In-depth characterization of stem cell potency and genotoxicity for clinical-scale ex vivo CRISPR-Cas9 gene editing.

Translating CRISPR-Cas9-based homology-directed repair (HDR) strategies into clinical application remains a major challenge due to limited standardization, concerns over safety, and efficacy issues. Here, we present a comprehensive and clinically compliant preclinical framework for the ex vivo correction of Wiskott-Aldrich syndrome (WAS) using a CRISPR-Cas9-AAV6 platform targeting hematopoietic stem and progenitor cells (HSPCs). In this study, we established a clinical-compatible platform enabling large-scale manufacturing while preserving HSPC viability, stemness, and multilineage functionali

Missense mutations in the SNCA gene: Molecular mechanisms and clinical implications.

The SNCA gene on chromosome 4 encodes the alpha-synuclein (αSyn) protein, which plays a central role in the pathogenesis of synucleinopathies, including Parkinson's disease (PD), dementia with Lewy bodies (DLB), and multiple system atrophy (MSA). While αSyn has established roles in synaptic vesicle dynamics and neuronal signaling, alterations in SNCA regulation and sequence contribute to protein misfolding, aggregation, and loss of function. Alterations in secondary and tertiary structure, as well as protein aggregation, affect biochemical interactions, ultimately leading to pathogenesis. This

Characterization and Evaluation of the Pathogenicity of a Novel Alpha Family Chaperone-Usher Pilus Present on Porcine Enterotoxigenic Escherichia coli.

Enterotoxigenic Escherichia coli (ETEC) is a leading cause of diarrhea in piglets, imposing significant economic burdens on swine production worldwide. Despite its impact, detailed characterization of intestinal colonization and comprehensive genomic profiling of virulent neonatal porcine ETEC strains remain limited. In this study, we isolated a β-hemolytic ETEC strain, PT4357, from a diarrheic neonatal piglet and conducted whole-genome sequencing (WGS) coupled with experimental infection in neonatal piglets to define its pathogenicity. The PT4357 belonged to serotype O8:H7 and sequence type S

Conventional In Vitro Fertilization and Intracytoplasmic Sperm Injection.

In vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI) are assisted reproductive technologies used to help individuals or couples conceive a child. IVF involves combining an oocyte and sperm outside the body in a laboratory to create embryos. Following ovarian stimulation, oocytes retrieved through a minor surgical procedure are mixed with sperm in the laboratory. If fertilization occurs, resulting embryos are cultured for several days, and one (or more) embryo(s) is (are) then transferred into the uterus (either "fresh" or after cryopreservation) in hopes of implantation and

Chromosome-Scale Genome Analysis Reveals Locus-Specific Disruption of the Citrinin-Associated Region in a Furu-Derived Monascus ruber Strain BC20.

Monascus species are widely used in traditional fermented foods for pigment and flavor formation, but citrinin contamination remains a major safety concern that limits broader food applications. Therefore, this study aimed to evaluate the citrinin risk of a furu-derived Monascus ruber strain, BC20, by integrating phenotypic screening across food-relevant matrices with genome-resolved analysis. After 14 days of cultivation across eight matrices, including fungal media as well as dairy-, cereal-, and bran-based substrates, citrinin was not detected by immunoaffinity cleanup combined with HPLC-FL

Enhancement Effects of α4/α6-Targeted Inhibitors on Eg5-Microtubule Interaction.

Eg5 is one kind of kinesin motor that participates in various cellular processes, especially in mitosis. The tetrameric structure of Eg5 can crosslink the antiparallel microtubules and generate forces on microtubules to separate chromosomes through walking on the microtubules. Inhibition of the activity of Eg5 leads to cell cycle arrest and apoptosis. Thus, Eg5 is a potential therapeutic target in anticancer drug development. Two inhibitor-binding sites have been discovered to date. The α2/L5/α3-targeted inhibitors can inhibit the chemical cycle of Eg5. However, drug resistance to the α2/L5/α3

Molecular genomic insights into melanoma associated proteins PRAME and BAP1.

Melanoma, a globally prevalent skin cancer with over 325,000 new cases annually, necessitates a comprehensive under- standing of its molecular components. This study looks at the PRAME (cutaneous melanoma-associated antigen) and BAP1 (gene controlling gene-environment interactions) proteins. Both PRAME and BAP1 are associated with critical genomic alterations that significantly influence melanoma progression and patient outcomes. PRAME is overexpressed in various cancers, especially uveal melanoma (UM), where high levels correlate with poor prognosis and genomic instability linked to chromosom

Arabidopsis Antiporter Genes as Targets of NO Signalling: Phylogenetic, Structural, and Expression Analysis.

Nitric oxide is a gaseous signalling molecule produced by plants. Slight changes in endogenous NO levels have significant biochemical and physiological consequences. We investigated the structural and functional properties of NO-responsive antiporter genes in Arabidopsis thaliana. Phylogenetic analysis of 50 antiporter genes classified them into four subgroups based on the presence of NHX and CPA domains and the evolutionary similarity of the protein sequences. Antiporters were found scattered across the five chromosomes with unique physico-chemical properties and subcellular localisation in t
