Educational attainment and diabetes risk: triangulation evidence from UK Biobank prospective cohort, NHANES 2011-2018, and cross-trait genomics analyses.

BACKGROUND: Lower educational attainment is associated with obesity and type 2 diabetes (T2D), but prospective evidence, external validation, mediator patterns, and genetic triangulation have rarely been integrated. METHODS: UK Biobank analyses included 501,932 participants at baseline and 474,659 participants without baseline diabetes prospectively. Logistic and Cox models estimated associations of higher educational attainment with prevalent and incident diabetes. T2D pathway analyses evaluated adiposity, health behaviors, and cardiometabolic biomarkers. NHANES 2011-2018 provided weighted c

Genome-wide association meta-analysis of human olfactory identification discovers sex-specific and sex-differential genetic variants.

Smelling is a human sense, expressing strong sexual dimorphisms. We aim to improve the knowledge of the genetics of human olfactory perception by performing an exploratory genome-wide association meta-analysis of up to 21,495 individuals of European ancestry. By sex-stratified and overall analysis of the identification of twelve odours and an identification score, we discovered ten independent loci, seven of them novel, with trait-wise genome-wide significance (p < 5 × 10-8) involving five odours. Seven of these loci, including four novel ones, are also significant using a stricter study-wide

NFKB1 and MANBA Confer Disease Susceptibility to Primary Biliary Cholangitis via Independent Putative Primary Functional Variants.

BACKGROUND & AIMS: Primary biliary cholangitis (PBC) is a chronic and cholestatic liver disease that eventually leads to cirrhosis and hepatic failure. We recently identified several susceptibility genes included NFKB1 and MANBA for PBC in the Japanese population by genome-wide association study. However, the primary functional variants in the NFKB1/MANBA region and the molecular mechanism for conferring disease susceptibility to PBC have not yet been clarified. METHODS: We performed high-density association mapping based on a single-nucleotide polymorphism (SNP) imputation analysis, using da…

Investigating the Mediating Role of Mental Disorders in the Relationship Between Early Sexual Intercourse and Intentional Self-Harm: A Two-Step and Multivariable Mendelian Randomization Study.

BACKGROUND: Epidemiological studies have established an association between early sexual intercourse and intentional self-harm. However, the causal mechanisms, particularly the role of mental health disorders, remain elusive. METHODS: In this Mendelian randomization (MR) study, we leveraged genome-wide association study data from multiple sources. Age at first sexual intercourse (AFSI) statistics were derived from the UK Biobank, encompassing up to 406,457 participants of European ancestry. Intentional self-harm (ISH) genome-wide association study (GWAS) data were obtained from the FinnGen Bi…

Genome-wide analysis of 53,400 people with irritable bowel syndrome highlights shared genetic pathways with mood and anxiety disorders.

Irritable bowel syndrome (IBS) results from disordered brain-gut interactions. Identifying susceptibility genes could highlight the underlying pathophysiological mechanisms. We designed a digestive health questionnaire for UK Biobank and combined identified cases with IBS with independent cohorts. We conducted a genome-wide association study with 53,400 cases and 433,201 controls and replicated significant associations in a 23andMe panel (205,252 cases and 1,384,055 controls). Our study identified and confirmed six genetic susceptibility loci for IBS. Implicated genes included NCAM1, CADM2, PH…

Cell-resolved genetic risk reveals hyperandrogenism-associated immune and metabolic rewiring in the PMOS follicular microenvironment.

BACKGROUND: Polyendocrine metabolic ovarian syndrome (PMOS) is a leading cause of anovulatory infertility. Although follicular developmental arrest is a defining feature of PMOS, the cell-type-specific mechanisms linking genetic susceptibility, endocrine imbalance and follicular microenvironmental dysfunction remain incompletely understood, particularly across clinical subtypes. METHODS: We performed single-cell RNA sequencing of preovulatory follicular-fluid cells from healthy controls and women with uncomplicated PMOS, hyperandrogenic PMOS and metformin-treated insulin-resistant PMOS. We in

Mendelian randomization identifies circulating miRNAs as causal mediators of gastric cancer susceptibility and survival outcomes.

Gastric cancer (GC) remains a leading cause of cancer mortality, yet the causal roles of microRNAs (miRNAs) in its pathogenesis are poorly characterized. While observational studies implicate miRNAs in GC progression, confounding biases and tissue-specific limitations hinder causal inference and clinical translation. We conducted a 2-sample Mendelian randomization (MR) analysis using genetic instruments derived from plasma miRNA expression quantitative trait loci (eQTLs). Summary-level data for miRNA-eQTLs were obtained from a study by Huan et al. (involving 5239 individuals and 280 miRNAs), w

Mendelian Randomization Identified SLC2A9 as a Novel cis-eQTL-Mediated Susceptibility Gene in Suppressing Renal Cancer and Its Related Metabolic Mechanisms.

OBJECTIVE: Mendelian randomization (MR) analysis has currently been widely used in combination with expression quantitative trait loci (eQTLs) or cis-eQTLs for identifying novel drug targets in various diseases. Hence in this article, we first integrated MR analysis with eQTLs and cis-eQTLs to reveal the causal effects among SLC2A9 and renal cancer (RC), along with its related metabolic mechanisms. METHODS: Related genome-wide association study (GWAS) data were obtained from online datasets. Two-sample MR and summary-data-based MR study (SMR) analyses were applied to assess the causal effects…

Understanding the bidirectional relationship between chronic respiratory disease and cardiovascular disease using genetic evidence.

BACKGROUND: Chronic respiratory diseases (CRDs) and cardiovascular diseases (CVDs) are leading global health burdens. Despite being common, CRD and CVD comorbidity is often underestimated due to overlapping symptoms and risk factors. Consequently, their relationship remains unclear. AIMS AND OBJECTIVES: To determine the bidirectional genetic relationship between CRD and CVD and explore smoking and inflammation as potentially shared joint risk factors. METHODS: We conducted bidirectional Mendelian randomisation (MR) to explore CRD-CVD relationships. Summary statistics from genome-wide associa

The causal effect of matrix metalloproteinase 9 on interstitial lung disease risk mediated by effector memory double negative T cells: A mediation Mendelian randomization study.

Matrix metalloproteinases (MMPs) have been implicated in the pathogenesis of interstitial lung disease (ILD). However, the causal role of MMPs in ILD pathogenesis remains to be elucidated. This study aimed to investigate the causal effects of circulating MMPs on ILD risk and to evaluate whether immune cell phenotypes mediate this relationship. A comprehensive Mendelian randomization (MR) and mediation analysis was conducted using publicly available data of genome-wide association study (GWAS) summary statistics. Genetic instruments for 6 MMPs (MMP-1, MMP-2, MMP-3, MMP-7, MMP-9, and MMP-12) and…

Exome sequencing reveals rare loss-of-function mutations in FLG and immune genes in patients with multiple food allergies.

BACKGROUND: Studies of twins, parents, and families have demonstrated a strong heritable role in allergy and food allergy in particular (60%-80%). However, genetic loci identified in previous genome-wide association studies focused on common variants, have not withstood validation, are not located inside protein coding exons, do not explain familial allergy, or have a low effect size. OBJECTIVE: We sought to apply forward genetics of families and exome sequencing to identify the frequency of Mendelian gene mutations for food allergy with emphasis on rare coding high-risk variants. METHODS: T

The causal relationship between neuroimaging and Parkinson's disease: A two-sample Mendelian randomization study.

Parkinson's disease (PD) is a neurodegenerative disorder whose etiology and progression remain not fully understood, posing significant challenges for both diagnosis and treatment. This study leverages the 2-sample Mendelian Randomization (TSMR) approach to systematically examine the possible causal connections involving neuroimaging characteristics and the risk of developing PD. Making use of genetic variations from extensive genome-wide association studies (GWAS) as instrumental factors, this analysis focuses on neuroimaging traits related to cortical and subcortical gray matter volumes. AI-guided multi-omics analysis identifies NPC1-modulated susceptibility to SARS-CoV-2 infection under PM2.5 exposure.

Exposure to airborne fine particulate matter (PM2.5) has been linked to increased risk of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection, yet the underlying mechanisms remain unclear. Here, by leveraging a fine-tuned foundation model of single-cell transcriptomics, we uncover shared transcriptional signatures between PM2.5 exposure and SARS-CoV-2 infection. We further validate this association using population-level epidemiological analyses and perform genome-wide association studies (GWAS) to identify genetic variants that modulate infection risk under PM2.5 exposu

Assessment of the Genetic Relationship Between Circulating Cytokines and Calcific Aortic Valvular Stenosis Using a Bidirectional Mendelian Randomization Analysis.

Available studies have found associations between several cytokines and calcific aortic valvular stenosis (CAVS). To better comprehend the causal link between circulating cytokines and CAVS, we performed a bidirectional Mendelian randomization (MR) study. Genetic variants associated with 41 cytokines were obtained from public genome-wide association study (GWAS), and summary statistics for GWAS of CAVS were obtained from the FinnGen consortium. Forward MR analysis was conducted to determine the impacts of 41 cytokines on CAVS risk and reverse MR analysis was used to determine whether genetic s…

Body mass index and type 2 diabetes liability in osteomyelitis and cellulitis: A cross-biobank Mendelian randomization study.

Previous Mendelian randomization (MR) studies have linked obesity and type 2 diabetes (T2D) liability to osteomyelitis and skin or soft-tissue infection, but the reproducibility of these associations across independent outcome biobanks remains uncertain. We conducted cross-biobank univariable MR and multivariable MR analyses using European-ancestry genome-wide association study summary statistics. FinnGen Release 13 osteomyelitis (2519 cases and 471,012 controls) was designated as the primary clinical outcome, whereas cellulitis (10,223 cases and 468,418 controls) served as a comparative infec…

Decoding Causal Associations Between Neuropsychiatric Disorders and Rotator Cuff Tendinopathy: A Two-Sample Mendelian Randomization Study.

BACKGROUND: Rotator cuff tendinopathy (RCT) is a major contributor to over 30 million surgeries which are conducted to treat shoulder overuse injuries worldwide annually. Accumulating evidence indicates that neuropsychiatric disorders (ND) share pathogenic pathways with tendinopathy. However, unclarified causal relationships between these two disease spectra undermine the individualized design of treatment strategies benefiting patients with ND while minimizing the RCT risk. We aimed to unveil whether ND were genetically associated with increased RCT occurrence by conducting a two-sample Mende

A Cis-Regulatory Duplication in a Hox Hotspot Implicated in Mimetic Convergence in the Bumble Bee Bombus flavifrons.

Several species of North American bumble bees spanning the Pacific Coastal and Rocky Mountain regions converge onto distinct mimetic abdominal colour forms for each region by switching abdominal coloration from black to red. Previous genome-wide association studies (GWAS) of red and black transitions in two mimics (Bombus melanopygus and Bombus vancouverensis) revealed that black forms were generated by independently deleting a portion of the same cis-regulatory region near the Hox gene Abdominal-B (Abd-B). Here, we test the genetic basis of these mimetic colour forms in a third co-mimic, Bomb
