Guben Qingyuan Herbal Medicine delays enzalutamide resistance in prostate cancer by alleviating tumor hypoxia and reprogramming energy metabolism in cancer stem cells.

ETHNOPHARMACOLOGICAL RELEVANCE: Enzalutamide resistance is a major clinical bottleneck limiting the therapeutic efficacy of castration-resistant prostate cancer (CRPC). Enzalutamide treatment can exacerbate tumor hypoxia, which is a critical contributor to the development of its resistance. The traditional Chinese formula Guben Qingyuan Herbal Medicine (GBQY), coupled with enzalutamide, has demonstrated potential in delaying enzalutamide resistance. Nevertheless, its interaction with the tumor microenvironment (TME) remains insufficiently investigated. AIM OF THE STUDY: This study aims to exp

Comprehensive Profiling of ac4C RNA Modification Identifies CERCAM in Cancer-Associated Fibroblasts as a Key Prognostic and Microenvironmental Regulator in Colorectal Cancer.

BACKGROUND: N4-acetylcytidine (ac4C) RNA modification has emerged as a critical epigenetic regulator in tumorigenesis and progression. However, the comprehensive landscape of ac4C modification in colorectal cancer (CRC), particularly its orchestration of the tumor microenvironment (TME) and stromal crosstalk, remains largely unexplored. AIMS: This study aimed to comprehensively delineate the landscape of N4-acetylcytidine (ac4C) RNA modification in colorectal cancer (CRC) to uncover its role in shaping the tumor microenvironment and clinical outcomes. Integrated bioinformatic and in vitro ana

Spatial transcriptomics in the human adult ovary: insights into key signalling pathways during follicular atresia.

STUDY QUESTION: Are key signalling pathways WNT, TGFβ/BMP, NOTCH, and HH involved in follicular atresia in the human adult ovary? SUMMARY ANSWER: In this study, we used spatial transcriptomics to investigate the progression of follicular atresia, focusing on genes of interest associated with steroidogenesis and key signalling pathways WNT, TGFβ/BMP, NOTCH, and HH. WHAT IS KNOWN ALREADY: While extensive research has focused on the mechanisms driving follicular growth, much less is known about the process of follicular atresia, despite its relevance for ovarian aging and reproductive longevity

Multicenter Phase 1b/2a Clinical Trial of Radioprotectant BIO 300 Oral Suspension for Patients With Non-Small Cell Lung Cancer Receiving Concurrent Chemoradiotherapy.

PURPOSE: Radiation therapy is part of the standard treatment regimen for non-small cell lung cancer (NSCLC). Although radiation therapy is an effective tool to manage NSCLC, it can be associated with significant dose-limiting toxicities. These toxicities can lead to treatment interruption or early termination and worsening clinical outcomes in addition to reductions in patient quality of life. Based on preclinical efficacy for radioprotection of normal tissues, we evaluated the clinical utility of BIO 300 Oral Suspension (BIO 300; synthetic genistein nanosuspension) in patients with NSCLC. The expression and localization of estrogen receptor beta in hyperplastic and neoplastic prostate lesions.

BACKGROUND/AIM: Benign acini in benign prostatic hyperplasia (BPH) are lined with pseudostratified cylindrical epithelium with a continuous basal cell layer. Adenocarcinoma of the prostate is the most common cancer in men. High gradus-prostatic intraepithelial neoplasia (HGPIN) lesions precede invasive cancer. Prostate adenocarcinoma (PCa) implies a complete absence of basal cells and stromal invasion by malignant acini. Estrogen receptor (ER) is located in nuclei of acinar basal and secretory cells and partially in stromal cells. The aim of this research was to demonstrate and localize ER in

[Ultrasonography methods in the diagnosis of renal osteodystrophy].

Bone disease, i.e. renal osteodystrophy, is commonly seen in patients with chronic renal failure. It encompasses all the disorders of mineral and bone metabolism associated with chronic renal insufficiency, i.e. secondary hyperparathyroidism, retention and accumulation of beta 2 microglobulin and aluminum. The most frequent cause of renal osteodystrophy is secondary hyperthyroidism, with a consequence of high turnover bone disease. Secondary hyperparathyroidism, i.e. increased parathyroid hormone (PTH) secretion and parathyroid gland hyperplasia, develops early in the course of chronic renal i

Mucopolysaccharidosis type II, Hunter's syndrome.

Hunter syndrome is caused by deficiency of the lysososmal enzyme iduronate-2-sulphatase that cleaves O-linked sulphate moieties from dermatan sulphate and heparan sulphate and leads to accumulation of GAGs. The disease is a X-linked condition affecting males and rarely females, clinically divided into severe (2/3) and attenuated types. Children with severe form, diagnosed at 12-36 months, have coarse facial feature, short stature, joint stiffness, short neck, broad chest, large head circumference, watery diarrhea, skeletal changes, progressive and profound mental retardation, retinal degenerat

Revitalizing GIP: Therapeutic Potential in Metabolic and Neurodegenerative Disorders.

Glucose-dependent insulinotropic polypeptide (GIP), once the overlooked sibling of the incretin family, is now experiencing a research renaissance. Historically, its therapeutic development was hindered by a seemingly diminished insulinotropic effect in type 2 diabetes (T2DM), its paradoxical stimulation of glucagon during hyperglycemia, and translational gaps between rodent and human physiology. This review highlights the renewed interest in GIP, driven by a deeper understanding of its pleiotropic actions. GIP stimulates glucose-dependent insulin secretion and, uniquely, also stimulates gluca

The OVAREX study: Establishment of ex vivo ovarian cancer models to validate innovative therapies and to identify predictive biomarkers.

BACKGROUND: Ovarian cancer is the first cause of death from gynecological malignancies mainly due to development of chemoresistance. Despite the emergence of PARP inhibitors, which have revolutionized the therapeutic management of some of these ovarian cancers, the 5-year overall survival rate remains around 45%. Therefore, it is crucial to develop new therapeutic strategies, to identify predictive biomarkers and to predict the response to treatments. In this context, functional assays based on patient-derived tumor models could constitute helpful and relevant tools for identifying efficient t

How bones learn to listen: In vivo models for investigating mechanosensitivity at bony interfaces in the ageing mouse.

Beyond the well-documented loss of bone volume, the aged skeletal environment is marked by structural decay and the degenerated network of osteocytes, the bone cells responsible for orchestrating bone formation and resorption. Recent findings point towards the loss of mechanosensitivity and thus the 'deafened' adaptive response to mechanical loading as the driver of decreased bone formation. This imbalance results in a net loss of bone mass and, over time, increases skeletal fragility and fracture incidence. Although the responsiveness of bones to mechanical loading is typically diminished wit

The efficient prediction of inflammatory osteolysis caused by polylactic acid through network toxicology and molecular docking strategy.

Polylactic acid (PLA), as a bioplastic, is extensively utilized in bone tissue engineering for its biocompatibility, adaptability and affordability. However, the toxicological research of PLA is still limited. The hydrolysis products of PLA induced inflammatory response which caused inflammatory osteolysis mediated by oxidative damage through the recruitment of macrophages and the accumulation of foreign body multinucleated giant cells, ultimately leading to the failure of bone tissue regeneration. The lack of effective treatments highlights the importance of finding new therapies. This study

Future regenerative medicine developments and their therapeutic applications.

Although the currently available pharmacological assays can cure most pathological disorders, they have limited therapeutic value in relieving certain disorders like myocardial infarct, peripheral vascular disease, amputated limbs, or organ failure (e.g. renal failure). Pilot studies to overcome such problems using regenerative medicine (RM) delivered promising data. Comprehensive investigations of RM in zebrafish or reptilians are necessary for better understanding. However, the precise mechanisms remain poorly understood despite the tremendous amount of data obtained using the zebrafish mode

Multidrug resistance in epilepsy: rats with drug-resistant seizures exhibit enhanced brain expression of P-glycoprotein compared with rats with drug-responsive seizures.

Medical intractability, i.e. the absence of any response to anti-epileptic drug (AED) therapy, is an unresolved problem in many patients with epilepsy. Mechanisms of intractability are not well understood, but may include alterations of pharmacological targets and poor penetration of AEDs into the brain because of increased expression of multiple drug-resistance proteins, such as P-glycoprotein (Pgp; ABCB1), capable of active brain extrusion of various drugs, including AEDs. Increased expression of Pgp has been reported in brain tissue of patients with refractory epilepsy, but there is a lack

Htra2-beta 1 stimulates an exonic splicing enhancer and can restore full-length SMN expression to survival motor neuron 2 (SMN2).

Spinal muscular atrophy (SMA), a common motor neuron disease in humans, results from loss of functional survival motor neuron (SMN1) alleles. A nearly identical copy of the gene, SMN2, fails to provide protection from SMA because of a single translationally silent nucleotide difference in exon 7. This likely disrupts an exonic splicing enhancer and causes exon 7 skipping, leading to abundant production of a shorter isoform, SMN2Delta7. The truncated transcript encodes a less stable protein with reduced self-oligomerization activity that fails to compensate for the loss of SMN1. This report des

Novel drug-free cascaded nanoparticles induce tumor-specific ROS storms via multimodal synergistic anticancer therapy.

Reactive oxygen species (ROS), generated by sonosensitizers, play a pivotal role in tumor cell apoptosis during sonodynamic therapy (SDT), particularly for tumors located deep within tissues. Nevertheless, conventional sonosensitizers present limitations including inadequate ROS generation, insufficient tumor-specific accumulation, and associated adverse effects, significantly restricting their clinical applicability. To address these limitations, novel drug-free multifunctional nanoparticles (designated HGMP NPs) were synthesized. These NPs consist of mesoporous polydopamine (MPDA)-loaded pro

From "time is brain" to "time is collaterals": updates on the role of cerebral collateral circulation in stroke.

BACKGROUND: Acute ischemic stroke (AIS) remains the leading cause of mortality and disability worldwide. While revascularization therapies-such as intravenous thrombolysis (IVT) and endovascular thrombectomy (EVT)-have significantly improved outcomes, their success is strongly influenced by the status of cerebral collateral circulation. Collateral vessels sustain cerebral perfusion during vascular occlusion, limiting infarct growth and extending therapeutic windows. Despite this recognized importance, standardized methods for assessing collateral status and integrating it into treatment strate
