[Antibiotic therapy of intra-abdominal infections in the era of multiresistance].

Recommendations for the treatment of intra-abdominal infections (IAI) caused by drug-resistant bacteria often fail to mention the bacteria o … ntered approach is mandatory to evaluate the optimal antimicrobial treatment regimen. Especially in gram-negative bacteria, which are the predominant cause only a few options remain for treatment. Clinical data with a high level of evidence are very limited. Future studies should focus on pharmacokinetic and pharmakodynamic aspects in critically ill patients, in the sen …

Studies on the phyllosphere microflora of tapioca (Manihot utilissima Pohl).

Studies on the phyllosphere microflora of tapioca (Manihot utilissima Pohl.) revealed that incidence of bacteria, fungi, and actinomycetes is indipendent of the variety, but dependent on the age. The bacteria and fungi increased with increasing age of the leaf and were more on mature leaves, whereas no such difference was noticed in the occurrence of actinomycetes. The large majority of the isolates, that were Gram-negative bacteria, was also amino acid-requiring. The chromogenic types occurred in greater abundance. The phyllosphere mycoflora consisted mainly of species of Asperigullus, Penici…

Eradication of a multiple drug resistant Salmonella paratyphi A causing meningitis with ciprofloxacin.

A previously healthy breast-fed baby was admitted at 10 days of age to a hospital in the north of Pakistan with diarrhoea and fever. He was … ital, Karachi. Examination of CSF there confirmed the diagnosis of pyogenic meningitis and revealed Gram-negative bacteria. Cultures of CSF and faeces yielded Salmonella paratyphi A but the blood culture was negative. The isolate was found to be multiple antimicrobially resistant but sensitive to ciprofloxacin. Treatment with this drug was therefore started 3 days after …

Loss of the Ap4A hydrolase YqeK impairs stress adaptation and virulence gene expression in Staphylococcus aureus.

UNLABELLED: The nucleotide diadenosine tetraphosphate (Ap4A) accumulates during stress across organisms and cell types and is widely hypothesized to be an alarmone or second messenger. While gram-negative bacteria use ApaH-family hydrolases to degrade Ap4A and other dinucleoside tetraphosphates (Ap4Ns), gram-positive bacteria, including Staphylococcus aureus, use YqeK. Inactivation of Ap4A hydrolases and corresponding Ap4A accumulation cause diverse phenotypic effects in gram-negative and gram-positive species, ranging from increased antimicrobial sensitivity to reduced virulence. However, the…

Integrating synthesis, pharmacological evaluation, and molecular dynamics simulation of novel 8-substituted theophylline hybrids as potential PDE-4B inhibitors, bronchodilators and antibacterial.

Methylxanthines, particularly theophylline, have long served as effective bronchodilators for severe asthma. Emerging evidence links bacterial infection to asthma pathogenesis, motivating the search for multifunctional therapeutic agents. In this study, a series of 8-substituted 1,3-dimethylxanthines bearing aryl and heteroaryl groups were synthesized and structurally characterized by NMR, elemental analysis, and HR-ESI-MS. Most compounds exhibited significant bronchodilator activity in an acetylcholine-induced guinea pig model, surpassing theophylline. The compounds demonstrated potent in vit

Effects of parental omega-3 fatty acid intake on offspring microbiome and immunity.

The "Western diet" is characterized by increased intake of saturated and omega-6 (n-6) fatty acids with a relative reduction in omega-3 (n-3) consumption. These fatty acids can directly and indirectly modulate the gut microbiome, resulting in altered host immunity. Omega-3 fatty acids can also directly modulate immunity through alterations in the phospholipid membranes of immune cells, inhibition of n-6 induced inflammation, down-regulation of inflammatory transcription factors, and by serving as pre-cursors to anti-inflammatory lipid mediators such as resolvins and protectins. We have previou

Genetic variations in toll-like receptor 4 in Mexican-Mestizo patients with intra-abdominal infection and/or pneumonia.

Sepsis is a leading cause of death around the world, and 73-83% of all sepsis cases requiring attention in intensive care units are linked to intra-abdominal infection (IAI) or pneumonia. The activation of innate immunity is central to the manifestation of sepsis, and toll-like receptor (TLR) 4 plays an important role in this activation process. The 299G and 399I alleles of TLR4 have been linked with an increased risk of Gram-negative bacteria (GNB) infections and septic shock in some populations. This case-control study evaluated the prevalence of D299G/T399I polymorphisms in Mexican patients…
