The dengue vector Aedes aegypti contains a functional high mobility group box 1 (HMGB1) protein with a unique regulatory C-terminus.

The mosquito Aedes aegypti can spread the dengue, chikungunya and yellow fever viruses. Thus, the search for key molecules involved in the mosquito survival represents today a promising vector control strategy. High Mobility Group Box (HMGB) proteins are essential nuclear factors that maintain the high-order structure of chromatin, keeping eukaryotic cells viable. Outside the nucleus, secreted HMGB proteins could alert the innate immune system to foreign antigens and trigger the initiation of host defenses. In this work, we cloned and functionally characterized the HMGB1 protein from Aedes aeg…

Protection from secondary dengue virus infection in a mouse model reveals the role of serotype cross-reactive B and T cells.

The four dengue virus (DENV) serotypes cause dengue fever and dengue hemorrhagic fever/dengue shock syndrome. Although severe disease has been associated with heterotypic secondary DENV infection, most secondary DENV infections are asymptomatic or result in classic DF. The role of cross-reactive immunity in mediating cross-protection against secondary heterotypic DENV infection is not well understood. DENV infection of IFN-α/β and IFN-γ receptor-deficient (AG129) mice reproduces key features of human disease. We previously demonstrated a role in cross-protection for pre-existing cross-reactive…

Serological evaluation of suspected West Nile virus human cases following its introduction during a dengue outbreak in Puerto Rico in 2007.

A laboratory testing algorithm was evaluated to confirm West Nile virus (WNV) infection in human serum following the introduction of the virus in Puerto Rico in 2007. This testing algorithm used two standard diagnostic assays, the IgM antibody capture enzyme-linked immunosorbent assay (MAC ELISA) and real-time reverse transcriptase PCR (RT-PCR), along with two nonconventional assays, the nonstructural protein 1 (NS1) ELISA and a 90%-plaque-reduction neutralization test (PRNT(90)) with IgG depletion for dengue virus (DENV) and WNV. A total of 2,321 serum samples from suspected WNV human cases w…

Both viremia and cytokine levels associate with the lack of severe disease in secondary dengue 1 infection among adult Chinese patients.

Secondary dengue infections are frequently associated with increased risk for dengue hemorrhagic fever and dengue shock syndrome. Surprisingly, we observed no dengue hemorrhagic fever cases among 353 hospitalized dengue-infected patients including 212 with primary, and 141 with secondary dengue 1 infection in China. To explore virological and immunological mechanisms which may account for this unexpected clinical observation, we assessed dengue viremia, type I interferon and inflammatory cytokine levels in these patients. While the levels of viremia and inflammatory cytokines are indistinguish…

Molecular epidemiology of dengue virus serotype 2 in the Taiwan 2002 outbreak with envelope gene and nonstructural protein 1 gene analysis.

The genetic relationships among dengue virus serotype 2 (DEN-2) isolates from the Taiwan 2002 epidemic were studied by sequence analysis of the envelope (E) and nonstructural protein 1 (NS1) genes. A 0-0.4% divergence among 10 isolates revealed an epidemic strain in the outbreak. Phylogenetic study demonstrated that the 2002 Taiwan isolates were of the Cosmopolitan genotype, which is different from the Asian 1 and Asian 2 genotypes of Taiwan DEN-2 isolates from 1981 to 1998 and the American/Asian genotype of 2005 Taiwan isolates. Although grouping results from both E and NS1 gene sequence anal…

Comparison of rapid centrifugation assay with conventional tissue culture method for isolation of dengue 2 virus in C6/36-HT cells.

A rapid centrifugation assay was compared with conventional tube cell culture for dengue virus isolation in both sera and autopsy samples from dengue and dengue hemorrhagic fever/dengue shock syndrome fatal cases. The rapid centrifugation assay allowed isolation of virus from 16.6% more samples than the conventional method, and it shortened the time for dengue virus detection. Finally, it allowed the isolation of dengue 2 virus in 42.8% of tissue samples from five fatal cases. Our results suggest that the rapid centrifugation assay may be useful for detection of dengue virus in clinical specim…

Cross-sectional study on the association between seroprevalence among children and their parents' knowledge, attitudes, and practices in a dengue-endemic city in Bangladesh.

INTRODUCTION: Bangladesh is experiencing a drastic increase in dengue infections since 2019, with a record number of cases exceeding 300,000 in 2023 and 102,082 cases in 2025. Control of dengue, spread by Aedes mosquitoes, requires an in-depth understanding of knowledge, attitudes, and practices (KAP) regarding prevention strategies and their association with the risk of infection. This association has been rarely examined in adults and even more rarely in relation to the risk of infection in children. This study aimed to investigate the association between parents' knowledge, attitudes, and p…

Epidemiological Insights into Dengue Infection with Preliminary Emphasis on Serotype Distribution and Risk Factors in Karbi Anglong, Assam: A Cross-Sectional Study.

Dengue is a viral illness primarily transmitted by Aedes aegypti and Aedes albopictus, which thrive in tropical and densely populated urban environments. Its rising global incidence has been linked to factors such as climate change, inadequate sanitation, and expanding urbanization. A dengue outbreak in Diphu, Assam, from 2022 to 2024 prompted this study, which aims to identify the circulating serotypes and assess associated risk factors. The study involved 7,805 symptomatic patients from Diphu Medical College and Integrated Disease Surveillance Program field visits. Patients with malaria or o…

Development and optimization of an mRNA-vectored single-chain IgA1 isotype monoclonal antibody with potential to treat or prevent dengue virus infection.

Dengue virus (DENV) is a rapidly expanding infectious disease threat that causes an estimated 100 million symptomatic infections every year. A barrier to preventing DENV infections with traditional vaccines or prophylactic monoclonal antibody (mAb) therapies is the phenomenon of Antibody-Dependent Enhancement (ADE), wherein sub-neutralizing levels of DENV-specific IgG antibodies can enhance infection and pathogenesis rather than providing protection from disease. Fortunately, IgG is not the only antibody isotype capable of binding and neutralizing DENV, as DENV-specific IgA1 isotype mAbs can b…

Genomic hotspots in the DENV-2 serotype (E, NS4B, and NS5 genes) are associated with dengue disease severity in the endemic region of India.

Dengue virus (DENV) infection manifests a wide spectrum of clinical outcomes, ranging from mild fever to severe and potentially fatal disease, yet the factors driving this variability remain poorly understood. This study aims to unravel the relationship between clinical manifestations of dengue and the genetic diversity of the virus, providing insights into the genomic variability driving disease severity. To achieve this, serum samples were collected during a dengue outbreak in National Capital Region-Delhi, India, from June to November 2023. Serotyping of RNA isolated from 4,045 clinical ser…

Dynamics of vector competence for dengue virus type 2 in rural and urban populations of Aedes albopictus: implications for infectious disease control.

BACKGROUND: Understanding the intrinsic factors that influence mosquito vector competence (VC) to pathogens is crucial for assessing the risk of disease transmission in both rural and urban environments. We assessed the VC of Aedes albopictus mosquitoes from urban (dengue-endemic) and rural (dengue-free) areas in Guangzhou, China, for dengue virus-2 (DENV-2), while also examining intrinsic factors such as Wolbachia and immune-related gene expression influencing VC. METHODS: Adult females of rural, urban, and laboratory (control) populations of Ae. albopictus were orally exposed to a freshly p…

Antioxidant Response as a Candidate Prognostic Factor for Dengue Hypotensive and Hemorrhagic Complications: Results from a Nested Case-Control Study in Colombia.

Dengue infection has been associated with oxidative stress (OS) induction; however, whether such a response predicts the development of complications remains unknown. We conducted a case-control study (1:2 ratio) nested within a cohort of febrile patients with a presumptive or confirmed diagnosis of dengue. Incident cases were patients who developed hypotension or severe bleeding during the follow-up, whereas controls did not. Total antioxidant status (TAS), superoxide dismutase (SOD), and glutathione peroxidase activity (GPx) were quantified in serums obtained ≤96 h from disease onset. The as…

The role of serum ferritin in predicting plasma leakage among adults and children with dengue in Sri Lanka: a multicentre, prospective cohort study.

BACKGROUND: Early prediction of plasma leakage helps in timely management of dengue. Currently, there are no defined early predictors of plasma leakage, and identified parameters are late predictors. Raised ferritin is associated with severe dengue, but its clinical utility early in the disease to predict severe dengue is not previously reported. We studied the efficacy of day 3 or 4 serum ferritin in predicting plasma leakage among adults and children with dengue in Sri Lanka. METHODS: We conducted a prospective cohort study in four hospitals in Sri Lanka from June 2022 to June 2023. Consecu…

Automation for lateral flow rapid tests: Protocol for an open-source fluid handler and applications to dengue and African swine fever tests.

Lateral flow rapid diagnostic tests (RDTs, RTs) are cost-effective with low infrastructure requirements for limited-resource settings, and in any setting can represent a bridge between early disease monitoring at outbreak onset and fully-scaled molecular testing for human or animal diseases. However, the potential of RTs to handle higher throughput testing is hampered by the need for manual processing. Here we review dengue virus and African swine fever virus rapid tests, and present a novel protocol that employs an open-source fluid handler to automate the execution of up to 42 RTs per run. A…
