An open-label Phase 2a study of fasudil in amyotrophic lateral sclerosis: safety and exploratory endpoints.

OBJECTIVES: The primary objective was to assess the safety of oral fasudil in amyotrophic lateral sclerosis (ALS) patients. Changes in serum neurofilament light (NfL) levels and the ratio of phosphorylated to total AKT (pAKT/tAKT) were exploratory endpoints. METHODS: This was a multicenter, open-label study. Two 31-patient cohorts were sequentially enrolled and treated with either 180 mg or 300 mg per day of oral fasudil for 24 weeks. The primary endpoint was safety. Secondary endpoints evaluated changes in the ALS functional rating scale-revised (ALSFRS-R), slow vital capacity, and muscle st…

Healthy diet is associated with better lung function, especially in individuals with COPD: findings from the Lifelines cohort study.

OBJECTIVE: To investigate whether the association between diet and lung function (LF) differs according to asthma or COPD status in the Lifelines cohort study. METHODS: We included adults from Lifelines with valid dietary and spirometry data. Dietary quality was calculated at baseline using the Lifelines diet score (LLDS) (range 0-48, where higher scores indicate a healthier diet). LF was assessed via pre-bronchodilator spirometry (forced expiratory volume in 1 s (FEV1), forced vital capacity (FVC) and FEV1/FVC) at baseline and two follow-ups (∼10 years). Cross-sectional associations between …

Cannabidiol as a Modulator of the Gut-Liver Axis: Clinical and Pharmacological Insights into Hepatic and Metabolic Disorder Therapies.

A non-intoxicating substance produced from Cannabis sativa, cannabidiol (CBD) has shown promise as a treatment for metabolic and hepatic diseases, primarily due to its capacity to alter the gut-liver axis. A vital bidirectional communication pathway, the gut-liver axis is where substances produced from the liver affect gut homeostasis and gut-derived microbial products and metabolites influence liver health. Conditions including alcoholic liver disease (ALD), metabolic syndrome, and non-alcoholic fatty liver disease (NAFLD) are mostly caused by dysregulation of this axis. According to preclini

Effects of dietary indole-3-acetate sodium on intestinal morphology, nutrient absorption, and inflammatory responses in weaned piglets.

Weaning stress can severely damage the piglets' intestines. Microbial tryptophan catabolites play a vital role in maintaining the health of the intestinal mucosa. Indole-3-acetic acid (IAA), an indole derivative with known anti-inflammatory properties, has not yet been studied for its impact on piglets' intestinal health. Twenty-four weaned crossbred piglets (Duroc × Yorkshire × Landrace, weighing 6.58 ± 0.07 kg) were randomly allocated to receive diets containing 0, 120, or 240 mg/kg indole-3-acetate sodium (IAA-Na). Although dietary IAA-Na did not significantly impact growth performance or d

Designing a novel vaccine against COVID-19 based on spike SARS-Cov-2 notable mutations using immunoinformatics approaches.

The rapid emergence of SARS-CoV-2 variants with spike protein mutations undermines the effectiveness of current vaccines, necessitating innovative strategies to ensure broad and lasting immunity. This study leverages an immunoinformatics approach to design two multi-epitope vaccine constructs Cov19-B (649 amino acids, 74 kDa) and Cov19-T (465 amino acids, 48 kDa) specifically targeting mutations in the spike protein observed in the Alpha, Beta, Gamma, and Omicron variants. Using sequence data retrieved from NCBI, GISAID, and UniProt, we predicted a range of epitopes, including linear B-cell, c

Transforming Surgical Training: The Evolution of the COSECSA eLogbook Through a Theory of Change Framework.

INTRODUCTION: Surgical training logbooks are vital for tracking trainee experience and training quality. As the College of Surgeons of East, Central and Southern Africa (COSECSA) expanded across 24 countries, its paper-based system outgrew the capacity of paper logbooks. In partnership with the Royal College of Surgeons in Ireland, COSECSA developed an electronic logbook (eLogbook) to enhance data collection, trainee monitoring, and quality assessment. This study reviews its evolution using a theory of change framework to inform future surgical training needs in sub-Saharan Africa and beyond.

Enhancement of community productivity and water‑air‑interface greenhouse gas uptake by plant growth-promoting rhizobacteria inoculation in submerged macrophyte communities.

Enhancing lake carbon sequestration is a vital nature-based climate solution, whereas the degradation of submerged macrophytes seriously threatens this potential. Plant growth-promoting rhizobacteria (PGPR) are capable of accelerating the stress recovery of submerged macrophytes and enhancing the greenhouse gas (GHG) absorption for single-species macrophyte. Here, four-species submerged macrophyte communities (Potamogeton wrightii, Myriophyllum spicatum, Vallisneria denseserrulata, and Hydrilla verticillata) were established under simulated underwater low-light conditions and inoculated with a

Salubrinal-activated integrated stress response protects against doxorubicin-induced cardiotoxicity via activating transcription factor 4-mediated antioxidant defense and glutathione homeostasis.

Doxorubicin is an effective chemotherapeutic agent; however, its use is limited by cardiotoxicity. Mitochondrial dysfunction is a central driver of doxorubicin-mediated cardiotoxicity. The role of the integrated stress response (ISR), a mitochondria-to-nucleus signaling pathway and crucial cellular defense mechanism in doxorubicin-induced cardiotoxicity, remains unclear. We investigated the pharmacological ISR activator salubrinal, a selective inhibitor of eukaryotic initiation factor 2α dephosphorylation with potential cardioprotective properties, to elucidate the molecular mechanisms underly
