OBJECTIVE: Microbiological analysis might not be able to identify the causative pathogen in one-third of brain abscess patients. Proteomics analysis of brain abscesses can aid in microbiological diagnosis. This could help tailor the antimicrobial therapy, give possible insights into the source of the abscess, and aid in developing targeted antimicrobial agents.
METHODS: This was a single-center prospective study including patients with brain abscess recruited over a 1-year period. Proteome-based analysis was used to identify the causative pathogen, and the results were compared using conventi

Porcine circovirus type 2 (PCV2) is an important swine pathogen that causes significant economic losses to the pig industry. PCV2 interacts with host cellular factors to regulate its replication. High-mobility-group box 1 (HMGB1) protein, a major nonhistone protein in the nucleus, was recently discovered to participate in viral infections. Here, we demonstrate that nuclear HMGB1 negatively regulated PCV2 replication as shown by overexpression of HMGB1 or blockage of its nucleocytoplasmic translocation with ethyl pyruvate. The B box domain was essential in restricting PCV2 replication. Nuclear

Tick-borne pathogen infections are an increasing occurrence globally, yet many aspects of pathogen maintenance and host-tick interactions remain poorly understood. Here we consider the potential role of eastern cottontails (Sylvilagus floridanus) in the enzootic cycles of tick-borne pathogens of medical importance in Virginia. Over a 3-year period, ticks and blood were collected from rabbits acquired through passive surveillance in 21 counties in Virginia. Seven hundred seventy ticks were collected from 90 of the 121 rabbits examined in this study. Tick species collected from the rabbits inclu

The escalating crisis of antimicrobial resistance necessitates the exploration of new treatment strategies, particularly against priority pathogens such as Staphylococcus aureus (S. aureus). While interferon-beta (IFN-β) is known for its immunomodulatory effects, its potential direct antibacterial activity remains poorly understood. This study uncovers a new, non-immune mechanism by which IFN-β inhibits S. aureus by directly blocking thymidine kinase (TK), a key enzyme in the nucleotide salvage pathway. We first identified a strong, spontaneous interaction between IFN-β and TK through fluoresc

Several lines of evidence suggest that Acanthamoeba interact with bacteria, which may aid in pathogenic bacterial transmission to susceptible hosts, and these interactions may have influenced evolution of bacterial pathogenicity. In this study, we tested if Gram-negative Pseudomonas aeruginosa and Gram-positive Corynebacterium spp. can associate/invade and survive inside Acanthamoeba castellanii trophozoites and cysts, as well as non-phagocytic human brain microvascular endothelial cells. The results revealed that both Corynebacterium spp. and P. aeruginosa were able to associate as well as in

In this study, the human immune response mechanisms against Sporothrix brasiliensis and Sporothrix schenckii, two causative agents of human and animal sporotrichosis, were investigated. The interaction of S. brasiliensis and S. schenckii with human monocyte-derived macrophages (hMDMs) was shown to be dependent on the thermolabile serum complement protein C3, which facilitated the phagocytosis of Sporothrix yeast cells through opsonization. The peptidorhamnomannan (PRM) component of the cell walls of these two Sporothrix yeasts was found to be one of their surfaces exposed pathogen-associated m

Mammalian hosts interface intimately with commensal and pathogenic bacteria. It is increasingly clear that molecular interactions between the nervous system and microbes contribute to health and disease. Both commensal and pathogenic bacteria are capable of producing molecules that act on neurons and affect essential aspects of host physiology. Here we highlight several classes of physiologically important molecular interactions that occur between bacteria and the nervous system. First, clostridial neurotoxins block neurotransmission to or from neurons by targeting the SNARE complex, causing t

Pathogenic Gram-negative bacteria use specialized secretion systems that translocate bacterial proteins, termed effectors, directly into host cells where they interact with host proteins and biochemical processes for the benefit of the pathogen. lpg1496 is a previously uncharacterized effector of Legionella pneumophila, the causative agent of Legionnaires disease. Here, we crystallized three nucleotide binding domains from lpg1496. The C-terminal domain, which is conserved among the SidE family of effectors, is formed of two largely α-helical lobes with a nucleotide binding cleft. A structural

The host defense against emerging respiratory pathogens begins with the induction of cell-intrinsic, interferon-mediated antiviral responses. The interferon response induces innate effector and adaptive cellular responses crucial for viral clearance and the establishment of long-lasting immune memory. Although these antiviral processes are primarily characterized at the transcriptional level, the epigenetic mechanisms that orchestrate the cellular transcriptional output during infection remain understudied. Technological advances in systems immunology and virology have revealed dynamic changes

Oral dysbiosis, particularly through periodontal disease, links strongly to cardiovascular risks by driving chronic inflammation and microbial translocation. Key pathogens invade vascular tissues, triggering systemic cytokines and metabolites that damage endothelial function and promote atherosclerosis. This creates a vicious cycle where oral inflammation worsens heart disease progression. Clinical interventions like intensive periodontal therapy show promise, reducing blood pressure and inflammatory markers in at-risk patients, much like established lifestyle changes. Salivary microbial profi

Influenza A viruses (IAVs) represent a persistent threat to global public health because of their capacity to cross species barriers and rapidly adapt to new hosts. Posttranslational modifications (PTMs) are known to regulate viral protein function, yet the evolutionary dynamics of PTM sites across influenza strains remain largely uncharacterized. In this study, we employed MusiteDeep to computationally predict potential PTM sites across influenza A virus proteins from H1N1, H5N1, and H7N9. We modeled predicted PTM states as discrete evolutionary traits using Bayesian phylogenetic methods to e

Porcine reproductive and respiratory syndrome virus (PRRSV) continues to pose a significant threat to global swine production. In China, PRRSV type 2 (PRRSV-2) is the predominant strain, and the inadequate cross-protection provided by existing vaccines highlights the urgent need for novel antiviral agents capable of effectively controlling PRRSV-2 infections. The host membrane-associated RING-CH 8 protein (MARCH8) has been demonstrated to play diverse roles in viral replication, either facilitating or inhibiting various viruses. However, its specific function in PRRSV infection and its broader

Specific targeting of host genes by plant viruses leads to the induction of distinct disease symptoms with devastating consequences for agriculture. However, it is unknown whether plants have evolved genes to specifically inhibit the development of viral disease symptoms. Here we identified the causal gene in 2 Arabidopsis thaliana mutants that develop prominent systemic necrosis after infection with both wild-type cucumber mosaic virus (CMV) and its mutant (CMV-Δ2b), which is defective in the suppression of antiviral RNA interference. We found that antiviral RNAi 5 (AVI5) is located in a larg

Respiratory syncytial virus (RSV) remains a major cause of severe respiratory disease worldwide, particularly affecting young children and immunocompromised individuals, highlighting the need for additional therapeutic strategies. In this study, the antiviral activity of the Petasites hybridus extract Ze 339 was investigated in RSV-infected cell-culture models. Antiviral efficacy was assessed using plaque reduction assays, reporter virus analyses, and proteomic profiling to elucidate potential mechanisms of action. Ze 339 potently reduced the infectivity of both RSVA and RSVB in vitro and reta

African swine fever virus (ASFV) infection induces cellular stress that activates the unfolded protein response (UPR), a key pathway for restoring endoplasmic reticulum (ER) homeostasis. However, the mechanisms by which ASFV modulates the UPR remain incompletely understood. Here, we identify the ASFV protein MGF_110-8L as a key regulator of the UPR, leading to the dissociation and activation of the UPR sensors PERK, IRE1α, and ATF6, which subsequently restore ER homeostasis. Moreover, MGF_110-8L triggers UPR-dependent autophagy, which in turn contributes to the suppression of type I interferon

African swine fever (ASF) is a viral disease of domestic pigs and wild boars with a high fatality rate. The causative agent of this disease is the African swine fever virus (ASFV), which continues to represent a major threat to the global swine industry, for which therapeutic options remain scarce. Among ASFV's structural proteins, p54 plays a crucial role in host-virus interactions, although its involvement in host cell stress responses and apoptosis remains incompletely elucidated. In the present study, we examined the functional characteristics of the ASFV p54 protein and assessed a nucleic

African swine fever (ASF) is an acute hemorrhagic disease caused by African swine fever virus (ASFV), with mortality rates approaching 100% in domestic pigs. Because its genome is large and structurally complex, the functions of many ASFV encoded genes remain poorly understood. In this study, ASFV I9R was identified as a relatively conserved early transcribed gene. To investigate its biological role, an I9R deletion recombinant virus (ASFV-ΔI9R) was generated by replacing the I9R gene with an enhanced green fluorescent protein (EGFP) expression cassette. In primary porcine alveolar macrophages

The ubiquitin-proteasome system (UPS) plays a pivotal role in the precise regulation of innate immunity. Within the spatiotemporal framework of coronavirus (CoV) infection, the UPS emerges as a critical arena and a molecular switch, dynamically mediating the balance between host antiviral defense and viral replication alongside immune evasion. The host leverages the UPS to activate interferon (IFN) signaling pathways and to directly target viral proteins for degradation. Conversely, CoVs have developed sophisticated mechanisms to exploit this system, either by degrading antiviral proteins or b

BACKGROUND: Chronic Hepatitis B virus (HBV) infection affects more than 250 million individuals worldwide, causing up to 1 million deaths yearly. Despite the significant disease burden, no curative therapy against HBV is currently available. Accumulating evidence indicate a strong and complex relationship between HBV and the host-cell lipid metabolism. HBV hijacks host lipoprotein transport pathways to gain access to hepatocytes and uses cellular cholesterol and sphingolipids for morphogenesis, while viral replication induces diverse and profound alterations of the hepatic lipid metabolism dri

Rotavirus (RV) replication occurs within viroplasms (VMs) and is initiated by two RV non-structural proteins NSP2 and NSP5. Viruses exploit host cellular components for their replication and assembly; however, information on the roles of host proteins in VM dynamics and RV replication is limited. Thus, in the current study, we used proteomics to identify host proteins that interact with NSP5 during RV infection to delineate their role in virus replication. A large number of host proteins (n = 128) were found to interact with NSP5, and Gene Ontology enrichment analysis revealed the enrichment o
