IPEC-J2 as a cellular model for studying intestinal mucus.

Mucus in the proximal small intestine serves critical protective and transport functions, regulating nutrient absorption to enterocytes. The … ometry revealed that ~ 8% of IPEC-J2 cells expressed MUC2 in 5PSAg, comparable to the proportion of jejunum's goblet cells in vivo (~ 5%). This study demonstrated that IPEC-J2 cells can differentiate into mucus-secreting cells under specific culture conditions, and they are a suitable in vitro model for investigating interactions between mucus and food co …

Assessing seromuscular layer and serosa removal on intestinal permeability measurements in weaned piglet everted sac segments.

The integrity of the intestinal barrier is crucial for regulating the passage of pathogens and toxins, while facilitating nutrient absorption. The everted gut sac technique, an ex-vivo technique, can be used to study interventions on barrier function. This cost-effective approach utilizes relatively large gut segments to study specific intestinal regions. Typically, intact (non-stripped) intestinal segments are used, but their use may underestimate permeability due to the medial positioning of blood vessels relative to the seromuscular layer and serosa. However, removing these layers risks phy

Is duodeno-jejunal bypass liner superior to pylorus preserving bariatric surgery in terms of complications and efficacy?

PURPOSE: Based on recent scientific evidence, bariatric surgery is more effective in the management of morbid obesity and related comorbidities than conservative therapy. Pylorus preserving surgical procedures (PPBS) such as laparoscopic single-anastomosis duodeno-jejunal or duodeno-ileal bypass with sleeve gastrectomy are modified duodenal switch (DS) surgical techniques. The duodeno-jejunal bypass liner (DJBL) is a novel surgical method in the inventory of metabolism focused manual interventions that excludes duodeno-jejunal mucosa from digestion, mimicking DS procedures without the risk of

Preservation and Processing of Intestinal Tissue for the Assessment of Histopathology.

The intestine is often examined histologically in connection with allergies and in search for pathological changes. To be able to examine the intestine histologically with a microscope, it must be sampled and processed correctly. For microscopic analysis, the samples have to be cut into thin sections, stained, and mounted on slides. Since it is not possible to cut fresh samples without damaging them, they must first be fixed. The most common method, which is described herein, is the fixation in formalin with subsequent embedding in paraffin and staining of the slides with hematoxylin and eosin

Protective effects of a cystine and theanine mixture against acute radiation injury in rats.

This study aims to examine the effects of cystine and theanine (CT), which increases glutathione biosynthesis, on the survival rate and acute radiation injury of the small intestine and bone marrow using a rat model. CT pre-treatment (280 mg/kg for 5 days) significantly improved weight loss and survival rate of rats as compared with the control group after 5 Gy. CT pre-treatment significantly increased the rate of mucosa and crypt length, and decreased the number of apoptotic cells, TUNEL and cleaved caspase-3 positive cells, while increasing the number of mitotic cells and Ki-67 positive cell

Study on the interactive effect of deoxynivalenol and Clostridium perfringens on the jejunal health of broiler chickens.

With global warming and ban on antibiotics, it occurs occasionally that deoxynivalenol (DON) together with Clostridium perfringens impairs the gut health of broiler chickens. However, the interactive effect of DON and C. perfringens on intestinal health is still unknown. A total of 120 one-day-old Arbor Acres broilers were randomly distributed to 4 groups. Birds were gavaged with C. perfringens (8 × 108 CFU/d per bird) or sterile medium and fed a DON diet (0 or 5 mg of DON per kg diet) to investigate the interactive effects. The main effect analysis showed that DON diet significantly downregul

Use of encapsulated L-lysine-HCl and DL-methionine improves postprandial amino acid balance in laying hens.

The supplementation of dietary limiting amino acids (AA) with crystalline AA makes the use of low-protein diets an option in poultry product … sion levels of AA transporters CAT-1, y+LAT1, b0,+AT, B0AT, rBAT, EAAT3, and PepT1 in the duodenum, jejunum, and ileum were not influenced (P > 0.05) by dietary treatment. There was an interaction of dietary AA treatment and time (P < 0.05) and the 80CLM hens exhibited higher concentrations of Lys (P < 0.05) than the controls at 2-h time point. In contras …

Uridine/UMP metabolism and their function on the gut in segregated early weaned piglets.

Uridine monophosphate (UMP) is a major nucleotide analogue in mammalian milk and uridine (UR) is its gastro-intestinal metabolite in vivo. This study aims to investigate the functional effects of UMP and UR on the gut in vitro and in vivo. Twenty-one piglets were randomly allotted into three groups, the control group, UMP group and UR group, and orally administered UMP or UR for 10 days. Results showed that UMP and UR supplements improved the ADG of piglets, and decreased the diarrhea rate. UR increased the jejunum villus length/crypt depth ratio, Claudin-3 and E-cadherin expression, and the p…

Recurrent intestinal obstruction in a patient of Peutz-Jeghers syndrome.

Peutz-Jeghers syndrome is a rare hamartomatous polyposis syndrome characterized by the presence of intestinal polyps and mucocutaneous melanotic pigmentations. It is associated with various gastrointestinal and extraintestinal malignancies. This case report deals with the clinical presentation, investigations, operative findings, and outcome of a patient harboring this disease. A 45-year-old female presented to us with intermittent colicky abdominal pain for the last 6 months and a single episode of melena 1 month back. She had a previous history of resection of a jejunal growth 22 years back.

Effect of body condition on intestinal permeability in horses.

OBJECTIVE: To investigate effects of body condition on permeability of intestinal mucosa in horses. ANIMALS: 13 horses (7 obese and 6 lean) from 8 to 15 years of age. PROCEDURES: Body condition score was assessed, and an oral sugar test (OST) was performed to evaluate glucose and insulin dynamics. Horses were allowed a 2-week diet acclimation period and were then euthanized. Tissue samples were collected from the jejunum, ileum, cecum, pelvic flexure, right dorsal colon, and rectum. Mucosal permeability was assessed by measuring transepithelial resistance and lipopolysaccharide (LPS) flux ac…

New Modality of Treatment for Malnutrition After Laparoscopic Roux-en-Y Gastric Bypass: the "Small Bowel Limb Transposition" Technique.

The technique of "small bowel limb transposition" consists in leaving the gastro-jejunal anastomosis (GJA) intact. It involves stapling (sectioning) of the alimentary limb (AL) at the level of the gastric antrum and creating a new anastomosis between the AL and the antrum. The "small bowel limb transposition" is an easy, simple, and interesting technique to be performed for the correction and treatment of severe malnutrition after laparoscopic Roux-en-Y gastric bypass, but this single case is not sufficient to prove that this is a standard option.

A Synbiotic with Tumor Necrosis Factor-α Inhibitory Activity Ameliorates Experimental Jejunoileal Mucosal Injury.

Despite the recent development of biological modifiers for inflammatory bowel diseases (IBD), there continues to be considerable interest in fermented medicines because of its negligible adverse effects. We previously showed that the synbiotic Gut Working Tablet (GWT) alleviates experimental colitis. Here we show that GWT is capable of ameliorating jejunoileal mucosal injury, which is frequently seen with IBD. We created experimental jejunoileal mucositis in rats by injection of methotrexate (MTX) which increases intestinal permeability, a hallmark finding of IBD. Administering GWT to MTX-inje
