Productive infection of CD4+ and CD8+ mature human T cell populations and clones by human herpesvirus 6. Transcriptional down-regulation of CD3.

The susceptibility to infection by human herpes-virus 6 (HHV-6) of mature human T lymphocytes belonging to the two major subpopulations (i.e., CD3+ CD4+ CD8- and CD3+ CD4- CD8+) was investigated by using CD4+ or CD8+ T cell populations and clones derived from normal adult peripheral blood. Productive HHV-6 infection was observed in both CD4+ and CD8+ T cells. By days 2 to 6 after infection, increasing numbers of cells exhibited characteristic morphologic alterations, becoming enlarged, uniformly rounded and refractile as a consequence of the virus-induced cytopathic effect. During the course o

Robust CD4+ CAR T cell expansion is associated with non-ICANS neurotoxicities after ciltacabtagene autoleucel in patients with multiple myeloma.

Nonimmune effector cell-associated neurotoxicity syndrome neurotoxicities (NINTs) are serious, atypical toxicities associated with ciltacabtagene autoleucel (cilta-cel), a US Food and Drug Administration chimeric antigen receptor T cell (CAR T cell) therapy approved for relapsed/refractory multiple myeloma (RRMM). Risk factors contributing to the development of NINTs are poorly understood. In a cohort of 109 patients with RRMM treated with cilta-cel, we identify predisposing risk factors and propose strategies to mitigate NINTs. We show that high-peak absolute lymphocyte count is a strong NINT

Lung mucosal and systemic responses at single-cell resolution in an aerosolized Mycobacterium bovis BCG human challenge model.

Tuberculosis (TB) vaccine development is hindered by limited understanding of human immune responses to Mycobacterium tuberculosis. Using an aerosolized Mycobacterium bovis BCG human challenge model, we perform single-cell and bulk immune profiling of bronchoalveolar lavage and blood samples to define the temporal immune dynamics in BCG-naive individuals. Rapid changes in cellular composition and gene expression occur in both compartments, with TB-associated gene signatures evident on days 2 and 7 post-challenge. T cell receptor clones that expand in blood at day 7 persist in the lung mucosa u

Characterization of Human CD4 T Cells Specific for a C-Peptide/C-Peptide Hybrid Insulin Peptide.

Hybrid Insulin Peptides (HIPs), which consist of insulin fragments fused to other peptides from β-cell secretory granule proteins, are CD4 T cell autoantigens in type 1 diabetes (T1D). We have studied HIPs and HIP-reactive CD4 T cells extensively in the context of the non-obese diabetic (NOD) mouse model of autoimmune diabetes and have shown that CD4 T cells specific for HIPs are major contributors to disease pathogenesis. Additionally, in the human context, HIP-reactive CD4 T cells can be found in the islets and peripheral blood of T1D patients. Here, we performed an in-depth characterization

Clinicopathological analysis of primary cutaneous CD4-positive small/medium pleomorphic T-cell lymphoproliferative disorder: a retrospective study of 22 patients.

BACKGROUND: Primary cutaneous CD4-positive small/medium pleomorphic T-cell lymphoproliferative disorder has been defined as a type of lymphoproliferative disorder with indolent clinical course and excellent prognosis, yet a precise diagnosis is still hard to reach. METHODS: A retrospective analysis of 22 patients including 16 females and six males was performed. RESULTS: The age of patients ranged from 5 to 79 years. The average age of all patients was 43.5, and the median age of all patients was 44.5. Two patients had multiple lesions, and others were presented with a solitary asymptomatic

CD4+ CADM1+ cell percentage predicts disease progression in HTLV-1 carriers and indolent adult T-cell leukemia/lymphoma.

We recently took advantage of the universal expression of cell adhesion molecule 1 (CADM1) by CD4+ cells infected with HTLV-1 and the downregulation of CD7 expression that corresponds with the oncogenic stage of HTLV-1-infected cells to develop a flow cytometric system using CADM1 versus CD7 plotting of CD4+ cells. We risk-stratified HTLV-1 asymptomatic carriers (AC) and indolent adult T-cell leukemia/lymphoma (ATL) cases based on the CADM1+ percentage, in which HTLV-1-infected clones are efficiently enriched. AC and indolent ATL cases were initially classified according to their CADM1+ cell p

Therapeutic hotline: Primary cutaneous CD4 + small/medium-sized pleomorphic T cell lymphoma coexisting with myelodysplastic syndrome transforming into chronic myelomonocytic leukemia successfully treated with cyclophosphamide.

Cutaneous T cell lymphomas other than mycosis fungoides, Sezary syndrome, and primary cutaneous CD30+ lymphoproliferations constitute less than 10% of all cutaneous T cell lymphomas. Primary cutaneous small/medium CD4+ T cell lymphoma is a member of this third group of cutaneous lymphomas, separated out as provisional entity in the World Health Organization classification - European Organization for Research and Treatment of Cancer (WHO-EORTC) classification. It still awaits development of more precise diagnostic criteria and optimal therapy. We report a case of primary cutaneous CD4 + small/m

Viral load of the human immunodeficiency virus could be an independent risk factor for endothelial dysfunction.

BACKGROUND: Recent reports of myocardial infarction in young persons infected with human immunodeficiency virus (HIV) who are receiving protease inhibitor therapy have raised concerns about premature coronary artery disease in this population. However, endothelial dysfunction, hypercoagulability, hypertriglyceridemia, and abnormal coronary artery pathology have been observed in association with HIV infection prior to the availability of protease inhibitor therapy. HYPOTHESIS: The study was undertaken to determine the association between endothelial function, viral load, CD4+ count, and other

A Human-Based Skin-Lymphoreticular Model-on-Chip to Emulate Inflammatory Skin Conditions.

Despite fast-paced advances in bioengineering human-based models, emulating fine-tuned and interdependent immunoregulations of the human body remains challenging. While many bioengineered models contain immune cells, secondary lymphoid tissues are typically missing. Here, we report a self-assembled, minimally functional lymphoreticular unit composed of fibroblastic reticular, lymphatic endothelial and CD4+ T cells that emulate structural and functional interactions with the skin under inflammatory conditions. Following the optimization of the culture conditions, a layer-by-layer approach combi

ZEB2 orchestrates a transcriptional program to safeguard the integrity of human CD4+ Th1 effector memory cells.

CD4+ Th1 cells migrate to sites of inflammation, where they are indispensable for eliminating intracellular pathogens. The lineage-defining transcription factor T-bet establishes the T-helper 1 (Th1) transcriptional program, directing IFN-γ to drive effector responses and inducing subordinate transcription factors to shape the Th1 phenotype. Aberrant Th1 cell activity drives the pathogenesis of multiple autoimmune diseases, but the detailed mechanisms by which Th1 cells maintain or lose their integrity remain largely uncharacterized. Using immunogenomics and high-resolution immune phenotyping

Butyrate epigenetically licenses sustained epithelial-T cell crosstalk for intestinal immune tolerance.

Intestinal epithelial cells (IEC) are the primary cell type in direct contact with stimuli from the luminal microbiota, playing a critical role in host-microbe interactions. However, how IECs communicate with underlying immune cells to maintain homeostasis remains poorly understood. In addition, the mechanisms by which IECs sense microbiota-derived stimuli to initiate this crosstalk are not yet fully understood. Here, we demonstrate that oral administration of the gut microbiota metabolite butyrate induces sustained IL-10 production in CD4⁺ T cells, conferring long-lasting protection against i

Limosilactobacillus reuteri alleviates proinflammatory T-cell-mediated liver injury and transcriptomic changes in immunocompromised mice.

BACKGROUND: A deficiency of immunosuppressive regulatory T cells, as seen in scurfy (SF) mice or in IPEX syndrome in humans, can lead to multiorgan inflammation. Oral administration of the probiotic Limosilactobacillus reuteri Deutsche Sammlung von Mikroorganismen und Zellkulturen GmbH (DSM) 17938 prolongs survival and reduces Th1- and Th2-associated inflammation in SF mice. It remains unclear how DSM 17938-educated SF-CD4+ T cells modulate T-cell-liver communication. METHODS: To characterize CD4+ T cells from SF mice orally administered DSM 17938 (Prob-SF-CD4+ T cells) and to compare them wi

The kallikrein-kinin system modulates CD4+ T cell function and contributes to T cell migration into the brain after stroke.

Ischemic stroke elicits a sustained thromboinflammatory response that critically shapes secondary brain injury. The kallikrein-kinin system (KKS) has been identified as the interface of vascular injury and inflammatory processes; however, its role in adaptive immunity remains poorly defined. Here, we show that the KKS is associated with pathogenic CD4+ T cell activation and trafficking. In both ischemic stroke patients and mice subjected to transient middle cerebral artery occlusion (tMCAO), circulating CD4+ T cells exhibited a conserved, activated, and trafficking-competent phenotype characte

Th2 skewing in patients with disseminated coccidioidomycosis.

BACKGROUNDDisseminated coccidioidomycosis (DCM) is an often fatal and otherwise intractable condition requiring lifelong antifungal treatment. We have previously shown that a deranged polarization of CD4+ T cells toward a Th2 phenotype can exist in the context of DCM. Here, we studied a large population to determine the frequency of abnormal Th2 skewing of CD4+ T cells in patients with coccidioidomycosis and to identify underlying genetic mechanisms supporting this phenotype.METHODSWe collected PBMCs from 204 patients with coccidioidomycosis, including 96 patients with disseminated disease. World free HIV: the novel therapeutic approaches for eliminating latent HIV infection.

Antiretroviral therapy (ART) is an intervention aimed at preventing the development and spread of HIV. The persistence of proviruses in the CD4 + memory T cells, which are transcriptionally inactive and other drug-resistant reservoirs, makes treatment of chronic HIV-1 infection through pharmacological means complex. The major measures to deal with the disease involve reactivating the inactive viruses then clearing them off by using virus-specific cytotoxic effect and also by host immune responses. Broadcast screening and mechanism-based techniques have revealed a huge number of medications tha

Methaneseleninic acid, a circadian-modulating agent, reactivates latent HIV-1 infection without cellular activation or proliferation.

Human immunodeficiency virus (HIV)-1 transcriptional activity is under circadian control in virally suppressed people with HIV (PWH), and circadian transcription factors can positively affect HIV transcription in vitro. We aimed to determine whether pharmacologic modulation of cell-autonomous circadian cycles can reverse HIV latency. We examined the effects of a known circadian modulator, the organic selenium compound, methaneseleninic acid (MSA), on viral reactivation, and cellular activation and proliferation in near-full-length latently infected cell lines and primary peripheral blood CD4+

The HIV-2 reservoir: lessons from HIV-1 and knowledge gaps.

HIV-2 establishes a persistent viral reservoir despite its lower replication capacity and attenuated clinical course compared with HIV-1. Some studies suggest that the HIV-2 cellular reservoir is substantially smaller, with total HIV-2 DNA levels in peripheral blood mononuclear cells typically 1-2log10 lower than those observed in HIV-1, even in individuals on long-term suppressive antiretroviral therapy. Importantly, HIV-2 proviral DNA remains detectable for many years, indicating durable persistence despite low-level replication. Detailed analyses of reservoir composition suggest that HIV-2

Vitamin D boosts HIV-1 resistance in female genital epithelial cells by enhancing antiviral cathelicidin expression.

INTRODUCTION: Elevated levels of serum Vitamin D (VitD) and increased expression of its receptor on peripheral blood cells have been reported in individuals who have been exposed to HIV-1 yet remain seronegative (HESN). This study aimed to investigate the potential antiviral role of VitD in the female genital epithelium, which constitutes the first physical barrier against HIV-1. We hypothesized that VitD may modulate susceptibility to HIV-1 infection by influencing epithelial functions such as tight junction integrity, production of antiviral peptides, and secretion of pro-inflammatory mediat

Lymph node remodelling underlies the attenuated form of HIV/AIDS in people with HIV-2.

HIV-2 infection is associated with low-to-undetectable plasma viral load, broad and sustained specific antibody and T cell responses, and a slow course of CD4 T cell decline, even in the absence of antiretroviral therapy (ART). Here, we investigated the secondary lymphoid organs of people with HIV-2 (PWH2) as compared to people with HIV-1 (PWH1) and seronegative subjects. We found active HIV-2 replication and major disruption of lymph node architecture in PWH2, also attested by the alterations that we demonstrated in the blood counterparts of follicular T and B cells. These findings were corro
