Sexual maturation in female rhesus macaques was studied after surgical isolation of the medial basal hypothalamus [complete hypothalamic disconnection (CHD); n = 4] or after creation of amygdaloid lesions (AMYG; n = 6). In four animals, CHD at 8 months did not affect the age when menarche occurred (30 months) but did result in a significant (P less than 0.01) advancement in age at first ovulation (35.8 +/- 1.1 vs. 43.7 +/- 1.1 months for the five controls). We measured the production rates and metabolic clearance, disappearance and excretion rates of FSH and LH as well as plasma testosterone and delta 4-androstenedione in males with precocious and delayed puberty. In precocious puberty, we found modestly elevated FSH production early in puberty, reaching adult levels by midpuberty and remaining constant thereafter. Examination of 16 adult men operated in childhood for hypospadias revealed signs of moderate injury of the male genital system. It appears that accessory sexual glands were mainly responsible for the observed abnormalities. Since testicular position and size, serum testosterone and FSH levels were normal it seems that hypospadias and the accompanied disturbances are connected with reduction of responsiveness of target tissues to androgen stimuli rather, than with lesion of the testicle. Antepartum pituitary infarction occurs only in insulin-dependent diabetic patients. It is manifested by severe headache, followed by decreasing insulin requirements. Delivery is frequently premature, with high fetal wastage and increased maternal mortality. Reproductive and endocrine function was investigated in 22 women with Hodgkin's disease who had bilateral mid-line oophoropexies performed at staging laparotomy. The operation was followed in 12 cases by "inverted Y" pelvic lymph node irradiation and in 4 cases by para-aortic lymph node irradiation. Fourteen girls and one boy with isosexual precocious puberty were submitted to LHRH stimulation tests, during and without therapy with cyproterone acetate. In addition, fourteen girls with isosexual precocious puberty not receiving any therapy were tested and served as controls. It was found that cyproterone acetate induces suppression of the responsiveness of the pituitary gland to secrete LH on LHRH stimulation.
