We developed a bottom-up, data-driven approach for constructing a time-informed adverse outcome pathway (AOP) network for environmental chemical-induced cardiotoxicity. Mechanistic end points were systematically extracted from 339 in vitro and in vivo studies, yielding 1759 Key Event (KE) entries and 4938 Key Event Relationship (KER) entries, together with study metadata and essentiality evidence from intervention experiments. After quality filtering and minimum support thresholding, 62 unique KEs and 256 unique KERs were retained for network construction. Network analysis highlighted oxidativ

Coronary microvascular dysfunction (CMD) is a key contributor to myocardial ischemia and cardiovascular diseases. It is characterized by abn … B) signaling pathway. We initially outline how the NRG-1/ErbB pathway affects endothelial function, ventricular remodeling, oxidative stress, and myocardial angiogenesis, highlighting its potential as a therapeutic target. In addition, we explore emerging evidence that traditional Chinese medicine (TCM) interventions may regulate the NRG-1/ErbB axis to improve microvasc …

ST-segment elevation on electrocardiography is classically associated with acute myocardial infarction and often prompts urgent invasive evaluation. However, non-ischemic causes may complicate diagnostic decision-making, particularly in elderly patients with acute non-cardiac illnesses. We report a 95-year-old woman admitted with COVID-19-associated pneumonia and aspiration pneumonia, whose admission electrocardiogram demonstrated ST-segment elevation with T-wave inversion in the lateral precordial leads, mimicking ST-elevation myocardial infarction. She had no chest pain, no elevation of card

Atrial fibrillation is a common cause of suboptimal cardiac resynchronization therapy (CRT) delivery because intrinsic atrioventricular conduction reduces effective biventricular capture. We performed an updated systematic review and meta-analysis evaluating atrioventricular junction ablation (AVJA) versus no AVJA or pharmacological rate control in CRT recipients with atrial fibrillation. PubMed, Scopus, and Cochrane Library were searched through February 16, 2026. Sixteen studies were included, comprising 14 observational cohorts and 2 randomized trials. AVJA significantly increased biventric

The BTBR T+ Itpr3tf/J (BTBR) strain is a widely used model of neurodevelopmental disorders, characterized by an altered neuroanatomy, including a full-penetrant agenesis of the corpus callosum. While the adult BTBR brain has been studied thoroughly, less is known about how its brain develops prenatally and about when neurodevelopmental trajectories begin to differ from neurotypical strains. Here, we conducted a comparative histological analysis of fetal brain development across multiple developmental stages in BTBR and C57BL/6J (B6) mice, focusing on neocortical and midline development. Pulmonary hypertension (PH) is a complex disorder associated with significant morbidity and mortality. PH is characterized by an elevated pulmonary vascular load, leading to right ventricular dysfunction and remodeling, with right heart failure and premature death if left untreated. PH is classified into five groups (WHO Groups 1-5) based on etiology and pathophysiology. Group 1 PH, known as pulmonary arterial hypertension (PAH), has seen recent therapeutic advances with life-extending treatments, including a new therapeutic option that targets the underlying vascular abnormalities. PAH treatm

Autoimmune myocarditis is a severe inflammatory heart disease that can progress to dilated cardiomyopathy and heart failure, while disease-modifying therapies remain limited. Dysregulated activation of antigen-presenting cells, particularly dendritic cells (DCs), disrupts immune tolerance and drives pathogenic cardiac inflammation. Here, we report a nanoparticle-enabled CD40 silencing strategy for immune tolerance programming in experimental autoimmune myocarditis. Using a polymer-lipid hybrid siRNA formulation, we achieved sustained CD40 silencing in myeloid antigen-presenting cells and induc

Disruption of circadian blood pressure (BP) rhythm is a key pathophysiological feature and an important marker of cardiovascular risk. Ambulatory BP monitoring, the gold standard for assessing BP circadian patterns, has revealed that non-dipper and reverse-dipper phenotypes are strong independent predictors of coronary artery disease, atrial fibrillation, heart failure, and all-cause mortality. Notably, even when daytime BP is adequately controlled, abnormal circadian BP profiles impose sustained cardiovascular stress, accelerating target organ damage, including left ventricular hypertrophy, a

Various mature tissue-resident cells exhibit progenitor characteristics following injury. However, the existence of endogenous stem cells with multiple lineage potentials in the adult spinal cord remains a compelling area of research. In this study, we present a cross-species investigation that extends from development to injury. We used single-nucleus transcriptomic sequencing and genetic lineage tracing to characterize neural cells in the spinal cord. Our findings show that ciliated ependymal cells lose neural progenitor gene signatures and proliferation ability following the differentiation

Cardiometabolic heart failure with preserved ejection fraction (HFpEF) is a heterogenous metabolic disease, which in the heart presents as left ventricle diastolic dysfunction, ventricular stiffness, and myocardial structural remodeling. Deleterious changes in cardiac metabolism are central to HFpEF pathophysiology, and proposed treatments for the disease have focused on repairing these defects. In this study, we used a preclinical mouse model that recapitulates cardiometabolic HFpEF to elucidate the molecular mechanisms driving cardiac dysfunction, and tested whether recombinant Adropin (a li

Pulmonary arterial hypertension (PAH) is a progressive obstructive pulmonary vasculopathy characterized by pathological vascular remodeling driven by excessive cellular proliferation, apoptosis resistance, chronic inflammation, fibrotic deposition, and dysregulated vasoconstriction. As the disease advances, these vascular abnormalities culminate in right heart failure (RHF). The right ventricular (RV) initially undergoes compensatory hypertrophy but ultimately decompensates, manifesting as chamber dilation, impaired contractility, and progressive fibrosis. Despite the critical determinant role

Myocardial infarction (MI) results in oxidative stress to the myocardium and frequently leads to heart failure (HF). There is an unmet clinical need to develop therapeutics that address the inflammatory stress response and prevent negative left ventricular remodeling. Here, the Keap1/Nrf2 protein-protein interaction is specifically targeted, as Nrf2 activation is known to mitigate the inflammatory response following MI. This is achieved using a Nrf2-mimetic protein-like polymer (PLP) to inhibit the Keap1-Nrf2 interaction. The PLP platform technology provides stability in vivo, potent intracell…

Pulmonary veno-occlusive disease (PVOD) is a rare but severe form of pulmonary hypertension characterized by the obstruction of pulmonary arteries and veins, causing increased pulmonary artery pressure and leading to right ventricular (RV) heart failure. PVOD is often resistant to conventional pulmonary arterial hypertension (PAH) treatments and has a poor prognosis, with a median survival time of 2-3 years after diagnosis. We previously showed that the administration of a chemotherapy agent mitomycin C (MMC) in rats mediates PVOD through the activation of the eukaryotic initiation factor 2 (e

Sudden cardiac death (SCD) accounts for a substantial proportion of mortality in heart failure with reduced ejection fraction (HFrEF), frequently triggered by ventricular arrhythmias (VA). This review aims to analyze the pathophysiological mechanisms underlying VA and SCD in HFrEF and evaluate the effectiveness of guideline-directed medical therapy (GDMT) in reducing SCD. Beta-blockers, angiotensin receptor-neprilysin inhibitors, and mineralocorticoid receptor antagonists have shown significant efficacy in reducing SCD risk. While angiotensin-converting enzyme inhibitors and angiotensin recept
