Transglutaminases (transamidases; endo-gamma-glutamine: zeta-lysine transferases) are calcium-dependent enzymes, which cross-link proteins by introducing covalent zeta-(gamma-glutaminyl)lysine pseudopeptide bonds between the molecules. The distribution and characteristics of transglutaminase in both human and rabbit eyes have been studied. Transglutaminase activity was measured with an isotope technique based on the enzyme catalysed incorporation of 14C-putrescine into casein. Electrophoretic characteristics were studied using agarose gel electrophoresis combined with a specific fluorescent ac

A kinetic study of inhibition of Polyporus versicolor laccase activity by fluoride-, chloride- and bromide ions has been carried out. It has been found that the fluoride ion is a non-competitive inhibitor with respect to the electron donor and a mixed inhibitor with respect to oxygen. However, the chloride and bromide ions are competitive inhibitors with respect to the electron donor. The constants of inhibition of the enzyme activity by both chloride and fluoride ions and the catalytic constant were found to be pH-dependent. Based on the pH-dependence of the catalytic constant, an existence o

Among strains of Drosophila melanogaster each derived from a single fertilized female taken from natural populations, there is variation in both alcohol dehydrogenase (ADH) activity and the amount of ADH protein. The correlation between ADH activity and number of molecules over all strains examined is 0.87 or 0.96 in late third instar larvae depending on whether the substrate is 2-propanol or ethanol. With respect to the two common electrophoretic allozymic forms, F and S, segregating in these populations, the FF strains on the whole have higher ADH activities and numbers of ADH molecules than

Clinical and hematological studies were performed on ten homozygous and seven heterozytous individuals with pyruvate kinase deficiency, aged between 2 and 71 years. Five of the homozygotes were splenectomized. With the exception of a decreased enzyme activity between 41 and 55 per cent and minor changes in their red cell metabolism the heterozygotes showed no abnormal results. In the homozygotes the following results could be demonstrated: 1. Pyruvate kinase activity was decreased to 11 to 35 per cent of normal enzyme activity. 2. There is no relation between the severity of hemolysis and the

Fabry disease is one of the most common lysosomal storage disorders caused by mutations in the gene encoding lysosomal α-galactosidase A (α-Gal A) and resultant accumulation of glycosphingolipids. The sugar mimetic 1-deoxygalactonojirimycin (DGJ), an orally available pharmacological chaperone, was clinically approved as an alternative to intravenous enzyme replacement therapy. The decision as to whether a patient should be treated with DGJ depends on the genetic variant within the α-galactosidase A encoding gene (GLA). A good laboratory practice (GLP)-validated cell culture-based assay to inve

N-nitrosodimethylamine (NDMA) is a volatile environmental contaminant formed from diverse precursor chemicals widely present in byproducts of industrial processes, personal care products, and pharmaceuticals. Therefore, NDMA is frequently detected in both indoor and outdoor air. However, its potential risk from inhalation exposure has been largely underestimated and the mechanisms underlying its toxicity remain insufficiently studied. To address this knowledge gap, we investigated the effects of NDMA on primary human bronchial epithelial (HBE) cells differentiated in air-liquid interface cultu

Tuberculosis, the deadliest human lung disease caused by Mycobacterium tuberculosis, continues to be a global health threat, and finding new drugs and drug targets seems an ongoing battle. The cytochrome P450 CYP125A1 enzyme of M. tuberculosis H37Rv, which is involved in cholesterol metabolism, is a well-established target for drug development. Research is ongoing to identify new compounds that target this enzyme. Understanding the structure-activity relationship of CYP125 family members is crucial for developing a specific and efficient inhibitor. In this direction, this study analyzed 21 cry

The Lyme disease spirochete Borrelia burgdorferi exhibits dramatic changes in gene expression as it transits between its tick vector and vertebrate host. A major hurdle to understanding the mechanisms underlying gene regulation in B. burgdorferi has been the lack of a functional assay to test how gene regulatory proteins and sigma factors interact with RNA polymerase to direct transcription. To gain mechanistic insight into transcriptional control in B. burgdorferi, and address sigma factor function and specificity, we developed an in vitro transcription assay using the B. burgdorferi RNA poly

Curcumin (bis-α,β-unsaturated β-diketone), the chief constituent of turmeric plant (Curcuma longa), plays significant role in prevention of various diseases including diabetes. The research objective in the current study was to synthesize novel anti-diabetic curcumin derivatives with inhibitory properties against α-amylase (α-Amy) and α-glucosidase (α-Gls), as these two carbohydrate-hydrolysing enzymes are known to be important molecular targets for attenuation of postprandial hyperglycemia. The curcumin-based pyrano[2,3-d]pyrimidine derivatives were synthesized in the presence of curcumin, ba

This work reports a rapid and sensitive organophosphates (OPs) amperometric biosensor based on acetylcholinesterase (AChE) immobilized on CdS-decorated graphene (CdS-G) nanocomposite. The as-prepared biosensor shows high affinity to acetylthiocholine (ATCl) with a Michaelis-Menten constant (K(m)) value of 0.24 mM. A rapid inhibition time (2 min) is obtained due to the integration of the CdS-G nanocomposite. Based on the inhibition of OPs on the enzymatic activity of the immobilized AChE, and used carbaryl as the model compound, the resulting biosensor exhibits excellent performance for OPs det

Salvicine is a novel topoisomerase II inhibitor possessing significant antitumor activity, both in vitro and in vivo. The antitumor effect of salvicine is associated with its ability to induce tumor cell apoptosis. Telomerase plays an important role in the apoptotic pathway. However, little is known about the mechanisms of telomerase regulation during apoptosis induced by anticancer drugs. This study investigated the regulation of telomerase activity in salvicine-induced human leukemia HL-60 cell apoptosis. Salvicine treatment resulted in HL-60 cell apoptosis and down-regulation of telomerase

In our mutation analyses of bilirubin UDP glycosyltransferase (UGT1A1) gene, we encountered six patients with Crigler-Najjar syndrome type II who were double homozygotes for G71R and Y486D, a patient with Gilbert's syndrome who was a single homozygote for G71R and six patients with Gilbert's syndrome who were single heterozygote for G71R. To clarify the role of each mutation in the occurrence of the two syndromes, we made four mutant expression models. Relative UGT1A1 activity of a single homozygous model of G71R was 32.2+/-1.6% of normal, that of a single homozygous model of Y486D was 7.6+/-0

Intracellular mediators regulating the initiation of parturition are not fully understood. This study was designed to determine the possible mechanism of oxytocin-induced uterine contractility during labour. In-vitro isometric contraction studies were performed with longitudinal strips of human pregnant myometrium in the presence and absence of the protein kinase C inhibitors, staurosporine and RO 31-8220, and the tyrosine kinase inhibitor, genistein. Phospholipase D activity was measured by employing the transphosphatidylation reaction. Staurosporine significantly reduced oxytocin-stimulated

1. Delayed neuronal cell death elicited by excess excitatory amino acid concentrations has been strongly implicated in many neurological disorders including head trauma, stroke, motor neurone disease and Huntington's disease. We have used the neurotoxin, L-2-chloropropionic acid (L-CPA) to model cellular events in vivo leading to delayed neuronal cell loss which is confined to the cerebellar cortex and can be prevented by inhibitors of nitric oxide synthase such as NG-nitro-L-arginine methyl ester. 2. Experiments were performed to determine whether the constitutive nitric oxide synthase (NOS)

Histamine has positive inotropic and chronotropic effects on the heart which are not abolished by beta adrenergic-blocking agents. Since the positive inotropic and chronotropic effects of other hormones on the heart are thought to be mediated by cyclic 3',5'-AMP, we examined the effect of histamine on adenyl cyclase in particulate preparations of guinea pig, cat, and human myocardium. Histamine at the peak of its dose-response curve, 3 x 10(-4)moles/liter, produced approximately a 300% increase in cyclic 3',5'-AMP accumulation in the guinea pig, 60% in the cat, and 90% in the human heart parti

Many viruses express their proteins in the form of large polyproteins comprising structural and non-structural (e.g. enzymatic) units that are released from the precursor through ordered proteolysis. Proteolytic processing of polyproteins is an indispensable regulatory step for virus maturation and replication that is carried out by the virus-encoded and/or cellular proteases. The activity of a viral protease that is expressed as a part of a polyprotein is controlled in part by the self-cleavage (autoprocessing) from the precursor. The mechanism of protease precursor processing has been establ

BACKGROUND: Cardiac fibrosis is a pathological remodeling process that contributes to the development and progression of heart failure. Although glucose-dependent insulinotropic polypeptide (GIP) may exert antifibrotic effects, how GIP receptor activation regulates cardiac fibrogenesis and modulates cardiac function in heart failure remains unclear. This study investigated whether GIP receptor agonism suppresses cardiac fibroblast activity and improves heart failure and explored the underlying mechanisms by using cellular and animal models.
METHODS: Human cardiac fibroblasts were treated with

BACKGROUND: Early platelet dysfunction is a key component of trauma-induced coagulopathy. Studies have suggested that excess catecholamine following shock conditions may be implicated through proteolytic shedding of key platelet surface receptors. This study investigates the impact of epinephrine exposure on platelet receptor shedding and its subsequent effects on platelet activation, aggregation, and adhesion in an in vitro model.
METHODS: Whole blood samples were exposed to increasing concentrations of epinephrine for 5 and 30 minutes. Enzyme-linked immunosorbent assay was then used to quan

Brucella-induced neuroinflammation represents a key mechanism in the development of neurobrucellosis. The objective of this investigation was to clarify the molecular pathways through which the BvrR contributes to neuroinflammation and cognitive dysfunction. Human microglial clone 3 (HMC3) cells were transfected with pcDNA3.1-BvrR-His to examine the effects of BvrR from Brucella abortus on endoplasmic reticulum (ER) function and the activation of activating transcription factor 2 (ATF2) and nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) p65. Transmembrane protease serine (TMPRSS)2 is a cell surface host protease, which plays a decisive role in viral infections. This trypsin-like enzyme cleaves the spike protein of coronaviruses [e.g. severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2)] and the hemagglutinin protein of influenza viruses, enabling viral membrane fusion and subsequent viral cell entry. Consequently, TMPRSS2 is an attractive therapeutic target, and it is of utmost importance to produce wildtype, enzymatically active recombinant TMPRSS2 for drug development purposes. We present the first successful strategy fo
