Disseminated cryptococcosis and recurrent oral candidiasis was presented in a-heterosexual AIDS patient. Candida tropicalis (C.tropicalis) was isolated from the oral pseudomembranous plaques and Cryptococcus neoformans (C. neoformans) was isolated from maculopapular lesions on body parts (face, hands and chest) and body fluids (urine, expectorated sputum, and cerebrospinal fluid). In vitro drug susceptibility testing on the yeast isolates demonstrated resistance to fluconazole acquired by C. tropicalis which was a suggestive possible root cause of recurrent oral candidiasis in this patient.

Paracoccidioidomycosis is a systemic disease endemic to subtropical areas in Central and South America caused by a dimorphic fungus known as Paracoccidioides brasiliensis. Central nervous system involvement is a severe complication of the systemic disease, and has been found in approximately 13% of patients. This paper describes the case of a patient whose computed tomography scan and magnetic resonance imaging showed a single tumor-like lesion in the brainstem. Histopathological and mycological examinations of stereotactic biopsy smears showed the characteristic yeast cells that confirmed the

A young human immunodeficiency virus-infected man was admitted in unconsciousness state. Few papular skin lesions resembling molluscum contagiosum (MC) were present on the face. He was found to have cryptococcal meningitis using India ink preparation of cerebrospinal fluid. Histopathological examination of skin lesion revealed the presence of capsulated yeast forms of Cryptococcus within the lesion of MC. The co-existence of MC and cryptococcosis in the same cutaneous lesion is rare, and it could be because of immunosuppression (low CD4 count).

Trichosporon beigelii is widely distributed in nature and is classically associated with white piedra, a mycosis that may involve the hair of the human body. Intramammary infections caused by T beigelii may be fatal in cows; the prevalence in affected dairy herds may be high. Affected cows may have hyperthermia, swelling of the udder, and substantially decreased milk production or agalactia. Intramammary infections caused by yeast, including T beigelii, may also be associated with high bacterial counts in bulk-tank milk.

The emergence of new fungal pathogens such filamentous fungi (Scedosporium, Fusarium) or yeast (Trichosporon) is a real problem for doctors who treat immunocompromised patients. These fungi present in our environment (Scedosporium, Fusarium) can also be on the skin (Trichosporon). They are responsible for invasive infections cause of morbidity and mortality because patients are immunocompromised and the fungi are resistant to antifungal treatment. Clinical manifestations are seldom specific, but cutaneous localisations are frequent. They are either the portal of entry of the infection or a met

Leukocyte recruitment to the site of infection by the encapsulated yeast Cryptococcus neoformans is critical for clearance of the infection. We review data from our lab that chemokines, such as the CC chemokines MCP-1 and MIP-1alpha, are important mediators of leukocyte recruitment during C. neoformans infection. In addition, studies in CC chemokine receptor knockout mice have demonstrated that CCR2 and CCR5 are required not only for leukocyte recruitment but also for other aspects of immune response development and innate imunity to C. neoformans.

Recovery of yeast cells after exposure to ionizing radiation was found in 1957. During the first decade, i.e. in the "premolecular period" of studying the phenomenon, its basic features were revealed: dependence on ploidy of cells, on their energy exchange, on radiation LTE, and others. A mathematical model of recovery was proposed; the damages causing death of irradiated haploid and diploid cells were shown to be double strand breaks of DNA. The concepts of universal biological importance of the cell property to repair genetic damages were formulated.

A peptide-polysaccharide, a peptide-rhamnomannan, was isolated from the pathogenic yeast form of the fungus Sporothrix schenckii. This substance, which may play a role in fungal virulence, was tested in an animal model of systemic disease, and depression of the immune response was observed in the animals between the 4th and 6th week of infection. Concomitantly, this compound showed mitogenic activity when challenged with normal lymphocytes and was also found to be involved in the inflammatory response. These results provide further information for the understanding of fungal implantation in ti

We report the identification of Blastomyces dermatitidis by microscopic examination of a direct faecal smear from a dog with pulmonary blastomycosis. A simultaneously obtained faecal culture grew Blastomyces dermatitidis. The fungus was also cultured from a transtracheal sample from this same dog. This report suggests that yeast-phase cells of B. dermatitidis may be recovered in the stool of dogs with pulmonary blastomycosis following transit through the gastrointestinal tract of swallowed infected sputum. Implications regarding the ecology of Blastomyces dermatitidis are discussed.

Recombinant DNA technology has enabled the large scale production of human hemoglobin in bacteria and yeast. This has opened up a way to produce a hemoglobin-based blood substitute which could replace conventional blood transfusion in some situations. Using our understanding of the structure-function relationships and evolutionary history of hemoglobin it has been possible to improve the oxygen transport properties of the molecule and solve a number of problems associated with the use of natural hemoglobin as a cell-free blood substitute.

Adherence of Malassezia yeast cells to human keratinocytes was assessed by a novel technique using double-sided Sellotape. Although adherence using double-sided Sellotape is still merely a model for in vivo adherence, it approximates to the conditions found on the skin surface. There were no differences in adhesive properties to human keratinocytes between Malassezia strains originating from HIV-positive and HIV-negative patients with seborrhoeic dermatitis, nor was there a relationship between the severity of seborrhoeic dermatitis and in vitro adherence to human keratinocytes.

Paracoccidioides brasiliensis probably uses many different mechanisms to establish itself in the host and cause disease. In this work, we assess an in vitro model system which uses cultured mammalian cells to investigate the virulence factors of P. brasiliensis. We were able to demonstrate an invasion process of the yeast form of this fungus in Vero cell cultures. We deduced that the overall invasive process involved three steps: adhesion, followed by invasion of individual epithelial cells and spread to adjacent cells.

The prevalence of Paracoccidioides brasiliensis infection in Panamanian children was demonstrated to be 12.73% (14/110) by skin reactivity to 50 micrograms of yeast derived antigen. Boys were infected more frequently than girls - 1.45:1. The earliest age of infection was 2 years, and a direct relationship was noted between age and frequency of infections (as the children became older, more of them became infected); boys more so than girls. Thirteen of 14 (93%) of the infected juvenile population live in a rural environment.

Pityriasis (tinea) versicolor is a common disorder of adults. We cared for five infants (four males and one female) with the disease. Diagnosis was confirmed by potassium hydroxide preparation demonstrating the filaments of Malassezia furfur and/or Pityrosporum orbiculare, the yeast form. Three patients had lesions in the neonatal period. The mother of one baby had pityriasis versicolor. Two patients were siblings. One baby had associated atopic dermatitis and two had a positive family history of atopy.

The technique is offer for rapid quantitative definition of sterols having a system of delta 5,7-conjugated double bonds which conditions biological activity of D vitamin--the result of photoisomerisation of the sterols. The technique bases on the analysis of UV absorption spectra of unsaponifiable lipid fraction of several kinds of mutant yeast strains and the model mixtures of sterols. The results obtained concerning the estimation of delta 5,7-sterols containing in total fraction are confirmed by gas-liquid-chromatography and mass-spectrometry data.

Acute acalculous cholecystitis due to Torulopis glabrata, an opportunistic yeast, developed postoperatively in a 70-year-old man who had an extremely complicated course after resection of an abdominal aortic aneurysm. The infection first appeared as an acute surgical abdomen, three days after resumption of solid food intake subsequent to a prolonged ileus and after 31 days of parenteral hyperalimentation. The condition was successfully treated by cholecystostomy; at the time of writing, six months after cholecystostomy, there are no gastrointestinal symptoms.

Post-fertilization embryo abortion is a major barrier to distant hybridization in lily, but the molecular mechanisms underlying hybrid embryo viability remain unclear. In this study, we examined embryo development in the cross between Asiatic hybrids 'Feixiang Tiancheng' (♀) and Lilium davidii var. unicolor (♂), and found that embryo abortion occurred at 40 days after pollination. Comparative transcriptome analysis of embryos differing in developmental stage and fate identified LdFUS3, encoding a nuclear B3-domain transcriptional activator, as a candidate regulator preferentially expressed in

Protein chimeras enable new biological functions by combining otherwise independent protein elements, but identifying insertion sites that preserve host protein function remains largely empirical. Here, we introduce CRISPR-guided protospacer adjacent motif (PAM) scanning, a scalable strategy for mapping chimera-permissive sites directly in living cells. PAM scanning uses CRISPR-addressable positions across coding sequences to systematically install functional payloads and measure insertion tolerance. In yeast, PAM accessibility provided near-comprehensive coverage across proteins of diverse si

Rapid, low-cost strain-level typing is essential for source attribution and microbiological control in clinical and industrial settings. Fourier-transform infrared (FT-IR) spectroscopy using IR Biotyper method was previously optimized to achieve an identification level equivalent to that of whole-genome sequencing (WGS) discrimination of Wickerhamomyces anomalus. However, the species of yeast studied were limited; further, studies that have used the results of WGS as a reference for strain identification in yeast remain limited. Therefore, here, the generalizability of this approach to Candida

Protein homeostasis is essential for maintaining normal cellular function. However, protein homeostasis efficiency declines with age, leading to the accumulation of aberrant protein structures associated with neurodegenerative diseases such as Parkinson's disease (PD). PD is characterized by the aggregation of alpha-synuclein (αSyn) into cytoplasmic inclusions. This process is accompanied by elevated phosphorylation at serine 129 (S129). The accumulation of αSyn into aggregates and their propagation disrupts key proteostasis pathways, including the ubiquitin-proteasome system (UPS) or autophag
