Use of parasite antigen detection to monitor macrofilaricidal therapy in Brugia malayi-infected jirds.

Improved methods are needed to evaluate new treatments for filarial infections. We have recently developed a monoclonal antibody-based enzym … level antigenemia was detected in 2 of 6 animals at 20 wk, perhaps suggesting incomplete killing of parasites or incomplete clearance of antigen. Parasite antigen levels were stable throughout the study in control animals. These preliminary results suggest that parasite antigen detection is useful as a means of noninvasively monitoring the efficacy of anti- …

[Epizootiology and prevention of helminthiasis of game animals in Czechoslovakia and its relationship to the parasitic diseases of cattle].

Decades of surveying helminthic diseases in domestic and free-living ruminants in the CSR evidenced extensive infestation by lungworms, gastrointestinal nematodes, and liver flukes. In some years, parasitic invasion caused up to 50% of total losses of roe deer. Programmes to control systematically dictyocaulosis and fascioliasis were carried out with cattle and sheep in 1970-1980, with deer in 1981-1985. In the eighties, the incidence of F. hepatica in cattle and sheep dropped to less than 1%, of D. viviparus in cattle to 0.2%; no D. filaria is to be encountered in sheep presently. The share o

Prevalence and seasonal fluctuations of intestinal parasitic infections in the Nablus area, West Bank of Jordan.

A total of 22,970 stool specimens collected from patients attending the Central Medical Laboratory in the city of Nablus in the period of 1981-1986 were examined for intestinal parasites. Of these 7412 (32.3%) were positive. Entamoeba histolytica (22.9%), Giardia lamblia (7.3%), and Ascaris lumbricoides (5.7%) were the most prevalent intestinal parasites found. Other intestinal parasites present included Hymenolepis nana, Trichomonas hominis, Trichuris trichiura, Taenia saginata, Enterobius vermicularis and Strongyloides stercoralis. Seasonal occurrence of intestinal parasites in the West Bank…

Attempts to culture the parasitic stage of Dermatobia hominis (L. Jr.) in vitro (Diptera: Cuterebridae).

Dermatobia hominis larvae were cultured in a semidefined liquid medium. First-instar larvae (L1) grew well up to 44 days; 29.1% molted in a mean period of 8.62 days. Two larvae reached the third instar but lived only 1 and 18 days, respectively, after the second molt. The increase in size, measured in 4 larvae, was about 10-fold. Second- and third-instar larvae, obtained from the skin of cattle, survived and grew in the medium for up to 2 mo; 39.0% of the L2 molted while 77.3% of the L3 pupated, and some produced flies when transferred to sand after 14.84 +/- 10.08 days in the culture medium.

Identification and partial characterization of a parasite antigen in sera from humans infected with Wuchereria bancrofti.

The purpose of this study was to identify and characterize soluble parasite antigens present in sera from humans infected with the filarial nematode Wuchereria bancrofti. Affinity chromatography and immunoblot methods were used to demonstrate a 200,000 m.w. circulating parasite antigen in sera from infected humans which corresponded to an antigen released by adult W. bancrofti during in vitro culture. Two monoclonal antibodies were produced to this antigen by immunizing mice with antigens from Dirofilaria immitis, a filarial parasite that is closely related to W. bancrofti, and screening cell

Circulating parasite antigen(s) in lymphatic filariasis: use of monoclonal antibodies to phosphocholine for immunodiagnosis.

Hybridoma cell lines producing monoclonal antibodies (MAb) against a 200 kD antigen found circulating in the sera of microfilaremic patients infected with Wuchereria bancrofti were obtained by immunizing mice with a partially purified antigen preparation. A sensitive MAb (CA101)-based ELISA for measuring circulating parasite antigen was capable of detecting antigen in the sera of 93% of patients with microfilaremia, 46% of those with lymphatic obstruction, and 56% of patients with tropical pulmonary eosinophilia syndrome. Circulating antigen was absent from sera of normal controls, and "false

[Experimental filariasis in Proechimys oris by Dipetalonema dessetae. 5. Effect of parasitism on metabolism of diethylcarbamazine].

The metabolism of diethylcarbamazine (DEC) in healthy and filaria-infected Proechimys oris Rodents was investigated. DEC and 14C labelled DEC were orally administrated (100 mg/kg). The drug was quickly and intensively metabolised. About 90% of the dose administered was eliminated in the urine within 24 hours. Monodeethylation was the major pathway. The major urinary metabolites were ethylcarbamazine and its N-oxide. Traces of N methylpiperazine 2,5 dione and their N-oxide were also found. Elimination was faster in filarial than in healthy animals, but metabolites were the same.

[Toxoplasma gondii pneumopathy in a patient with the acquired immunodeficiency syndrome: demonstration of the parasite by bronchioloalveolar lavage].

Toxoplasmosis is an infection frequently observed in patients with acquired immunodeficiency syndrome. Its first manifestations usually are cerebromeningal symptoms, but others may occur. A pulmonary lesion is sometimes present; it is discovered at post-mortem examination in most cases. The authors report a case of pulmonary toxoplasmosis diagnosed by bronchoalveolar lavage (BAL). Three points are emphasized: toxoplasmosis of the lung may produce an interstitial pneumonia similar to that of pneumocystosis; diagnosis can be made non-invasively by BAL, and provided an appropriate treatment is ad

Enhanced mosquito blood-finding success on parasitemic hosts: evidence for vector-parasite mutualism.

The generalized hematopathology frequently found in animals infected with vector-borne pathogens may maximize transmission by enhancing the ability of vectors to locate blood in infected hosts. We tested this idea of mutualism between parasite and vector by comparing duration of probing of mosquitoes feeding on noninfected and on malaria-(Plasmodium chabaudi) or arbovirus-(Rift Valley fever virus) infected animals. We found that median duration of probing (blood location) on infected rodents was reduced by at least 1 min as compared to noninfected rodents. This enhanced ability of vectors to l

Leishmaniasis in Greece I. Isolation and identification of the parasite causing human and canine visceral leishmaniasis.

Three human and 19 canine leishmanial stocks were typed according to their excreted factor serotype and the electrophoretic mobility of their MDH, GPI, G6PDH and 6PGDH and shown to be identical with regard to these characters and, thus with Leishmania donovani infantum. This verifies the opinion of earlier researchers, who suggested that the parasites which cause human and canine visceral leishmaniasis in Greece are the same organism and that dogs are the reservoir for the human infection. The complexities raised by the co-existence of human cutaneous leishmaniasis in Greece caused by L. tropi…

Evidence for immunological cross-reaction between sporozoites and blood stages of a human malaria parasite.

Malaria parasites (Plasmodium spp.) show a complex pattern of development in the mammalian host and many studies support the view that the surface of the sporozoite, injected by the mosquito, has no antigens in common with the erythrocytic stage of development. For example, immunization with the erythrocytic parasites generates antisera with negligible titre by indirect immunofluorescence to the sporozoite surface. Although monoclonal antibodies prepared against erythrocytic stages were reported to show cross-reaction to the sporozoite stage, this appeared to be due to cytoplasmic antigens exp…

Cholinephosphotransferase and ethanolaminephosphotransferase activities in Plasmodium knowlesi-infected erythrocytes. Their use as parasite-specific markers.

CDPcholine: 1,2-diacylglycerol cholinephosphotransferase (EC 2.7.8.2) and CDPethanolamine: 1,2-diacylglycerol ethanolaminephosphotransferase (EC 2.7.8.1) activities were investigated in Plasmodium knowlesi-infected erythrocytes obtained from Macaca fascicularis monkeys. Disrupted infected erythrocytes possess a cholinephosphotransferase activity (1.3 +/- 0.2 nmol phosphatidylcholine/10(7) infected cells per h) 1.5-times higher than the ethanolaminephosphotransferase activity. Optimal activities of both enzymes were observed in the presence of 12 mM MnCl2, which was about 3-times as effective a

IgE production in vitro by peripheral blood mononuclear cells of patients with parasitic helminth infections.

Helminth parasites induce production of high levels of IgE antibodies but the immunoregulatory mechanisms determining this IgE biosynthesis are poorly understood. To investigate these mechanisms, peripheral blood mononuclear cells were obtained from six normal controls, six atopic patients and eight patients with parasitic helminth infections (three with schistosomiasis, two with loiasis, three with onchocerciasis). Cells were cultured at 1 X 10(6) cells/ml for 8 days in the presence of media alone or media supplemented with pokeweed mitogen (PWM) or cycloheximide; the supernatant fluids from …
