Targeting DDR1 with novel synthesized antagonists delays cellular aging and enhances wound healing in mesenchymal stem cells.

Mesenchymal stem cells (MSCs) are central to regenerative medicine; however, their clinical utility is constrained by reduced proliferative capacity and replicative senescence during ex vivo expansion. Here, we report on the rational design and biological characterization of novel phthalimide-based inhibitors targeting discoidin domain receptor 1 (DDR1). Structure-activity relationship optimization efforts led to the identification of AC-4067 and AC-4061 as potent DDR1 inhibitors, with IC₅₀ values of 30.9 nM and 53.6 nM, respectively. AC-4067 demonstrated 18-fold selectivity for DDR1 over DDR2

Revealing the antibacterial mechanism of dielectric barrier discharge cold plasma and lactic acid synergy against Pseudomonas fragi based on molecular docking and oxidative damage.

Slaughter process of broiler is a major source of microbial contamination and chilling is one of the most important steps for carcasses' sterilization. However, limited research has focused on the carcasses' sterilization technology. Furthermore, the use of sodium hypochlorite in chilling operations can pose risks to human health. Therefore, we hypothesized that synergistic application of dielectric barrier discharge cold plasma (DBD) combined with lactic acid (LA) (DBD + LA) can effectively inhibit the growth of proliferation of specific spoilage bacteria in carcasses. Based on this hypothesi

Fabric-Templated Co-Deposition of Nanofiber-Reinforced Composite Films with Ordered Surface Arrays for Ultrasensitive Wearable Sensing.

Fabricating polymeric films with ordered surface microarrays is of great significance for wearable sensing applications, yet it remains challenging to achieve scalable, cost-effective, and biocompatible architectures. Herein, we report a fabric-templated co-deposition strategy to construct nanofiber-reinforced composite films with well-ordered surface arrays. The approach utilizes woven cotton fabrics as soft templates, where capillary action directs the deposition of chitosan/poly (vinyl alcohol-co-ethylene) nanofiber mixtures within the microscale grooves, enabling direct replication of the

Functional redundancy and structural flexibility as contrasting plankton strategies under macrotidal pulses.

Macrotidal estuaries are hydrodynamic pulse systems that continuously reconfigure coastal environments. While seasonal dynamics are well documented, the short-term mechanisms enabling plankton to persist in these hyperdynamic habitats remain poorly understood. We investigated how planktonic communities at a tropical Amazonian Ramsar site respond to high-frequency tidal oscillations. Integrating diel sampling with random forest and SHAP modeling revealed that extreme tidal energy acts as a severe ecological filter, driving a temporal decoupling between nutrient pulses during ebb flows and phyto

Disease-stage-associated transcriptional responses underlying bacterial wilt resistance in tobacco cultivar Yunyan 87 caused by Ralstonia solanacearum.

Bacterial wilt caused by Ralstonia solanacearum severely limits tobacco production, yet the molecular basis of resistance to vascular wilt diseases remains poorly understood. We integrated field phenotyping, pathogen isolation, and transcriptomic analyses across different disease stages to investigate root responses of the resistant tobacco cultivar Yunyan 87 at healthy, moderately infected, and severely infected stages. Disease progression was accompanied by marked root biomass loss, with fresh and dry weights reduced by approximately 60% and 70%, respectively. Transcriptomic analyses identif

De novo Genes in Plants: Origins, Mechanisms, and Functional Implications.

De novo genes originate from previously non-coding genomic regions. They provide an important source of lineage-specific innovation. In plants, these genes may contribute to adaptation, trait diversity and crop evolution. This review summarizes recent progress in plant de novo gene research. It first discusses major routes of gene birth, including transcription-first, open reading frame (ORF)-first and concurrent models. It also examines how nascent loci acquire regulatory control and enter existing biological networks. The review then summarizes their evolutionary features, including weak ear

Current Single-cell Proteins as Ingredients: Functions, Challenges, and Regulatory Frameworks.

Single-cell proteins (SCPs) broadly comprise microbial biomass or biomass-derived proteinaceous products produced from bacteria, yeasts, filamentous fungi, or microalgae. In the broader field of cell-derived alternative proteins, animal- and plant-cell-derived platforms are also being explored because they can reproduce the structural, compositional, and sensory equality of conventional food materials. In contrast, microbial SCPs are distinguished by diverse cellular structures, metabolites, cell-wall components, and bioactive fractions that may confer functionality beyond nutritional suppleme

ChIPmentation-based mapping of histone modifications in aging: Experimental design and analytical considerations.

Aging is accompanied by progressive alterations in chromatin organization that contribute to transcriptional dysregulation, genomic instability, and loss of cellular identity. Among the epigenetic mechanisms implicated, histone modifications play a central role in regulating chromatin accessibility and gene expression during aging. Changes in key histone marks, including H3K9me3, H3K27me3, H4K20me3, and histone acetylation signatures, are associated with heterochromatin loss, increased transcriptional noise, and altered cellular function across diverse tissues and organisms. These modification

Crosstalk between integrin signaling and NAD⁺ biosynthetic pathways promotes glycolysis, proliferation, survival, and tumor growth in triple-negative breast cancer.

Triple-negative breast cancer (TNBC) is an aggressive breast cancer subtype with limited therapeutic options. Here, we investigated how integrin-dependent signaling pathways regulate tumor metabolism and therapeutic vulnerability in TNBC. Pharmacological inhibition of the integrin/FAK axis and/or BRD4 induced cell cycle arrest, autophagy, and senescence in highly proliferative cells, consistent with a metabolic stress phenotype. Metabolomic analyses using [U-¹³C]-glucose revealed a marked suppression of glycolytic carbon flux, accompanied by an approximately 30-47% reduction in intracellular N

Foliar metal micronutrients reshape rhizosphere soil multifunctionality by filtering microbial life-history strategies.

Foliar application of metal micronutrients is increasingly adopted in intensive cultivation systems, yet its potential ecological risks to rhizosphere functions remain poorly understood. Here, using the medicinal plant Panax notoginseng as a model, we conducted a gradient foliar amendment experiment with iron (Fe), zinc (Zn), and copper (Cu) to evaluate how aboveground metal inputs regulate rhizosphere soil multifunctionality (MF) through microbial life-history strategies. By integrating 16S rRNA amplicon sequencing, metagenomics, root transcriptomics, and a newly developed quantitative Yield-

PDA-Based Nanoplatform with High Ligustilide Loading for Simultaneous Elimination of Breast Cancer Cells and Stem Cells Via BAX/BCL-2/Caspase-3 Pathway Activation.

Breast cancer (BC) remains the most prevalent and deadly malignancy among women worldwide. While photothermal therapy (PTT) and chemodynamic therapy (CDT) offer tumor-specific and minimally invasive advantages, their standalone efficacy is limited. Ligustilide (LIG) can exert antitumor effects by generating •O2-, but its bioavailability is poor, thus necessitating improved delivery strategies. Here, we reported a multifunctional nanoplatform (FeD@LIG) integrating PTT, CDT, and chemotherapy for the simultaneous elimination of breast cancer stem cells (BCSCs) and non-BCSCs. FeD@LIG was synthesiz

Clemastine ameliorates ulcerative colitis-induced cognitive impairment by restoring PI3K/Akt/GSK-3β signaling and suppressing ferroptosis.

Ulcerative colitis (UC) is a refractory inflammatory bowel disease with ongoing colonic inflammation and extra-intestinal manifestations, including cognitive impairment. In this study, we evaluated the peripheral and central effects of clemastine on cognitive impairment induced by UC using an acetic acid (1 ml, 4% v/v) model with emphasis on the role of PI3K/Akt/GSK-3β signaling, ferroptosis, and autophagy in its possible mediated neuroprotection. Clemastine revealed a dose-dependent improvement of colonic damage linked with UC, as demonstrated by inhibition of TNF-α, IL-1β, and caspase-3 leve
