In vivo and in vitro studies with agents that cause quasi-morphine withdrawal syndromes.

Compounds that elicit a "quasi-morphine withdrawal syndrome" (QMWS) after acute administration to rats constitute a new class of behavior-modifying agent. In studying the pharmacological bases of quasi-opiate withdrawal syndromes, we found that the dihydrocodeinone, RX 336-M, a standard QMWS-inducing agent, caused a naloxone-insensitive increase in twitch tension on the field-stimulated rat was deferens preparation. In contrast, IBMX (3-isobutyl-1-methylxanthine), the historical inducer of quasi-abstinence in rats, gave a naloxone-insensitive decrease in twitch tension. Tolerance developed to

On the interaction of diazepam with human, rat and mouse plasma proteins and erythrocytes.

Binding of diazepam to the blood fractions (erythrocytes and plasma proteins) in man, rat and mouse was studied. Only little dependence of binding on total drug concentration was found. The main binding fraction of plasma in studied species is albumin, but interspecies differences are both in the amount of diazepam bound to albumin and in that bound to other components of plasma. The determination of the binding of diazepam with erythrocytic mass and with blood plasma demonstrates the proportion of these bonds in total distribution of the drug under study in blood and the importance of thus ex

A study on the in vivo production of thrombosis in rat mesenteric arterioles and action of prostaglandin (PG) I2 on the thrombosis.

We have investigated reproducible and quantitative thrombus formation in rat mesenteric arterioles using electrical stimulation under an inverted microscope. In vivo thrombi were formed in arterioles of 30-40 micrometers diameter by electrical stimulation using a tungsten electrode with a tip of 5 micrometers diameter. A "threshold" response was defined as one in which platelets adhered to the injured site of vascular wall and formed a white body which flowed away intermittently. The thrombus formations were classified into three stages: stage I, thrombi covering up less than 20% of the vessel

Cardiovascular and respiratory effects of beta-endorphin in anesthetized and conscious rats.

This study deals with the effects of beta-endorphin on blood pressure, heart rate, and respiratory frequency in anesthetized and conscious rats after both peripheral and central administration. Intravenous injection of beta-endorphin (40 and 160 micrograms/kg) in urethane-anesthetized rats resulted in a prolonged decrease in blood pressure, which, after the higher dose, was accompanied by bradycardia. In the pentobarbitone-anesthetized animals the same doses of beta-endorphin caused a small rise in blood pressure without affecting heart rate. No effect on respiratory frequency was observed. In

Correlation of the level of beta-citryl-L-glutamic acid with spermatogenesis in rat testes.

beta-Citryl-L-glutamic acid, which is known to be highly concentrated in the brains of immature animals, is preferentially localized in the testes of various adult animals, including mammals, amphibians and fish, mainly in the germinal cells. In young rats, the citrylglutamate concentration increases with age and coincides with the development of late spermatocytes into early spermatids. Rats with seminiferous tubule failure induced by ductuli efferentes ligation and experimental cryptorchidism are infertile as a result of germ cell depletion, especially spermatocytes and early spermatids. In

Comparison of enhancement of GGTase-positive foci and induction of ornithine decarboxylase in rat liver by barbiturates.

The indication of ornithine decarboxylase (ODC) by barbiturates and the ability of barbiturates to enhance neoplastic progression of chemically initiated cancer was examined in rat liver. All seven barbiturates induced ODC with barbital (7.7 fold increase) and phenobarbital (5.7 fold increase) demonstrating the most potent activity. Maximum induction of ODC by phenobarbital was obtained in 18 h. Barbital (500-5000 p.p.m.) and phenobarbital (500 p.p.m.) administered in the drinking water enhanced the appearance of diethylnitrosamine (DENA)-initiated gamma- glutamyltranspeptidase (GGTase)-positi

Isolation and characterization of the sulfated gangliotriaosylceramide from rat kidney.

A sulfoglycosphingolipid containing N-acetylgalactosamine has been isolated from the lipid extract of rat kidney. The isolation procedure involved extraction of lipids with chloroform/methanol, alkaline methanolysis, and column chromatographies with DEAE-Sephadex and Silica beads. The components were N-acetylgalactosamine, galactose, glucose, sphingoid bases, fatty acids, and sulfate in equimolar amounts. The yield of this sulfoglycolipid was about 24 nmol/g of tissue, which was about 13% of that of galactosylceramide 3-sulfate from rat kidney. By infrared spectroscopy, proton magnetic resonan

Molecular interaction of acrylonitrile and potassium cyanide with rat blood.

The interaction of acrylonitrile (VCN) with rat blood has been investigated at the molecular level in an attempt to understand the possible mechanism of its toxicity. The results obtained were compared to those with potassium cyanide (KCN), a compound known to liberate cyanide (CN-) in biologic conditions. The radioactivity derived from K14CN was eliminated faster than that from [1-14C]VCN. Up to a maximum of 94% of 14C from VCN in erythrocytes was detected covalently bound to cytoplasmic and membrane proteins, whereas 90% of the radioactivity from KCN in erythrocytes was found in the heme fra

Studies on the cyanamide-ethanol interaction. Dimethylcyanamide as an inhibitor of aldehyde dehydrogenase in vivo.

Administration of dimethylcyanamide (DMC) to rats caused a marked elevation in ethanol-derived blood acetaldehyde (AcH) and depressed the specific activity of the low Km mitochondrial aldehyde dehydrogenase (AIDH) by 90% at 12-24 hr, coincident with depletion of hepatic glutathione levels. Comparison of the relative efficacy of DMC and cyanamide in elevating blood AcH measured at 2 hr and 1 hr post-drug treatment, respectively, indicated that DMC was at least one-fifth as active as cyanamide. However, since the comparison was not made at optimal times for DMC (12-24 hr), it is likely that its

Cyanide-insensitive and clofibrate enhanced beta-oxidation of dodecanedioic acid in rat liver. An indication of peroxisomal beta-oxidation of N-dicarboxylic acids.

The beta-oxidation rate of dodecanedioic acid in rat liver homogenates (600 X g supernatant fraction) was determined by simultaneous measurements of the C6-C12-dicarboxylic acids, i.e., adipic, suberic, sebacic and dodecanedioic acids, in relation to time in assays incubated with dodecanedioic acid. Measurements were performed by a combined gas chromatographic-mass spectrometric technique, i.e., selected ion-monitoring. The beta-oxidation rate was registered as the consumption rate of dodecanedioic acid and as the initial rise in the concentrations of C6-C10-dicarboxylic acids. The beta-oxidat

Implantation and later fetal development in immature rats given a superovulatory dose of pregnant mare's serum gonadotropin, later neutralized by antiserum.

In an earlier experiment, 29-day-old female rats were superovulated with 40 IU pregnant mare's serum gonadotropin (PMSG) and very few blastocysts were recovered from the uterus on Day 5. Administration of a PMSG antiserum (a/s) prior to ovulation resulted in recovery of blastocysts in all rats and the present set of experiments was undertaken to investigate the later development of these blastocysts. Implantation was found to occur in only approximately 50% of superovulated (SOV) a/s-treated animals regardless of whether or not estrogen was given on Day 4. It is suggested that the exposure to

Modification of two models of Arthus reaction in the rat by various drugs.

The action of several steroidal and non-steroidal antiinflammatory agents, immunosuppressives, and antirheumatics (levamisole, chloroquine, sodium aurothiopropanol sulphonate, D-penicillamine) was studied in two models of Arthus-passive reaction in the rat: paw oedema induced by an anti-ovalbumin serum, and pleurisy induced by an anti-bovine-albumin serum. The steroidal antiinflammatory agents reduced both types of reaction. In pleurisy, they acted on exudate and the number of neutrophils. The non-steroidal compounds were not active on the inflammatory reaction of the paw, but they decreased t

Changes in the levels of prostaglandins and thromboxane and their roles in the accumulation of exudate in rat carrageenin-induced pleurisy--a profile analysis using gas chromatography-mass spectrometry.

Injection of lambda-carrageenin into the pleural cavity of rats caused the accumulation of the pleural exudate. When levels of prostaglandins (PGs) and thromboxane (TX) B2 were quantified by gas chromatography-mass spectrometry as their methyl ester (ME)-dimethylisopropylsilyl (DMiPS) ether or ME-methoxime-DMiPS ether derivatives, 6-keto-PGF1 alpha reached the maximum at 1 hr after carrageenin, then PGE2 and TXB2 showed peaks at 3 hr and waned off before 9 hr. The PGF/ alpha level was kept low, but PGD2, PGE1 and PGF1 alpha were not detected. Aspirin (100 mg/kg, i.p.) significantly decreased t

Chromatographic resolution of amino acid adducts of aliphatic halides.

Numerous xenobiotics are known to be bioactivated and to covalently and to proteins, but the resulting amino acid adducts (AAAs) are unknown. In this study the AAAs of twelve 14C-labeled aliphatic halides were examined after formation in an in vitro microsomal system. After exhaustive solvent extraction of the precipitated microsomal protein, the AAAs were isolated by Pronase digestion, followed by filtration through a 500 mol. wt. exclusion membrane. The liberated AAAs were applied to a constant flow DC-4A cation exchange column, resolved by stepwise buffer elution, collected and counted for

Effect of dose level and pregnancy on the distribution and toxicity of intravenous lead in rats.

Female Wistar rats were injected intravenously with tracer levels of 210Pb, alone or combined with carrier Pb(NO3)2 at 5 or 25 mg/kg body weight at 9 or 15 days of gestation (dg). Tissue 210Pb distribution and retention, and lead excretion, were measured several times during the first 30 h and at 20 dg. Toxic effects following the administration of 25 mg/kg (a teratogenic dose) included an early decrease in hematocrit, hematuria, gastrointestinal hemorrhage, and diarrhea, as well as an eventual loss of body weight and an increase in spleen and kidney weights. The stage of pregnancy at injectio

Normal sexual behavior and penile reflexes in long-term diabetic male rats.

Male rats made diabetic by streptozotocin injection were tested intermittently for copulation and penile reflexes. Eight to nine months later their bodies, penises, and seminal vesicles weighed significantly less than those of control-injected males, although serum androgen as well as penile and vas deferens norepinephrine levels were not significantly depressed. None of the measures of sexual motivation, performance, or penile responsivity was depressed in the diabetic rats, suggesting that rats are not a fruitful animal model for diabetic impotence.

The anorectic action of naloxone is attenuated by adaptation to a food-deprivation schedule.

Recent studies have shown that naloxone and other opiate antagonists can reduce the amounts of food and water consumed by laboratory animals, a finding consistent with a role for endogenous opioids in the control of appetite. Because there have also been some failures to observe an anorectic action of naloxone, a study was carried out in which the effects of the drug on food intake were investigated using two different experimental procedures. In naive rats deprived of food for 24 h, both naloxone (0.1, 1.0 and 10.0 mg/kg) and fenfluramine (1.0, 3.0 and 10.0 mg/kg) produced dose-related decrea

Modulation of [3H]-dopamine binding by cholecystokinin octapeptide (CCK-8).

Cholecystokinin-octapeptide (CCK-8) is a putative neurotransmitter which has been demonstrated previously to occur in midbrain dopamine neurones. We observe that CCK-8 causes changes in both the affinity and density of binding sites for [3H]-dopamine in rat striatal homogenates, in vitro, upon incubation with the peptide at a concentration of 1 micromolar. A dose-response study of the competetion of CCK-8 with [3H]-dopamine binding indicates an IC50 for the peptide of 450 nM; desulfated CCK-8 and the related peptide caerulin are at least 4-fold less active than CCK-8. CCK-8 was also administer

Effects of phenobarbital and sodium salicylate on cytochrome P-450 mixed function oxygenase and glutathione S-transferase activities in rat brain.

The effects of acute and therapeutic doses of phenobarbital and sodium salicylate on cytochrome P-450 mixed function oxygenase (EC 1.14.14.1) and glutathione S-transferase (EC 2.5.1.18) activities have been studied in rat brain and compared with those of rat liver. P-450 enzymic activity was assayed by N-demethylation of p-chloro-N-methylaniline and 1-chloro-2,4-dinitrobenzene was used as substrate for glutathione S-transferase activity. The acute effects of a single daily dose of phenobarbital (75 mg/kg/day; i.p.) and sodium salicylate (500 mg/kg/day; i.p.) for 3 days increased cytochrome P-4
