METTL3-m6A-STAT1/NF-κB axis: a key switch for the immunosuppressive capacity of mesenchymal stem cells.

BACKGROUND: Owing to their immunosuppressive nature, mesenchymal stem cells (MSCs)—a type of multipotent stem cell—hold considerable promise for therapeutic applications. Functioning as the primary catalytic subunit of the RNA N6-methyladenosine (m6A) methyltransferase complex, methyltransferase-like 3 (METTL3) plays extensive roles in numerous biological processes. It is hypothesized that METTL3 participates in governing the immunomodulatory functions inherent to MSCs. However, the exact mechanisms governing METTL3’s control over MSCs’ immunosuppressive capacity are poorly defined. METHODS: M

Chemical Precursors of Flocs in Sweetened Beverages: Mechanisms of Formation, Analytical Methods, and Industrial Strategies.

Flocs, visible particles formed in sugar-sweetened beverages, reduce clarity and consumer acceptance of products. Their presence can be caused not only by different types of trace impurities in the sugar but also by interactions among beverage components. In this review, scientific reports on acid beverage flocs (ABFs) and alcohol flocs are summarized, the main pathways for their formation are described, and practical options for detecting them and preventing their formation in beverages are compiled. Isoquinoline alkaloids enhance growth performance through multifaceted modulation of the bacterial-fungal microbiome, CAZyme profiles, gut health, and neuroendocrine function in broilers.

The bacterial-fungal microbiome and its carbohydrate-active enzyme (CAZyme) capacity play critical roles in regulating gut health and growth performance in broiler chickens. This study evaluated the effects of dietary isoquinoline alkaloids (IQ) on growth performance, gut microbiome composition, CAZyme profiles, and the microbiome-gut-neuroendocrine axis in broilers. A total of 400 Ross 308 (1-day-old) chicks were randomly assigned to either a Basal diet (CON) or IQ supplemented diet (IQ). Dietary IQ supplementation significantly increased final body weight and cumulative body weight gain (P <

Modulation of experimental colitis by Enterococcus faecalis, Enterococcus faecium, and Bacillus subtilis: insights from infectious and chemical models.

The roles of Enterococcus faecium (E. faecium), Enterococcus faecalis (E. faecalis), and Bacillus subtilis (B. subtilis) in colitis remain poorly defined, with existing literature presenting inconsistent and often contradictory evidence. This study aimed to elucidate their contributions to inflammatory bowel disease (IBD) using both infectious (non-typhoidal Salmonella enterica serovar Typhimurium, S. typhimurium) and chemical (dextran sulfate sodium, DSS)-induced colitis models. Colitis was induced in C57BL/6 mice via a single oral dose of S. typhimurium (ATCC14028) and/or administration of D

Differentiation and functionality of human bronchial epithelial cells in an air-liquid interface culture are modified by irradiation exposure.

INTRODUCTION: We aimed to investigate the effects of α-particle and X-ray irradiation on a human bronchial epithelium model, representing environmentally and medically relevant exposure. Our focus was on non-cancer outcomes, namely mucociliary transport (MCT) and epithelial barrier function, both of which are crucial for cancer risk assessment and therapeutic efficacy. MATERIALS AND METHODS: Basal stem cells were irradiated and terminally differentiated under air-liquid interface conditions into all epithelial cell types. Clonogenic survival assays were used to determine iso-effective doses.

Compositional analysis and physicochemical characterization of a purified tremella fuciformis polysaccharide-rich fraction and its intestinal barrier- and microbiota-related effects in DSS-induced colitis.

Tremella fuciformis is a widely consumed edible fungus and a source of structurally diverse polysaccharides, but the intestinal biofunctionality of a purified Tremella polysaccharide-rich fraction remains insufficiently defined. In this study, a purified T. fuciformis polysaccharide-rich fraction (TPS) was obtained by hot-water extraction, ethanol precipitation, anion-exchange chromatography, and gel filtration. TPS was characterized using carbohydrate/protein analysis, gel permeation chromatography (GPC), monosaccharide composition analysis, Fourier-transform infrared spectroscopy (FTIR), X-r

SR9009 regulates macrophage polarization via the NR1D1/NF-κB axis to ameliorate obesity-associated ulcerative colitis.

OBJECTIVE: The incidence of ulcerative colitis (UC) and obesity has risen in recent years, potentially linked through metabolic dysregulation and chronic inflammation. The nuclear receptor NR1D1 is pivotal in regulating circadian rhythms and plays a significant role in inflammation and metabolism. This study investigates the therapeutic effects and mechanisms of the NR1D1 agonist SR9009 on obesity-related UC. METHODS: We established a mouse model of obesity-induced UC utilizing a high-fat diet alongside dextran sulfate sodium (DSS). 32 male C57BL/6 mice were divided into four groups: control

Ocular Tissue Distribution of Ocular Surface Immunoglobulin Following Topical and Subconjunctival Administration: A Novel Strategy for Mitigation of Antibody-Drug Conjugate-Associated Ocular Toxicity.

PURPOSE: Antibody-drug conjugate (ADC)-associated ocular toxicity is an emerging, sometimes dose-limiting complication. Macropinocytosis has been implicated in off-target ADC uptake by corneal epithelial cells. This study evaluated whether ocular surface immunoglobulin (OSIG) inhibit macropinocytosis in human corneal epithelial cells (HCE-T) and characterized ocular biodistribution and tolerability following topical and subconjunctival administration. METHODS: Macropinocytosis was quantified in corneal epithelial cells after exposure to vehicle or increasing concentrations of OSIG. Ocular pha

Cold-Induced Elevation of 3-Hydroxypropionate Exacerbates Colitis by Remodeling Gut Microbiota and Impairing Mitochondrial Respiration in Intestinal Epithelial Cells.

BACKGROUND/OBJECTIVES: Inflammatory bowel disease (IBD) is a chronic gastrointestinal disorder influenced by environmental factors including cold stress. While cold exposure exacerbates intestinal inflammation, the specific microbial metabolites linking environmental stress to colitis remain unclear. 3-Hydroxypropionate (3-HPA) is a gut microbial metabolite elevated following cold exposure, but its pathogenic role in intestinal inflammation has not been investigated. This study aimed to determine whether 3-HPA contributes to colitis progression and to characterize its effects on gut microbiota

Electronic cigarette aerosols disrupt airway barrier function via MMP-dependent E-cadherin cleavage: findings from cell culture and murine models.

Electronic cigarette (e-cigarette) use continues to rise, yet the toxicological mechanisms by which inhaled aerosols impair airway epithelial integrity remain poorly defined. E-cadherin, a key adherens junction protein required for epithelial cohesion, can undergo proteolytic cleavage to generate soluble E-cadherin (sE-cad), a mediator of epithelial barrier dysfunction and inflammation. We hypothesized that e-cigarette aerosol exposure promotes MMP-dependent E-cadherin cleavage, resulting in sE-cad release and epithelial barrier disruption. Using differentiated primary normal human bronchial e

The protein extracted from fermented Zanthoxylum bungeanum seeds mitigates DSS-induced ulcerative colitis by orchestrating inflammatory cytokine modulation, counteracting oxidative stress, and safeguarding gut microbiota homeostasis.

Zanthoxylum bungeanum seeds (ZBS) are protein-rich by-products with limited high-value utilization, and fermentation may provide an effective strategy to improve the functional properties and bioactivity of their proteins. In this study, fermented ZBS protein (FZP) was prepared, and its physicochemical properties, antioxidant activity, gastrointestinal digestibility, and protective effects against colitis were systematically investigated. FZP exhibited a high purity of 94.37% and was predominantly composed of proteins with molecular weights ranging from 15 to 25 kDa. Ultrafiltration revealed t

Persisting Region-Specific Microglia Activation and Synaptic Imbalance During Remission in a DSS-Induced Colitis Model.

Ulcerative colitis (UC) patients experience cycles of active gut inflammation and remission, with neuropsychiatric comorbidities persisting even during clinical remission. While the dextran sulfate sodium (DSS) mouse model was previously applied to study gut-brain interactions, those studies focused on acute protocols missing the chronic, relapsing-remitting nature of human UC, when patients continue to experience central nervous system symptoms. Thus, we employed a chronic DSS treatment regimen comprising three cycles of active intestinal inflammation followed by remission phases to investiga

The gut-brain axis mediates precocious puberty induced by environmentally relevant low-dose endocrine-disrupting chemical mixtures.

BACKGROUND: The global rise in precocious puberty (PP) is increasingly linked to exposure to endocrine-disrupting chemicals (EDCs). However, the mechanisms by which environmentally relevant, low-dose mixtures of EDCs influence PP remain inadequately explained by direct endocrine disruption. OBJECTIVE: This systematic review evaluates a novel hypothesis: that disruption of the gut-brain axis (GBA) serves as a pivotal mechanism in EDC mixture-induced PP. METHODS: We synthesized evidence from 87 studies (45 human, 32 animal, 10 in vitro) following PRISMA 2020 guidelines. An exploratory Random F

Membrane Raft Redox Signaling Mediates Trimethylamine N-Oxide-Induced NLRP3 Inflammasome Activation and Endothelial Dysfunction.

Trimethylamine-N-oxide (TMAO), a gut microbiota-derived metabolite, is a known risk factor for cardiovascular disease. We previously showed that TMAO induces NLRP3 inflammasome activation and contributes to endothelial dysfunction. However, the upstream mechanisms linking TMAO to inflammasome activation and barrier injury remain unclear. In the present study, we examined whether membrane raft (MR) redox signaling contributes to TMAO-induced NLRP3 inflammasome activation and endothelial dysfunction. MR clustering, colocalization of MRs with NADPH oxidase subunits, inflammasome formation, and ju

Hemp seed oil ameliorates DSS-induced ulcerative colitis in mice by altering transcriptomics and metabolomics.

Ulcerative colitis (UC) is a chronic relapsing inflammatory bowel disease with complex pathogenesis and limited treatments. Hemp seed oil, with its unique ω-6/ω-3 polyunsaturated fatty acid ratio and phenolic antioxidants, exhibits anti-inflammatory and gut barrier-enhancing properties, though its mechanisms in UC are not fully understood. Here, we applied an integrated transcriptomics and metabolomics approach to investigate the effects of hemp seed oil in dextran sulfate sodium (DSS)-induced UC mice. Treatment with hemp seed oil (1 g/kg) significantly alleviated disease activity, suppressed
