Chemical structure of corticosteroids and its relationship with their acute induction of lordosis in the female rat.

The intravenous injection of several corticosteroids in the spayed estrogen-primed rat resulted, 5 min later, in a remarkable induction of lordosis response for deoxycorticosterone and 11-deoxycortisol. A lower but important effect was induced with corticosterone and 17 alpha-hydroxyprogesterone. A chemical structure-biological effect analysis showed that C-21 hydroxylation (e.g., deoxycorticosterone, 11-deoxycortisol, and corticosterone) induces the behavioral effect. However, C-11 or C-17 hydroxylation alone promotes the lordosis response to a lesser degree (corticosterone and 17 alpha-hydro

Comparing Empirical and Physics-Based Models of Intermolecular Dispersion and Repulsion Energies.

Understanding molecular recognition, crystal packing and biological processes including protein-ligand binding, protein folding, and macromolecular structure all depend on accurate modeling of noncovalent interactions. In this study, we introduce and evaluate two complementary models─empirical and physically grounded ─for computing exchange-repulsion, and dispersion energies. Once combined, these two terms form van der Waals (vdW) potentials allowing the computation of total vdW interaction energies. A set of 21 atom-type-specific vdW parameters of a buffered 7-14 potential was derived via a l

Bioactive Compounds of Sideritis Species in Inflammation, Neuroprotection and Cardiometabolic Health: A Review.

Sideritis species (Lamiaceae), the aromatic mountain teas of the Mediterranean and Balkans, accumulate a phytochemically rich array of bioactive compounds-phenylethanoid glycosides (notably verbascoside), flavonoid aglycones and glycosides, phenolic acids, and terpenoids-whose pharmacological relevance is increasingly defined at the molecular level. This review synthesizes current evidence on the molecular mechanisms through which these constituents act in human health and disease, integrating in vitro, in vivo, in silico, and clinical data. Anti-inflammatory activity is attributed to modulati

Preparation, molecular docking, and biological evaluation of radioiodinated cefaclor for inflammation detection.

BACKGROUND: Inflammation serves as a natural defense mechanism; however, its persistence can lead to chronic diseases with serious clinical consequences. Early detection of inflammation is therefore critical to slowing disease progression and improving therapeutic outcomes. METHODS: In this study, cefaclor (Cefa) was successfully radiolabeled with iodine-131 via electrophilic substitution to facilitate the detection of infected and inflamed muscles in mouse models. The labeling reaction was carried out with 100 µg of Cefa and 100 µg of iodogen, using glass frits as the oxidizing system at pH 7

Phenylalanine Transport through LAT1: Insights from Molecular Dynamics, Steered Molecular Dynamics, and Targeted Molecular Dynamics.

L-type amino acid transporter 1 (LAT1) is a heterodimeric membrane protein that primarily facilitates the transport of phenylalanine, along with other amino acids such as valine, leucine, isoleucine, tryptophan, and tyrosine across the cell membrane. It is predominantly expressed at the blood-brain barrier. While LAT1 has been extensively studied in the context of drug delivery, its specific transport pathway for phenylalanine and the related conformational dynamics remains largely unexplored. Investigation of the transport pathway of phenylalanine via LAT1 is also very important, as excessive

Cell-Type-Dependent Responses to Oleuropein in MDA-MB-231 Breast Cancer Cells and HUVECs.

Breast cancer progression is associated with dysregulation of cell survival, apoptosis, and cell-cycle control. Oleuropein (OL), a major secoiridoid constituent of olive-derived products, has attracted interest because of its diverse biological activities and potential relevance to cancer research. This study evaluated the cellular and molecular responses to OL in MDA-MB-231 triple-negative breast cancer cells and non-malignant human umbilical vein endothelial cells (HUVECs). Cell viability was assessed following exposure to 10-250 µg/mL OL for 24, 48, and 72 h. OL produced an exposure-time-de

IFI44L modulates chemoradiotherapy sensitivity and CD8+ T cell infiltration in colorectal cancer.

OBJECTIVE: Chemoradiotherapy (CRT) represents a cornerstone treatment for colorectal cancer (CRC), yet heterogeneous responses to CRT in treatment response remain a major clinical challenge. Increasing evidence suggests that the tumor immune microenvironment (TME) especially intratumoral CD8+ T cells plays an important role in determining CRT efficacy; however, the molecular mechanisms linking tumor-derived factors to immune-mediated treatment sensitivity remain poorly understood. Here, we aimed to identify and characterize regulators of CRT response and the underline mechanisms in CRC. METHO

Integrating machine learning and structure-based approaches for repurposing potent tyrosine protein kinase Src inhibitors to treat inflammatory disorders.

Tyrosine-protein kinase Src plays a key role in cell proliferation and growth under favorable conditions, but its overexpression and genetic mutations can lead to the progression of various inflammatory diseases. Due to the specificity and selectivity problems of previously discovered inhibitors like dasatinib and bosutinib, we employed an integrated machine learning and structure-based drug repurposing strategy to find novel, targeted, and non-toxic Src kinase inhibitors. Different machine learning models including random forest (RF), k-nearest neighbors (K-NN), decision tree, and support vec

Meis transcription factors regulate cardiac conduction system development and adult function.

AIMS: The cardiac conduction system (CCS) is progressively specified during development by interactions among a discrete number of transcription factors (TFs) that ensure its proper patterning and the emergence of its functional properties. Meis genes encode homeodomain TFs with multiple roles in mammalian development. In humans, Meis genes associate with congenital cardiac malformations and alterations of cardiac electrical activity; however, the basis for these alterations has not been established. Here, we studied the role of Meis TFs in cardiomyocyte development and function during mouse d
