Blood vessels from bone marrow.

Lengths of silastic tubing were inserted into the peritoneal cavity of rats or rabbits. By two weeks the free-floating implants had become covered by a capsule consisting of several layers of "macrophage"-derived myofibroblasts and collagen matrix overlaid by a single layer of mesothelial cells. The tubing was removed from the harvested implant and the tissue everted. This now resembled an artery with an inner lining of mesothelial cells (the "intima"), a "media" of myofibroblasts, and an outer collagenous "adventitia." The tube of living tissue was grafted by end-to-end anastomoses into the t

Peripheral lymphocyte subsets and other immune aspects in Rett syndrome.

A possible role of the immune system in the pathogenesis of some neurologic disorders, including infantile autism, was recently postulated. This observation prompted the authors to investigate some immunologic aspects in a group of patients with Rett syndrome, a disorder still not completely clarified but with some points of commonality with infantile autism. Humoral and cell-mediated immunity were investigated in 20 females with Rett syndrome. Peripheral lymphocyte subsets revealed a reduced percentage of CD8+ suppressor-cytotoxic cells in all of the patients with Rett syndrome, resulting in

Parathyroid hormone--quo vadis?

Parathyroid hormone is the most important calcium regulating hormone, especially in the process of aging. As a compensation for progressive calcium deficiency in aging, PTH secretion progressively rises. Heterogeneity of PTH in peripheral blood indicates the importance of understanding the degradation mechanism throughout the life cycle of the hormone. In addition to cathepsin B and D which degrades PTH in the liver, kidney and parathyroid gland, a new neutral PTH ase was found in the cytosolic fraction of rat kidney cells. PTH responsive bone and kidney cell lines, UMR-106 and OK cells, were

[Accumulation of mu-type opiate receptor ligands during the culture of phytohemagglutinin-stimulated lymphocytes].

Substances displacing labelled ligands from opiate receptors of the rat brain membrane fractions were found in the extracts of human blood lymphocytes. Pronase treatment has shown that at least part of the opiate receptor ligands present in lymphocyte extracts are of peptide nature. Opiate mu-receptor ligands content was identical in the extracts of the total population, T-lymphocyte and B-cell-enriched population. However, opiate delta-receptor ligands concentration in T-cell population is 15 times lower than in B-cell-enriched population. Opiate receptor ligands content is not changed during

Studies on amino acid incorporation in isolated toad bladder epithelial cells. Seasonal changes in protein synthesis.

Amino acid incorporation in isolated toad bladder epithelial cells was investigated by examining the rate of radiolabel incorporation into trichloroacetic acid-insoluble material. The results demonstrated that during the summer months, a large fraction of the radiolabel incorporated was not inhibited by cycloheximide and did not seem to correspond to incorporation of the radioactive amino acid into elongating peptides. A similar effect was not observed in other organs of the toad (e.g. liver, spleen). During the winter months, however, virtually all the acid-insoluble radiolabel appears to cor

The mutagenicity and cytotoxicity of selenite, "activated" selenite and selenate for normal and DNA repair-deficient human fibroblasts.

At doses varying from 8 x 10(-5) to 3 x 10(-3) M sodium selenite (Na2SeO3) induced DNA fragmentation, DNA-repair synthesis, chromosome aberrations and a mitotic inhibition in cultured human fibroblasts. The response of DNA repair-deficient xeroderma pigmentosum (XP) fibroblasts to selenite is comparable to that of control cells. Incubation with mouse liver S-9 microsomal fraction increased the capacity of selenite to induce chromosome aberrations, DNA-repair synthesis and a lethal effect. XP cells behaved as control cells when treated with activated selenite. Sodium selenate (Na2SeO4) at doses

Self-replication of somatostatin cells in the antral mucosa of rodents.

The possibility that antral somatostatin cells have a self-replicating activity has been studied in three species of rodents: mice, rats and guinea-pigs, after a flash tritiated thymidine injection. The immunocytochemical staining of somatostatin cells, using specific antiserum, was combined with radioautographic procedures. The labelling index for somatostatin cells--and for gastrin cells identified on serial sections--was established after counting a large number of cells at the optical microscope level, on parallel tissue strips removed throughout the entire antrum. A significant percentage

Surface membrane changes in lymphocytes from patients with infectious mononucleosis.

Peripheral blood lymphocytes from 20 patients with acute infectious mononucleeosis (IM) were studied for cell aggregation and for cap formation by concanavalin A (Con A). The lymphocytes from these patients showed 5.2+/-1.5% cells with a Con-A-induced cap and a high degree of cell aggregation without Con A, compared to 27.7+/-3.2% caps and a low degree of cell aggregation with normal lymphocytes. The lymphocytes from IM patients were fractionated to enrich for T and B cells. There was a low frequency of cap formation in both T and B cells, but the high degree of celll aggregation without Con A

MiR-22-3p in exosomes increases the risk of heart failure after down-regulation of FURIN.

Heart failure (HF) is often the inevitable manifestation of myocardial ischemia. Hypoxia can induce cardiomyocytes to express many microRNAs (miRNAs), which are highly expressed in exosomes. In addition, miR-22-3p is a marker in heart failure. Therefore, miR-22-3p was taken as the research object to explore its role and mechanism in HF. HF differentially expressed miRNAs were screened by bioinformatic analysis. The HF rats model was constructed and identified by detecting serum brain natriuretic peptide (BNP) and ultrasound analysis [left ventricular ejection fraction (LVEF) and left ventricul

Separation of leucocytes: improved cell purity by fine adjustments of gradient medium density and osmolality.

This paper briefly reviews commonly used procedures for separation of mononuclear cells (MNC) and granulocytes from human blood with X-ray contrast media as gradient material, and also presents new and modified procedures for leucocyte preparation. Standard techniques for human blood do not always yield satisfactory results with blood from other species. In general pure MNC are easily obtained (top fraction), but often the granulocyte fraction has a low purity, due to contamination with MNC that move to the bottom during centrifugation and contaminate the granulocyte suspension. Obviously the

New-Onset Acute Heart Failure With Reduced Ejection Fraction Associated With Severe Microcytic Anemia.

Severe microcytic anemia may serve as a potentially reversible precipitating factor in heart failure with reduced ejection fraction (HFrEF). This case highlights the diagnostic complexity and hemodynamic consequences of severe, microcytic anemia with undetermined etiology in a patient presenting with newly diagnosed heart failure. A 60-year-old Hispanic woman presented with new-onset acute dyspnea, chest pain, and bilateral edema. Investigations revealed severe microcytic anemia (hemoglobin 4.5 g/dL, MCV 60.8 fL) and an ejection fraction of 33%. Iron studies (iron 40 μg/dL, ferritin 14 ng/mL,

Dynamic bidirectional coupling of membrane morphology and rod organization in flexible vesicles.

The ordering of rod-like particles in soft, deformable containers emerges from the interplay of anisotropic interactions, geometric confinement, and boundary compliance. This competition couples internal particle organization to container morphology, producing behavior distinct from both rigid confinement and bulk systems. Such coupling is also relevant to biological contexts in which filamentous structures are confined by deformable membranes. Using a minimal model combining experiments and simulations of colloidal rods encapsulated in lipid vesicles, we show that soft confinement drives a bi

Curcumae Radix polysaccharides alleviate sepsis in rats: Isolation and characterization of an active homogeneous polysaccharide.

Curcumae Radix (CR), a traditional Chinese herbal medicine, has shown potential in the treatment of sepsis, yet its active substances and mechanisms of action remain unclear. In this study, CR was separated into polysaccharide and non-polysaccharide fractions. Both fractions exhibited significant anti-inflammatory and anticoagulant effects in a rat model of sepsis induced by lipopolysaccharide (LPS) and carrageenan. These effects were mediated by sphingosine kinase 1 (SPHK1) signaling pathway inhibition, which protected the vascular endothelial barrier and consequently alleviated liver and lun

Efficacy of Radiotherapy in Patients with Relapsing Primary Rosai-Dorfman Disease of the Nasal Cavity.

Rosai-Dorfman disease (RDD) is a rare proliferative disorder characterized by an overproduction of a type of immune cell called histiocytes, with unknown etiology. Although extranodal involvement is not uncommon, it is rarely seen within the nose. The available data are limited, and currently, there are no established guidelines for managing RDD. Different therapeutic options have been described, including corticosteroids, surgery, radiotherapy, or chemotherapy. This study aims to evaluate the potential benefit of radiotherapy in patients with nasal Rosai-Dorfman disease to improve the current

Thermoregulation Effects of Phoneutria nigriventer Isolated Toxins in Rats.

Body temperature is primarily regulated by the hypothalamus, ensuring proper metabolic function. Envenomation by Phoneutria nigriventer can cause symptoms such as hypothermia, hyperthermia, sweating, and shivering, all related to thermoregulation. This study aims to analyze and identify components of the venom that affect thermoregulation and to evaluate possible mechanisms. Rats were used for thermoregulation analysis, venom fractionation by gel filtration and reverse-phase chromatography (C18), and sequencing by Edman degradation. The venom exhibited hypothermic effects in rats, while its fr

Mechanism of the wine pomace tannin in hyperpigmentation inhibition: Impact on signaling pathways, cell proliferation, and tyrosinase activity.

After winemaking, tannins with high polymerization remain in the pomace. Utilizing these tannin fractions is a concern for the wine industry. While tannins show potential in treating hyperpigmentation, their mechanisms in vivo and at the cellular level are unclear. Herein, pomace tannin fractions (PTFs) were isolated post-winemaking. Nuclear magnetic resonance spectroscopy and mass spectrometry analysis showed PTFs were composed of (epi)catechin gallate and (epi)catechin as terminal and extensional units, with polymerization degrees of 10, 16, and 35. In vivo studies demonstrated that PTFs rem

Impact of Evanescence Process on Three-Dimensional Sub-Diffusion-Based Molecular Communication Channel.

In most of the existing works of molecular communication (MC), the standard diffusion environment is taken into account where the mean square displacement (MSD) of an information molecule (IM) scales linearly with time. On the contrary, this work considers the sub-diffusion motion that appears in crowded and complex (porous or fractal) environments (movement of the particles in the living cells) where the particle's MSD scales as a fractional order power law in time. Moreover, we examine an additional evanescence process resulting from which the molecules can degrade before hitting the boundar

iASPP protects the heart from ischemia injury by inhibiting p53 expression and cardiomyocyte apoptosis.

Currently, there remains a great need to elucidate the molecular mechanism of acute myocardial infarction in order to facilitate the development of novel therapy. Inhibitor of apoptosis-stimulating protein of p53 (iASPP) is a member of the ASPP family proteins and an evolutionarily preserved inhibitor of p53 that is involved in many cellular processes, including apoptosis of cancer cells. The purpose of this study was to investigate the possible role of iASPP in acute myocardial infarction. The protein level of iASPP was markedly reduced in the ischemic hearts in vivo and hydrogen peroxide-exp

Serum proteases prevent bacterial biofilm formation: role of kallikrein and plasmin.

Biofilm formation is a general strategy for bacterial pathogens to withstand host defense mechanisms. In this study, we found that serum proteases inhibit biofilm formation by Neisseria meningitidis, Neisseria gonorrhoeae, Haemophilus influenzae, and Bordetella pertussis. Confocal laser-scanning microscopy analysis revealed that these proteins reduce the biomass and alter the architecture of meningococcal biofilms. To understand the underlying mechanism, the serum was fractionated through size-exclusion chromatography and anion-exchange chromatography, and the composition of the fractions that
