.

Caveolin-1 (Cav-1) is a scaffolding protein and a major component of caveolae/lipid rafts. Previous reports have shown that endothelial dysfunction in Cav-1-deficient (Cav-1-/-) mice is mediated by elevated oxidative stress through endothelial nitric oxide synthase (eNOS) uncoupling and increased NADPH oxidase. Oxidant stress is the net balance of oxidant generation and scavenging, and the role of Cav-1 as a regulator of antioxidant enzymes in vascular tissue is poorly understood. Extracellular SOD (SOD3) is a copper (Cu)-containing enzyme that is secreted from vascular smooth muscle cells/fib

Multi-Autoantibody Signature and Clinical Outcome in Membranous Nephropathy.

BACKGROUND AND OBJECTIVES: Patients with membranous nephropathy can have circulating autoantibodies against membrane-bound (phospholipase A2 receptor 1 [PLA2R1] and thrombospondin type-1 domain containing 7A [THSD7A]) and intracellular (aldose reductase, SOD2, and α-enolase) podocyte autoantigens. We studied their combined association with clinical outcomes. DESIGN, SETTING, PARTICIPANTS, & MEASUREMENTS: Serum levels of anti-PLA2R1, anti-THSD7A, anti-aldose reductase, anti-SOD2, and anti-α-enolase autoantibodies were determined in 285 patients at diagnosis and during follow-up using standardi

Local injection of mesenchymal stem cells protects testicular torsion-induced germ cell injury.

INTRODUCTION: Testicular torsion is a urological emergency and infertility is a common complication due to ischemic injury. Surgical reduction and orchiopexy is indicated, but to date there is no effective method for restoration of spermatogenesis. The effects of mesenchymal stem cells (MSCs) on acute tissue injury have been demonstrated, and the abilities of paracrine support, differentiation and immune-modulation may benefit to testicular torsion-induced infertility. We investigate the therapeutic efficacy and the mechanisms of MSCs in testicular torsion-induced germ cell injury when injecte

Immunization with a Myelin-Derived Antigen Activates the Brain's Choroid Plexus for Recruitment of Immunoregulatory Cells to the CNS and Attenuates Disease Progression in a Mouse Model of ALS.

Amyotrophic lateral sclerosis (ALS) is a devastating fatal motor neuron disease, for which there is currently no cure or effective treatment. In this disease, local neuroinflammation develops along the disease course and contributes to its rapid progression. In several models of CNS pathologies, circulating immune cells were shown to display an indispensable role in the resolution of the neuroinflammatory response. The recruitment of such cells to the CNS involves activation of the choroid plexus (CP) of the brain for leukocyte trafficking, through a mechanism that requires IFN-γ signaling. He

Fasudil, a rho kinase inhibitor, limits motor neuron loss in experimental models of amyotrophic lateral sclerosis.

BACKGROUND AND PURPOSE: Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disorder with no effective treatment. Fasudil hydrochloride (fasudil), a potent rho kinase (ROCK) inhibitor, is useful for the treatment of ischaemic diseases. In previous reports, fasudil improved pathology in mouse models of Alzheimer's disease and spinal muscular atrophy, but there is no evidence in that it can affect ALS. We therefore investigated its effects on experimental models of ALS. EXPERIMENTAL APPROACH: In mice motor neuron (NSC34) cells, the neuroprotective effect of hydroxyfasudil (M3), an

Sensitivity of liver injury in heterozygous Sod2 knockout mice treated with troglitazone or acetaminophen.

Recently, it was reported that the intraperitoneal administration of 30 mg/kg/day troglitazone to heterozygous superoxide dismutase 2 gene knockout (Sod2+/-) mice for twenty-eight days caused liver injury, manifested by increased serum ALT activity and hepatic necrosis. Therefore, we evaluated the reproducibility of troglitazone-induced liver injury in Sod2+/- mice, as well as their validity as an animal model with higher sensitivity to mitochondrial toxicity by single-dose treatment with acetaminophen in Sod2+/- mice. Although we conducted a repeated dose toxicity study in Sod2+/- mice treate

Ro5-4864 promotes neonatal motor neuron survival and nerve regeneration in adult rats.

The translocator protein (18 kDa; TSPO), formerly known as the peripheral benzodiazepine receptor, is an outer mitochondrial membrane protein that associates with the mitochondrial permeability transition pore to regulate both steroidogenesis and apoptosis. TSPO expression is induced in adult dorsal root ganglion (DRG) sensory neurons after peripheral nerve injury and a TSPO receptor ligand, Ro5-4864, enhances DRG neurite growth in vitro and axonal regeneration in vivo. We have now found that TSPO is induced in neonatal motor neurons after peripheral nerve injury and have evaluated its involve

Suppression of mitochondrial oxidative stress provides long-term neuroprotection in experimental optic neuritis.

PURPOSE: Axonal loss is thought to contribute to the persistence of visual loss in optic neuritis and multiple sclerosis (MS). The mechanisms of injury are poorly understood. The authors investigated the contribution of mitochondrial oxidative stress and the effects of modulating mitochondrial antioxidant gene expression in the optic nerves of mice induced with experimental allergic encephalomyelitis (EAE), with a focus on long-term neuroprotection. METHODS: Optic nerves from mice with EAE were probed for reactive oxygen species (ROS) with the use of dichlorofluorescein diacetate (DCFDA), dih

Selective vulnerability and pruning of phasic motoneuron axons in motoneuron disease alleviated by CNTF.

Neurodegenerative diseases can have long preclinical phases and insidious progression patterns, but the mechanisms of disease progression are poorly understood. Because quantitative accounts of neuronal circuitry affected by disease have been lacking, it has remained unclear whether disease progression reflects processes of stochastic loss or temporally defined selective vulnerabilities of distinct synapses or axons. Here we derive a quantitative topographic map of muscle innervation in the hindlimb. We show that in two mouse models of motoneuron disease (G93A SOD1 and G85R SOD1), axons of fas

The expression of the chemorepellent Semaphorin 3A is selectively induced in terminal Schwann cells of a subset of neuromuscular synapses that display limited anatomical plasticity and enhanced vulnerability in motor neuron disease.

Neuromuscular synapses differ markedly in their plasticity. Motor nerve terminals innervating slow muscle fibers sprout vigorously following synaptic blockage, while those innervating fast-fatigable muscle fibers fail to exhibit any sprouting. Here, we show that the axon repellent Semaphorin 3A is differentially expressed in terminal Schwann cells (TSCs) on different populations of muscle fibers: postnatal, regenerative and paralysis induced remodeling of neuromuscular connections is accompanied by increased expression of Sema3A selectively in TSCs on fast-fatigable muscle fibers. White Feces Syndrome is closely linked to impaired antioxidant capacity, the aquaculture environment, and microbial dysbiosis in Litopenaeus vannamei reared in small-pond culture system.

BACKGROUND: White feces syndrome (WFS) is a major health problem affecting intensive shrimp aquaculture, but its underlying mechanisms remain unclear. Increasing evidence suggests that WFS may involve complex interactions among host physiological status, intestinal microbial dysbiosis, and environmental microbial communities. However, the relationships among these factors within commercial shrimp culture systems remain poorly understood. OBJECTIVES: This study aimed to investigate the associations among intestinal antioxidant capacity, intestinal microbiota, sediment microbial communities, an

Feeding Effect and Antioxidant Capacity of Fermentation Broths Based on Strain-Specific Utilization of Poria cocos Polysaccharide Derivatives.

This study investigated the prebiotic properties of the supernatant from gluconic acid-hydrolyzed alkali-soluble Poria cocos polysaccharide (s-GA-PCP). s-GA-PCP directly promoted the proliferation of Bifidobacterium and Lactobacillus strains. Among the s-GA-PCP-utilizing strains, strain L10 (Bifidobacterium pseudolongum) exhibited the broadest feeding capability, as its fermentation broth supported the growth of all six non-utilizing strains. Analyses revealed that L10 produced a more diverse oligosaccharide profile and accumulated higher levels of amino acid and peptide metabolites. Furthermo

LcRAV2 regulates drought resistance in litchi (Litchi chinensis Sonn) through a potential LcSnRK1-mediated phosphorylation.

Drought stress severely affects the flowering and fruit set of litchi, while the molecular mechanism of the RAV transcription factor in litchi drought resistance remains unclear. In this study, we systematically evaluated the leaf drought tolerance characteristics of 32 litchi cultivars and identified highly dehydration tolerant cultivars including 'Hongdenglong' and 'Nuomici', a moderately dehydration tolerant cultivar 'Qingrenli', as well as weakly dehydration tolerant cultivars 'Nanjuhong' and 'Hongxiuqiu'. Physiological analysis showed that litchi leaves cope with drought by increasing the

Perfluorohexane sulfonate (PFHxS)-induced energy metabolism disruption in Chlorella pyrenoidosa: Integrating physiological and transcriptomic insights.

Perfluorohexane sulfonate (PFHxS), a widely used alternative to perfluorooctane sulfonate (PFOS), has been prevalent in aquatic environments. However, its toxic mechanisms in algae remain poorly understood. In this study, a 12-day exposure experiment was conducted to systematically investigate the toxicity of PFHxS in Chlorella pyrenoidosa through physiological parameter measurements and transcriptomic analysis. The results revealed that significant growth inhibition (9%) was observed at 1 mg/L PFHxS on day 12, accompanied by altered photosynthetic responses, including decreases in chlorophyll

Recycling waste, recycling risk: cypermethrin-contaminated poultry litter triggers sublethal toxicity in Folsomia candida at realistic environmental concentrations.

This study for the first time investigated the effects of poultry litter containing cypermethrin (740 μg kg-1 d.w.) and metals on Folsomia candida, assessing lethality, behavior, reproduction, and oxidative stress biomarkers, which were combined into an Integrated Biomarker Response (IBR) assessment. The sublethal effects covered application rates equivalent to those reported for different cropping systems (2.0-25.0 t ha-1). Compared to control soil, poultry litter showed elevated concentrations in mg kg-1 of Cu (27.5), Zn (36.6), Mn (37.7), and Cr (0.93). The median lethal concentrations (LC5

The hypothalamus is an early site of mitochondrial failure and neuro-immune circuit disruption in amyotrophic lateral sclerosis.

BACKGROUND: Metabolic dysfunction is a defining feature of amyotrophic lateral sclerosis (ALS), emerging early and strongly associated with disease progression and prognosis. While systemic hypermetabolism is well documented, the central mechanisms underlying energy imbalance remain poorly understood. The hypothalamus, a key regulator of whole-body energy homeostasis, has recently been implicated in ALS, but its mechanistic contribution to metabolic failure and disease progression remains unclear. METHODS: We analyzed the hypothalamus SOD1-G93A mouse model using proteomics (ProteomeXchange ID

The evaluation to combined toxic effects of polypropylene microplastics and benzo[a]pyrene on macrophage cells.

Microplastics (MPs) pollution has emerged as a significant concern for human health, and their toxicological mechanisms help to better understand their potential risks. Previous studies paid main attention on the toxicity of MPs themselves, but in recent years, there has been growing awareness of their combined effects with other environmental pollutants. This study investigated the toxic effects of polypropylene microplastics (PP MPs) prepared via nonsolvent induced phase separation method in conjunction with benzo[a]pyrene (BaP) stimulation in vitro to macrophagocytes based on toxicoproteomi

Six new 8-O-4'-linked sesquineolignans from the stems of Kadsura longipedunculata and their neuroprotective effects against 6-OHDA-induced injury in PC12 cells.

Six previously undescribed sesquineolignans, longipedunlignans A-F (1-6), were isolated from the stems of Kadsura longipedunculata. Their structures were elucidated by extensive spectroscopic analysis, including HRESIMS and 1D and 2D NMR data. Compounds 1-6 possess closely related sesquineolignan frameworks composed of three C6-C3 phenylpropanoid units connected through two consecutive 8-O-4' ether linkages. All isolates were evaluated for neuroprotective effects against 6-OHDA-induced injury in PC12 cells. Compounds 3 and 4 exhibited the most pronounced protective effects and attenuated oxida

LhCAT2-mediated crosstalk between reactive oxygen species homeostasis and anthocyanin biosynthesis enhances zinc tolerance in Liriodendron hybrid.

Excessive zinc (Zn) accumulation in soils impairs plant growth and threatens ecosystem sustainability. Although catalase (CAT) enzymes are central to antioxidant defense, their coordination with secondary metabolism in woody plants under heavy metal stress remains poorly understood. Here, we characterized LhCAT2, a Zn-inducible catalase gene from Liriodendron hybrid, using three Arabidopsis overexpression lines and two Liriodendron hybrid overexpression lines under controlled Zn treatments. Under Zn stress, LhCAT2 overexpression increased Arabidopsis germination rate by 22%, primary root lengt
