Differential gene expression induction by TRAIL in B chronic lymphocytic leukemia (B-CLL) cells showing high versus low levels of Zap-70.

Among 14 peripheral blood samples obtained from patients affected by B chronic lymphocytic leukemia (B-CLL) at initial stages (Rai 0-1) of the disease, 6 showed intermediate/high levels of Zap-70 while 8 displayed low/absent levels of Zap-70. Although Zap-70(high) and Zap-70(low) B-CLL samples displayed similar levels of surface death receptor TRAIL-R2, recombinant TRAIL induced cytotoxicity only in a subset of Zap-70(low) B-CLL samples while Zap-70(high) were completely resistant to TRAIL. The gene expression profiling was next analyzed in all B-CLL samples treated with either chlorambucil or

Identification of dysregulated genes in cutaneous squamous cell carcinoma.

Carcinogenesis is a multi-step process resulting from the accumulation of genetic mutations and subsequently leading to dysregulated genes, but the number and identity of differentially expressed genes in cutaneous squamous cell carcinoma (SCC) is unknown at present. In order to identify dysregulated genes, we examined the relative mRNA expression present in cutaneous SCC and its precursor lesion actinic keratosis (AK) by comparison to normal skin. Snap frozen biopsies from 20 specimens of normal skin, 10 AK, and 10 cutaneous SCC were examined. Total-RNA was extracted, reversely transcribed, a

Expression profiling of autoimmune regulator AIRE mRNA in a comprehensive set of human normal and neoplastic tissues.

Defects in the autoimmune regulator (AIRE) gene cause the monogenic autoimmune disease autoimmune polyendocrinopathy syndrome type 1 (APS-1), which is characterized by a loss of self-tolerance to multiple organs. In concordance with its role in immune tolerance, AIRE is strongly expressed in medullary thymic epithelial cells (mTECs). Data on mechanisms controlling AIRE activation and the expression of this gene in other tissues are fragmentary and controversial. We report here AIRE mRNA expression profiling of a large set of normal human tissues and cells, tumor specimen and methylation defici

Differential expression of NF-kappaB pathway genes among peripheral T-cell lymphomas.

Nuclear factor kappa B (NF-kappaB) is one important pathway in T-cell proliferation and survival. In a previously reported microarray study, we found NF-kappaB pathway genes differentially expressed between peripheral (PTCL) and lymphoblastic lymphomas. Here, we investigated the expression of NF-kappaB pathway genes using cDNA microarrays in a group of 62 PTCL and in reactive lymph nodes. We found two different subgroups of PTCL based on the expression of NF-kappaB pathway genes. One-third of PTCL showed clearly reduced expression of NF-kappaB genes, while the other group was characterized by

Expression of NOTCH1 Is Correlated with Expression of Cancer Stem Cell Markers and miR-150 in Oral Epithelial Dysplasia.

NOTCH1 is associated with various tumors, including oral squamous cell carcinoma (OSCC), with a complex role depending on cellular contexts. Our aim was to analyze the expression of NOTCH1, several stem cell markers, and selected microRNAs in preneoplastic lesion of the oral cavity, oral epithelial dysplasia (OAD). Our study included formalin-fixed paraffin-embedded biopsy samples of 36 cases of OAD and 15 cases of normal oral mucosa. Expression of NOTCH1, stem cell markers (AGR2, KLF4, NANOG, OCT4, SOX2), and miR-27a, miR-34a, miR-128, miR-145, miR-150, and miR-335 was analyzed by quantitativ

Decoding the Oncogenic Role of GNG10 in Colorectal Cancer: A Non-Canonical Wnt Pathway-Driven Mechanism.

Guanine nucleotide-binding protein gamma 10 (GNG10) is implicated in various biological processes, yet its specific oncogenic role in colorectal cancer (CRC) remains poorly defined. This study aimed to elucidate the expression patterns, biological functions, and underlying mechanisms of GNG10 in CRC progression. We integrated TCGA datasets with tissue microarray immunohistochemistry and multivariate Cox regression models to evaluate the clinical significance of GNG10. Functional impacts on CRC malignant phenotypes and cancer stemness were assessed through gain- and loss-of-function models in v

P-Element-Induced Wimpy Testis (PIWI)-Interacting RNA-823/PIWIL1/DNMT3B/CDH1 as Potential Axis to Drive EMT, Stemness, and Tumor Aggressiveness in Ovarian Cancer Tissue Samples: An Integrative Computational and Clinical Insights.

Ovarian cancer (OC) remains the leading cause of death among gynecologic cancers. Most women diagnosed with OC at advanced stages eventually develop relapse and chemoresistance, leading to poor clinical outcomes. While piRNAs have emerged as critical regulators of gene expression and tumor biology, their specific roles in OC remain to be fully elucidated. This study integrated clinical and computational analyses to investigate the expression pattern and functional relevance of P-element-induced wimpy testis (PIWI)-interacting RNA-823 (piR-823) and its associated protein piwi-like RNA-mediated

SLC22A3 regulates ferroptosis in the mesenchymal subtype of triple-negative breast cancer by modulating histone H3K4 serotonylation.

Triple-negative breast cancer (TNBC) remains a challenging and clinically aggressive subtype due to its heterogeneity and high mortality rate. Recent molecular subtyping has identified distinct TNBC subgroups with varying therapeutic responses, highlighting the need for targeted therapeutic strategies. The mesenchymal (MES) subtype is characterized by low immune cell infiltration, cancer stem cell-like features, and resistance to multiple drugs. Ferroptosis, a form of iron-dependent cell death, has emerged as a promising therapeutic strategy in TNBC due to the abundance of iron and lipids in t

Mitochondrial reverse electron transport regulates glioma stemness through Sirt3-HIF1α-SOX2 signaling.

Glioblastoma multiforme (GBM) is an aggressive primary brain tumor characterized by high recurrence and resistance to current therapies, largely driven by glioma stem cells (GSCs). While mitochondrial dysfunction has been implicated in GBM, the specific mitochondrial mechanisms that sustain GSC maintenance remain poorly defined. Here, we identify mitochondrial reverse electron transport (RET) as a key metabolic program enriched in GSCs relative to differentiated glioma cells. Pharmacological inhibition of RET markedly suppresses GSC sphere formation by downregulating the stemness-associated tr

Loss of cold shock domain-containing protein E1 (CSDE1) function enhances mRNA stability of interleukin-6 and promotes malignant transformation in endometrial cancer cells.

BACKGROUND: RNA-binding proteins (RBPs) are indispensable effectors of post-transcriptional gene regulation and are increasingly recognized as pivotal modulators of oncogenic signaling pathways. Cold shock domain-containing protein E1 (CSDE1) has been reported to be dysregulated or mutated in several malignancies; however, its functional significance and the mechanistic basis of its involvement in endometrial cancer (EC) progression remain poorly understood. METHODS: Bioinformatic analysis of The Cancer Genome Atlas (TCGA) was employed to characterize CSDE1 expression and mutation profiles in

RelB drives integrin-mediated stress tolerance and relapse in high-grade serous ovarian cancer.

High-grade serous ovarian cancer (HGSOC) often relapses after chemotherapy due to chemoresistant cancer stem-like cells (CSCs). NF-κB signaling, previously shown to enhance stemness traits, was investigated for its role in post-treatment tumor regrowth. Here, we found that NF-κB subunits RelA and RelB jointly regulate extracellular matrix genes but differentially control integrin subunits: RelA regulates ITGAV (αV) and RelB regulates ITGB3 (β3). Integrin αVβ3 expression is upregulated on CSCs, and this is partially driven by NF-κB activation. In vivo models demonstrate that αVβ3+ cells have hi

Post-transcriptional control of HIF-1α by MBNL1 restrains hypoxia-driven stemness in GBM.

MBNL1 binds the HIF-1α 3'UTR to promote rapid mRNA decay, thereby limiting HIF-1 activity and hypoxia-induced stemness in glioblastoma. Using patient-derived glioma stem cells, we show that MBNL1 loss stabilizes HIF-1α mRNA and increases HIF-1α protein, HRE reporter activity, and target gene expression under hypoxia; MBNL1 knockout also prolongs target gene expression after reoxygenation, indicating enhanced hypoxia "memory." MBNL1 depletion markedly elevates stemness markers (KLF4, SOX2, GLI1) and clonogenic growth, and re-expression of MBNL1 reverses these effects. These results identify a p

LINC01116 promotes nascent protein synthesis and cellular stemness through the miR-432-5p/FKBP7/14 regulatory axis in melanoma.

Skin cutaneous melanoma (SKCM) is a highly aggressive and malignant neoplasm. A wealth of evidence highlights the critical role of LINC01116 in tumorigenesis and progression. Additionally, numerous small molecules within the FK506-binding protein (FKBP) family have been implicated in cancer development. Herein, we interrogated the functional significance of lncRNA LINC01116 and FKBP7/14 in melanoma pathogenesis. This study aim to the expression of long non-coding RNA (lncRNA) LINC01116 in melanoma and its mechanistic role in promoting de novo protein synthesis and cancer stemness through the m

Role of TGF-β in metastatic colon cancer: it is finally time for targeted therapy.

Colorectal cancer (CRC) is one of the most frequent tumor types in Western countries. Approximately 20 % of patients show metastasis at the time of diagnosis, with the liver being one of the most affected organs. Transforming growth factor-beta (TGF-β) plays a regulatory role not only in the physiology of the normal colon but also in the development of CRC and its metastatic process. In this review, we analyze the molecular mechanisms leading to TGF-β dysregulation in tumor and stroma cells and the modification of the microenvironment that fosters CRC metastasis. Recent genomic studies have id

8-Bromo-7-methoxychrysin-blocked STAT3/Twist axis inhibits the stemness of cancer stem cell-like cell originated from SMMC-7721 cells.

Signal transducer and activator of transcription 3 (STAT3) is a member of the family of latent cytoplasmic transcriptional factors that could regulate cell proliferation, survival, and development. It has been reported that Twist is a target gene of STAT3, and STAT3/Twist signaling plays an important role in regulating cancer progress. Here, to explore whether 8-bromo-7-methoxychrysin (BrMC) inhibits liver cancer stem-like cell (LCSLC) properties via disrupting STAT3/Twist signaling, we cultured SMMC-7721 cells in vitro, and evaluated the effects of BrMC on the stemness of spheroids by determi

Persistent exposure to Mycoplasma induces malignant transformation of human prostate cells.

Recent epidemiologic, genetic, and molecular studies suggest infection and inflammation initiate certain cancers, including those of the prostate. The American Cancer Society, estimates that approximately 20% of all worldwide cancers are caused by infection. Mycoplasma, a genus of bacteria that lack a cell wall, are among the few prokaryotes that can grow in close relationship with mammalian cells, often without any apparent pathology, for extended periods of time. In this study, the capacity of Mycoplasma genitalium, a prevalent sexually transmitted infection, and Mycoplasma hyorhinis, a myco
