Mutations in the human GJB3 gene that codes for Connexin31 (Cx31), a protein subunit of gap junction channels, have recently been reported to cause deafness and the skin disorder erythrokeratodermia variabilis. To study the function of this gene in mice, we generated animals with targeted replacement of the Cx31 gene (Gjb3) by a lacZ reporter gene. Although homozygous Cx31-deficient adult mice (Gjb3(-/-)) were found among the offspring of heterozygous Cx31-deficient parents (Gjb3(+/-)), 60% of the animals expected according to Mendelian inheritance were lost between ED 10.5 and 13.5. It is well known that exposure to intense acoustic stimuli can induce permanent damage to the tissues of the inner ear. In this study the technique of ABR measurement and the experimental animal model of acoustic trauma were presented. In forty 3 months old guinea pigs the hearing threshold was assessed using the ABR technique (Amplaid mk 10 system). The hearing thresholds ranged from 40 to 50 dB-pe-SPL (mean 45.26 dB, sd. 4.73). The mean latencies of the four most prominent peaks in the ABR pattern were also measured. Fourteen of animals were then exposed to a 1 kHz pure tone at different dur

Guinea pigs were exposed to a 4.5 kHz pure-tone at 104 dB for 10 min during artificial ventilation with either carbogen gas (95% O2/5% CO2) or normal air. Mean N1 response amplitudes to tone bursts at 32 test frequencies extending from 2.1 kHz through 30 kHz were measured at standardized intervals before and after the acoustic overstimulation. All animals received normal air during recovery. Significant reduction of N1 response amplitude depression within a 3/8 to 1 octave frequency domain above the exposure frequency was found in the group which received the carbogen gas. Those frequencies fo

BACKGROUND: Good hearing is a fundamental skill that allows children to develop properly, both socially and intellectually. In contrast to defects in inner ear function, however, auditory processing disorders (APDs)-which can affect up to 2-3% of school-children-are not easily identified with basic screening programs and must be diagnosed using special tests. Although such psychoacoustic tests are available, the scores achieved depend highly on the social, cultural, and linguistic characteristics of the population, and norms must be established for each population separately. Age-related neurodegenerative diseases and age-related hearing loss are increasing in prevalence globally. Immune cells are novel mediators linking these diseases through shared mechanisms of chronic inflammation and maladaptive repair. In the cochlea, resident macrophages and infiltrating immune cells respond to injury, noise exposure, and aging by adopting activated phenotypes that disrupt synaptic integrity, impair myelination, and promote hair cell and neuronal loss. Similarly, in the central nervous system, microglia, the resident immune cells of the brain undergo age- and disease-associa

Age-related hearing loss (ARHL) is a prevalent neurosensory impairment. Traditional audiometry is post-symptomatic, fundamentally failing to capture the molecular deterioration that precedes clinical hearing decline by years. To address this gap, the clinical paradigm is urgently shifting toward precision medicine. This review summarizes emerging molecular biomarkers enabling the early detection, patient stratification, and targeted prevention of ARHL. We systematically evaluate the clinical measurability and diagnostic value of biomarkers across core pathological axes: cellular senescence, in

Background/Objectives: A Cochlear implantation (CI) is well-established auditory rehabilitation for severe to profound sensorineural hearing loss (SNHL), yet outcomes vary widely among implantees. Even with advancements in surgical methods and device technology, CI is still commonly applied as a generally uniform procedure, with limited attention to the underlying cause of SNHL. This review aims to summarize current evidence supporting etiology-based personalization of CI and to examine how etiology influences electrode selection, implantation timing, and clinical outcomes. Methods: We reviewe

Hereditary hearing loss is frequently linked to hair cell defects, even when the associated genes are expressed in the inner ear and other tissues including the brain. An example of this scenario is the clain-1 gene, associated with progressive loss of hearing in Usher syndrome type IIIA (USH3A). While mouse studies demonstrate that clarin-1 is an essential hair cell gene and early cochlear gene therapy shows promise, a comprehensive understanding of clarin-1's role in the auditory system may lead to better therapeutic and rehabilitation strategies for USH3A. Menière's disease is a chronic inner ear disorder characterized by recurrent vertigo attacks, tinnitus, and fluctuating hearing loss, which may progress to severe sensorineural hearing impairment in a subset of patients. Since 2015, international consensus criteria distinguish between definite and probable disease. Current therapeutic strategies follow a stepwise approach, ranging from non-ablative to ablative interventions. Cochlear implantation (CI) has emerged as an effective option for auditory rehabilitation and has also shown beneficial effects on vertigo and tinnitus. Combined strategie

Tinnitus is the perception of sound without a corresponding external sound source. This condition affects approximately 14% of adults, with approximately 2% experiencing severe symptoms. Underlying mechanisms of tinnitus suggest involvement of both peripheral and central processes, in which cochlear injury and deafferentation may trigger maladaptive plasticity, increased central gain, and thalamocortical dysrhythmia, modulated by limbic and salience networks. Neuroinflammation, somatosensory-auditory coupling and other factors, such as stress, may contribute to chronicity. Clinical expression

Von Hippel-Lindau disease (VHL) is a rare autosomal dominant hereditary tumour predisposition syndrome affecting multiple organ systems, including the retina, central nervous system, kidneys, pancreas, adrenal glands, and inner ear. It is caused by pathogenic variants in the VHL gene, resulting in loss of functional pVHL, abnormal stabilization of hypoxia-inducible factor (HIF)-α subunits, and activation of downstream transcriptional programmes involved in angiogenesis, metabolism, cell survival, and tumour growth. Clinically, VHL is characterized by a spectrum of benign and malignant neoplasm

Ménière's disease (MD) and vestibular migraine (VM) are two common vestibular disorders with significant clinical overlap in their symptomatic presentations, including vertigo, hearing loss, tinnitus, and aural fullness. Although distinct diagnostic criteria exist for each, this symptomatic similarity often makes early-stage differentiation challenging. While recent studies have found potential biomarkers for MD and VM, their diagnostic utility remains limited by small sample sizes and lack of standardized validation protocols. This necessitates continued reliance on a synthesis of established

Aquaporin-2 (AQP2) is a critical protein involved in water metabolism. It is primarily located in the renal collecting duct cells' apical plasma membranes and intracellular vesicles and regulates the movement of water into and out of cells. It plays a pivotal role in maintaining fluid balance, and its dysregulation is associated with conditions such as hypertension and heart failure, which contribute to cardiovascular disease progression. AQP2 has garnered significant attention as an emerging therapeutic target. Traditional Chinese medicine (TCM) has demonstrated efficacy in treating water bal

INTRODUCTION: Growing evidence implicates vitamin D in inner ear homeostasis, though its association with Ménière's disease (MD) remains incompletely characterized. This study aimed to compare serum 25-hydroxyvitamin D (25(OH)D) levels between patients with MD and healthy controls and to assess its correlation with hearing thresholds in MD cohort.
METHODS: In this cross-sectional study, 49 patients with MD and 250 matched healthy controls were enrolled at our institution between January 2023 and January 2025. Groups were matched for demographics, comorbidities, exercise habits, and seasonal b

Vogt-Koyanagi-Harada syndrome (VKH) is a rare autoimmune condition affecting melanocyte-containing tissues such as the eyes, skin, hair, inner ear, and meninges. The disease progresses through four phases: prodromal, uveitic, convalescent, and recurrent. Symptoms include headaches, photophobia, ocular inflammation, and auditory disturbances. Although the etiology behind VKH is unknown, it is believed to be triggered by viral infections such as the Epstein‒Barr virus or Cytomegalovirus in genetically predisposed individuals. It is diagnosed by confirming bilateral ocular involvement and ruling

Noise-induced hearing loss (NIHL) is a significant and urgent global public health concern, arising from prolonged exposure to elevated levels of noise. This auditory impairment harms delicate inner ear structures, particularly the essential hair cells transmitting auditory signals to the brain. Recognized by the World Health Organization as a major contributor to worldwide hearing loss, NIHL requires a comprehensive examination of its molecular and cellular mechanisms. Animal models emerge as indispensable tools for unraveling these intricacies, allowing researchers to simulate and study the
