Airborne transmission-mediated delivery of a live-attenuated influenza virus vector-based intranasal COVID-19 vaccine protects Syrian hamsters against SARS-CoV-2 challenge.

Given their critical role in eliciting respiratory immunity, mucosal SARS-CoV-2 vaccines receive significant research attention. Our previous phase 1/2 trial revealed that aerosolized live-attenuated vectored influenza virus COVID-19 vaccine likely elicited specific cellular immunity and weak s-IgA responses in placebo recipients. Here, we demonstrated that viral vector vaccines undergo contact or airborne transmission in various animal models. Vaccines have difficulty spreading in the mouse model, but can effectively spread through the air in more sensitive ferrets. However, the virus transmi…

Superconducting gap of H3S measured by tunnelling spectroscopy.

Several hydrogen-rich superconductors have been found to show unprecedentedly high critical temperatures1-4, stimulating investigations into the nature of the superconductivity in these materials. Although their macroscopic superconducting properties are established1,5-7, microscopic insights into the pairing mechanism remains unclear. Here we characterize the superconducting gap structure in the high-temperature superconductor H3S and its deuterium counterpart D3S by performing tunnelling spectroscopy measurements. The tunnelling spectra reveal that H3S and D3S both have a fully gapped struct

Change in novel plasmin activated clotting time assay predicts hyperfibrinolysis, massive transfusion, and mortality in trauma in under 3 minutes.

BACKGROUND: Hyperfibrinolysis after trauma increases mortality, yet rapid identification remains a challenge. Prior work demonstrates liver transplantation is an idealized model of fibrinolysis. Early resource mobilization and antifibrinolytic therapy improves outcomes in trauma, but efficacy is lost with delays in care. We set out to develop a novel plasmin-activated clotting time (PACT) assay to rapidly diagnose hyperfibrinolysis. METHODS: Whole blood from adult liver transplant(n = 40) and trauma patients(n = 32) underwent PACT assay alongside traditional and tPA-challenged thromboelas

Independent nuclear and organellar mechanisms determine apicoplast fate in malaria parasites.

The apicoplast organelle of the malaria parasite, Plasmodium falciparum, is essential for parasite replication, though its cell cycle regulation remains poorly understood. We developed a dynamic live-imaging platform with analytical capabilities to track subcellular structures throughout the parasite's 48-h intraerythrocytic life cycle. Our analysis revealed four distinct morphological stages in apicoplast development that correlate with nuclear replication. We identified a critical "Crown" morphology stage required for nucleus-apicoplast attachment, where the apicoplast stretches across multi

Screening of an Antibacterial and Coagulation Multifunctional Peptide and Its Application in Wound Healing.

As a new type of drug developed in the era of antibiotics, antimicrobial peptides (AMPs) have great potential in antibacterial, antiviral and antitumor aspects. Different from the principle of action of antibiotics, AMPs have a better effect on resisting drug-resistant bacteria. We synthesized a peptide library using solid-phase synthesis and successfully screened the antimicrobial peptide RKGAK. AMPs provide effective defense against both bacteria and fungi. Research has found that the lowest inhibitory concentrations of RKGAK against S. aureus, E. coli, and C. albicans are 75 μg/mL, 100 μg/m

Initial development of skill with a reversed bicycle and a case series of experienced riders.

Riding a bicycle is considered a durable skill that cannot be forgotten. Here, novice participants practiced riding a reversed bicycle, in which a reversing gear inverted the handlebar's rotation. Although learning to ride the reversed bicycle was possible, it was slow, highly variable, implicit, and followed an S-shape pattern. In the initial learning phase, failed attempts to ride the normal bicycle indicated strong interference between the two bicycle skills. While additional practice decreased this interference effect, a subset of learners could not ride either bicycle after eight sessions

Micromechanism for Copper-Deficiency Enhanced Stability: A Quasi In Situ TEM Study of Electric-Induced Structural Evolution in Cu2-x(S, Se) Liquid-Like Thermoelectric Materials.

Cu-based liquid-like thermoelectric materials have garnered tremendous attention due to their inherent ultralow lattice thermal conductivity. However, their practical application is hampered by stability issues under a large current or temperature gradient. It has been reported that introduction of copper vacancies can enhance the chemical stability, whereas the micromechanism behind this macroscopic improvement still remains unknown. Here, we have established a quasi in situ TEM method to examine and compare the structural evolution of Cu2-xS0.2Se0.8 (x = 0, 0.05) under external electric fiel

Terahertz Néel spin-orbit torques drive nonlinear magnon dynamics in antiferromagnetic Mn2Au.

Antiferromagnets have large potential for ultrafast coherent switching of magnetic order with minimum heat dissipation. In materials such as Mn2Au and CuMnAs, electric rather than magnetic fields may control antiferromagnetic order by Néel spin-orbit torques (NSOTs). However, these torques have not yet been observed on ultrafast time scales. Here, we excite Mn2Au thin films with phase-locked single-cycle terahertz electromagnetic pulses and monitor the spin response with femtosecond magneto-optic probes. We observe signals whose symmetry, dynamics, terahertz-field scaling and dependence on sam

Molecular assembly of carbon nitride-based composite membranes for photocatalytic sterilization and wound healing.

Polymeric carbon nitride (pCN) has attracted increasing interest as a metal-free photocatalyst because of its high efficiency in reactive oxygen species (ROS) generation. However, due to poor solubility, compounding pCN at the molecular level into more advanced nanocomposites remains a challenge. Herein, we report the dissolution of pCN in polyphosphoric acid (PPA) for the first time and fluid-phase assembly with carbon nanotubes (CNTs) into a flexible free-standing membrane. Mechanism and generality studies disclosed that the coordination of the acidity, viscosity, and adsorption energy of th

A biobased magnetic dual-dummy-template molecularly imprinted polymer using a deep eutectic solvent as a coporogen for highly selective enrichment of organophosphates.

An eco-friendly magnetic dual-dummy-template molecularly imprinted polymer (MDDMIP) was prepared by a "one-pot" green synthesis using mixed-valence iron hydroxide as the magnetic material, a deep eutectic solvent as the coporogen, and caffeic acid and glutamic acid as binary monomers. The adsorption properties toward organophosphorus pesticides (OPPs) were investigated. High adsorption capacities (269.65-304.93 mg g-1), quick adsorption times (20 s), and high imprinting factors (2.28-3.83) were obtained. The proposed MDDMIP was utilized for magnetic solid phase extraction (MSPE) of OPPs prior

The Autonomous Parvovirus Minute Virus of Mice Localizes to Cellular Sites of DNA Damage Using ATR Signaling.

Minute Virus of Mice (MVM) is an autonomous parvovirus of the Parvoviridae family that replicates in mouse cells and transformed human cells. MVM genomes localize to cellular sites of DNA damage with the help of their essential non-structural phosphoprotein NS1 to establish viral replication centers. MVM replication induces a cellular DNA damage response that is mediated by signaling through the ATM kinase pathway, while inhibiting induction of the ATR kinase signaling pathway. However, the cellular signals regulating virus localization to cellular DNA damage response sites has remained unknow

ATR protects ongoing and newly assembled DNA replication forks through distinct mechanisms.

The ATR kinase safeguards genomic integrity during S phase, but how ATR protects DNA replication forks remains incompletely understood. Here, we combine four distinct assays to analyze ATR functions at ongoing and newly assembled replication forks upon replication inhibition by hydroxyurea. At ongoing forks, ATR inhibitor (ATRi) increases MRE11- and EXO1-mediated nascent DNA degradation from PrimPol-generated, single-stranded DNA (ssDNA) gaps. ATRi also exposes template ssDNA through fork uncoupling and nascent DNA degradation. Electron microscopy reveals that ATRi reduces reversed forks by in…
