Immediate versus one-stage restoration of small-diameter implants for a single missing maxillary lateral incisor: a 3-year randomized clinical trial.

BACKGROUND: The aim of this study was to compare the bone loss pattern and soft tissue healing of immediately versus one-stage loaded 3.0-mm-diameter implants in cases involving a single missing lateral maxillary incisor. METHODS: Sixty patients with a missing lateral incisor in the maxilla were randomized to one of the treatments: 30 patients in the immediate-restoration group and 30 patients in the one-stage group. All implants were placed in healed sites and had to be inserted with a torque >25 Ncm. The implants in the immediate-restoration group were fitted with a non-occluding temporary

Challenges and Solutions in Delivering Preventive Periodontal Care to Older Adults in Community and Long-Term Residential Care Settings: A Scoping Review.

BACKGROUND: Preventive periodontal care is essential for older people in the community to reduce the risk of tooth loss, systemic diseases, and decreased quality of life. Despite its importance, older people in assisted living, home-based care (HBC), daycare, and other community settings face barriers to effective preventive periodontal care, including mobility, cognitive, and caregiver knowledge issues. Although these challenges are being recognized more and more, the evidence on barriers and solutions for delivering consistent preventive periodontal care remains scattered across different st…

Citrate Transporter NaCT and Enamel Mineralization: The Slc13a5R337* Mouse Model.

Solute Carrier Family 13 Member 5 (SLC13A5) encodes the sodium-dependent citrate cotransporter NaCT, which mediates citrate transport across cell membranes. Pathogenic variants in SLC13A5 cause developmental and epileptic encephalopathy 25 with amelogenesis imperfecta, DEE25; OMIM #615905, a debilitating autosomal recessive disorder. To better define the role of NaCT in ameloblast function and enamel mineralization, we used CRISPR/Cas9 genome editing to generate Slc13a5R337* knock-in mice that terminate NaCT translation at the Arg337 codon, which is homologous to the human SLC13A5R333* variant

Protein Interactomes of Streptococcus mutans YidC1 and YidC2 Membrane Protein Insertases Suggest SRP Pathway-Independent- and -Dependent Functions, Respectively.

Virulence properties of cariogenic Streptococcus mutans depend on integral membrane proteins. Bacterial cotranslational protein trafficking involves the signal recognition particle (SRP) pathway components Ffh and FtsY, the SecYEG translocon, and YidC chaperone/insertases. Unlike Escherichia coli, S. mutans survives loss of the SRP pathway and has two yidC paralogs. This study characterized YidC1 and YidC2 interactomes to clarify respective functions alone and in concert with the SRP and/or Sec translocon. Western blots of formaldehyde cross-linked or untreated S. mutans lysates were reacted w

Clinical efficacy and patient-reported outcomes of coconut oil as adjunctive periodontitis treatment: triple-blinded pilot randomized trial.

BACKGROUND: Periodontitis is a prevalent chronic disease commonly managed with nonsurgical therapy and adjunctive antimicrobial rinses, though their side effects can hinder long-term use. Coconut oil has emerged as a potential natural alternative due to its antimicrobial and anti-inflammatory effects. This study aimed to evaluate the clinical and patient-reported outcomes of periodontal patients treated with nonsurgical therapy with or without coconut oil as an adjunctive method. METHODS: Thirty participants with stage II-III periodontitis were randomly assigned to three groups: coconut oil,

PAR1 Activation, via LMBR1/BMP Axis, Promotes the Osteogenesis of Periodontal Ligament Stem Cells (PDLSCs) and Alleviates the Inhibitory Effect of Sodium Butyrate on PDLSCs Osteogenesis.

BACKGROUND: Periodontitis is the leading cause of tooth loss and can exacerbate various systemic inflammatory conditions. Periodontal ligament stem cells (PDLSCs) stand out as prominent and favorable candidates for promoting periodontal tissue regeneration. This study aimed to investigate whether the protease-activated receptor type 1 (PAR1) can mitigate the sodium butyrate (NaB)-induced PDLSCs osteogenesis inhibition and unravel the underlying mechanism. METHODS: Public datasets from the Gene Expression Omnibus (GEO) were utilized to analyze differentially expressed genes (DEGs) in periodont…

Site-Specific Profiling of the Dental Mycobiome Reveals Strong Taxonomic Shifts during Progression of Early-Childhood Caries.

Dental caries is one of the most common diseases worldwide. Bacteria and fungi are both commensals in the oral cavity; however, most researc … emains a global public health concern. Untreated cavities are painful and expensive and can lead to tooth loss and a lower quality of life. Caries are driven by acid production via microbial fermentation of dietary carbohydrates, resulting in enamel erosion. While caries is a well-studied disease, most research has focused on bacterial impacts, even though f …

Gene therapy improves dental manifestations in hypophosphatasia model mice.

BACKGROUND AND OBJECTIVE: Hypophosphatasia is a rare inherited skeletal disorder characterized by defective bone mineralization and deficiency of tissue non-specific alkaline phosphatase (TNSALP) activity. The disease is caused by mutations in the liver/bone/kidney alkaline phosphatase gene (ALPL) encoding TNSALP. Early exfoliation of primary teeth owing to disturbed cementum formation, periodontal ligament weakness and alveolar bone resorption are major complications encountered in oral findings, and discovery of early loss of primary teeth in a dental examination often leads to early diagnos
