Magnetosome-incorporated silk fibroin-based magnetic hydrogel for efficient tumor magnetic hyperthermia.

We developed a magnetosome-incorporated silk fibroin-based magnetic hydrogel (MSMH) for efficient tumor magnetic hyperthermia. The MSMH was fabricated by integrating biologically synthesized magnetosomes with glycidyl methacrylate-modified silk fibroin (SFGMA), enabling injectable deployment and rapid photo-crosslinking. The as-fabricated MSMH exhibits excellent magnetothermal conversion performance, achieving prominent temperature rises under alternating magnetic fields (AMF). MSMH can also serve as a T₂-weighted MRI contrast agent (r2 = 173.57 mM-1 s-1 at 0.5 T), enabling imaging-guided ther

Thermal bubble single-cell printing chip: High-throughput, wide-field, and efficient.

Single-cell printing technology has arisen as a potent instrument for investigating cell biology and disease pathophysiology. Nonetheless, current single-cell printing methodologies are hindered by restricted throughput, a limited field of view, and diminished efficiency. We present an innovative single-cell printing chip that utilizes thermal inkjet technology for single-cell printing, therefore addressing these constraints. We have accomplished high-throughput, wide-field, and efficient single-cell printing by merging a high-density thermal foam-based inkjet nozzle array on a chip with high-…

Highly Efficient Room-Temperature Nonvolatile Magnetic Switching by Current in Fe3GaTe2 Thin Flakes.

Effectively tuning magnetic state by using current is essential for novel spintronic devices. Magnetic van der Waals (vdW) materials have shown superior properties for the applications of magnetic information storage based on the efficient spin torque effect. However, for most of known vdW ferromagnets, the ferromagnetic transition temperatures lower than room temperature strongly impede their applications and the room-temperature vdW spintronic device with low energy consumption is still a long-sought goal. Here, the highly efficient room-temperature nonvolatile magnetic switching is realized

Energy-efficient picosecond spin-orbit torque magnetization switching in ferro- and ferrimagnetic films.

Electrical current pulses can be used to manipulate magnetization efficiently via spin-orbit torques. Pulse durations as short as a few picoseconds have been used to switch the magnetization of ferromagnetic films, reaching the terahertz regime. However, little is known about the reversal mechanisms and energy requirements in the ultrafast switching regime. In this work we quantify the energy cost for magnetization reversal over seven orders of magnitude in pulse duration, in both ferromagnetic and ferrimagnetic samples, bridging quasi-static spintronics and femtomagnetism. To this end, we dev

Phospholipid Scramblase 1 Controls Efficient Neurotransmission and Synaptic Vesicle Retrieval at Cerebellar Synapses.

Phospholipids (PLs) are asymmetrically distributed at the plasma membrane. This asymmetric lipid distribution is transiently altered during calcium-regulated exocytosis, but the impact of this transient remodeling on presynaptic function is currently unknown. As phospholipid scramblase 1 (PLSCR1) randomizes PL distribution between the two leaflets of the plasma membrane in response to calcium activation, we set out to determine its role in neurotransmission. We report here that PLSCR1 is expressed in cerebellar granule cells (GrCs) and that PLSCR1-dependent phosphatidylserine egress occurred a

Oral administration of probiotic spore ghosts for efficient attenuation of radiation-induced intestinal injury.

Radiation-induced intestinal injury is the most common side effect during radiotherapy of abdominal or pelvic solid tumors, significantly impacting patients' quality of life and even resulting in poor prognosis. Until now, oral application of conventional formulations for intestinal radioprotection remains challenging with no preferred method available to mitigate radiation toxicity in small intestine. Our previous study revealed that nanomaterials derived from spore coat of probiotics exhibit superior anti-inflammatory effect and even prevent the progression of cancer. The aim of this work is

A miniature and efficient colorimetric system assisted by the microplasma jet array for Cr (VI) detection.

Microplasma jet (MPJ), serving as excitation sources in miniaturized systems, has been widely used for contaminant detection. However, the employment of the MPJ is conditioned due to the limit generation of the radicals. Here, a novel 4-channel microplasma jet array (MPJA) is designed for 3,3,5,5-tetramethylbenzidine (TMB) rapid oxidation without oxidizing reagents and catalysts. The jet array, generated from a ceramic tube, allows direct contact with the sample. A colorimetric system containing the MPJA has been developed for the detection of hexavalent chromium. Compared to the MPJ, the empl

Discovery of potent and novel dual NAMPT/BRD4 inhibitors for efficient treatment of hepatocellular carcinoma.

The NAPRT-induced increase in NAD+ levels was proposed as a mechanism contributing to hepatocellular carcinoma (HCC) resistance to NAMPT inhibitors. Thus, concurrently targeting NAMPT and NAPRT could be considered to overcome drug resistance. A BRD4 inhibitor downregulates the expression of NAPRT in HCC, and the combination of NAMPT inhibitors with BRD4 inhibitors simultaneously blocks NAD+ generation via salvage and the PH synthesis pathway. Moreover, the combination of the two agents significantly downregulated the expression of tumor-promoting genes and strongly promoted apoptosis. The pres

ER-mitochondria distance is a critical parameter for efficient mitochondrial Ca2+ uptake and oxidative metabolism.

IP3 receptor (IP3R)-mediated Ca2+ transfer at the mitochondria-endoplasmic reticulum (ER) contact sites (MERCS) drives mitochondrial Ca2+ uptake and oxidative metabolism and is linked to different pathologies, including Parkinson's disease (PD). The dependence of Ca2+ transfer efficiency on the ER-mitochondria distance remains unexplored. Employing molecular rulers that stabilize ER-mitochondrial distances at 5 nm resolution, and using genetically encoded Ca2+ indicators targeting the ER lumen and the sub-mitochondrial compartments, we now show that a distance of ~20 nm is optimal for Ca2+ tra…

Efficient turn-on fluorescent probe cooperated by cascade response for disclosing the fluctuation of cysteine in cells.

BACKGROUND: The research on cysteine (Cys) determination is deemed as a hot topic, since it has been reported to be connected with various physiological processes and disease prediction. However, existing Cys-responding probes may expose some defects such as long reaction time, disappointing photostability, and suboptimal sensitivity. Under such a circumstance, our team has proposed an efficient fluorescent probe with novel sensing mechanism to perfectly cope with the above-mentioned drawbacks. RESULTS: A novel cascade reaction-based probe 9-(2,2-dicyanovinyl)-2,3,6,7-tetrahydro-1H,5H-pyrido[

Ultrasound-responsive theranostic platform for the timely monitoring and efficient thrombolysis in thrombi of tPA resistance.

There is no effective and noninvasive solution for thrombolysis because the mechanism by which certain thrombi become tissue plasminogen activator (tPA)-resistant remains obscure. Endovascular thrombectomy is the last option for these tPA-resistant thrombi, thus a new noninvasive strategy is urgently needed. Through an examination of thrombi retrieved from stroke patients, we found that neutrophil extracellular traps (NETs), ε-(γ-glutamyl) lysine isopeptide bonds and fibrin scaffolds jointly comprise the key chain in tPA resistance. A theranostic platform is designed to combine sonodynamic and

Efficient strabismus diagnosis from small samples: Harnessing spatial features for improved accuracy.

Strabismus is a common ophthalmological condition, and early diagnosis is crucial to preventing visual impairment and loss of stereopsis. However, traditional methods for diagnosing strabismus often rely on specialized ophthalmic equipment and trained personnel, limiting the widespread accessibility of strabismus diagnosis. Computer-aided strabismus diagnosis is an effective and widely used technology that assists clinicians in making clinical diagnoses and improving efficiency. To address this, we designed an efficient strabismus diagnosis model, RIS-MLP, based on a small number of samples de…

Optimizing Mucoadhesive Film-Forming Spray for Efficient Oral Delivery of Fluconazole in Candidiasis Treatment.

Buccal candidiasis has become increasingly prevalent in recent years, with Candida albicans being the primary causative organism. While systemic fluconazole is an effective treatment, its use is associated with adverse effects such as gastric upset, hepatic failure, and potential drug interactions. Therefore, the development of local fluconazole treatment presents a promising solution to these challenges. This study aimed to formulate an efficient local mucoadhesive film-forming spray for the targeted delivery of fluconazole in the treatment of oral candidiasis. The investigation involved the

SPLICER: a highly efficient base editing toolbox that enables in vivo therapeutic exon skipping.

Exon skipping technologies enable exclusion of targeted exons from mature mRNA transcripts, which have broad applications in medicine and biotechnology. Existing techniques including antisense oligonucleotides, targetable nucleases, and base editors, while effective for specific applications, remain hindered by transient effects, genotoxicity, and inconsistent exon skipping. To overcome these limitations, here we develop SPLICER, a toolbox of next-generation base editors containing near-PAMless Cas9 nickase variants fused to adenosine or cytosine deaminases for the simultaneous editing of spli
