K-bZIP Mediated SUMO-2/3 Specific Modification on the KSHV Genome Negatively Regulates Lytic Gene Expression and Viral Reactivation.

SUMOylation is associated with epigenetic regulation of chromatin structure and transcription. Epigenetic modifications of herpesviral genomes accompany the transcriptional switch of latent and lytic genes during the virus life cycle. Here, we report a genome-wide comparison of SUMO paralog modification on the KSHV genome. Using chromatin immunoprecipitation in conjunction with high-throughput sequencing, our study revealed highly distinct landscape changes of SUMO paralog genomic modifications associated with KSHV reactivation. A rapid and widespread deposition of SUMO-2/3, compared with SUMO

Glc7-Reg1 phosphatase signals to Yck1,2 casein kinase 1 to regulate transport activity and glucose-induced inactivation of Saccharomyces maltose permease.

The Saccharomyces casein kinase 1 isoforms encoded by the essential gene pair YCK1 and YCK2 control cell growth and morphogenesis and are linked to the endocytosis of several membrane proteins. Here we define roles for the Yck1,2 kinases in Mal61p maltose permease activation and trafficking, using a yck1delta yck2-2(ts) (yck(ts)) strain with conditional Yck activity. Moreover, we provide evidence that Glc7-Reg1 phosphatase acts as an upstream activator of Yck1,2 kinases in a novel signaling pathway that modulates kinase activity in response to carbon source availability. The yck(ts) strain exh

Understanding protein ISGylation, a multifaceted posttranslational modification.

ISG15 represents a key ubiquitin-like modifier induced primarily by interferon signaling. ISG15 is synthesized as a precursor, processed to a mature form, and covalently conjugated to substrates through a dedicated E1-E2-E3 enzymatic cascade involving UBE1L, UBE2L6, and E3 ligases such as HERC5, TRIM25, and ARIH1. This modification is reversed by deISGylases, particularly the highly specific protease USP18, which also negatively regulates interferon signaling. ISGylation impacts diverse molecular and cellular processes, including protein stability and function, protein-protein interaction, aut

Molecular mechanisms in rare proteasomopathies.

Proteasomopathies comprise rare interferonopathy-related syndromes caused by genetic defects in proteasomal subunits or their assembly factors resulting in failed proteasome biogenesis and/or function. The concomitant proteasome impairment leads to imbalanced protein homeostasis by dysfunctional ubiquitin-mediated protein degradation. Two distinct clinical phenotypes have been characterised in proteasomopathies so far: (i) proteasome-associated autoinflammatory syndromes and (ii) proteasome-associated neurodevelopmental disorders. Despite these differences, both syndromes show molecular simila

Translophagy-A potential link between autophagy impairment and translational errors.

Maintenance of protein homeostasis requires coordination between protein synthesis and degradation, yet whether autophagy directly controls translational machinery remains unclear. Here we explored whether autophagy maintains translational fidelity. Proteomic analysis of ubiquitin-enriched fractions and p62-associated proteins in autophagy-deficient tissue confirmed previous studies identifying ribosomal proteins and RNA-binding factors among candidate autophagy cargo. Pharmacological or genetic impairment of autophagy increased translational errors, particularly during oxidative stress. Compo

Identification of Novel Amino Acid-based PROTACs Exhibiting Broad-Spectrum Antiviral Activity Against Enteroviruses.

Enteroviruses threaten children's health, yet no antiviral drugs are available. We developed a series of AATacs by conjugating acylthiourea with basic or hydrophobic amino acids. Unlike PROTACs, AATacs hijack host endogenous E3 ligases to exert their function. The representative compound N-6 targets the EV-A71 3D polymerase for degradation via the ubiquitin-proteasome and autophagy-lysosomal pathways, thereby blocking viral replication. N-6 exhibits broad-spectrum activity against multiple enteroviruses with nanomolar to picomolar potency. In vivo, N-6 protected 60% of the infected mice from m

Covalent warheads as a versatile platform for molecular glue development.

Targeted protein degradation (TPD) has become a novel therapeutic modality for diseases in drug discovery. Most progress on TPD has focused on the ubiquitin proteasome system; however, only a handful of >600 human genome-encoded E3 ligases have been successfully applied. Expansion of the E3 ligase toolbox with novel E3 ligases and their ligands will broaden the potential applications of TPD. Besides that, covalent molecular glue degraders (MGDs) provide a promising direction for TPD. Covalent MGDs may offer several advantages, including increased selectivity, broad applicability to traditional

UBE2C-Mediated CD147-CTLA-4 Axis Promotes T Cell Exhaustion and Immunosuppressive Microenvironment in Bladder Cancer.

Despite immune checkpoint inhibitors (ICIs) having benefited bladder cancer (BCa) patients, their limited response rates urge elucidation of intrinsic resistance mechanisms. In this study, we demonstrated that ubiquitin conjugating enzyme E2C (UBE2C) promotes the proliferation, invasion, and migration of BCa and inhibits DNA damage, accompanied by a decrease in T cell abundance. In syngeneic C57BL/6 BCa models and BCa-CD8+ T-cell cocultures, UBE2C silencing reduced CD147 and CTLA-4 abundance, restored IFN-γ, TNF-α, and TGF-β secretion, expanded CD3+CD8+ subsets, and attenuated tumor growth, in

Proteolysis Targeting Chimeric-Based Technology in Myeloma and Lymphoma.

Proteolysis-targeting chimeras (PROTAC) leverage the ubiquitin-proteasome system to selectively degrade oncogenic proteins, including those previously seen as undruggable. Recent preclinical studies indicate that PROTACs may represent a novel therapeutic strategy in lymphoma and myeloma. Indeed, preclinically, PROTACs have shown high efficacy and remarkable selectivity, a favorable safety profile, and lower toxicity compared with conventional therapies. Their catalytic, reusable mechanism enables drug dosing and offers the perspective of long-term low-dose treatment. PROTACs have demonstrated

DNAJB9 promotes epithelial-mesenchymal transition in the nasal epithelium of chronic rhinosinusitis by enhancing TRIM22-mediated IκBα degradation.

This study investigates the functional role of DNAJB9 in the pathogenesis of chronic rhinosinusitis with nasal polyps (CRSwNP). Using quantitative proteomics alongside human tissue analysis and murine models, we found that DNAJB9 is significantly upregulated in CRSwNP nasal epithelium and closely mirrors epithelial-mesenchymal transition (EMT) progression. Functional assays in human epithelial cells demonstrated that DNAJB9 actively drives the EMT phenotype. Mechanistically, DNAJB9 interacts with and stabilizes the E3 ubiquitin ligase TRIM22 by inhibiting its autoubiquitination. This stabilize…

Otud6a Knockout Leads to Male Subfertility in Mice.

Ovarian tumor ubiquitinating 6A (OTUD6A) is a deubiquitinating enzyme whose aberrant expression has been linked to various diseases, including inflammation and prostate cancer. Research indicates that deubiquitinating enzymes (DUBs) play a significant role in spermatogenesis in mice. However, the role of OTUD6A in spermatogenesis remains unclear. To investigate the function of OTUD6A in mouse spermatogenesis, we generated Otud6a-knockout mice using the CRISPR/Cas9 system. Our results showed that OTUD6A is predominantly expressed in the testis and localized to the cytoplasm of spermatogonia and

Hypoxia-induced conversion of sensory Schwann cells into repair cells is regulated by HDAC8.

After a peripheral nerve injury, Schwann cells (SCs), the myelinating glia of the peripheral nervous system, convert into repair cells that foster axonal regrowth, and then remyelinate or re-ensheath regenerated axons, thereby ensuring functional recovery. The efficiency of this mechanism depends however on the time needed for axons to regrow. Here, we show that ablation of histone deacetylase 8 (HDAC8) in SCs accelerates the regrowth of sensory axons and sensory function recovery. We found that HDAC8 is specifically expressed in sensory SCs and regulates the E3 ubiquitin ligase TRAF7, which d

Autosomal recessive hypertrophic cardiomyopathy associated with variants in TRIM63.

BACKGROUND: Hypertrophic cardiomyopathy (HCM) is typically linked to dominant variants in sarcomeric genes, but rare minor genes, including TRIM63 coding for an E3 ubiquitin-protein ligase, have recently emerged as potential causes of recessive HCM. METHODS AND RESULTS: Among 517 adult patients with clinical HCM who underwent next-generation sequencing, we found six index cases carrying biallelic TRIM63 variants-four homozygous and two compound heterozygous. They presented with early-onset disease, marked concentric hypertrophy, diffuse myocardial fibrosis, and progressive left ventricular dy

Targeting MDM2 affects spastin protein levels and functions: implications for HSP treatment.

Spastin is a microtubule (MT) severing enzyme that regulates several cell functions associated with MT dynamics. A reduction in spastin protein levels is responsible for approximately 40% of cases of Hereditary Spastic Paraplegia (HSP), a neurodegenerative disease. Currently, there is no cure for HSP but strategies to induce a recovery of spastin levels are emerging as potential therapeutic approaches. Here, we show that MDM2 interacts with spastin MT-interacting and trafficking (MIT) domain. By biochemical and functional experiments, we demonstrate that MDM2 binds spastin and regulates its le

Is the SPLUNC1-Orai1 axis a critical determinant of lung health?

Short palate lung and nasal epithelial clone 1 (SPLUNC1; gene name BPIFA1) is a secreted protein that is highly expressed in the nasopharyngeal and pulmonary systems. By data mining, we found that SPLUNC1 is also expressed in other organs, including the kidneys and the pituitary gland. SPLUNC1 is an asthma and cystic fibrosis gene modifier that also inversely correlates with the severity of bronchiectasis. Orai1 is a plasma membrane Ca2+ channel that is an essential regulator of the immune system. We previously found that SPLUNC1 binds to Orai1, causing it to be ubiquitinated, internalized and

Incidence and characterization of polyglucosan bodies in the cerebella of montserrat orioles (Icterus oberi).

Polyglucosan bodies are accumulations of insoluble glucose polymers and proteins that form intracytoplasmic inclusions in the brain, large numbers of which can be indicative of neurodegenerative diseases such as Lafora disease. Montserrat orioles (Icterus oberi) are an icterid passerine endemic to Montserrat with conservation populations maintained in captivity abroad. We demonstrate that polyglucosan bodies are unusually abundant in the cerebellar molecular and Purkinje cell layers and cerebellar peduncles of captive-bred and wild-caught Montserrat orioles. The bodies are periodic acid-Schiff

Proteasome associated function of UCH37 is evolutionarily conserved in Plasmodium parasites.

Ubiquitin C-terminal hydrolase 37 (UCH37 also known as UCHL5) is a conserved deubiquitinating enzyme (DUB) with dual roles in proteasomal degradation and chromatin remodeling in humans. Its Plasmodium falciparum ortholog, PfUCH37, is unusual in that it possesses both DUB and deneddylating activities. While PfUCH37 is enriched in proteasome preparations, its direct interaction and broader functions in Plasmodium remain unclear, particularly given the absence of the chromatin remodeling complex INO80 homologs. This study utilizes transgenic parasites and proteomics to identify PfUCH37-associatin

Lycorine protects motor neurons against TDP-43 proteinopathy-induced degeneration in cross-species models with amyotrophic lateral sclerosis.

Aggregation of TAR-DNA binding protein-43 (TDP-43) is a pathological feature present in nearly 97 % cases of amyotrophic lateral sclerosis (ALS), making it an attractive target for pathogenic studies and drug screening. Here, we have performed a high-throughput screening of 1500 compounds from a natural product library and identified that lycorine, a naturally occurring alkaloid, significantly decreases the level of TDP-43A315T in a cellular model. We further demonstrate that lycorine reduces the level of TDP-43A315T both through inhibiting its synthesis and by promoting its degradation by the
