SREBF1-associated hereditary mucoepithelial dysplasia (HMD) is a rare autosomal dominant disorder involving the hair, skin, mucosa, and eyes. We report a 5-year-old boy with recurrent non-scarring alopecia beginning at 18 months of age, together with blepharokeratoconjunctivitis, dry eye, corneal ulceration, recurrent oral ulcers, and nail pitting. Hair loss repeatedly worsened during or shortly after febrile episodes, most prominently in the occipital scalp, and was followed by gradual regrowth. Clinical exome sequencing identified a heterozygous SREBF1 NM_004176.5:c.1580G>A (p.Arg527His) var

Focal segmental glomerulosclerosis (FSGS) is a common histopathological diagnosis among adult patients presenting with nephrotic syndrome. This pattern is associated with diverse underlying pathophysiological mechanisms, which may influence disease management, therapeutic response, and prognosis. Genetic forms of FSGS are caused by variants in genes encoding proteins expressed in podocytes or the glomerular basement membrane. Multiple genes have been implicated in the pathogenesis of genetic FSGS, and clinical presentation can vary depending on the specific gene involved. We report the case of

BACKGROUND: May-Hegglin anomaly, a rare autosomal dominant disorder caused by MYH9 mutations, is characterized by the classic "triad" of thrombocytopenia, giant platelets, and granulocyte cytoplasmic inclusion bodies; some patients also present non-hematological symptoms. RESULTS: We reported a family of MYH9-related disease. The proband had microscopic hematuria, proteinuria, and thrombocytopenia on physical examination; blood smear re-examination showed giant platelets and Döhle-like bodies. His medical history included childhood epistaxis; his father had unexplained thrombocytopenia/protein

Tuberous sclerosis complex (TSC) is an autosomal dominant multisystem disorder distinguished by hamartomatous lesions involving multiple organs, most notably the brain, skin, kidneys, and oral cavity. Considerable variability in clinical expression is often observed even among affected members of the same family. We describe two siblings, aged 12 and 8 years, who presented for dental evaluation and were found to have systemic and orofacial features consistent with TSC. Both children displayed multiple cutaneous manifestations and demonstrated subependymal nodules on magnetic resonance imaging

Transcriptome analysis can improve the diagnostic yield for neurodevelopmental disorders. We applied RNA-seq using urine-derived cells (UDCs) to a family with two affected brothers with a muscular dystrophy-dystroglycanopathy and dilated cardiomyopathy. Exome sequencing identified a maternally inherited heterozygous B3GALNT2 splice-site variant (NM_152490.5:c.261-2A>G) but no second pathogenic allele. UDCs RNA-seq with OUTRIDER detected markedly decreased expression of B3GALNT2 in the proband and a milder decrease in the father, suggesting a paternally inherited non-coding variant. Genome sequ

This study investigated the relationship between early childhood experiences and eating disorder related core beliefs in the context of cognitive behavioural models of eating disorders highlighting the role of core beliefs or schema. A non-clinical sample of 736 participants (87 % female) was recruited and completed online self report questionnaires. Given the complexity of the relationship between the independent (parental bonding) and dependent (eating disorder related core belief) variables canonical correlation analysis was used to analyse the relationship between the two variable sets. This paper revisits the very well-known episode of Humphry Davy's electrical detection, isolation, naming, announcement, and publication of potassium and sodium. His initial experiments were made in the basement laboratory of the Royal Institution during the autumn of 1807. This paper draws on recently digitised Davy manuscripts as well as two letters (published as an appendix) written by the medical student Henry Holland to his father at the end of November 1807 which all provide significant new insights into this episode. The letters also illustrate the sociability and clubbable mores of the

Each new mammalian life begins with the fusion of an oocyte and a sperm to produce a fertilized egg containing two sets of genomes, one from the mother and one from the father. Androgenesis, a way for producing offspring solely from male genetic material, is limited in mammals, presumably due to barriers arising from genomic imprinting, an epigenetic mechanism leading to monoallelic gene expression. Here, we report adult mammalian offspring derived from the genetic material of two sperm cells. These mice, which we refer to as androgenetic mice, were produced via targeted DNA methylation editin

Leukocyte adhesion deficiency type III (LAD-III) is a rare autosomal recessive disorder characterized by immune dysfunction and bleeding tendencies. The condition arises from mutations in the FERMT3 gene, which disrupts integrin activation on leukocytes and platelets. This case study focuses on a family with consanguineous parents and multiple affected individuals spanning two generations, all diagnosed with LAD-III due to a novel homozygous mutation in the FERMT3 gene (c.1683-22_1683-19del). Clinical manifestations ranged from mild ecchymosis to severe bleeding necessitating transfusions. The

INTRODUCTION: Congenital mirror movement disorder refers to involuntary movements on one side of the body that mimic the deliberate movements on the opposite side. Congenital mirror movement is primarily associated with mutations in the DCC netrin-1 receptor (DCC) gene.
CASE PRESENTATION: A 3-year-old child had been involuntarily grasping with one hand and then the other from infancy. His neuromotor development corresponded with that of his contemporaries. Identical unusual movements were also observed in his father, uncle, and grandmother within his family heritage. In the family where ident

NM_000141.5: FGFR2 c.1032G>A is a pathogenic variant that causes Crouzon syndrome through activation of a new donor splice site. This clinical report highlights the intrafamilial variability that can exist with this specific variant. The proband is a 4-year-old boy who initially presented with concern for seizures. Computed tomography and magnetic resonance imaging revealed pancraniosynostosis and Chiari 1 malformation. Papilledema on fundoscopic exam and copper beaten appearance of the skull on imaging both suggested the presence of elevated intracranial pressure. Genetic testing identified c

OBJECTIVE: To explore the genetic basis for a boy affected with Cohen syndrome.
METHODS: A boy admitted to Children's Hospital of Nanjing Medical University in January 2021 was selected as the study subject. Genome DNA was extracted from peripheral blood samples from the child and his parents. Whole exome sequencing (WES) was carried out. And candidate variants were verified by Sanger sequencing. This study was approved by the Medical Ethics Committee of the hospital (Ethics No.: 202106060-1).
RESULTS: WES revealed that the child has harbored compound heterozygous variants of the VPS13B gene

Familial hypocalciuric hypercalcemia (FHH) is a rare genetic disorder caused by inactivating variants of the calcium-sensing receptor gene (CASR), GNA11, and AP2S1 genes, which lead to decreased intracellular receptor activity. Typical biochemical features include hypercalcemia and hypocalciuria, while PTH serum levels may be inappropriately normal or high. Despite the finding of hypercalcemia, the disorder has a benign course and patients usually do not require any therapeutical intervention. We describe a man with FHH caused by a previously unreported missense variant, c.496A>C, p.(Ser166Arg

Childhood emotional and behavioural difficulties tend to co-occur and often precede diagnosed neuropsychiatric conditions. Identifying shared and specific risk factors for early-life mental health difficulties is therefore essential for prevention strategies. Here, we examine how parental risk factors shape their offspring's emotional and behavioural symptoms (e.g. feelings of anxiety, and restlessness) using data from 14,959 genotyped family trios from the Norwegian Mother, Father and Child Cohort Study (MoBa). We model maternal reports of emotional and behavioural symptoms, organizing them i

Rabson-Mendenhall syndrome (RMS) is a rare autosomal recessive disorder caused by mutations in the insulin receptor gene (INSR), leading to severe insulin resistance. Clinical manifestations of RMS include hypertrichosis and acanthosis nigricans. A 3-yr-old male patient presented with darkened skin on the neck, without any apparent precipitating factors, and did not exhibit symptoms of polyuria or polydipsia. Both the patient and his older sister displayed signs of hypertrichosis and acanthosis nigricans. Laboratory investigations revealed significantly elevated levels of insulin and C-peptide
