Hypoadiponectinaemia in South Asian women during pregnancy: evidence of ethnic variation in adiponectin concentration.

AIMS: People of South Asian descent face an increased risk of Type 2 diabetes mellitus (DM) and coronary artery disease (CAD) compared with other ethnic groups. One candidate factor underlying this risk may be adiponectin, as circulating levels of this adipocyte-derived protein are reduced in both Type 2 DM and CAD. In a recent study, we assessed the relationship between adiponectin and gestational diabetes (GDM), a potential model of early events in the natural history of Type 2 DM. Here, we report the impact of ethnicity on plasma adiponectin concentration in that study. METHODS: A cross-se

Alternative splicing variants of dual specificity tyrosine phosphorylated and regulated kinase 1B exhibit distinct patterns of expression and functional properties.

The dual specificity tyrosine phosphorylated and regulated kinase (DYRK) family of protein kinases is a group of evolutionarily conserved pr … T1-7 and also in differentiated, adipocyte-like 3T3-L1 cells, but not in undifferentiated 3T3-L1 preadipocytes. A comparison of the mouse and human Dyrk1b genomic and cDNA sequences defined the alternative splicing events that produce the variants of DYRK1B. In DYRK1B-p75, transcription starts with exon 1B instead of exon 1A, generating a new translation sta …

Leptin-replacement therapy for lipodystrophy.

BACKGROUND: The adipocyte hormone leptin is important in regulating energy homeostasis. Since severe lipodystrophy is associated with leptin deficiency, insulin resistance, hypertriglyceridemia, and hepatic steatosis, we assessed whether leptin replacement would ameliorate this condition. METHODS: Nine female patients (age range, 15 to 42 years; eight with diabetes mellitus) who had lipodystrophy and serum leptin levels of less than 4 ng per milliliter (0.32 nmol per milliliter) received recombinant methionyl human leptin (recombinant leptin). Recombinant leptin was administered subcutaneousl

Analysis of the lamin A/C gene as a candidate for type II diabetes susceptibility in Pima Indians.

AIMS/HYPOTHESIS: Lamin A/C (LMNA) is located within a region on chromosome 1q that has been linked with Type II (non-insulin-dependent) diabetes mellitus in Pima Indians. Rare mutations in exon 8 of LMNA underlie Dunnigan-Type familial partial lipodystrophy, a disease characterized by regional adipocyte degeneration and frequently accompanied by insulin resistance, glucose intolerance, and diabetes. A more common variant in exon 10 (3408C/T) has recently been associated with obesity in non-diabetic aboriginal Canadian subjects. Because obesity is a strongly predisposing factor for Type II diab

The peroxisome proliferator-activated receptors at the cross-road of diet and hormonal signalling.

The peroxisome proliferator-activated receptors (PPARs) are members of the steroid/thyroid nuclear receptor superfamily of ligand-activated transcription factors. To date, three isotypes have been identified, alpha, beta and gamma, encoded by three different genes. The alpha isotype is expressed at high levels in the liver where it has a role in lipid oxidation. Its expression and activity follow a diurnal rhythm that parallels the circulating levels of corticosterone in the bloodstream. The gamma isotype on the other hand, is mainly expressed in adipose tissue and has a critical role in adipo

Insulin resistance and diabetes due to different mutations in the tyrosine kinase domain of both insulin receptor gene alleles.

Mutations in the insulin receptor gene can lead to in vivo and in vitro insulin resistance and can be the cause of diabetes mellitus in selected patients. We have studied a 22-year-old diabetic woman with Type A insulin resistance and acanthosis nigricans. Insulin binding to the patient's erythrocytes, monocytes, adipocytes, fibroblasts, and transformed lymphocytes was decreased. Receptor autophosphorylation and tyrosine kinase activity toward an exogenous substrate were reduced in partially purified insulin receptors from the proband's transformed lymphocytes. Determination of the nucleotide …

Metabolic Reprogramming by Time-Restricted Feeding Confers Resistance to PM2.5-Induced Metabolic Disturbances in Adipose Tissue.

Recent research indicates that exposure to fine particulate matter (PM2.5) not only impairs the respiratory system but also disturbs adipose tissue homeostasis, contributing to rising obesity rates and systemic metabolic disturbances. Time-restricted feeding (TRF), a dietary approach that confines daily food consumption to specific time windows, has demonstrated significant potential in preventing and managing metabolic disorders. However, it remains unclear if TRF can alleviate PM2.5-induced adipose tissue dysfunction, and the mechanisms involved require investigation. We established a PM2.5-

Variation in Intrinsic Aerobic Capacity Reveals Distinct Differences in the Maintenance of Rat Knee Joint Integrity.

The objective was to test how intrinsic aerobic capacity affects the articular cartilage, infrapatellar fat pad (IFP), subchondral bone, and synovial membrane of the rat knee joint. Thirty-five 24-week-old male N:NIH rats selectively bred for high aerobic capacity (high-capacity-runner [HCR]; n = 17) and low aerobic capacity (low-capacity-runner [LCR]; n = 18) were fed either a high-fat/high-sucrose (HFHS) diet or a chow diet for 12 weeks. Alterations in articular cartilage, subchondral bone microarchitecture, IFP hypertrophy, and synovial membrane cytokine levels were quantified. Body composi

Atrial fibrillation modifies the association between CT-derived epicardial adipose tissue volume and left atrial remodeling in valvular heart disease.

BACKGROUND: Epicardial adipose tissue volume (EATv) is associated with atrial fibrillation (AF), but whether AF modifies the association between EATv and atrial remodeling in valvular heart disease remains unclear. OBJECTIVE: To determine whether AF modifies the association between computed tomography (CT)-derived EATv and left atrial remodeling. METHODS: This retrospective valvular surgery cohort included 436 patients (218 with AF and 218 with sinus rhythm [SR]) after 1:1 propensity score matching. Multivariable linear and logistic regression analyses assessed the associations of EATv with

Clinical evidence for fat-binding-mediated fecal fat excretion by RODLTM (Red Okra-Diospyros lotus botanical combination) under a prescribed restricted diet: a randomized, double-blind, placebo-controlled trial.

Preclinical studies have demonstrated that a botanical combination of red okra (Abelmoschus esculentus) and Diospyros lotus (RODLTM) exerts dual anti-obesity actions by both inhibiting intestinal fat absorption and stimulating adipocyte lipid metabolism. However, its clinical efficacy in promoting fecal fat excretion under controlled dietary conditions has not been validated in humans. A randomized, double-blind, placebo-controlled clinical trial was conducted in 70 healthy adults under a prescribed restricted diet containing 130-150 g/day of fat. Participants were randomly assigned to receive

Britanin improved high-fat diet-induced obesity by regulating the MAPK signaling pathway.

BACKGROUND: The escalating prevalence of obesity has made it a critical public health concern. There is an urgent need to identify naturally derived compounds with anti-obesity potential. Britanin (BRI), a bioactive sesquiterpene lactone derived from Inula species, has shown promise in metabolic disorder management, but its anti-obesity mechanisms remain uncharacterized. OBJECTIVES: Combining animal experiments and network pharmacology analysis to explore the effect of BRI in high-fat diet-induced obesity. METHODS: C57BL/6J male mice were used for experiment. Effects of a phenolic-rich grape pomace extract on intestinal barrier, gut microbiota, and adipose tissue in high-fat diet-fed mice.

Obesity is associated with impaired intestinal barrier integrity, gut dysbiosis, and adipose tissue dysfunction, leading to endotoxemia and metabolic disturbances. We previously observed that grape pomace extract (GPE), rich in phenolic compounds, protected mice from high-fat diet (HFD)-induced endotoxemia and hepatic steatosis. This study aimed to evaluate the effect of GPE on intestinal permeability, gut microbiota composition, and the associated metabolic alterations in HFD-fed mice. Mice were assigned to four experimental groups and fed for 13 weeks with either a standard diet or HFD (appr

Characterization of Isoorientin and Paeoniflorin as Botanical Glucocorticoid Receptor Modulators from White Peony and Chasteberry.

BACKGROUND/OBJECTIVES: Botanical supplements are increasingly investigated for their potential to address women's health concerns. Compounds that modulate progesterone receptor (PR) signaling may help manage gynecologic disorders such as endometriosis, uterine hyperplasia, and preterm birth. Because PR ligands often cross-react with the glucocorticoid receptor (GR), this study examined two botanical compounds, paeoniflorin from Paeonia lactiflora (white peony root) and isoorientin from Vitex agnus-castus (chasteberry), that were identified as modulators of GR or PR signaling. METHODS: Lucifer

Identifying microRNA predictors of weight loss after bariatric surgery.

BACKGROUND: Bariatric surgery is an effective treatment for obesity. However, the degree of postoperative weight loss can vary greatly between patients, and the rationale behind such disparity remains unclear. Recent studies have suggested that genetics can influence the response to bariatric surgery, but little is known about the role of microRNAs (miRNAs) in this setting. OBJECTIVES: To identify miRNA candidates that predict weight loss after bariatric surgery. SETTING: Academic medical center. METHODS: RNA was isolated from patients' blood collected at initial preoperative consult visits

"Aptamer-guided therapeutics and diagnostics for metabolic syndrome: Design, delivery, and translation of aptamer-based systems".

Metabolic syndrome (MetS) is a composite cardiometabolic condition defined by visceral obesity, dyslipidemia, hypertension, and insulin resistance, and it promotes the worldwide prevalence of type 2 diabetes and atherosclerotic cardiovascular disease. Nucleic-acid aptamers have developed as programmable recognition ligands that integrate antibody-like specificity with chemical synthesis, modular functionalization, low batch variability, and usually a low immunogenicity. Their translational significance in MetS is not solely in target binding, but in the capacity to establish disease-relevant a

Postnatal overfeeding induces gut microbiota disturbances and impairs GPR43/FIAF/LPL pathway in the rat model of PCOS.

Women with polycystic ovary syndrome (PCOS) has high incidence of metabolic dysfunction-associated steatotic liver disease (MASLD). The development of PCOS-associated MASLD is accelerated by prepubertal obesity, therefore, we analyzed the impact of postnatal overfeeding-induced obesity on the gut microbiota and hepatic lipid metabolism in the PCOS rat model. Wistar rats were divided into 4 groups, where treatment with 5α-dihydrotestosterone (5α-DHT) stimulated hyperandrogenemia (DHT groups), whereas litter size reduction induced early postnatal overfeeding and obesity (SL groups).
