Tics and heredity. A study of the relatives of child tiqueurs.

The limited literature on the pedigrees of tiqueurs, including those with Gilles de la Tourette's syndrome, is reviewed. Most statistical analyses have been restricted to affected family members without specifying the unaffected ones. The present statistical analysis of a series of child tiqueurs, including 91 probands and 1293 first- and second-degree relatives, 46 of whom were tiqueurs, predicts the odds on being a tiqueur for individuals, and establishes how those odds are affected by certain explanatory variables using log-linear models. The data do not confirm a familial pattern beyond re

Genetic overlap of chronic pain, musculoskeletal-specific pain, substance use disorders and substance use consumption: Common addiction and substance-specific effects.

BACKGROUND AND AIMS: Chronic pain and substance use disorders frequently co-occur and may share general addiction-related and substance-specific genetic pathways. Additionally, substance consumption and problematic use may differ in their genetic associations with pain subtypes. We applied genomic structural equation modeling to assess shared genetic and neurological enrichment between general and musculoskeletal-specific chronic pain to general addiction liability, substance use disorder-specific risk and substance use. DESIGN: Cross-trait genomic structural equation modeling using genome-wi

Genetic landscape and phenotypic correlations of lissencephaly: prenatal and postnatal insights.

Lissencephaly (LIS) is a spectrum of cortical malformations including agyria, pachygyria and subcortical band heterotopia, which arises from aberrant neuronal migration and is associated with severe neurodevelopmental impairments. Despite advancements in prenatal imaging, diagnosing LIS remains challenging. Genetic factors play a crucial role in LIS, involving multiple genes and signalling pathways, yet research on prenatal diagnosis and the genetic basis is still limited. This study aimed to assess the diagnostic yield of whole exome sequencing (WES) in LIS and to examine genotype-phenotype c

Embryos created for research purposes.

The creation of embryos for research use has drawn a great deal of criticism. It is difficult to defend an ethical distinction between what one can do to "spare" embryos and what one can do to "Исследование" embryos. The strongest ground on which to argue against the creation of embryos for research is a symbolic one, having to do with respect for human life. Ronald Dworkin's work in Life's Dominion on the symbolic meaning of the abortion debate throws a helpful light on this dispute. By understanding the basic question to be, Does the creation of research embryos weaken or insult our communal res

Sulfolobus acidocaldarius, half a century of archaeal research.

Since its isolation from a Yellowstone hot spring in 1972, Sulfolobus acidocaldarius has become one of the most important model organisms for archaeal biology. Initially studied for its remarkable adaptation to high temperature and low pH, it has evolved into a genetically tractable system that has contributed substantially to our understanding of archaeal physiology, molecular biology, and evolution. Here, we summarize more than 5 decades of research on S. acidocaldarius, highlighting key developments in genetic and microscopy tools and their impact on our understanding of archaeal cell biolo

Skin Picking in Prader-Willi Syndrome: Independent of Sensory Processing, Maladaptive Behaviors, and Genetic Subtype.

Skin picking, a prevalent and clinically significant issue, can lead to chronic wounds, infections, scarring, and a substantial burden on patients with Prader-Willi syndrome (PWS) and caregivers. However, the psychological and behavioral correlates of skin picking in PWS are poorly understood, and its relationship with sensory processing abnormalities and maladaptive behaviors has not been systematically examined. We conducted a retrospective cross-sectional study involving individuals with genetically confirmed PWS. Skin picking severity was assessed using the Skin Picking Scale (SPS). Sensor

Exploring the Causality Between Mood Swings and Neurological Diseases: A Mendelian Randomization Study.

BACKGROUND: Associations between genetically predicted mood swings and neurological diseases have been suggested. However, the causal relationships between these factors remain undefined. METHOD: We used a Mendelian randomization (MR) study to examine the causal relationship of genetically predicted mood swings with the risk of neurological diseases, including stroke, Alzheimer's disease (AD), multiple sclerosis (MS), epilepsy (EP), and headache. The single-nucleotide polymorphisms (SNPs) that exhibited genetic associations with mood swings were utilized as instrumental variables (IV) in the

The importance of dopamine levels and single-nucleotide polymorphism within COMT, DRD1 and DRD2 genes in obstructive sleep apnoea.

BACKGROUND: Obstructive sleep apnoea (OSA) is a prevalent sleep disorder that contributes to serious cardiovascular comorbidities. While the mechanical aspects of OSA are well-studied, its neurobiological underpinnings remain underexplored. In this study, we investigated the role of dopamine and its genetic modulators in OSA pathophysiology. PATIENTS AND METHODS: Serum dopamine levels were assessed in a cohort of 153 participants (96 OSA patients and 57 controls), and single-nucleotide polymorphisms (SNPs) in dopamine-related genes, including COMT, DRD1 and DRD2, were analysed in a cohort of

Bioinformatics and omics data management in genetic diagnosis.

Next Generation Sequencing (NGS) encompasses a range of technologies that have transformed genomic research since the 2000s. By allowing the sequencing of large DNA fragments at a significantly lower cost than Sanger sequencing, NGS has become an indispensable tool in molecular laboratories, particularly in the field of molecular genetics. Its high efficiency and speed make it a first-line technique in genetic analysis. A crucial step in achieving a diagnosis is bioinformatics analysis. Short-read sequencing technology generates raw data that must be processed to extract meaningful and interpr

Diagnostic and prognostic perspectives of Fabry disease via fiber evanescent wave spectroscopy advanced by machine learning.

Fabry disease (FD) is a rare disorder resulting from a genetic mutation characterized by the accumulation of sphingolipids in various cells throughout the human body, leading to progressive and irreversible organ damage, particularly in males. Genetically-determined deficiency or reduced activity of the enzyme (alpha - Galactosidase; α-Gal) leads to the accumulation of sphingolipids in the lysosomes of various cell types, including the heart, kidneys, skin, eyes, central nervous system, and digestive system, triggering damage, leading to the failure of vital organs, and resulting in progressiv

Patient-derived induced pluripotent stem cells to study non-canonical splicing variants associated with Hypertrophic Cardiomyopathy.

Hypertrophic cardiomyopathy (HCM) is the most prevalent inherited cardiomyopathy and a leading cause of sudden death. Genetic testing and familial cascade screening play a pivotal role in the clinical management of HCM patients. However, conventional genetic tests primarily focus on the detection of exonic and canonical splice site variation. Oversighting intronic non-canonical splicing variants potentially contributes to a proportion of HCM patients remaining genetically undiagnosed. Here, using a non-integrative reprogramming strategy, we generated induced pluripotent stem cell (iPSC) lines

Chronic eosinophilic leukaemia-Not otherwise specified: Clinical features, genomic insight and therapeutic strategies.

Chronic eosinophilia leukaemia-not otherwise specified (CEL-NOS) is a rare myeloproliferative neoplasm characterized by persistent clonal hypereosinophilia. Recent advances in genetics have refined diagnostic criteria, leading to the identification of CEL subtypes with specific cytogenetic and molecular abnormalities now classified as myeloid/lymphoid neoplasms with eosinophilia and tyrosine kinase gene fusions, which may benefit from targeted therapies. In contrast, CEL-NOS lacks specific genetic drivers and intervention points to halt leukemogenesis. Molecular techniques have also enabled th

GoldVariants, a resource for sharing rare genetic variants detected in bleeding, thrombotic, and platelet disorders: Communication from the ISTH SSC Subcommittee on Genomics in Thrombosis and Hemostasis.

The implementation of high-throughput sequencing (HTS) technologies in research and diagnostic laboratories has linked many new genes to rare bleeding, thrombotic, and platelet disorders (BTPD), and revealed multiple genetic variants linked to those disorders, many of them being of uncertain pathogenicity when considering the accepted evidence (variant consequence, frequency in control datasets, number of reported patients, prediction models, and functional assays). The sequencing effort has also resulted in resources for gathering disease-causing variants associated with specific genes, but f

A Focus Group Study of Perceptions of Genetic Risk Disclosure in Members of the Public in Sweden: "I'll Phone the Five Closest Ones, but What Happens to the Other Ten?".

This study explores perceptions and preferences on receiving genetic risk information about hereditary cancer risk in members of the Swedish public. We conducted qualitative content analysis of five focus group discussions with participants (n = 18) aged between 24 and 71 years, recruited from various social contexts. Two prominent phenomena surfaced around the interplay between the three stakeholders involved in risk disclosure: the individual, healthcare, and the relative at risk. First, there is a genuine will to share risk information that can benefit others, even if this is difficult and

Violence exposure in an urban city: A GxE interaction with aggressive and impulsive behaviors.

BACKGROUND: Previous research has demonstrated a reciprocal relationship between community violence exposure and disruptive behavior problems among youth. No study to date, however, has explored the potential interaction between violence exposure in early adolescence and genetics. METHODS: We explore the gene x environment interaction's impact on teacher-rated aggressive and impulsive behaviors. Violence exposure during the middle school years was assessed using self-report. Genetic data collection occurred in emerging adulthood. A polygenic score was created using findings from a conduct dis

The Emerging Role of Epigenetics in Autoimmune Thyroid Diseases.

Autoimmune thyroid diseases (AITD) are a group of both B cell- and T cell-mediated organ-specific autoimmune diseases. Graves' disease and Hashimoto thyroiditis are the two main clinical presentations of AITD. Both genetic and environmental factors have important roles in the development of AITD. Epigenetics have been considered to exert key roles in integrating those genetic and environmental factors, and epigenetic modifications caused by environmental factors may drive genetically susceptibility individuals to develop AITD. Recent studies on the epigenetics of AITD have provided some novel

Hypocretin (orexin) biology and the pathophysiology of narcolepsy with cataplexy.

The discovery of hypocretins (orexins) and their causal implication in narcolepsy is the most important advance in sleep research and sleep medicine since the discovery of rapid eye movement sleep. Narcolepsy with cataplexy is caused by hypocretin deficiency owing to destruction of most of the hypocretin-producing neurons in the hypothalamus. Ablation of hypocretin or hypocretin receptors also leads to narcolepsy phenotypes in animal models. Although the exact mechanism of hypocretin deficiency is unknown, evidence from the past 20 years strongly favours an immune-mediated or autoimmune attack

Regulation of aldosterone secretion: from physiology to disease.

Arterial hypertension is a major cardiovascular risk factor that affects between 10 and 40% of the population in industrialized countries. Primary aldosteronism (PA) is the most common form of secondary hypertension with an estimated prevalence of around 10% in referral centers and 4% in a primary care setting. Despite its high prevalence until recently, the underlying genetic and molecular basis of this common disease had remained largely obscure. Over the past decade, a number of insights have been achieved that have relied on in vitro cellular systems, wild-type and genetically modified in
