Endurance training in the spontaneously hypertensive rat: conversion of pathological into physiological cardiac hypertrophy.

The effect of endurance training (swimming 90 min/d for 5 days a week for 60 days) on cardiac hypertrophy was investigated in the spontaneously hypertensive rat (SHR). Sedentary SHRs (SHR-Cs) and normotensive Wistar rats were used as controls. Exercise training enhanced myocardial hypertrophy assessed by left ventricular weight/tibial length (228+/-7 versus 251+/-5 mg/cm in SHR-Cs and exercised SHRs [SHR-Es], respectively). Myocyte cross-sectional area increased approximately 40%, collagen volume fraction decreased approximately 50%, and capillary density increased approximately 45% in SHR-Es

Xenon and hypothermia combine to provide neuroprotection from neonatal asphyxia.

Perinatal asphyxia can result in neuronal injury with long-term neurological and behavioral consequences. Although hypothermia may provide some modest benefit, the intervention itself can produce adverse consequences. We have investigated whether xenon, an antagonist of the N-methyl-D-aspartate subtype of the glutamate receptor, can enhance the neuroprotection provided by mild hypothermia. Cultured neurons injured by oxygen-glucose deprivation were protected by combinations of interventions of xenon and hypothermia that, when administered alone, were not efficacious. A combination of xenon and

IL-1β promotes IL-17A production of ILC3s to aggravate neutrophilic airway inflammation in mice.

IL-17A-producing group 3 innate lymphoid cells (ILC3s) have been found to participate in the development of various phenotypes of asthma, however, little is known about how ILC3s mediate neutrophilic airway inflammation. Elevated IL-1β has been reported in neutrophilic asthma (NA) and IL-1β receptor is highly expressed on lung ILC3s. Therefore, we hypothesize that IL-1β aggravates neutrophilic airway inflammation via provoking IL-17A-producing ILC3s. We sought to determine the pathological roles of the IL-1β-ILC3-IL-17A axis in neutrophilic airway inflammation. Lung ILC subsets were measured i

Enhanced synergistic antitumor effect of a DNA vaccine with anticancer cytokine, MDA-7/IL-24, and immune checkpoint blockade.

BACKGROUND: MDA-7/IL-24 cytokine has shown potent antitumor properties in various types of cancer without exerting any significant toxicity on healthy cells. It has also been proved to encompass pro-immune Th1 cytokine-like behavior. Several E7 DNA vaccines have developed against human papillomavirus (HPV)-related cervical cancer. However, the restricted immunogenicity has limited their clinical applications individually. To address this deficiency, we investigated whether combining the E7 DNA vaccine with MDA-7/IL-24 as an adjuvant would elicit efficient antitumor responses in tumor-bearing m

Neuroprotective role of sphingolipid rheostat in excitotoxic retinal ganglion cell death.

The glutamate excitotoxicity has been suggested as a factor involved in the loss of retinal neuronal cells, including retinal ganglion cell (RGC), in various retinal degenerative diseases including ischemia-reperfusion injury, diabetic retinopathy, and glaucoma. Excitotoxic RGC death is caused not only by direct damage to RGCs but also by indirect damage due to the inflammation of retinal glial cells. Sphingosine 1-phosphate (S1P) and ceramides are bioactive sphingolipids which have been shown to possess important physiological roles in cellular survival and apoptosis, and the balance between

Acacetin alleviates pyroptosis, inflammatory response, and oxidative stress in renal tubular epithelial cells caused by kidney stone via targeting GPR35.

Nephrolithiasis, particularly calcium oxalate (CaOx) stones, remains a global health burden with limited pharmacological options for recurrence prevention. In this study, we investigated the therapeutic mechanism of acacetin (ACA) using both in vitro (COM-stimulated HK-2 cells) and in vivo (glyoxylate-induced mice) models of CaOx kidney stone. Our results demonstrated that ACA treatment dose-dependently attenuated renal injury, reduced the expression of pro-inflammatory cytokines (IL-1β, IL-6, and TNF-α), and activated the Nrf2/HO-1/NQO1 antioxidant pathway. Moreover, ACA significantly suppres

Capsicum annuum Regulates Tumor Growth Through Modulation of TLR4/PI3K Signaling in a Lewis Lung Carcinoma Mouse Model.

Since lung cancer remains a leading cause of cancer-related mortality, effective adjunct strategies against this disease are needed. This study evaluated an extract prepared from organically cultivated Capsicum annuum treated with deep-seawater-derived ion minerals during cultivation (ODSW-CE) in Lewis lung carcinoma (LLC1) cells and LLC1 tumor-bearing mice. Targeted HPLC-DAD analysis identified and quantified lutein and β-carotene as representative carotenoids. ODSW-CE reduced MTT-derived cell viability and colony-forming ability in vitro and suppressed tumor growth in vivo. In tumor-bearing

Repurposing acetyldigitoxin as a potential EZH2 inhibitor for non-small cell lung cancer: a computational and experimental approach.

Non-small cell lung cancer (NSCLC) remains a leading cause of cancer-related mortality worldwide. The epigenetic regulator EZH2 is a promising therapeutic target due to its role in tumor progression and therapy resistance. This study combined computational and experimental methods to repurpose FDA-approved drugs as EZH2 inhibitors. Virtual screening and molecular dynamics simulations identified acetyldigitoxin (ADT) as a potent EZH2 inhibitor, demonstrating superior binding affinity (-10.90 kcal/mol) and complex stability compared to the known inhibitor GSK126. ADT formed robust hydrogen bonds

Eriodictyol in Cancer Therapy: Reviewing Mechanistic Insights and Translational Opportunities.

Eriodictyol, a naturally occurring flavanone, has appeared as a biologically versatile compound with increasing relevance in biomedical research, especially in cancers. Evidence over the past few decades indicates that eriodictyol influences cancer cell fate through coordinated modulation of cell-cycle control, survival, and regulated cell death pathways. Eriodictyol appears to reshape oncogenic signaling networks, including PI3K/Akt/mTOR and associated kinase cascades, thereby restricting proliferative capacity and lowering resistance thresholds. Studies consistently report cell-cycle arrest

Targeting SQLE-Mediated Cholesterol Metabolism to Promote Oxidative Stress and Attenuate Drug Resistance in Osteosarcoma.

High-grade osteosarcoma presents a significant clinical challenge due to unpredictable therapeutic responses and aggressive progression. This study aimed to identify the critical molecular pathways driving chemotherapy resistance and aggressive phenotypes in osteosarcoma patients. Through transcriptomic and bioinformatic analyses, we identified cholesterol biosynthesis as a key upregulated metabolic pathway in poor chemotherapy responders, where squalene epoxidase (SQLE) emerged as an exploratory candidate hub gene whose elevated expression significantly correlates with shortened survival in t

Endogenous Amplification of Apoptosis via p53 Regulation using a Cascade Nanocatalytic Medicine.

Nanocatalytic therapy is an emerging strategy that leverages in situ catalytic reactions within the tumor microenvironment to convert endogenous substrates into cytotoxic species, achieving spatially confined cancer cell killing with reduced systemic toxicity. However, the lack of durable, DNA-focused cytotoxic mechanisms hampers the translational efficacy of nanocatalytic therapy. Herein, we proposed a cascade nanocatalysis-mediated strategy for endogenous amplification of apoptosis, achieved by an engineered metal organic framework nanomedicine (MOF-Au-L-Arginine, abbreviated as MAL). The MO

Anti-Cancer Effect of a New 5-FU Derivative Containing Triazole-Bearing Mannose (5-FUD-MAN) Against Human Breast Cancer Cells Through LC3B-Mediated Cell Death.

Breast cancer remains one of the most common malignancies affecting women worldwide. Despite the effectiveness of traditional chemotherapeutic agents such as 5-fluorouracil (5-FU), their lack of selectivity often results in damage to healthy tissues, leading to undesirable adverse effects. The aim of this study was to modify 5-FU with mannose containing 1,2,3-triazole compound to reduce its toxic effect, and to investigate the anticancer properties of the resulting 5-FU derivative (5-FUD-Man) in ER-positive MCF-7 breast cancer cells. Our results demonstrated that while 5-FU caused significant

Oncolytic Vaccinia Virus-HSP70-shRNA Amplifies Viral Replication, ROS/Autophagy, and Immunity to Fight Colorectal Cancer.

Colorectal cancer persists as a major contributor to cancer mortality, with immunotherapy offering limited efficacy in immunologically "cold" tumors. To address this issue, we engineered an oncolytic vaccinia virus to express HSP70 shRNA within its TK gene locus (oncoVV-shHSP70). In vitro, oncoVV-shHSP70 significantly suppressed HSP70 expression, enhanced viral replication, and induced apoptosis in CRC cells via Caspase-7/9 activation and Bcl-2 downregulation. OncoVV-shHSP70 also triggered reactive oxygen species (ROS) accumulation by suppressing the antioxidant regulator Nrf2 and stimulated a
