Does Aldosterone Play a Significant Role for Regulation of Vascular Tone?

Besides the well-known renal effects of aldosterone, the hormone is now known to have direct vascular effects. Clinical observations underline substantial adverse effects of aldosterone on cardiovascular function. The source of systemic circulating aldosterone is the adrenal gland zona glomerulosa cells through stimulus-secretion coupling involving depolarization, opening of L- and T-type calcium channels and aldosterone synthase activation. Local formation and release in peripheral tissues such as perivascular fat is recognized. Where does aldosterone affect the vasculature? Mineralocorticoid

Pathologic findings in malignant hyperthermia: a case report and review of literature.

Malignant hyperthermia (MH) is a rare, potentially lethal disorder of skeletal muscle calcium homeostasis characterized by muscle contracture and life-threatening hypermetabolic crisis following exposure to halogenated anesthetics and depolarizing muscle relaxants. Susceptibility to MH results from mutations in calcium channel proteins that mediate excitation-contraction coupling, with the ryanodine receptor calcium release channel (RyR1) representing the major locus. The mode of inheritance appears to be autosomal dominant with variable penetrance. The authors report the death of a 60-year-ol

CXCL8 as a Potential Biomarker for Crohn's Disease is Associated with Immune Infiltration and RNA Methylation.

TRP channels have an important role in regulating the function of gastrointestinal epithelial cells. The aim of this study was to investigate the molecular mechanisms of genes associated with TRP channels in Crohn's disease (CD) by bioinformatics approach and to identify potential key biomarkers. In our study, we identified TRP channel-related differentially expressed genes (DEGs) based on the GSE95095 dataset and the TRP channel-related gene set from the GeneCards database. Hub genes (CXCL8, HIF1A, NGF, JUN, IL1A) were identified by the PPI network and validated by the external GSE52746 datas

Transient receptor potential (TRP) channels: an emerging therapeutic target for the treatment of cardiac hypertrophy.

INTRODUCTION: Cardiac hypertrophy (CH) is a major contributor to cardiovascular disease (CVD), affecting millions worldwide. While initially an adaptive response, sustained hypertrophic stimuli lead to maladaptive outcomes, including heart failure (HF), contractile dysfunction, and ventricular remodeling. Key pathophysiological processes involve myocardial fibrosis, cardiomyocyte death, mitochondrial impairment and Ca2+ dysregulation. Despite available therapies, treatment of CH is symptomatic with no approved targeted options. Many interventions have only temporary benefits, may cause adverse

Sphingosylphosphorylcholine induces itch via activation of TRPM3 and TRPA1 in mice.

Itch is a prevalent symptom in atopic dermatitis (AD), often leading to a strong urge to scratch. Elevated levels of sphingosylphosphorylcholine (SPC) are found in the stratum corneum of AD patients, and while SPC is known to induce itch, its molecular targets are not well understood. This study aims to identify the signaling pathway of SPC-induced itch under AD conditions. We demonstrate that SPC specifically activates the Transient Receptor Potential Melastatin 3 (TRPM3) channel in sensory neurons. In HEK293T cells expressing TRPM3, SPC treatment caused a significant increase in intracellula

Calcium-Signalling in Human Glaucoma Lamina Cribrosa Myofibroblasts.

Glaucoma is one of the most common causes of treatable visual impairment in the developed world, affecting approximately 64 million people worldwide, some of whom will be bilaterally blind from irreversible optic nerve damage. The optic nerve head is a key site of damage in glaucoma where there is fibrosis of the connective tissue in the lamina cribrosa (LC) extracellular matrix. As a ubiquitous second messenger, calcium (Ca2+) can interact with various cellular proteins to regulate multiple physiological processes and contribute to a wide range of diseases, including cancer, fibrosis, and gla

COVID-19 and hairy-cell leukemia: an EPICOVIDEHA survey.

The high incidence of disability and fatality rates associated with chronic diabetic wounds are difficult problems in the medical field. The steady-state and regular changes of the microenvironment in and around the wound provide good conditions for wound healing and achieve a dynamic and complex process of wound healing. The pH value and ionic environment composed of a variety of ions in wound are important factors affecting the wound microenvironment, and there are direct or indirect connections between them. Abnormalities in pH, ion concentrations, and channels in skin tissue may be one of t

Stability and Safety of Inhibitor Cystine Knot Peptide, GTx1-15, from the Tarantula Spider Grammostola rosea.

Inhibitor cystine knot (ICK) peptides are knotted peptides with three intramolecular disulfide bonds that affect several types of ion channels. Some are proteolytically stable and are promising scaffolds for drug development. GTx1-15 is an ICK peptide that inhibits the voltage-dependent calcium channel Cav3.1 and the voltage-dependent sodium channels Nav1.3 and Nav1.7. As a model molecule to develop an ICK peptide drug, we investigated several important pharmaceutical characteristics of GTx1-15. The stability of GTx1-15 in rat and human blood plasma was examined, and no GTx1-15 degradation was

[Characteristic of age-dependent changes in pial arterial vessels endothelium-dependent hyperpolarization in normotensive and spontaneously hypertensive rats.].

A comparative study of changes in the contribution of endothelium-dependent hyperpolarization (EDH) induced by activation of the intermediate-conductance calcium-activated potassium channels (IKса) channels and the contribution of NO in pial arterial vessels dilation to acetylcholine (ACh) in normotensive (WKY) and spontaneously hypertensive (SHR) rats at the age of 4 and 18 months was conducted. It was found that in WKY, the EDH is mainly expressed in the group of small pial arterial vessels. During aging, the contribution of EDH to the dilatation of small (diameter less than 20 microns) and

Use of Levosimendan in Intensive Care Unit Settings: An Opinion Paper.

Levosimendan is an inodilator that promotes cardiac contractility primarily through calcium sensitization of cardiac troponin C and vasodilatation via opening of adenosine triphosphate-sensitive potassium (KATP) channels in vascular smooth muscle cells; the drug also exerts organ-protective effects through a similar effect on mitochondrial KATP channels. This pharmacological profile identifies levosimendan as a drug that may have applications in a wide range of critical illness situations encountered in intensive care unit medicine: hemodynamic support in cardiogenic or septic shock; weaning f

Persistent Bradycardia with the Long-term Use of Phenytoin and Oxycodone: First Case Report.

Phenytoin is a medication that is used primarily in the treatment of epilepsy as well as generalized tonic-clonic seizures and status epilepticus. Phenytoin is also considered a class IB antiarrhythmic medication by shortening the duration of the action potential and increasing myocardial conduction. The neurologic adverse effects of phenytoin are well-documented and include altered mental status, ataxia, and nystagmus. Some adverse cardiac manifestations have also been reported, including arrhythmias, hypotension, and respiratory arrest. Oxycodone is an opioid that exerts its effects by bindi

Blockade by NNC 55-0396, mibefradil, and nickel of calcium and exocytotic signals in chromaffin cells: implications for the regulation of hypoxia-induced secretion at early life.

Adrenal chromaffin cells (CCs) express high-voltage activated calcium channels (high-VACCs) of the L, N and PQ subtypes; in addition, T-type low-VACCs are also expressed during embryo and neonatal life. Effects of the more frequently used T channel blockers NNC 55-0396 (NNC), mibefradil, and Ni2+ on the whole-cell Ba2+ current (IBa), the K+-elicited [Ca2+]c transients and catecholamine secretion have been studied in adult bovine CCs (BCCs) and rat embryo CCs (RECCs). NNC, mibefradil, and Ni2+ blocked BCC IBa with IC50 of 1.8, 4.9 and 70 μM, while IC50 to block IBa in RECCs were 2.1, 4.4 and 41

Characterization of stretch-activated ion currents in isolated atrial myocytes from human hearts.

To explore further the mechanisms that may underlie cardiac arrhythmia, we analysed stretch-activated ion currents in human atrial myocytes. Longitudinal stretch of freshly isolated atrial myocytes prolonged the duration of action potentials, depolarized the resting membrane potential and caused extra action potentials. Under voltage-clamp conditions, the amplitude of stretch-induced transmembrane currents increased reversibly with the intensity of stretch. Stretch-activated currents ( I(SAC)) had a reversal potential of 0 mV and were insensitive to substitution of Cl(-) with aspartate ions in

Mutation in pre-mRNA adenosine deaminase markedly attenuates neuronal tolerance to O2 deprivation in Drosophila melanogaster.

O2 deprivation can produce many devastating clinical conditions such as myocardial infarct and stroke. The molecular mechanisms underlying the inherent tissue susceptibility or tolerance to O2 lack are, however, not well defined. Since the fruit fly, Drosophila melanogaster, is extraordinarily tolerant to O2 deprivation, we have performed a genetic screen in the Drosophila to search for loss-of-function mutants that are sensitive to low O2. Here we report on the genetic and molecular characterization of one of the genes identified from this screen, named hypnos-2. This gene encodes a Drosophil

Novel structure having antagonist actions at both the glycine site of the N-methyl-D-aspartate receptor and neuronal voltage-sensitive sodium channels: biochemical, electrophysiological, and behavioral characterization.

A novel series of N-substituted 4-ureido-5,7-dichloro-quinolines were synthesized to contain pharmacophores directed at voltage-sensitive sodium channels (VSNaCs) and N-methyl-D-aspartate (NMDA) receptors. These compounds were shown to act in a use-dependent manner as antagonists of VSNaCs and to act as selective competitive antagonists at the strychnine-insensitive glycine recognition site of NMDA receptors. These agents had little or no effect on alpha-adrenergic receptors, other glutamate receptors, or sites other than the glycine site on the NMDA receptor, and did not block voltage-sensiti

Release of substance P from peripheral sensory nerve terminals.

It is postulated that release of SP from peripheral terminals of sensory neurons contributes to neurogenic inflammation and possibly mediates, in part, the inflammatory response in a variety of diseases. This review addresses the Ca2+-dependent release of SP from peripheral nerve terminals in response to antidromic electrical stimulation of peripheral nerves and noxious thermal and chemical stimuli. Initial studies suggested that SP release is induced via an axon reflex, which involves antidromic impulses travelling along the peripheral nerve terminal. Further investigation indicated that SP r

[Early after-depolarization in polymorphic ventricular tachycardia without QT prolongation].

Endocardial monophasic action potentials (MAP) were recorded at different sites of the right ventricle in a patient with a history of syncope and recurrent polymorphic ventricular tachycardia (pVT) without QT prolongation. The MAP recording from the infero-septal wall demonstrated persisting early afterdepolarizations (EAD) during sinus rhythm with no cycle length dependence. Programmed ventricular stimulation at this site provoked a pVT rapidly degenerating into ventricular fibrillation. Iodine-123-meta-iodobenzylguanidine (MIBG) scintigraphy indicated a defective sympathetic innervation in i

Immunopathology of the Lambert-Eaton myasthenic syndrome.

LEMS is an antibody-mediated autoimmune disease that can occur in isolation, or as a paraneoplastic disorder in association with SCLC (60% of patients). The underlying defect is a reduction in the quantal release of the neurotransmitter ACh from the presynaptic nerve terminal at the neuromuscular junction. Experimental evidence indicates the autoantibodies are directed against nerve terminal VGCCs causing down-regulation in the number of functional channels by cross-linkage. Functional VGCCs have been detected in SCLC cell lines. In cancer-associated LEMS it appears likely that antibodies init

Protocatechualdehyde-functionalized BMSC-derived exosomes orchestrate calcium signaling in the immunoinflammatory microenvironment to promote periodontal regeneration.

The significant global burden of periodontitis has increased over the past few decades. However, the regeneration and reconstruction of periodontal tissue remain major challenges in clinical practice. Successful periodontal treatment relies on the regulation of calcium signaling. Bone mesenchymal stem cell-derived exosomes (BMSC Exo) hold great potential for promoting calcium influx to enhance osteogenic differentiation, modulate the inflammatory microenvironment, and promote angiogenesis, but their therapeutic efficiency is limited by inadequate stability. To overcome this problem, a mild and

Stimulation and inhibition of elastase release from human neutrophil-dependence on the calcium messenger system.

The neutral proteinase elastase is released from polymorphonuclear (PMN) leukocytes in various physiological and pathological conditions. Aim of the present study was to gain further insight into the mechanisms which govern the liberation of this proteinase. Therefore, the effects of the calcium ionophore A23187 and of the protein kinase C activator phorbol myristate acetate (PMA) on neutrophils were investigated in human whole-blood samples. Furthermore, the inhibitory effects of the calcium channel blocker verapamil and of the calmodulin blocker trifluoperazine were followed. A23187 induced
