Prenatal exercise regulates influence of parental body mass index on birth outcomes.

BACKGROUND: Offspring body mass index is often higher in parents with overweight or obesity, thereby increasing the risk of obesity later in life. As data has shown that exercise during pregnancy reduces gestational weight gain and offspring adiposity, we believe the intergenerational risk of obesity could be reduced. OBJECTIVE: This study aimed to test the influence of paternal and maternal overweight or obesity on neonatal body mass index and other birth measures, and whether exercise during pregnancy would improve outcomes. METHODS: Prenatal exercise (aerobic, resistance, or combination)

Impact of Maternal Diabetes on the Incidence of Critical Congenital Heart Disease in the United States.

BACKGROUND: Critical congenital heart disease (CCHD) represents a significant subset of congenital heart disease (CHD). While the association between maternal diabetes mellitus and offspring CHD is well established, the specific relationship between maternal diabetes and CCHD remains underexplored. OBJECTIVES: This study aims to investigate the association between maternal diabetes and the incidence of offspring CCHD. METHODS: We analyzed natality data from the Centers for Disease Control and Prevention Wide-Ranging Online Data for Epidemiologic Research (CDC WONDER) from 2016 to 2021. The d

Cytogenomic characterization of karyotypes with additional autosomal material.

Chromosomal rearrangements involving additional material in individuals with phenotypic alterations usually result in partial trisomy, often accompanied by partial monosomy. To characterize chromosomal rearrangements and analyze genomic characteristics in the breakpoint regions in 31 patients with additional material on an autosomal chromosome. Different tests were performed to characterize these patients, including karyotyping, chromosomal microarray analysis (CMA), and fluorescent in situ hybridization (FISH). In silico analyses evaluated A/B chromosomal compartments, segmental duplications,

Case Report: Heterozygous ADAR c.3019G>A pathogenic variant associated with variable neurological symptoms and incomplete penetrance in a four-generational family.

Heterozygous pathogenic variants in ADAR have been associated with dyschromatosis symmetrica hereditaria, while biallelic pathogenic variants have been associated with Aicardi-Goutières syndrome 6 (AGS6). However, the heterozygous variant c.3019G>A, (p.Gly1007Arg) has been described to cause neurological manifestations, which resemble AGS6 and are associated with an upregulation of interferon-stimulated genes. We report a four-generation family with two symptomatic family members and five unaffected carriers of the heterozygous pathogenic ADAR variant c.3019G>A. The index (patient 1) manifeste

Novel mutations of maternal effect gene thyroid hormone receptor interactor 13 involved in biparental complete hydatidiform mole.

RATIONALE: Biparental complete hydatidiform mole (BiCHM) is a rare form of molar pregnancy, frequently associated with familial recurrence, whose mechanism was historically unclear. It is classically diploid with biparental inheritance, but manifests as a complete hydatidiform mole (CHM). Cytogenetic studies suggested a link to maternal mutations in imprinted genes NOD-like receptor family, pyrin domain containing 7 or Kelch helper domain containing 3-like within the oocyte. This case reports a novel gene association. PATIENT CONCERNS: A 43-year-old woman presented with a history of recurrent

MicroRNA participates in embryo implantation by modulating endometrial tolerance in sows during peri-implantation period.

MicroRNAs (miRNAs) are a class of non-coding RNA. MiRNAs affect physiological processes by regulating messenger RNA (mRNA) translation of target genes. The peri-implantation period is the period with the most loss of pig embryos, during which the endometrium provides support for embryo selectivity. The effects of miRNAs during the porcine peri-implantation period include roles in pregnancy recognition, embryo adhesion, maternal vascular support, and immune system modulation. 1) During peri-implantation period, miRNAs regulates the synthesis and secretion of estrogen, progesterone and prostagla

Transient neonatal hyperparathyroidism caused by a monoallelic TRPV6 dominant negative variant.

Transient neonatal hyperparathyroidism (TNHP) is a rare disorder characterized by severe skeletal abnormalities in newborns, resulting from insufficient maternal-fetal calcium transport across the placenta. Transient receptor potential cation channel, subfamily V, member 6 (TRPV6) plays a significant role in placental calcium transport. While biallelic variants in the TRPV6 gene have been associated with TNHP, we present a case involving a neonate of Malay descent, born to non-consanguineous parents, who exhibited severe skeletal abnormalities at birth that resolved spontaneously within 6 wk.

DDOST-Congenital Disorder of Glycosylation: Defining the Clinical Spectrum and First Report of a Structural Variant.

Congenital disorders of glycosylation (CDG) are a heterogeneous group of metabolic diseases caused by defects in glycan biosynthesis, predominantly affecting the N-glycosylation pathway. Pathogenic variants in the DDOST gene, encoding a non-catalytic subunit of the oligosaccharyltransferase (OST) complex, underlie an ultra-rare CDG subtype with only three patients reported to date, complicating diagnosis and genotype-phenotype correlations. We describe a female patient with developmental delay, hypotonia, and dysmorphic features. Genetic analyses included chromosomal microarray, FMR1 testing,

Ecuador genetic mosaic: biological and adaptive variations across Mestizos, Native Amerindians, and Afro-Ecuadorians. Implications for public health and precision medicine.

INTRODUCTION: Understanding human biological diversity is fundamental to improving health and addressing inequities, yet most genomic and biomedical studies remain focused on European and Asian populations. Latin American groups-particularly mestizos, Native Amerindians, and Afro-descendants-are underrepresented, limiting the applicability of global findings. Ecuador, with its tri-hybrid ancestry shaped by Amerindian, European, and African lineages, provides a valuable model to explore how genetic, environmental, and sociocultural factors jointly influence adaptation and disease. OBJECTIVE: T

Risk score for non-vaccination of voluntary vaccines: The Japan Environment and Children's Study.

BACKGROUND: In Japan, as several important vaccines are still categorized as voluntary vaccines that require out-of-pocket payment, the vaccination coverage of voluntary vaccines is lower than that of routine vaccines. Thus, higher voluntary vaccination rates are desired. Herein, we used nationwide birth cohort data to create a voluntary vaccine risk score to identify high-risk individuals who were not vaccinated with voluntary vaccines. METHODS: The data from 74,733; 73,571; and 74,360 infants were analyzed for rotavirus, mumps virus, and influenza virus vaccinations, respectively. The risk

De novo variants predicting haploinsufficiency for DIP2C are associated with expressive speech delay.

The disconnected (disco)-interacting protein 2 (DIP2) gene was first identified in D. melanogaster and contains a DNA methyltransferase-associated protein 1 (DMAP1) binding domain, Acyl-CoA synthetase domain and AMP-binding sites. DIP2 regulates axonal bifurcation of the mushroom body neurons in D. melanogaster and is required for axonal regeneration in the neurons of C. elegans. The DIP2 homologues in vertebrates, Disco-interacting protein 2 homolog A (DIP2A), Disco-interacting protein 2 homolog B (DIP2B), and Disco-interacting protein 2 homolog C (DIP2C), are highly conserved and expressed w

Maternal Accommodation of Adolescent Body Dysmorphic Disorder: Clinical Correlates and Association with Treatment Outcomes.

Family accommodation (FA) is widely-recognised as an important clinical phenomenon in obsessive-compulsive disorder (OCD) and anxiety disorders, and is related to poorer treatment outcomes. However, FA has not been quantitatively explored in Body Dysmorphic Disorder (BDD). The aim of this study was to investigate the patterns and correlates of maternal accommodation in adolescent BDD, and its association with treatment outcomes. Participants were 131 adolescents with BDD and their mothers who completed the Family Accommodation Scale-Parent Report (FAS-PR) as part of routine clinical practice i
