Engineering Magnesium Oxide as a Mitochondrial Electron Transport Chain Inhibitor for Enhanced Photodynamic Therapy in Triple-Negative Breast Cancer.

Triple-negative breast cancer (TNBC) is a highly aggressive subtype with limited therapeutic options due to its resistance to conventional targeted therapies. TNBC cells heavily rely on oxidative phosphorylation (OXPHOS), making mitochondrial electron transport chain (ETC) inhibition a promising therapeutic approach. However, existing ETC inhibitors pose severe cytotoxicity risks, highlighting the need for biocompatible alternatives. In this study, we develop an MgO-ICG suspension (MgO-ICG@S), utilizing magnesium oxide (MgO), biocompatible pharmaceutical excipients that are already in widespre

Stigmatizing Language in Large Language Models for Alcohol and Substance Use Disorders: A Multimodel Evaluation and Prompt Engineering Approach.

OBJECTIVES: Large language models (LLMs) are increasingly used in health care communication but can inadvertently perpetuate stigmatizing language toward individuals with alcohol and substance use disorders. Despite growing interest in LLM performance, a focused evaluation of their propensity for SL and strategies to mitigate it remains lacking. METHODS: We generated 60 clinically relevant questions ["prompts"; 20 each for alcohol use disorder (AUD), alcohol-associated liver disease (ALD), and substance use disorder (SUD)] and tested 14 LLMs. Two physicians independently assessed all response

Differentiating Human Falls from Daily Activities Using Machine Learning Methods Based on Accelerometer and Altimeter Sensor Fusion Feature Engineering.

This paper presents a detailed analysis of signal data acquired from wearable sensors such as accelerometers and barometric altimeters for human activity recognition, with an emphasis on fall detection. This research addressed two types of activity recognition tasks: a binary classification problem between activities of daily living (ADLs) and simulated fall activities and a multiclass classification problem involving five different activities (running, walking, sitting down, jumping, and falling). By combining features derived from both sensors, traditional machine models such as random fores

Functional Group Engineering of Single-Walled Carbon Nanotubes for Anchoring Copper Nanoparticles Toward Selective CO2 Electroreduction to C2 Products.

Electroreduction of carbon dioxide (CO2) is a key strategy for achieving net-zero carbon emissions. Copper (Cu)-based electrocatalysts have shown promise for CO2 conversion into valuable chemicals but are hindered by limited C2+ product selectivity due to competing hydrogen evolution and ineffective dimerization of adsorbed CO intermediate (*CO). Here, a functional-group-directed strategy is reported to enhance selectivity using single-walled carbon nanotubes (SWCNTs) as supports. The catalytic performance of Cu nanoparticles is strongly influenced by the type and density of functional groups

Functional and structural insights into a thermostable (S)-selective amine transaminase and its improved substrate scope by protein engineering.

A (S)-selective amine transaminase from a Streptomyces strain, Sbv333-ATA, is a biocatalyst showing both high thermostability with a melting temperature of 85 °C and broad substrate specificity for the amino acceptor. This enzyme was further characterized both biochemically and structurally. The Sbv333-ATA is stable in the presence of up to 20% (v/v) of the water-miscible cosolvents methanol, ethanol, acetonitrile, and dimethyl sulfoxide, and in biphasic systems with petroleum ether, toluene, and ethyl acetate as an organic phase. The enzyme showed also a good activity toward different amino d

Hafnium-Based Chiral 2D Organic-Inorganic Hybrid Metal Halides: Engineering Polarity and Nonlinear Optical Properties via Para-Substituent Effects.

Two-dimensional (2D) organic-inorganic hybrid metal halides (OIMHs), characterized by noncentrosymmetric structures arising from the incorporation of chiral organic molecules that break inversion symmetry, have attracted significant attention. Particularly, chiral-polar 2D OIMHs offer a unique platform for multifunctional applications, as the coexistence of chirality and polarity enables the simultaneous manifestation of distinct properties such as nonlinear optical (NLO) effects, circular dichroism (CD), and ferroelectricity. In this study, we report the first synthesis of hafnium (Hf)-based

Functional Group Engineering Stabilizing Precursor Solution and Passivating Defects for Operationally Stable and Highly Reproducible Inverted Perovskite Solar Cells.

The instability of perovskite precursor solution induced by deprotonation of organic cations and oxidation of iodide ions substantially deteriorates the reproducibility and reliability of the photovoltaic performance of perovskite solar cells (PSCs). The above decomposition reactions can be conquered via the synergistic engineering of organic functional groups. However, how spatial conformation and type of weak acid functional groups impact the stability of perovskite precursor solution remains to be investigated. Herein, it is uncovered that the position of functional groups on the benzene an

Integrated genomic and transcriptomic Insights into methanol tolerance mechanisms in Methylobacterium extorquens AM1, identifying key targets for strain engineering.

Methanol is an attractive one-carbon feedstock for sustainable biomanufacturing because of its abundance, cost-effectiveness, and industrial compatibility. However, its cytotoxicity limits its biotechnological applications in native methylotrophs such as Methylobacterium extorquens AM1. In this study, we developed AM1-derived strains capable of sustained growth under elevated methanol concentrations through adaptive laboratory evolution (ALE). From the evolved population, five representative strains were isolated, exhibiting up to a 1.68-fold increase in specific growth rates compared with tho

Engineering IL-10 and rapamycin to bind collagen leads to improved anti fibrotic efficacy in lung and kidney fibrosis.

Fibrotic diseases are involved in 45% of deaths in the United States. In particular, fibrosis of the kidney and lung are major public health concerns due to their high prevalence and lack of existing treatment options. Here, we harness the pathophysiological features of fibrotic diseases, namely leaky vasculature and aberrant extracellular matrix (ECM) protein deposition (i.e. collagen), to target an anti-fibrotic biologic and a small molecule drug to disease sites of fibrosis, thus improving the therapeutic potential of both the biologic and small molecule in mouse models of both lung and kid

Engineering a Hydrogen Peroxide-Activated Hydrogen Sulfide Donor-Based Fluorescent Agent for Integrated Diagnosis and Therapy of Chronic Wounds.

Diabetic chronic wounds are one of the most severe complications of diabetes mellitus, which are pathologically characterized by persistent oxidative stress-inflammatory cascades. Thus, it is vital for effective diagnosis and treatment of diabetic chronic wounds by targeting the critical pathological oxidative stress feature in the tissue microenvironment. Herein, regarding the overexpressed hydrogen peroxide (H2O2) at the oxidative stress sites, we innovatively developed a H2O2-activated hydrogen sulfide (H2S) donor-based near-infrared fluorescent theragnostic agent (NDTA) to achieve synergis

Phosphorus-doped CoFe-prussian blue analogues (PBA)-Derived oxide with defect engineering for efficient peroxymonosulfate activation toward SMX degradation.

In this work, phosphorus-doped cobalt ferrite nanocubes (P-FCO) were synthesized from CoFe-PBA precursors via high-temperature oxidation and phosphorus doping. The P-FCO2/PMS system achieved 99 % removal of 20 ppm sulfamethoxazole (SMX) within 10 min, with an apparent rate constant of 1.52 min-1, outperforming most reported Co-Fe catalysts. Phosphorus doping increased oxygen vacancy concentration and enriched low-valence Co2+/Fe2+ species, enhancing PMS chemisorption. The formation of M(Co/Fe)-O-P bonds modulated the electronic structure, reduced electron transfer barriers, and promoted a dire

Numerical analysis of the MASnI3/CZT(Se1-x S x ) interface to boost the performance via band offset engineering.

This study investigates a tin-based perovskite solar cell (PSC) incorporating an inorganic hole transport layer, examined through simulations with the SCAPS simulator. The chosen CuZnSn(Se1-x S x ) compound emerges as a promising candidate for the hole transport layer, allowing for a tunable band gap via adjustments to the S/(S + Se) ratio. The band gap varies from 0.95 eV for Cu2ZnSnSe4 to 1.5 eV for Cu2ZnSnS4, achieved through strategic valence band offset engineering at the MAPbI3/CuZnSn(Se1-x S x ) interface. However, achieving an optimal Valence Band Offset (VBO) at MASnI3/CuZnSn(Se1-x S

Targeting and structural engineering of light-responsive nanoprobes for hierarchical therapy: construction, optimization, and applications in cancer stem cells.

Cancer stem cells (CSCs) possess the ability to self-renew and exhibit high differentiation potential, and they have been proven to be responsible for the maintenance, growth, and metastasis of tumors. As such, accurate identification and targeted therapy for CSCs are of great importance in clinical treatment. Here, a dual-targeted and light-responsive nanoprobe is presented, utilizing the reconstructed mesoporous SiO2 of a binary fatty acid eutectic mixture and a gold porous shell. The gold shell in the nanoprobe sustains a large absorption cross-section, providing a robust photothermal treat

Engineering and characterization of Hu3A4: A novel humanized antibody with potential as a therapeutic agent against myeloid lineage leukemias.

Leukemia stem cells (LSCs) play a critical role in the initiation, recurrence, and resistance to treatment of leukemia. Eradicating LSCs is crucial for the complete elimination of the disease. CD45RA is identified as an important marker for LSC subsets in acute myeloid leukemia (AML), providing a strategic target for therapy. In this report, we introduce Hu3A4, an innovative humanized CD45RA antibody devised to target LSCs expressing this antigen. Hu3A4 retains the antigen-recognition ability of its parental antibody while removing sequences from the variable region that could elicit human ant

Protocol for generation and engineering of thyroid cell lineages using CRISPR-Cas9 editing to recapitulate thyroid cancer histotype progression.

Thyroid carcinoma represents the first malignancy among the endocrine organs. Investigating the cellular hierarchy and the mechanisms underlying the initiation of thyroid carcinoma is crucial in thyroid cancer research. Here, we present a protocol for deriving thyroid cell lineage from human embryonic stem cells. We also describe steps for engineering thyroid progenitor cells utilizing CRISPR-Cas9 technology, which can be used to perform in vivo studies, thus facilitating the development of representative thyroid tumorigenesis models. For complete details on the use and execution of this proto

High-Specificity CRISPR-Mediated Genome Engineering in Anti-BCMA Allogeneic CAR T Cells Suppresses Allograft Rejection in Preclinical Models.

Allogeneic chimeric antigen receptor (CAR) T cell therapies hold the potential to overcome many of the challenges associated with patient-derived (autologous) CAR T cells. Key considerations in the development of allogeneic CAR T cell therapies include prevention of graft-vs-host disease (GvHD) and suppression of allograft rejection. Here, we describe preclinical data supporting the ongoing first-in-human clinical study, the CaMMouflage trial (NCT05722418), evaluating CB-011 in patients with relapsed/refractory multiple myeloma. CB-011 is a hypoimmunogenic, allogeneic anti-B-cell maturation an

Harnessing the potential of the NALT and BALT as targets for immunomodulation using engineering strategies to enhance mucosal uptake.

Mucosal barrier tissues and their mucosal associated lymphoid tissues (MALT) are attractive targets for vaccines and immunotherapies due to their roles in both priming and regulating adaptive immune responses. The upper and lower respiratory mucosae, in particular, possess unique properties: a vast surface area responsible for frontline protection against inhaled pathogens but also simultaneous tight regulation of homeostasis against a continuous backdrop of non-pathogenic antigen exposure. Within the upper and lower respiratory tract, the nasal and bronchial associated lymphoid tissues (NALT

Nature as blueprint: Global phenotype engineering of CHO production cells based on a multi-omics comparison with plasma cells.

Especially for the production of artificial, difficult to express molecules a further development of the CHO production cell line is required to keep pace with the continuously increasing demands. However, the identification of novel targets for cell line engineering to improve CHO cells is a time and cost intensive process. Since plasma cells are evolutionary optimized for a high antibody expression in mammals, we performed a comprehensive multi-omics comparison between CHO and plasma cells to exploit optimized cellular production traits. Comparing the transcriptome, proteome, miRNome, surfac
