Panax notoginseng flower extract ameliorates chronic unpredictable mild stress -induced depression-like behaviors in mice by reducing neuroinflammation.

OBJECTIVE: To evaluate whether Panax notoginseng flower extract (PNF) can alleviate depression-like behavior caused by chronic unpredictable mild stress (CUMS) in mice and to explore its relations to neuroinflammation. MATERIALS AND METHODS: C57BL/6J mice were subjected to CUMS for 7 weeks to induce depressive-like behaviors. Then PNF 1.7 or 3.4 g/kg was administered via intragastric gavage once a day for 4 consecutive weeks. After behavioral assessment, the systemic inflammation and neuroinflammation were investigated by detecting inflammatory factors in serum and brain with enzyme-linked im

Molecular mechanisms of blood-brain barrier dysfunction caused by obesity: pathophysiological understandings and treatment targets.

Chronic low-grade inflammation and systemic metabolic dysregulation are linked to obesity, a rapidly expanding worldwide health concern that affects the central nervous system (CNS) in addition to peripheral organs. Dysfunction of the blood-brain barrier (BBB), which impairs selective permeability, tight junction integrity, and neurovascular coupling, is one of its most serious neurobiological effects. The BBB, which is made up of pericytes, endothelial cells, and astrocytic end-feet, depends on specialized transporters and tight junction proteins like occludin, ZO-1, and claudin-5 to control

Molecular Changes in CSPG and Glial Scar Markers in Response to Subpial Chondroitinase ABC Treatment Following Spinal Cord Injury.

Chondroitin sulphate proteoglycans (CSPGs) are major contributors to the inhibitory microenvironment that hinders regeneration following spinal cord injury (SCI). Chondroitinase ABC (ChABC) mediated degradation of CSPGs is associated with enhanced regenerative potential following SCI. In this study, the molecular responses of Neurocan, NG2 and Phosphacan CSPGs, as well as cellular markers for pro-inflammatory and neuroprotective microglia, astrocytes, oligodendrocytes and neurons, were investigated following a single subpial injection of ChABC (0.2 U in 10 μL) 14 days after thoracic (Th9) spin…

Sphingosine-1-Phosphate Receptor 3 Mediated Neuroinflammation is Critically Involved in the Development of Brain Injury Following Cardiac Arrest in Mice.

Brain injury after cardiac arrest (CA) is a major cause of death and disability, with neuroinflammation increasingly recognized as a key driver. Although the sphingosine-1-phosphate receptor 3 (S1PR3)-a G protein-coupled receptor-has been linked to neurological disorders, its role in CA-induced brain injury remains unclear. We induced CA in mice via intravenous potassium chloride injection. S1PR3 expression and subcellular localization were assessed in cortex and hippocampus. Mice received intraperitoneal CAY10444 (a selective S1PR3 antagonist) alone or with Colivelin TFA (a Janus Kinase 2 (JA

Glial dynamics in brain aging: Cellular heterogeneity and regional vulnerability.

Brain aging involves progressive structural, functional, and molecular changes that impair cognition and increase vulnerability to neurodegenerative diseases. While neurons have traditionally received primary research focus, recent advances in single-cell transcriptomics, spatial genomics, and functional imaging reveal that glial cells-microglia, astrocytes, and oligodendrocytes-undergo profound, heterogeneous alterations during aging that actively drive brain dysfunction. These changes include microglial transition from homeostatic surveillance to inflammatory, dystrophic states; astrocyte sh…

Recombinant Hc-TeTx modulates TrkA-related signaling and preserves cholinergic integrity in the nucleus basalis magnocellularis after Aβ(25-35) injury.

The recombinant C-terminal domain of tetanus toxin (Hc-TeTx) has been reported to exert neurotrophic-like effects in experimental models of neurodegeneration, potentially through activation of signaling pathways associated with tropomyosin receptor kinase A (TrkA). However, its effects on basal forebrain cholinergic dysfunction induced by amyloid-β remain incompletely characterized. The present study evaluated whether administration of Hc-TeTx in the nucleus basalis magnocellularis (NBM) is associated with attenuation of Amyloid beta peptide 25-35 (Aβ(25-35))-induced alterations in cognitive f

Case Report: The molecular profile of granular cell astrocytoma predicts aggressive clinical behavior, independent of morphology.

BACKGROUND: Granular cell astrocytoma (GCA) is a rare, morphologically distinct variant of IDH-wildtype glioblastoma that can appear deceptively low-grade yet behave aggressively. Its molecular features remain poorly defined, and no methylation-based classification has previously been reported. METHODS: Two GCAs diagnosed in our clinical neuropathology department were described with the integration of clinical, intraoperative, histopathological, and molecular data, including DNA methylation profiling, a targeted next-generation sequencing panel, and, in one case, whole genome sequencing (WGS)

Stage-dependent associations of glial fibrillary acidic protein and chitinase-3-like protein 1 with white matter network integrity in Alzheimer's disease.

BackgroundAstrocytes become reactive in Alzheimer's disease (AD), and fluid markers of this response, including glial fibrillary acidic protein (GFAP) and chitinase-3-like protein 1 (YKL-40), increase across the AD continuum. However, it remains unclear whether astrocytic biomarker levels relate to preserved versus disrupted white matter (WM) integrity, and whether these relationships vary by amyloid-β (Aβ) burden and apolipoprotein E (APOE) ε4 status.ObjectiveTo examine associations of GFAP and YKL-40 with WM integrity, AD pathology, and memory across Aβ and APOE ε4 contexts.MethodsWe analyze…

YAP Regulates the Nrf2 Signaling Axis to Attenuate Oxidative Stress and Neuroinflammation in Retinal Ganglion Cell Degeneration.

PURPOSE: Oxidative stress is a key driver of retinal ganglion cell (RGC) degeneration after optic nerve injury. Yes-associated protein (YAP), a Hippo pathway effector, is known to reprogram stress responses, yet its role in regulating oxidative stress during RGC degeneration is unclear. METHODS: This study investigated the role of YAP in RGC injury using an in vivo optic nerve crush (ONC) model and an in vitro oxidative-stress model with primary RGCs. YAP expression was modulated pharmacologically and genetically. We assessed its effects on nuclear factor erythroid 2-related factor 2 (Nrf2) s

Short-chain fatty acids alleviated fluoride-induced neuroinflammation via the gut-brain axis in rats.

Neuroinflammation is a hallmark of central nervous system injury caused by fluorosis; however, the regulatory relationship between intestinal microbial and fluoride-induced neuroinflammation remains unclear. Here, we aimed to evaluate the alleviating effect of short-chain fatty acids (SCFAs) on fluoride exposure-induced neuroinflammation in rats. In this study, it is demonstrated that the composition of colonic microbiota was disrupted in fluoride-exposed rats at both phylum and genus levels, and fluoride exposure decreased the number of positive cells per crypt, inhibited mucin secretion, and

Worsened axonal injury and gliosis following diffuse brain injury in male and female mice with neurofilament light gene deletion.

Neurofilaments have been theorized to make neurons more vulnerable to damage with injury or disease. To investigate how neurofilaments influence traumatic brain injury (TBI) pathophysiology, we examined neuropathology and glial response in neurofilament light chain knockout (NFL-KO) mice. Given the critical role of neurofilaments in maintaining axonal structure, we hypothesized that NFL-KO mice would exhibit altered neuropathology and gliosis after diffuse TBI. Male and female NFL-KO and wild-type mice were subjected to midline fluid percussion injury. Brains were collected at 3 h, 1 day, or 3

Intranasal Application of Diluted Saline Alleviates Ischemic Brain Injury in Association with Suppression of Vasopressin Neurons.

INTRODUCTION: Cerebral swelling and brain injury in ischemic stroke are closely related to increased vasopressin (VP) secretion. How to alleviate ischemic brain injury by suppressing VP hypersecretion through simply available approaches remains to be established. METHODS: Using a rat model of middle cerebral artery occlusion (MCAO), testing effects of the intranasal application of low concentration saline-0.09% NaCl (IAL) on brain damage, VP neuronal activity, synaptic inputs, astrocytic plasticity, and olfactory bulb (OB) activity in immunohistochemistry, patch-clamp recording, Western blott

Exploratory Randomised Trial of Tranexamic Acid to Decrease Postoperative Delirium in Adults Undergoing Lumbar Fusion: A trial stopped early.

BACKGROUND: Postoperative delirium may be mediated by perioperative systemic- and neuro-inflammation. By inhibiting the pro-inflammatory actions of plasmin, tranexamic acid (TXA) may decrease postoperative delirium. To explore this hypothesis, we modified an ongoing randomised trial of TXA, adding measures of postoperative delirium, cognitive function, systemic cytokines, and astrocyte activation. METHODS: Adults undergoing elective posterior lumbar fusion randomly received intraoperative intravenous TXA (n=43: 10 mg kg-1 loading dose, 2 mg kg-1 h-1 infusion) or Placebo (n=40). Blood was coll

Crosstalk between lipocalin-2 and IL-6 in traumatic brain injury: Closely related biomarkers.

Clinical biomarkers are crucial for diagnosing and predicting outcomes in patients with traumatic brain injury (TBI). In this study, we performed an unbiased analysis of plasma proteins in acute TBI patients using bead-based multiplex assays and identified a strong positive correlation between LCN2 and IL-6 levels. Based on these findings, we hypothesized that LCN2 and IL-6 are closely related circulating biomarkers for TBI. Our previous and current studies demonstrate that the expression of LCN2, IL-6, and its receptors is upregulated in patients with chronic traumatic encephalopathy, in mous
