Frequency and Longitudinal Course of Behavioral and Neuropsychiatric Symptoms in Participants With Genetic Frontotemporal Dementia.

BACKGROUND AND OBJECTIVES: Behavioral and neuropsychiatric symptoms are frequent in patients with genetic frontotemporal dementia (FTD). We aimed to describe behavioral and neuropsychiatric phenotypes in genetic FTD, quantify their temporal association, and investigate their regional association with brain atrophy. METHODS: We analyzed data of pathogenic variant carriers in the chromosome 9 open reading frame 72 (c9orf72), progranulin (GRN), or microtubule-associated protein tau (MAPT) gene from the Genetic Frontotemporal dementia Initiative cohort study that enrolls both symptomatic pathogen

Histopathological changes underlying frontotemporal lobar degeneration with clinicopathological correlation.

We have investigated the pathological correlates of dementia in the brains from a consecutive series of 70 patients dying with a clinical diagnosis of frontotemporal lobar degeneration (FTLD). Clinical misdiagnosis rate was low with only 3 patients (4%) failing to show pathological changes consistent with this diagnosis; 1 patient had Alzheimer's disease and 2 had cerebrovascular disease (CVD). In the remaining 67 patients, the most common underlying histological cause was ubiquitin pathology with 24 (36%) cases so affected. In these, ubiquitin-positive inclusions were present in the cerebral

Population genetic structure analysis of Guangdong Black pigeon and validation of black plumage trait-associated loci based on genome-wide association study.

Plumage color is one of the key phenotypic traits for breed identification in poultry, and deciphering its genetic basis is of great significance for the conservation and sustainable utilization of germplasm resources. In this study, we performed whole-genome resequencing (WGR) on 127 Guangdong Black pigeons and 12 Tarim pigeons, and integrated existing resequencing data from four other pigeon breeds (40 individuals in total). Principal component analysis, population structure analysis, and phylogenetic tree construction revealed that Guangdong Black pigeons have a unique genetic background an

Profiling Extrachromosomal Circular DNA (ecDNA) in Cancer via Next-Generation Sequencing: A New Frontier in Tumor Evolution and Therapy Resistance.

INTRODUCTION: Extrachromosomal circular DNA (ecDNA) has emerged as a critical factor in cancer biology. Unlike chromosomal DNA, ecDNA exists as circular, non-centromeric molecules that can carry oncogenes, promoting tumor growth, genetic diversity, and resistance to therapy. METHODS: In this review, we explored recent findings on ecDNA in human cancers, highlighting its structural characteristics, formation mechanisms, and functional roles. We examined how next-generation sequencing (NGS) technologies-such as whole-genome sequencing, Circle-Seq, and single-cell approaches-have improved the id

Establishment of Sertoli Cell Lines from Giant Pandas (Ailuropoda melanoleuca).

Giant pandas (Ailuropoda melanoleuca) are a critically protected endangered species in China, but current understanding of their testicular physiology and regulatory mechanisms for spermatogenesis remains limited. Sertoli cells (SCs), the sole somatic cell type within seminiferous tubules, play essential roles in nutritional support, blood-testis barrier (BTB) formation, and hormonal regulation, indispensable for germ cell development in testes. However, primary cultured SCs are prone to undergo replicative senescence, which severely impedes their in-depth studies in vitro. In this study, we i

The role of methylation and structural variants in shaping the recombination landscape of barley.

Meiotic recombination is not only a key mechanism for sexual adaptation in eukaryotes but crucial for the accumulation of beneficial alleles in breeding populations. The effective manipulation of recombination requires, however, a better understanding of the mechanisms regulating the rate and distribution of recombination events in genomes. Here, we identified the genomic features that best explain the recombination variation among a diverse set of segregating populations of barley at a resolution of 1 Mbp and investigated how methylation and structural variants (SVs) determine crossover (CO)

From Tumor Suppressor to Tumor Promoter - The Dual Role of LIMCH1 in Cancer and Its Clinical Implications.

LIM and Calponin Homology Domain 1 (LIMCH1) is a multi-domain protein-coding gene located on human chromosome 4p13. Its encoded protein participates in the dynamic regulation of the actin cytoskeleton through an N-terminal calponin homology domain and a C-terminal LIM domain. LIMCH1 specifically binds to non-muscle myosin IIA (NM-IIA). This binding enhances the dual phosphorylation of myosin regulatory light chains. As a result, it increases the adenosine triphosphatase (ATPase) activity and assembly efficiency of NM-IIA, stabilizes stress fibers, promotes focal adhesion maturation, and inhibi

Cell-cycle-resolved Hi-C reveals unexpected plasticity of A/B compartments across interphase.

The spatial organization of chromatin into active (A) and inactive (B) nuclear compartments is fundamental to genome regulation, yet their cell-cycle dynamics remain largely unexplored. Most research on chromatin dynamics during the cell cycle has primarily focused on events surrounding mitosis, providing only limited insight into chromatin behavior during S-phase. To address this gap, we developed a simple, drug-free approach that combines the Fucci cell-cycle indicator with in situ Hi-C to comprehensively analyze A/B compartment dynamics throughout interphase in mouse embryonic stem cells (m

Optical genome mapping detects cryptic high-risk and targetable abnormalities in adult AML.

Acute myeloid leukaemia (AML) risk stratification relies on cytogenetic and molecular abnormalities defined by European LeukemiaNet (ELN) 2022. Conventional cytogenetic techniques, including chromosomal banding analysis (CBA) and fluorescence in situ hybridization, have limited resolution and may miss cryptic events. Optical genome mapping (OGM) is a genome-wide approach capable of detecting balanced and unbalanced structural variants with high resolution, potentially revealing cryptic abnormalities of diagnostic and prognostic relevance. We retrospectively studied 100 adults with newly diagno

Phage satellites induced by virulent phages are mobilized by natural competence leading to phage resistance in a new host.

A phage satellite (PS) typically resides within repeat regions (attL and attR sites) of a bacterial genome. Its genome ranges from 7 to 20-kb and includes genes encoding an integrase along with regulatory and DNA replication functions. However, it lacks genes associated with viral structural proteins. Streptococcus thermophilus (S.t.) is extensively used to produce yogurt and specialty cheeses. Intriguingly, the majority of S.t. strains harbor a PS while very few possess a complete prophage, suggesting that PSs may confer advantages to their hosts. In this study, we showed that PSs of S.t. can

RVB-1 and RVB-2 are stress responsive proteins in Neurospora crassa and RVB-1 interacts with the centromeric Shugoshin (SGO-1) protein.

The Neurospora crassa RVB-1 and RVB-2 proteins belong to the AAA+ (ATPases Associated with various cellular Activities) superfamily and are organized in a hetero-hexameric complex essential for multiple cellular processes. Although they have been described as associated with cancer, new molecular functions have recently been described for them. Here, we investigate their participation in the fungus stress response, and our results support that they are components of the regulatory mechanism involved in the cellular stress response. Results of gene and protein expression, as well as cellular lo

Mechanistic Perspectives From Genomics and Pangenomics of Medicinal and Aromatic Plants: Linking Genome Architecture to Phytochemical Diversity.

Medicinal and aromatic plants (MAPs) produce a remarkable diversity of specialized metabolites with significant pharmaceutical, nutraceutical, and industrial value. Although advances in long-read sequencing, chromosome-scale genome assembly, and pangenomics have greatly expanded genomic resources, the mechanistic links between genome architecture and phytochemical diversity remain incompletely understood. The present review synthesizes current evidence describing how structural genomic variation may contribute to phytochemical diversity, while acknowledging that many proposed genome-to-metabol

Rethinking genomics of facioscapulohumeral muscular dystrophy in the telomere-to-telomere era: pitfalls in the hidden landscape of D4Z4 repeats.

Facioscapulohumeral muscular dystrophy (FSHD) is genetically associated with reduction of the D4Z4 macrosatellite array at 4q35 on a permissive 4qA haplotype, a configuration that enables the stable expression of the DUX4 transcription factor. Current diagnostic and mechanistic models, however, rely heavily on the incomplete GRCh38/hg38 reference and assume that D4Z4 repeats are predominantly confined to 4q35 and 10q26 loci. Here we present a systematic re-analysis of the configuration of D4Z4-like repeats in the human genome using the Telomere-to-Telomere human genome assembly (T2T-CHM13 v2.0

The SlDUF789 Gene Family in Tomato: Genome-Wide Identification and Responsiveness to Hormonal and Abiotic Stress Signals.

The DUF789 protein family comprises conserved domain proteins in plants, most of which remain functionally uncharacterized despite their involvement in growth, development, and stress responses. Moreover, systematic identification of the DUF789 family in tomato (Solanum lycopersicum L.) has not been reported. In this study, 11 SlDUF789 members were identified from the tomato genome, unevenly distributed across seven chromosomes, with members within the same subgroup sharing similar exon-intron structures and conserved motifs. Promoter analysis revealed the enrichment of cis-acting elements ass

Ulomoides dermestoides as an Insect Pharmacological Resource of Antioxidant and Anti-Inflammatory Bioactive Substances: Chemical Basis, Mechanisms of Action, Pharmacological Evidence, and Translational Challenges.

Ulomoides dermestoides (Yangchong) is a tenebrionid beetle used in traditional medicine across Asia and Latin America. While crude extracts show effects on inflammation, oxidative stress, and other conditions, systematic integration of its bioactive substances, mechanisms, and translational potential is lacking. This review consolidates its chemical basis, comprising volatile benzoquinones, terpenes, and alkenes, alongside non-volatile fatty acids, proteins (antioxidant enzymes, glycoproteins), and phenolics. Pharmacological evidence indicates multi-target modulation of reactive oxygen species

Multi-omics of the endangered Acer miaotaiense reveal nervonic acid biosynthesis, population evolution, and environmental adaptation.

Climate change threatens global forest biodiversity, particularly affecting long-lived, endangered tree species. However, the molecular mechanisms underlying their population evolution and environmental adaptation remain unclear. Here, we employed multi-omics analyses, including pan-genomes, transcriptomics, metabolomics, population genomics, and epigenomics, to decipher nervonic acid biosynthesis and population evolutionary history of the endangered tree Acer miaotaiense. A high-quality, chromosome-level genome of A. miaotaiense was first assembled (645.94 Mb). Pan-genome characterization sho

Transcriptomic responses of kveik yeasts to heat and ethanol stress.

Kveik strains are a group of Saccharomyces cerevisiae strains notable for their exceptional tolerance to environmental stress. To investigate the molecular basis of this robustness, the transcriptomes of three kveik strains and a conventional ale strain were compared under non-stress and stress conditions using RNA-seq. Under non-stress conditions, kveik strains showed elevated basal expression of environmental stress response genes, suggesting that part of the stress-responsive transcriptional program is active irrespective of stress exposure. Catalase activity was elevated, providing physiol

Inflammatory signaling differentially changes chromatin accessibility and gene expression of the PD- associated kinase LRRK2 between human and mice.

The genomic locus that encodes the Leucine-rich repeat kinase 2 (LRRK2) gene is highly pleiotropic and associated with Parkinson’s disease (PD). Coding variants associated with risk of PD act as gain of function kinase mutations increasing phosphorylation of RAB substrates, and non-coding variants in the promoter region of LRRK2 increase expression of the gene, notably in immune cells. If regulation of LRRK2 expression is a causal contributor to PD, it is important to understand the mechanism(s) by which LRRK2 is regulated, particularly in the context of inflammation. Here, we show that interf

The senescence ecosystem in genomic and epigenomic evolution during inflammation-associated carcinogenesis.

Chronic inflammation promotes cancer through repeated tissue injury, incomplete repair, and selection of adapted cell clones, but the mechanisms that convert a reversible inflammatory response into a tumor-permissive state remain incompletely defined. Cellular senescence provides a plausible, context-dependent component of this process. Although senescence initially limits expansion of damaged cells, persistent senescent cells and their niches can alter DNA-damage exposure, epigenomic state, immune surveillance, stromal mechanics, vascular function, and the fitness landscape encountered by nei
