Symptomatic west syndrome secondary to glucose transporter-1(GLUT1) deficiency with complete response to 4:1 ketogenic diet.

Glucose transporter type 1 (GLUT-1) deficiency is a rare cause of preventable intellectual disability. Intellectual disability is due to refractory seizures in infancy and reduced supply of glucose to the brain. The authors report a third born male child of consanguineous parentage who presented with infantile spasms. Initially, he had refractory convulsions of focal, generalised, and myoclonic jerks, not responding to multiple anticonvulsants. He also had choreoathetoid movements. On examination he had microcephaly. MRI of brain was normal and EEG showing diffuse slowing. CSF glucose was low

Ingestion of proteoglycan fraction from shark cartilage increases serum inhibitory activity against matrix metalloproteinase-9 and suppresses development of N-nitrosobis(2-oxopropyl)amine-induced pancreatic duct carcinogenesis in hamster.

A water extract of shark cartilage was fractionated into acidic and basic fractions by preparative isoelectric focusing on the basis of the amphoteric nature of samples. The acidic fraction was further fractionated into ethanol-soluble and -precipitate fractions. After the carcinogenesis treatment using N-nitrosobis(2-oxopropyl)amine, hamsters received a diet containing each fraction or purified chondroichin sulfate to give 0.4% (w/w) for 50 days. Only administration of the acidic ethanol-precipitate-fraction-containing diet significantly increased serum inhibitory activity against matrix meta

Ivermectin as a rodent feed-through insecticide for control of immature sand flies (Diptera: Psychodidae).

Ivermectin was evaluated as a potential rodent feed-through for the control of immature stages of Phlebotomus papatasi. The survival of sand fly larvae fed feces of Syrian hamsters (Mesocricetus auratus) that had been fed a diet containing 0, 2, 6, 10, 20, 60, or 100 ppm ivermectin was measured. Sand fly larvae fed the feces of ivermectin-treated hamsters had significantly reduced survival, with 100% mortality of larvae fed feces of hamsters fed a diet containing 20, 60, and 100 ppm ivermectin. The results of this study suggest that a control strategy using rodent baits containing ivermectin t

Correction of ornithine accumulation prevents retinal degeneration in a mouse model of gyrate atrophy of the choroid and retina.

Deficiency of ornithine-delta-aminotransferase (OAT) in humans results in gyrate atrophy of the choroid and retina (GA), an autosomal recessive disorder characterized by ornithine accumulation and a progressive chorioretinal degeneration of unknown pathogenesis. To determine whether chronic, systemic reduction of ornithine can prevent this form of retinal degeneration, we used an arginine-restricted diet to maintain long term reduction of ornithine in a mouse model of OAT-deficiency (Oat(-/-)) produced by gene targeting. We evaluated the mice over a 12-month period by measurement of plasma ami

Morphological and biochemical changes in lenses of guinea pigs after vitamin-C-deficient diet and UV-B radiation.

The effect of ultraviolet B (UV-B) radiation and a vitamin-C-deficient (VCD) diet on guinea pig lenses was investigated. The initial lens changes in the VCD group were observed by slit-lamp examination 6 weeks after the start of the VCD treatment; after 12 weeks the changes in the posterior subcapsular region became more prominent, and the dissociation around the posterior suture became wider and slightly deeper toward the posterior cortex. The high concentration of lens oxidized glutathione (GSSG), and the low ratio of reduced glutathione (GSH) to oxidized glutathione (GSH/GSSG) on the lens p

The hemodynamic effect of dietary calcium supplementation on rat renovascular hypertension.

Mechanisms which could be responsible for the hypotensive effects of increasing calcium intake from 1.2 to 2.5% of diet were examined in two kidney, one clip renovascular hypertension (2K, 1C RHV) in male Sprague-Dawley rats. Plasma renin activity was elevated similarly in NCA and HCA rats. No evidence for volume or phosphate depletion was found in HCA animals. Cardiac output was not different. Decreased sensitivity was demonstrated to moderate doses of norepinephrine in HCA rats, while vascular reactivity to exogenously administered angiotensin II over the range of doses used was not differen

Magnesium-deficient myocardium demonstrates an increased susceptibility to an in vivo oxidative stress.

Previous studies in our laboratory have indicated free radical participation in magnesium deficiency cardiomyopathy. In this study, we examined the capacity of the magnesium-deficient animals to withstand an in vivo oxidative stress. Syrian hamsters were placed on either magnesium-deficient diet or a magnesium-supplemented control diet. Animals from each group also received vitamin E. After 14 days some of animals were given the catecholamine isoprenaline; 2 days later the animals were killed. The severity of the isoprenaline-induced injury was assessed by a morphometric analysis. The isoprena

Effect of varying levels of magnesium during gestation and lactation on humoral immune response and tissue minerals in rats.

Weanling rats were maintained on diets which were Mg-adequate (49 mmol Mg/kg), groups 1 and 3, or Mg-deficient (8 mmol Mg/kg), groups 2 and 4, for 8 weeks of growth. During breeding, gestation, and lactation, groups 3 and 4 received diets supplemented with 41 mmol Mg/kg. Femur and cardiac Mg in group 2 dams was reduced, confirming that Mg deficiency was present. Thymic weight from dams and pups and numbers of plaque-forming cells in pups in group 1 were higher than in other groups while splenic weight in pups was lowest in group 2. Results indicate that maternal magnesium deficiency impaired i

Calcinogenic activity of vitamin D3 and vitamin D3 palmitate in rat and rabbit.

Experimental hypervitaminosis D was produced in rabbits by feeding 25,000 I.U. vitamin D3 or the corresponding amount of vitamin D3 palmitate per kg of diet. Hypercalcemia and hyperphosphatemia was accompanied by increased CaBP activity and reduced weight gain in the vitamin D3 group as well as in the vitamin D3 ester group. The degree of calcification in the aorta and in the kidney also was similar in both groups. Increasing the vitamin intake by giving 10,000 I.U. vitamin D3 or vitamin D3 palmitate per day resulted in earlier and more widespread calcific deposits. In rats, receiving 50,000 I

Primary hyperparathyroidism: possible cause of primary hyperaldosteronism in a 60-year-old woman.

Hypertension and hypokalemia were found in a 60-yr-old woman suffering from primary hyperparathyroidism. Laboratory investigations in this patient disclosed 1) elevated levels of plasma aldosterone (PA) which could not be suppressed by a high sodium diet alone or in combination with fludrocortisone (Florinef); 2) a decline of the elevated PA levels after 4 h of ambulation; and 3) low PRA which was unresponsive to stimulation by a low sodium diet coupled with diuretic-induced volume depletion and 4 h of ambulation. These findings were consistent with the diagnosis of primary hyperaldosteronism.

Plant-based diet, metabolic signature, genetic susceptibility, and risk of musculoskeletal disorders: a large-scale population-based prospective cohort study.

AIMS: To explore the association between plant-based diets, diet-related metabolic signatures, and the risk of total and specific musculoskeletal disorders (MSDs), while accounting for genetic susceptibility. METHODS: We analyzed data from 107,983 participants in the UK Biobank. Plant-based diet indices [PDI, unhealthy PDI (uPDI), and healthy PDI (hPDI)] were calculated via 24-h dietary recalls. Elastic net regression was applied to generate PDIs-related metabolic signatures. Polygenic risk score (PRS) was constructed to quantify genetic susceptibility to MSDs. Cox proportional hazards regres

Racial disparities in systemic inflammation: The mediating role of diet and physical activity in α-1-acid glycoprotein levels in the U.S. population.

Racial and ethnic disparities in systemic inflammation contribute to differences in chronic disease risk. α-1-acid glycoprotein (AAG), an acute-phase inflammatory marker, has been associated with metabolic dysfunction and cardiovascular risk. Despite known racial differences in inflammatory biomarkers, the role of diet and physical activity as mediators of racial disparities in AAG levels remains unclear. This study investigates whether racial disparities in AAG are mediated by diet quality and physical activity in a nationally representative U.S. sample. We conducted a cross-sectional mediati

Association between protein diet score and colorectal adenomas risk: a prospective study.

OBJECTIVE: Given the significantly increased risk of colorectal adenoma in middle-aged and elderly populations, identifying modifiable risk factors remains a priority. While dietary protein is an essential nutrient in human metabolism, its relationship with colorectal adenoma remains controversial. With advances in nutritional science, contemporary dietary guidelines advocate increasing plant-based protein intake to achieve a more balanced protein consumption pattern. To provide new insights, we sought to investigate the association between colorectal adenoma risk and the Protein Diet Score, w

Modified Mediterranean diet effects on Parkinson's disease (MED-PARK): a single-centre randomised controlled trial protocol.

INTRODUCTION: Only symptomatic treatments are available for patients with Parkinson's disease (PD), the second most common chronic neurodegenerative disease worldwide, and it is therefore imperative to identify disease-modifying interventions that can alter the course of the disease. Epidemiological studies in PD patients suggest that a Mediterranean diet is associated with better motor and non-motor symptoms, slower progression and reduced mortality. Few interventional studies, however, investigated the relationship between diet and PD severity and progression. This study aims to determine wh

Intestine epithelial-specific hypoxia-inducible factor-1α overexpression ameliorates western diet-induced MASLD.

BACKGROUND: Intestine epithelial hypoxia-inducible factor-1α (HIF-1α) plays a critical role in maintaining gut barrier function. The aim of this study was to determine whether pharmacological or genetic activation of intestinal HIF-1α ameliorates western diet-induced metabolic dysfunction-associated steatotic liver disease. METHODS: Metabolic effects of pharmacological activation of HIF-1α by dimethyloxalylglycine were evaluated in HIF-α luciferase reporter (ODD-luc) mice. Male and/or female intestinal epithelial-specific Hif1α overexpression mice (Hif1αLSL/LSL;VilERcre) and wild-type litterm

Thick Liquids and Clinical Outcomes in Hospitalized Patients With Alzheimer Disease and Related Dementias and Dysphagia.

IMPORTANCE: Oropharyngeal dysphagia is common in hospitalized patients with Alzheimer disease and related dementias (ADRD). Although the use of thick liquids in patients with dysphagia has been shown to reduce aspiration on direct visualization, there is no clear evidence that this practice translates into improved clinical outcomes. OBJECTIVES: To determine whether a diet of thick liquids compared with thin liquids is associated with improved outcomes in hospitalized patients with ADRD and dysphagia. DESIGN, SETTING, AND PARTICIPANTS: This cohort study included adults aged 65 years and olde
