Genetic variation in human aquaporins and effects on phenotypes of water homeostasis.

Aquaporins (AQPs) are critical for the transport of water and small solutes. The 13 known human AQPs are divided into those that transport only water molecules, the "orthodox" AQPs, and those that transport glycerol and small solutes in addition to water, the aquaglyceroporins. In humans, genetic variation in AQPs can cause phenotypes of abnormal water homeostasis. Cellular and human studies of naturally-occurring and synthetic mutations have provided insight into the biology and phenotypes of these variants. Many AQPs have not been well-characterized in terms of the effect of genetic variatio

Management of fluid balance in continuous renal replacement therapy: technical evaluation in the pediatric setting.

Fluid overload control and fluid balance management represent very important factors in critically ill children requiring renal replacement therapy. A relatively high fluid volume administration in children and neonates is often necessary to deliver adequate amounts of blood derivatives, vasopressors, antibiotics, and parenteral nutrition. Fluid balance errors during pediatric continuous renal replacement therapy (CRRT) might significantly impact therapy delivery and have been described as potentially lethal. The aim of this study was to evaluate the accuracy of delivered vs. prescribed net ul

Sodium and water retention in heart failure and diuretic therapy: basic mechanisms.

The pathophysiology of sodium and water retention in heart failure is characterized by a complex interplay of hemodynamic and neurohumoral factors. Relative arterial underfilling is an important signal that triggers heart failure-related sodium and water retention. The response to perceived arterial underfilling is modulated by the level of neurohormonal activation, the degree of renal vasoconstriction, and the extent to which renal perfusion pressure is reduced. Sodium retention can also be exceeded by water retention, with the result being dilutional hyponatremia. Sodium and water retention

Activation of the ligand-mineralocorticoid receptor functional unit by ancient, classical, and novel ligands. Structure-activity relationship.

The mineralocorticoid effect on epithelial cells is the resultant of an intricate net of biochemical regulations that ultimately leads to the maintenance of electrolyte homeostasis. Two key protagonists in this plot are the ligand, which broadcasts the information, and the receptor, which functions as a receiver and transducer. Therefore, the responsibility for the final biological effect is not limited to each individual component but to both of them, so they constitute a functional unit. In addition, several prereceptor regulatory mechanisms are also determinant factors for the final biologi

Voltammetric studies of Cu-adriblastina complex and its effect on ssDNA-adriblastina interaction at in situ mercury film electrode.

Adriblastina, a cancerostatic anthracycline antibiotic, causes considerable oxidative damage to DNA molecules. The interaction of this compound with DNA was investigated using Osteryoung square wave stripping voltammetry (OSWSV) and cyclic voltammetry (CV) at an in situ mercury film electrode. It was found that the equilibrium constant of the bonded oxidized form of the drug was 63 times bigger more important than that of the bonded reduced form. Copper forms 1 metal: 2 drug stoichiometry complex which is highly stable compared to ssDNA-drug interaction and consequently inhibited the drug bioc

Influence of Al on growth, cell size and content of intracellular water of Arthrobacter sp. PI/1-95.

A distinct inhibition of the growth of a typical soil-born bacterium, Arthrobacter sp. PI/1-95, by aluminium was established. The content of intracellular water and (consequently) the cell size of the bacterium significantly increased when cells came into contact with aluminium. Both parameters were also increased when applying HCl, but Al caused the greater change, so that a distinct discrimination between the effects of aluminium on the one hand and a sole pH-effect on the other hand was possible. The presented results point to an osmo-regulative disorder connected with a malfunction of cell

[Does constitutional hypotension exist?].

Orthostatic hypotension is frequently related to severe insufficiency of the autonomic nervous system associated with neuropathy or systemic disease like diabetes. Inversely, pre-syncopal orthostatic symptoms associated with mild drop in orthostatic blood pressure is quite often a reason to seek medical care, but is relatively unrecognized in the literature. Recently a syndrome of mild orthostatic intolerance has been defined, and seem to be quite common among young subjects, characterized by frequent orthostatic presyncopal symptoms associated with orthostatic tachycardia and high plasma cate

[Hypertonic-hyperoncotic solutions: possibilities and limits].

Hypertonic-hyperoncotic solutions are a supplementary possibility in the acute treatment of hypovolemia. The main mechanism of action of this solution is, induced by the tonicity of the infusion (approximately 2400 mOsm/l), the transcapillary fluid shift from the intracellular and interstitial to the intravascular compartment. Additionally the fluidity of blood is improved (due to an increase in the diameter of the capillaries), the adherence of leucocytes to the endothelial wall is reduced, and the arteriolar vasomotion is restarted. Due to the available prehospital and clinical results it ca

Thirst in elderly subjects.

The hormonal changes associated with ageing, the occurrence of other medical condition and the use of medications increases the likelihood of conditions such as hyper- and hypo-natremia. Awareness of the homeostatic changes with age and the relations with disease and drugs will lead better management of fluid disorders in the elderly patient and hopefully a reduction in morbidity and mortality.

Disordered water channel expression and distribution in acquired nephrogenic diabetes insipidus.

A series of recent studies have demonstrated that expression of aquaporin-2 (AQP2), the vasopressin-regulated water channel of the kidney collecting duct, is greatly reduced in acquired forms of nephrogenic diabetes insipidus (NDI). In some forms of NDI, there is also impaired delivery of these channels to the apical plasma membrane, where they permit water reabsorption from the urine. The combination of these factors is likely to underlie the urinary concentrating defect that defines these conditions. Direct infusion of vasopressin causes an increase in AQP2 expression, probably via a rise in

A method of measuring fluid balance during transurethral resection of the prostate.

OBJECTIVE: To design a practical system for non-invasively monitoring fluid balance during transurethral resection of the prostate (TURP) and other endoscopic procedures. MATERIALS AND METHODS: Load cell transducers are incorporated into a platform placed under the operating table. Output is passed to a digital weighmeter and then to a portable computer. The raw data is filtered using software written by the authors (CAL) and the output displayed both numerically and graphically on the computer screen. The device was tested under laboratory conditions and then assessed in the clinical setting

[Anesthetic management of two patients with mitochondrial encephalopathy, lactic acidosis and stroke-like episodes (MELAS)].

MELAS is a type of the mitochondrial myopathy characterized by elevation of pyruvate and lactate levels in both the blood and cerebrospinal fluid. This syndrome frequently accompanies cerebral infarction like symptoms. Recently, we experienced two patients for anesthesia with MELAS (both 11-year-old girls). V-P shunt construction and IVH reservoir implantation were conducted, respectively. Anesthesia was induced with fentanyl and midazolam, and vecuronium was used to facilitate tracheal intubation. Volatile anesthetic was avoided, and anesthesia was maintained with fentanyl, midazolam, and nit

Schedule-induced polydipsia in rats with gastric fistulas.

We have investigated the development and maintenance of schedule-induced polydipsia (SIP) when ingested water (and food) was allowed to drain from the stomach. Fourteen male Long-Evans rats were prepared with permanent gastric cannulas and, after recovery, their body weight was reduced to 80%. Water intake was measured, with cannulas open or closed, during 42 daily 1-h sessions in which 45-mg food pellets were delivered one per minute. Allowing ingested material to drain from the stomach impaired the development of SIP and reduced the polydipsia in rats in which SIP had already been establishe
