Preliminary results of primary chemotherapy in retinoblastoma.

BACKGROUND: Eye retention rates after external beam therapy for advanced retinoblastoma are close to 90% with very few ocular side effects like radiation retinopathy or radiation induced optic neuropathy. Late complications, however, comprise a sixfold increase of secondary non-ocular cancers like soft tissue sarcomas demanding alternative treatment options in the management of advanced retinoblastoma. PURPOSE: A phase I study on primary chemotherapy using vincristine, cyclophosphamide, etoposide and carboplatin in advanced bilateral retinoblastoma was undertaken to evaluate the potential of

Comprehensive prediction of immune microenvironment and hot and cold tumor differentiation in cutaneous melanoma based on necroptosis-related lncRNA.

As per research, causing cancer cells to necroptosis might be used as a therapy to combat cancer drug susceptibility. Long non-coding RNA (lncRNA) modulates the necroptosis process in Skin Cutaneous Melanoma (SKCM), even though the precise mechanism by which it does so has yet been unknown. RNA sequencing and clinical evidence of SKCM patients were accessed from The Cancer Genome Atlas database, and normal skin tissue sequencing data was available from the Genotype-Tissue Expression database. Person correlation analysis, differential screening, and univariate Cox regression were successively u

Unlocking beta cell health: The clinical potential of extracellular vesicles in type 1 diabetes.

BACKGROUND: Type 1 diabetes (T1D) is a lifelong autoimmune disease characterised by progressive immune-mediated destruction of insulin-producing beta (β)T1D-cells, leading to permanent insulin dependence and increased risk of microvascular and macrovascular complications. Despite advances in autoantibody screening and immunotherapies, major clinical challenges persist in early detection, accurate disease staging, prediction of progression and monitoring of therapeutic response. Current biomarkers provide limited insight into real-time β-cell stress and immune activity, restricting opportunitie

Clinical Application of Inflammatory Vital Pulp Therapy in Immature Permanent Teeth: A Retrospective Case Series.

BACKGROUND: Management of immature permanent teeth with pulpal or periapical pathology remains challenging due to incomplete root development and open apices. Conventional treatments, such as calcium hydroxide apexification, require multiple appointments over extended periods and may weaken dentinal walls, whereas apical barrier techniques using mineral trioxide aggregate (MTA) provide apical closure but do not promote continued root development. Inflammatory vital pulp therapy (IVPT) has emerged as a biologically based approach that preserves stem cells from the apical papilla (SCAPs) to prom

Collagen remodeling in breast cancer progression: from molecular mechanisms to diagnostic and therapeutic opportunities.

Collagen remodeling is an emerging hallmark of breast cancer progression, offering profound insights into tumor biology and therapeutic vulnerability. While breast cancer has long been understood through the lens of genetic mutations and cellular deregulation, recent advances have emphasized the crucial role of the tumor microenvironment (TME), particularly the extracellular matrix (ECM), in directing cancer progression. As the most prevalent structural protein within the ECM, collagen plays a key role in tissue mechanics, cell signaling, and immune regulation. This review examines the current

Tailoring pH-responsive charge-conversional interfaces in ionizable lipid-based nanoliposomes for synchronized co-delivery and synergistic therapy of triple-negative breast cancer.

Combination chemotherapy is often limited by the distinct physicochemical properties and intracellular behaviors of individual drugs. This limitation leads to poorly coordinated delivery in tumor tissues. In this study, pH-responsive ionizable nanoliposomes were developed using the ionizable lipid DLin-DMA to co-deliver erlotinib and doxorubicin hydrochloride. This approach introduces a novel spatial and temporal synchronization strategy for two mechanistically distinct drugs within a single carrier. The resulting nanoliposomes formed stable nanoscale assemblies with an optimized particle size

Influence of Genetic and Epigenetic Factors on Periodontitis Susceptibility and Treatment Outcomes.

Periodontitis (PD) is a complex inflammatory condition driven by the interplay of microbial, genetic, epigenetic, and environmental factors. While bacterial biofilms are regarded as the primary cause, growing evidence underscores the significant role of genetic predisposition in determining susceptibility to the disease, its progression, and treatment outcomes. This review explores the genetic polymorphisms of PD, focusing on genes related to inflammatory mediators, immune responses, antimicrobial peptides, matrix metalloproteinases, and vitamin D receptor pathways. Key epigenetic mechanisms i

Nanoparticle-integrated smart hydrogel with H2S-triggered antibacterial escalation and sustained oxygen release for periodontal regeneration.

Periodontitis is a widespread chronic inflammatory disease characterized by pathogenic bacterial biofilms and a hypoxic microenvironment, which together impede effective treatment and tissue regeneration. To overcome these challenges, we developed an injectable, hydrogen sulfide (H2S)-responsive nanocomposite hydrogel, termed PDA@Hb/NOCG, for microenvironment-adaptive therapy. This hydrogel was formulated by incorporating hemoglobin-polydopamine nanoparticles (PDA@Hb NPs) as an oxygen-generating element and tetra-armed azide-terminated polyethylene glycol as an H2S-scavenging component into a

Current Mechanobiological Pathways and Therapies Driving Spinal Health.

Spinal health depends on the dynamic interplay between mechanical forces, biochemical signaling, and cellular behavior. This review explores how key molecular pathways, including integrin, yeas-associated protein (YAP) and transcriptional coactivator with PDZ-binding motif (TAZ), Piezo, and Wingless/Integrated (Wnt) with β-catenin, actively shape the structural and functional integrity of spinal tissues. These signaling mechanisms respond to physical cues and interact with inflammatory mediators such as interleukin-1 beta (IL-1β), interleukin-6 (IL-6), and tumor necrosis factor alpha (TNF-α),

Distribution of macrophages in the developing cochlea of the common marmoset, a primate model animal.

INTRODUCTION: Macrophages are essential immune cells in the cochlea that contribute to inflammation, tissue repair, and homeostasis. They also play an important role in local cochlear immunity. The developmental immigration and maturation of macrophages in the cochlea have been investigated and are considered essential for normal hearing acquisition. Most of our current knowledge regarding cochlear development is based on rodent models because of the ethical challenges of using human fetal samples for research. However, inter-species differences between rodents and humans have been reported. I

Energy Substrate Transporters in High-Grade Ovarian Cancer: Gene Expression and Clinical Implications.

Ovarian cancer is a non-homogenous malignancy. High-grade serous carcinoma (HGSC) is the most common subtype, and its drug resistance mechanisms remain unclear. Despite the advantages of modern pharmacotherapy, high-grade ovarian cancer is associated with a poor prognosis and research into targeted therapies is in progress. The aim of the study was to assess the dominant energy substrate transport mechanism in ovarian cancer cells and to verify whether genomic aberrations could predict clinical outcomes using the Cancer Genome Atlas (TCGA) dataset. Total RNA was extracted from HGSC frozen tiss

Mutation of Hashimoto's Thyroiditis and Papillary Thyroid Carcinoma Related Genes and the Screening of Candidate Genes.

Clinical studies have shown similarities in the genetic background and biological functional characteristics between Hashimoto's thyroiditis (HT) and papillary thyroid carcinoma (PTC), and that HT may increase risks of PTC. Here, we set to determine the gene expression specificity of HT and PTC by screening related genes or co-expressed genes and exploring their genetic correlation. Referencing the Oncomine database, HT-related genes were discovered to be expressed in many different types of thyroid cancer, such as TSHR that is highly expressed in thyroid cancer. An in-depth genetic analysis a

Defining recovery neurobiology of injured spinal cord by synthetic matrix-assisted hMSC implantation.

Mesenchymal stromal stem cells (MSCs) isolated from adult tissues offer tangible potential for regenerative medicine, given their feasibility for autologous transplantation. MSC research shows encouraging results in experimental stroke, amyotrophic lateral sclerosis, and neurotrauma models. However, further translational progress has been hampered by poor MSC graft survival, jeopardizing cellular and molecular bases for neural repair in vivo. We have devised an adult human bone marrow MSC (hMSC) delivery formula by investigating molecular events involving hMSCs incorporated in a uniquely desig

The renin-angiotensin-aldosterone system (RAAS) - physiology and molecular mechanisms of functioning.

Secretion of renin juxtaglomerular cells into bloodstream initiates activation of an enzymatic-hormonal cascade known as the RAAS (renin - angiotensin - aldosterone system). As a result, blood pressure is increased by the means several interrelated mechanisms. Mechanism of Zjednoczoaction of this system has been known for decades, but a few previously unknown components were recently added, such as ACE-2 and Ang(1-7), and their role often seems to be opposite to that of the conventional components. Local tissue systems also have important biological functions. They operate largely independentl
