Expression of ATM in ataxia telangiectasia fibroblasts rescues defects in DNA double-strand break repair in nuclear extracts.

Ataxia telangiectasia (A-T) is a human genetic disorder characterized by progressive cerebellar degeneration, hypersensitivity to ionizing radiation (IR), immunodeficiency, and high cancer risk. At the cellular level, IR sensitivity and increased frequency of spontaneous and IR-induced chromosomal breakage and rearrangements are the hallmarks of A-T. The ATM gene, mutated in this syndrome, has been cloned and codes for a protein sharing homology with DNA-PKcs, a protein kinase involved in DNA double-strand break (DSB) repair and DNA damage responses. The characteristics of the A-T cellular phe

p53 stabilization and functional impairment in the absence of genetic mutation or the alteration of the p14(ARF)-MDM2 loop in ex vivo and cultured adult T-cell leukemia/lymphoma cells.

Human T-cell lymphotropic virus type I (HTLV-I) transforms T cells in vitro, and the viral transactivator Tax functionally impairs the tumor suppressor p53 protein, which is also stabilized in HTLV-I-infected T cells. Thus, the functional impairment of p53 is essential to maintain the viral-induced proliferation of CD4+ mature T cells. However, in the CD4+ leukemic cells of patients with adult T-cell leukemia/lymphoma (ATLL), the viral transactivator does not appear to be expressed, and p53 mutations have been found only in a fraction of patients. We sought to investigate whether p53 function

Neuroimaging in bipolar disorder: what have we learned?

New technologies are offering increasingly powerful means to obtain structural, chemical, and functional images of the brain during life, often without the use of ionizing radiation. Bipolar disorder, with its clear physiologic features, would appear to be a prime candidate for the application of current brain imaging; however, only a modest number of studies have been reported to date, and most studies have small sample sizes and heterogeneous subject groups. Nonetheless, there are a few consistent findings among these studies, including the following: 1) Structural imaging studies suggest an

[Cancers in the scars of thermal burns (apropos of 67 cases)].

Burns are very frequent. Skin cancer on burns scars are one of the known complications. The mechanisms and the risk factors of this disease are not very well known. To review the risk factors and the mechanisms of transformation of burn scars into cancer, we analyzed 67 retrospective cases of Marjolin's ulcer to describe the epidemiological features of the disease in our practice and identify the factors of relapse. Our patients are young (means age 41), mainly male (54%), with disease localized on arms and legs (88%). The initial burn was from flames (54%), charcoal or hot cooking oil (19.5%)

Efficacy of sonography as a guidance technique for biopsy of abdominal, pelvic, and retroperitoneal lymph nodes.

OBJECTIVE: In the United States, CT is generally considered the guidance technique of choice for biopsy of abdominal, pelvic, and retroperitoneal lymph nodes. The aim of this study was to evaluate the efficacy of sonography for this purpose. SUBJECTS AND METHODS: We attempted 26 sonographically guided biopsies of five abdominal, six pelvic, and 12 retroperitoneal lymph nodes in 23 patients. The patients included 19 women and four men who were 26-76 years old (mean, 50 years old). The nodes were less than 2 cm in 16 patients, 2-3 cm in four patients, and greater than 3 cm in three patients. Bi

Relationships between G1 arrest and stability of the p53 and p21Cip1/Waf1 proteins following gamma-irradiation of human lymphoma cells.

We investigated temporal relationships between ionizing radiation-induced G1 arrest and induction of the p53-regulated genes GADD45, CIP1/WAF1, and MDM2 in a series of Burkitt's lymphoma and lymphoblastoid cell lines that differed in p53 gene status. Emphasis was placed on characterization of the EW36 cell line, which despite expressing wild-type p53 genes, is defective in G1 arrest following gamma-irradiation (P. M. O'Connor et al., Cancer Res., 53: 4776-4780, 1993). Induction of CIP1/WAF1, GADD45, and to a lesser extent MDM2 mRNA was observed in all wild-type p53 lines that arrested in G1. C

Oxygen radical detoxification processes during aging: the functional importance of melatonin.

That free radical destruction of macromolecules is a basis of aging and age-related diseases has considerable experimental support. Melatonin, a hormone produced in organisms as diverse as algae and humans, is believed to have evolved coincident with aerobic metabolism. In all organisms melatonin is produced primarily during the daily period of darkness, with only small amounts being synthesized during the day. In mammals including man, melatonin is produced by and secreted from the pineal gland during the night; however, the night-time production of melatonin falls markedly with aging such th

Methods and concepts in detecting abnormal reproductive outcomes of paternal origin.

There is conclusive evidence that exposures of human males to ionizing radiation or certain chemicals can diminish sperm production and reduce fertility. Of approximately 100 chemical agents and mixtures that have been evaluated in men by semen analysis, about half (mostly drugs and a few occupational exposures) reduced sperm quantity and quality; several of these agents also affected the fertility of exposed men. It is now well recognized that the importance of the father in reproduction goes beyond fertilization. Abnormalities in paternal chromosomes (structural and numeric) have been found

A review of forty-five years study of Hiroshima and Nagasaki atomic bomb survivors. Brain damage among the prenatally exposed.

Significant effects on the developing brain of exposure to ionizing radiation are seen among those individuals exposed in the 8th through the 25th week after fertilization. These effects, particularly in the most sensitive period, 8-15 weeks after fertilization, manifest themselves as an increased frequency of severe mental retardation (SMR), a diminution in IQ score and in school performance, and an increase in the occurrence of seizures. Of 30 SMR cases, 18 (60%) had small heads. About 10% of the individuals with small head sizes observed among the in utero clinical sample were mentally reta

Cytogenetics of secondary myelodysplasia (sMDS) and acute nonlymphocytic leukemia (sANLL).

76 cases of secondary myelodysplasia (sMDS) and acute non-lymphocytic leukemia (sANLL) were cytogenetically analyzed. Among the 36 sMDS patients, 13 (36%) had only normal karyotypes whereas 23 (64%) displayed clonal chromosomal abnormalities. The most common aberrations were -7, 5q-, -5, and +8. In 10 patients (43% of the cytogenetically aberrant cases), clones with only one anomaly, mostly 5q- or -7, were found. Of the 40 sANLL patients, normal karyotypes were detected in 10 (25%). Among the 30 (75%) abnormal cases, the most frequent aberrations were -7, -5, +8, 7q-, -17, and +21. 12 patients

Results of a North American survey of rapid-sequence MRI utilization to evaluate cerebral ventricles in children.

OBJECT: Growing concern about potential adverse effects of ionizing radiation exposure during imaging studies is particularly relevant to the pediatric population. To decrease radiation exposure, many institutions use rapid-sequence (or quick-brain) MRI to evaluate cerebral ventricle size. There are obstacles, however, to widespread implementation of this imaging modality. The purpose of this study was to define and quantify these obstacles to positively affect institutional and governmental policy. METHODS: A 9-question survey was emailed to pediatric neurosurgeons who were either members or

Mammary gland neoplasia in long-term rodent studies.

Breast cancer, the most frequent spontaneous malignancy diagnosed in women in the western world, is continuously increasing in incidence in industrialized nations. Although breast cancer develops in women as the result of a combination of external and endogenous factors such as exposure to ionizing radiation, diet, socioeconomic status, and endocrinologic, familial, or genetic factors, no specific etiologic agent(s) or the mechanisms responsible of the disease has been identified as yet. Thus, experimental models that exhibit the same complex interactions are needed for testing various mechani

Total body irradiation across conditioning intensities: historical context, dose-fractionation strategy, and the role of the chemotherapy backbone.

Total body irradiation (TBI) is a form of radiotherapy that provides a uniform dose of ionizing radiation to the entire body. It has been a mainstay of the conditioning regimen for allogeneic hematopoietic stem cell transplantation (allo-HSCT) for many decades. The clinical usefulness of TBI is based on the balance of its cytotoxic and immunosuppressive effects, which allow for the eradication of remaining malignant cells, including sanctuary sites, as well as the establishment of a successful transplant. Early "supra-lethal" single-dose schedules of TBI were highly effective but also came wit

A framework for assessing the safety of degraded pharmaceuticals for long-duration space missions - Part 2: framework application in four case studies.

Spaceflight environmental conditions can alter the rate of degradation for some drug products; however, only a limited number of drugs have been tested after prolonged exposure to spaceflight. Existing ground-based drug stability studies do not include key factors associated with spaceflight, such as levels of carbon dioxide (CO2) and ionizing radiation that are significantly higher than on Earth. The National Aeronautics and Space Administration (NASA) has recognized the use of expired or degraded drugs as a risk for space crews because the loss of active pharmaceutical ingredient (API) decre

Experimental models of chemical carcinogenesis: bridging toxicology and preclinical pharmacology.

Those experimental animals that tend to develop primary malignancies as a result of direct exposures to synthetic cancer agents can very well represent early stages of human cancers as compared to transplanted as well as genetically engineered tumor models. Generally, malignant growth resulting due to cancer agents tends to undergo three phases: initiation, promotion, as well as progression. On the basis of genetic alteration, agents can be divided into genotoxgents as well as non-genotoxgents. Carcinogens that tend to be complete, incomplete, as well as specified towards a target organ are cl

Defining and validating a high specificity MRI cut-off for the radiographic part of the modified New York criteria in patients with axial spondyloarthritis.

BACKGROUND: According to the modified New York criteria (mNYc), moderate-severe sacroiliac joint (SIJ) structural damage on radiography is necessary for classifying radiographic axial spondyloarthritis (r-axSpA). In contrast to radiography, MRI provides no ionizing radiation, higher sensitivity for structural damage, better inter-reader reliability, and is gradually replacing radiography. Therefore, we aimed to develop and validate a high specificity MRI cut-off to the radiographic component of the mNYc (rad-mNYc), defined as MRI findings that correspond to an mNY-positive radiograph. A selective BRG1 inhibitor targeting bromodomain sensitizes hepatocellular carcinoma to chemoradiotherapy by disrupting the DNA damage response.

BACKGROUND: The poor response to ionizing radiation and chemotherapies remains a significant clinical challenge for unresectable hepatocellular carcinoma (HCC). BRG1, the core ATPase subunit of switch/sucrose non-fermentable (SWI/SNF) complex, is often overexpressed in HCC and represents a potential therapeutic target. Mechanistically, BRG1 facilitates DNA damage repair by assisting the assembly of DNA damage response (DDR) complex via its bromodomain (BRD), which selectively binds to acetylated histone H3. Therefore, pharmacological targeting of the BRG1-BRD represents an innovative strategy
