Heterogenous expression of bcr-abl fusion mRNA in a patient with Philadelphia-chromosome-positive acute lymphoblastic leukaemia.

We performed molecular and cytogenetic analysis on a 56-year-old woman with Philadelphia chromosome (Ph1)-positive acute lymphoblastic leukaemia (ALL) having two types of major and minor bcr-abl (M-bcr-abl, m-bcr-abl/fusion mRNA at diagnosis. In the course of her disease, unexpected heterogenous bcr-abl fusion mRNA was detected by sequential analysis using the reverse transcription and polymerase chain reaction (RT-PCR). The RT-PCR analysis showed both M-bcr-abl (b2-a2 type) and m-bcr-abl at diagnosis. Although b2-a2 type M-bcr-abl disappeared after complete remission (CR) was achieved with in

Liposome-Supplemented Cell-free Synthesis of a Peptide that Forms β-barrel Nanopores.

Rational design of nanopore-forming proteins and peptides is a promising approach for single-molecule analysis by using custom-designed nanopores tailored to specific target molecules. One challenge in this approach is the expression or synthesis of the designed nanopores, due to their high hydrophobicity. We sought to streamline the synthesis of an artificial and highly aggregative nanopore-forming peptide using liposome-supplemented cell-free protein synthesis. By optimizing the liposomal lipid composition, we recovered yields of the expressed aggregative peptide that were suitable for funct

Axial Round Cell Sarcoma Harboring a Non-ETS EWSR1 Rearrangement: Diagnostic Challenges and Clinical Implications.

We present a rare case of a cervicothoracic epidural spindle and round cell sarcoma in a 59-year-old man, characterized by an EWSR1 gene rearrangement. The patient experienced progressive cervical pain and lower limb weakness due to an extradural mass at the C7-T2 level. Surgical resection and cervicothoracic fixation were performed, followed by radiotherapy (30 Gy/10 fractions) and Ewing-based chemotherapy (doxorubicin/ifosfamide). Histopathological analysis revealed a spindle and oval cell neoplasm with a Ki-67 index of 30%. The tumor was positive for CD99, SATB2, TLE1, cyclin D1, and focal

An Immunochromatographic Test Strip and Its Application in Rapid Screening of Pepper Mild Mottle Virus.

The quality and productivity of peppers have been severely impacted by the pepper mild mottle virus (PMMoV). To effectively control the transmission and damage caused by PMMoV, it is crucial to establish a simple, rapid, and field-applicable detection method. In this study, the coat protein of PMMoV was induced expression as an immunogen. After cell fusion and subcloning, a total of 10 hybridoma cell lines that stably secrete the PMMoV monoclonal antibody were screened. Through antibody pairing and screening, using monoclonal antibody 4D7 as the capture antibody and 3B9 as the detection antibo…

Hybrid Junction-Enabled Biomimetic Human Eye Structure for Large Dynamic Range Vision Sensor.

The responsive light intensity dynamic range (DR) of the human eye far exceeds that of existing visual systems, and the development of a biomimetic retinal detecting unit is currently an important challenge in the field of machine vision. Here, a two-terminal Au-contacted VO2/WSe2 heterojunction photodetector with the same adaptive DR as retinal cells is developed. It is revealed that the VO2/WSe2 heterojunction part-mimics the cone cell for strong light detection with photoresponsivity (R) of 320 mA W-1 and the Au/WSe2 Schottky contact part-mimics the rod cell for weak light detection with an

The ALS- and FTD-associated proteins annexin A11 and CHMP2B act sequentially in plasma membrane repair.

Maintenance of plasma membrane integrity is essential for compartmentalization of the cytosol and for cellular viability. Upon membrane damage, several factors including endosomal sorting complex required for transport-III (ESCRT-III) proteins, annexins, stress granules, lipids, and membrane fusion proteins are mobilized to orchestrate membrane repair. However, whether these factors operate independently or act together is unclear. Here, using human cell lines, we expose temporal differences and interdependencies in the recruitment of ESCRT-III and annexin proteins to sites of plasma membrane

Novel Aloe-Emodin Derivatives as Potential Anticancer Agents: Synthesis, Characterization and Cytotoxic Activity.

The fusion of heterocycles onto an anthraquinone scaffold represents a promising strategy to optimize anticancer activity. This study has the aim to synthesize and characterize novel anthra[1,2-b]furan compounds based on the natural product aloe-emodin. Six novel anthra[1,2-b]furans bearing phenyl, n-hexane, and methoxy carbonyl substituents were synthesized starting from aloe-emodin. The synthetic route employed involved acetyl protection of aloe-emodin, electrophilic aromatic halogenation, subsequent Castro-Stephens coupling, spontaneous intramolecular cyclization, and deprotection of hydrox

Cord blood natural killer cell-derived extracellular vesicles inhibit Zika virus infectivity through ITGB2/perforin-mediated envelope disruption in vitro and in vivo.

Zika virus (ZIKV) can traverse the placental barrier, leading to fetal microcephaly and congenital zika syndrome (CZS). The viral E protein mediates host-cell interactions and infection. Here, we demonstrated that cord blood natural killer cell-derived extracellular vesicles (CBNK-EVs) potently inhibit ZIKV infection in vitro without compromising cellular viability. Mechanistically, CBNK-EVs engage ZIKV through ITGB2, a surface-enriched integrin that interacts with the viral E protein, facilitating nanoparticle-virion contact or membrane fusion. This interaction triggers antiviral activity via

Genetic and regenerative contexts specify distinct soft tissue sarcoma identities from Twist2+ progenitors.

Soft tissue sarcomas (STS) comprise diverse mesenchymal malignancies whose developmental origins and mechanisms of subtype specification remain poorly understood. Whether distinct sarcoma subtypes can arise from a common progenitor through distinct oncogenic and tissue contexts has remained unresolved. Here, we show that skeletal muscle progenitors marked by expression of the Twist2 transcription factor generate distinct STS subtypes in response to defined genetic and microenvironmental cues. Activation of Kras together with Trp53 loss maintained a mesenchymal cell identity and drove highly pe

LFA-1 interaction with GBP-130 on Plasmodium falciparum-infected red blood cells mediates NK cell activation and parasite control.

Natural killer (NK) cells contribute to early immunity against Plasmodium falciparum by recognizing and eliminating infected red blood cells (iRBCs), a process mediated in part by the integrin LFA-1. However, the cognate parasite ligand for LFA-1 has remained unknown. Here, we identify glycophorin binding protein-130 (PfGBP-130) as a surface-expressed ligand on iRBCs that binds the I-domain of LFA-1 (LFA-1 αI). Using an LFA-1 αI-Fc fusion protein, we demonstrate stage-specific binding to iRBCs, and LC-MS/MS analysis of immunoprecipitates of αI-Fc bound to iRBC revealed PfGBP-130 as a high-conf

A Humanized Anti-gD Broadly Neutralizing Antibody Confers Complete Post-Exposure Protection Against Pseudorabies Virus.

Pseudorabies virus (PRV), an α‑herpesvirus, has recently been recognized as a potentially significant zoonotic pathogen, causing severe encephalitis and endophthalmitis in humans with high mortality and disability rates. Despite its growing public‑health threat, no effective therapeutics are available for human PRV infection. Here, we isolated a broadly neutralizing monoclonal antibody, designated 6F7, which targets the PRV glycoprotein D (gD) and neutralizes all tested PRV lineages. Mechanistic studies reveal that 6F7 does not impede viral attachment or internalization; instead, it specifical

Mapping the placental galectin-3 interactome identifies CD9 and ITGB1 as functional glycoprotein counterreceptors during syncytialization.

The fetomaternal interface is replete with glycan-binding proteins (GBPs) that can interact with cell surface glycoprotein counterreceptors to regulate placental function. Here, we interrogate the role of galectin-3, a GBP that controls placental trophoblast syncytialization, an important differentiation process where progenitor cytotrophoblast cells fuse to produce the multinucleated syncytiotrophoblast. The molecular mechanism of galectin-3-mediated fusion has not yet been elucidated in part due to the difficulty of studying glycan-GBP binding events in live cells. To overcome these challeng

Mitochondrial dynamics: Molecular mechanism and implications in endometriosis.

Endometriosis affects about 10 % of women of reproductive age, leading to a disabling gynecologic condition. Chronic pain, inflammation, and oxidative stress have been identified as the molecular pathways involved in the progression of this disease, although its precise etiology remains uncertain. Although mitochondria are considered crucial organelles for cellular activity, their dysfunction has been linked to the development of this disease. The purpose of this review is to examine the functioning of the mitochondrion in endometriosis: in particular, we focused on the mitochondrial dynamics

Coupling IL-2 with IL-10 to mitigate toxicity and enhance antitumor immunity.

Wild-type interleukin (IL)-2 induces anti-tumor immunity and toxicity, predominated by vascular leak syndrome (VLS) leading to edema, hypotension, organ toxicity, and regulatory T cell (Treg) expansion. Efforts to uncouple IL-2 toxicity from its potency have failed in the clinic. We hypothesize that IL-2 toxicity is driven by cytokine release syndrome (CRS) followed by VLS and that coupling IL-2 with IL-10 will ameliorate toxicity. Our data, generated using human primary cells, mouse models, and non-human primates, suggest that coupling of these cytokines prevents toxicity while retaining cyto

The glycosyltransferase POGLUT1 regulates muscle stem cell development and maintenance in mice.

Mutations in protein O-glucosyltransferase 1 (POGLUT1) cause a recessive limb-girdle muscular dystrophy (LGMDR21) with reduced satellite cell number and NOTCH1 signaling in adult patient muscles and impaired myogenic capacity of patient-derived muscle progenitors. However, the in vivo roles of POGLUT1 in the development, function, and maintenance of satellite cells are not well understood. Here, we show that conditional deletion of mouse Poglut1 in myogenic progenitors leads to early lethality, postnatal muscle growth defects, reduced Pax7 expression, abnormality in muscle extracellular matrix

YAP1::MAML2-Rearranged Poroid Squamous Cell Carcinoma Arising in a Non-sebaceous Lymphadenoma of the Parotid Gland.

Salivary gland lymphadenoma is an uncommon neoplasm and malignant transformation is even rarer. Reported tumor types include basal cell adenocarcinoma, sebaceous carcinoma, EBV-associated lymphoepithelial carcinoma, and undifferentiated carcinoma; a squamous phenotype has not been reported. Here we describe the case of a 62-year-old female who underwent total parotidectomy for a painful parotid gland mass of uncertain duration. On microscopic examination, a high grade lymphoepithelial carcinoma with squamous and poroid features arising in a non-sebaceous lymphadenoma was recognized. Both benig

Signatures of omicron-like adaptation in early SARS-CoV-2 variants and chronic infection.

Persistent severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infections are a source of new variants and can provide insight into evolutionary trajectories. Here, we observe upper airway-specific evolution of SARS-CoV-2, demonstrating a fusion peptide (FP) domain mutation (S:P812S) adjacent to the S2' cleavage site that emerged during a chronic infection. Indeed, this mutation had emerged previously and been transmitted in a delta variant lineage. P812S in a spike-pseudotyped virus did not impact entry efficiency. However, cleavage at S1/S2 was reduced, and molecular dynamics simula

Adamantinoma‑like Ewing Sarcoma of Head and Neck: A Clinicopathologic Study of Three Cases Including One EWSR1-Negative Case with NF1 Mutation.

Adamantinoma-like Ewing sarcoma (ALES) is a rare variant of Ewing sarcoma that predominantly occurs in the head and neck region, characterized by epithelial differentiation and overlapping morphology with other small round cell tumors. We present three cases of head and neck ALES (two in the parotid gland and one in the nasal cavity) in patients aged 23-42 years. Histologically, the tumors exhibited a spectrum of features, including small round blue cells and basaloid squamous morphologies. Immunohistochemically, the tumor cells variably expressed CKpan, p63, CD99, NKX2.2, FLI1, CD56, and Syn.

Taperin bundles F-actin at stereocilia pivot points enabling optimal lifelong mechanosensitivity.

Stereocilia are rod-like mechanosensory projections consisting of unidirectionally oriented actin filaments that extend into the inner ear hair cell cytoskeleton, forming dense rootlets. Taperin (TPRN) localizes to the narrowed-down base of stereocilia, where they pivot in response to sound and gravity. We show that TPRN-deficient mice have progressive deafness characterized by gradual asynchronous retraction and fusion of outer and inner hair cell stereocilia, followed by synaptic abnormalities. Stereocilia that lack TPRN develop warped rootlets with gradual loss of TRIOBP-5 and ANKRD24 from

The Mce2B recombinant protein was evaluated as an inducer of immune response The research involved naturally infected cattle with different immunological profiles. Moderate homology (≥ 40 %) between Mce2B of M. bovis and homologous proteins in non-tuberculous mycobacteria was corroborated, as well as the presence of epitopes restricted by the bovine leucocyte antigen class II. Despite this prediction, cell-mediated responses to Mce2B were
