Glycopeptide-resistant enterococci: a decade of experience.

Since their first description in 1988, glycopeptide-resistant enterococci (GRE) have emerged as a significant cause of nosocomial infections … cs. Although VanA resistance is detected readily in the clinical laboratory, the variable levels of vancomycin resistance associated with the other phenotypes makes detection less reliable. Under-reporting of VanB resistance as a result of a lower detection rate may account, in part, for the difference in the numbers of enterococci displaying VanA and VanB resistance r …

Performance evolution of the Nano Boron nitride enhanced bone cement composites.

Bone cement is a research hotspot and has been partially applied in the field of bone repair thanks to the good mechanical, physical and antibacterial properties. However, the easy wear and high temperature during curing characteristics would cause surrounding tissue necrosis, which seriously limits the wider application to some extent. In this work, the hexagonal boron nitride (h-BN) nano flakes were optimized to enhance the bone cement matrix (PMMA) via mechanical doping. The doping of h-BN into PMMA results in an improved mechanical (bending stress increased by 26%), thermal-conductivity (i

A mechanistic neural network model predicts both potency and toxicity of antimicrobial combination therapies.

Antimicrobial resistance poses a major global threat due to the diminishing efficacy of current treatments and limited new therapies. Combination therapy with existing drugs offers a promising solution, yet current empirical methods often lead to suboptimal efficacy and inadvertent toxicity. The high cost of experimentally testing numerous combinations underscores the need for data-driven methods to streamline treatment design. We introduce CALMA, an approach that predicts the potency and toxicity of multi-drug combinations in Escherichia coli and Mycobacterium tuberculosis. CALMA identified s

Proline-Modified RWn Peptides: Enhanced Antifungal Efficacy and Synergy with Conventional Antibiotics for Combating Resistant Fungal Infections.

Candida albicans (Ca) and Cryptococcus neoformans (Cn) infections pose a growing threat due to rising antifungal resistance. This study explores a new class of antifungal agents, the RWn series (n = 4, 6, 8) peptides. These synthetic peptides were evaluated for their ability to inhibit Ca and Cn growth. All peptides except RW4 displayed antifungal activity, with RW6 exhibiting exceptional potency against Cn. Importantly, the incorporation of a proline residue significantly reduced cytotoxicity while maintaining antifungal activity against Cn for all RWnP peptides. Notably, RW6P demonstrated br

Development of 4-[4-(Anilinomethyl)-3-phenyl-pyrazol-1-yl] Benzoic Acid Derivatives as Potent Anti-Staphylococci and Anti-Enterococci Agents.

From a library of compounds, 11 hit antibacterial agents have been identified as potent anti-Gram-positive bacterial agents. These pyrazole derivatives are active against two groups of pathogens, staphylococci and enterococci, with minimum inhibitory concentration (MIC) values as low as 0.78 μg/mL. These potent compounds showed bactericidal action, and some were effective at inhibiting and eradicating Staphylococcus aureus and Enterococcus faecalis biofilms. Real-time biofilm inhibition by the potent compounds was studied, by using Bioscreen C. These lead compounds were also very potent agains

Dual-emission fluorescence biosensing of vancomycin based on AIEgen-peptide conjugates and aptamer-modified Au nanoclusters.

Precise doses of antibiotics are necessary to prevent bacterial drug resistance. Although fluorescent sensors are promising for quantitative analyses of antibiotics, improvements in feasibility, selectivity, and sensitivity are needed. In this study, a dual-emission fluorescence biosensor platform was developed for simple, selective, and sensitive determination of vancomycin (Van) based on a peptide conjugated with blue-emitting aggregation-induced emission luminogens (AIEgen) and aptamer-modified red-emitting gold nanoclusters (AuNCs-apt). The peptide and aptamer together recognized Van with

Analysis of Four New Enterococcus faecalis Phages and Modeling of a Hyaluronidase Catalytic Domain from Saphexavirus.

Background: Phage therapy (PT), as a method to treat bacterial infections, needs identification of bacteriophages targeting specific pathogenic host. Enterococcus faecalis, a Gram-positive coccus resident in the human gastrointestinal tract, may become pathogenic in hospitalized patients showing acquired resistance to vancomycin and thus representing a possible target for PT. Materials and Methods: We isolated four phages that infect E. faecalis and characterized them by host range screening, transmission electron microscopy, and genome sequencing. We also identified and three-dimensional mode

Self-Propelled Active Photothermal Nanoswimmer for Deep-Layered Elimination of Biofilm In Vivo.

Increasing penetration of antibacterial agents into biofilm is a promising strategy for improvement of therapeutic effect and slowdown of the progression of antibiotic resistance. Herein, we design a near-infrared (NIR) light-driven nanoswimmer (HSMV). Under NIR light irradiation, HSMV performs efficient self-propulsion and penetrates into the biofilm within 5 min due to photothermal conversion of asymmetrically distributed AuNPs. The localized thermal (∼45 °C) and thermal-triggered release of vancomycin (Van) leads to an efficient combination of photothermal therapy and chemotherapy in one sy

Combating bacterial infection by in situ self-assembly of AIEgen-peptide conjugate.

Antibiotic abuse and the resulting resistance to antibiotics are serious problems faced by the world. Methods for fast and precise detection of bacterial infections are in urgent need. Here, we report a sensitive and selective probe for diagnosis and treatment of Gram-positive bacterial infection. The probe is made of self-assembling short peptide as the skeleton, a luminogen with aggregation-induced emission (AIEgen) as the responsive fluorescence turn-on motif and vancomycin as the targeting group. In vitro assembly of the probe can turn on its fluorescence and simultaneously enhance reactiv

Evaluation of TD test for analysis of persistence or tolerance in clinical isolates of Staphylococcus aureus.

Besides natural and acquired mechanisms of resistance, bacteria can cope with presence of antibiotics by using complex mechanisms such as persistence or tolerance. The main purpose of this study was to evaluate the suitability of newly developed Tolerance Disk Test (TDtest) (Gefen et al., 2017) to detect persistent or tolerant bacterial cells in clinical isolates of Staphylococcus aureus. The principle of the test is to resuscitate the subpopulation of persistent or tolerant bacterial cells following a disk diffusion test by glucose. Results of the TDtest were evaluated using time killing expe
