Chronic actinic keratopathy--a condition associated with conjunctival elastosis (pingueculae) and typified by characteristic extracellular concretions.

Morphologic observations on a peculiar type of corneal reaction with a predisposition for the superficial stroma of the interpalpebral portion of the cornea are reviewed. Histochemical evidence is provided which indicates that the corneal concretions, though not homogenous, are proteinaceous in nature and contain amino acids not normally detectable in the cornea. The corneal concretions were associated with conjunctival elastosis (pingueculae) in all 22 instances in which the eyes were sectioned in the horizontal plane. Identical concretions were identified within these associated pingueculae,

Distinct extracellular architectures shape mechanical persistence in GPVI and CLEC-2 receptor assemblies.

Glycoprotein VI (GPVI) and C-type lectin-like receptor 2 (CLEC-2) are platelet-activating receptors whose extracellular organization contributes to receptor association and signaling during thrombus formation, yet how alternative receptor architectures influence mechanical persistence remains unclear. We used molecular dynamics to compare a GPVI dimer based on 2GI7, a domain-swapped GPVI assembly derived from 7NMU, and a crystallographic CLEC-2 contact using the same pulling protocol and constant-force conditions. During the first 5 nm of imposed separation, reconstructed GPVI required less fo

Evolution, structural diversity and functional plasticity of plant derived multifunctional proteases and α-amylases inhibitors.

Plant derived multifunctional inhibitors (MFIs) are an important class of plant defense proteins that disrupt insect digestion by simultaneously impairing protein and carbohydrate catabolism. With the increasing knowledge of inhibitors of plant enzymes, MFIs have become a separate functional group. In this review we propose an extended definition that encompasses the classical inhibitors acting on structurally unrelated enzymes and also inhibitors acting on enzymes of the same substrate pathway, e.g. α-amylase/α-glucosidase and trypsin/chymotrypsin systems. Most of the MFIs consist of single p

Structure and functional properties of Dendrobium officinale polysaccharide-hemp seed protein gels for dysphagia diets as regulated by alkali concentration and temperature.

Dysphagia is highly prevalent among older adults and increases the risks of malnutrition, dehydration, and aspiration pneumonia, highlighting the need for safe and nutritionally adequate texture-modified foods. Polysaccharide-protein gels are promising candidates for this purpose, but precise control of their rheological properties and structural stability remains challenging. Here, we systematically investigated how alkali concentration (Na2CO3, 0.5-2.5%, w/v) and thermal treatment (25-85 °C) regulate the gelation, structure, and functional performance of Dendrobium officinale polysaccharide-

Cascade-responsive HA-decorated liposome/mesoporous silica nanoplatform co-delivering BMF-219 and 5-FU enhances gastric cancer chemotherapy through Menin- and p53-associated apoptotic signaling.

BACKGROUND: Our preliminary study found that Men1 is an oncogene and a potential therapeutic target in gastric cancer (GC). Chemotherapy remains a common therapeutic regimen for GC, but is frequently limited by multidrug resistance and poor intratumoral coordination of combination drugs. Targeting Menin may enhance chemotherapy sensitivity, but efficient co-delivery of Menin inhibitors and chemotherapeutics is hindered by the profound spatiotemporal pharmacokinetic decoupling of the drugs in vivo. METHODS: We first evaluated the synergistic antitumor effect and Menin/p53-associated mechanism

Lower mitochondrial DNA copy number in ATTRV50M Swedish carriers.

BACKGROUND: Hereditary transthyretin (TTR) amyloidosis caused by TTR p.Val50Met (ATTRV50M) is a progressive systemic disease caused by misfolding of the protein transthyretin (TTR). The conversion of TTR into an amyloidogenic state has recently been found to be strongly influenced by a non-native disulfide bond, thus also a redox environment. Variation in mitochondrial DNA (mtDNA) copy number serves as an indirect marker of redox imbalance and mitochondrial dysfunction. MtDNA is moreover strictly maternally inherited, supporting previous findings where maternal inheritance of the pathogenic va

Staged structural changes of fermented chicken breast myofibrillar protein: Conformational unfolding and aggregation.

Fermented chicken breast is promoted as a healthier alternative to traditional high-fat fermented meat products, yet its quality depends critically on the structural state of myofibrillar protein (MP). To clarify how MP deteriorates during extended natural fermentation, this study established a 150-day fermentation model and systematically monitored MP degradation, oxidation, conformational change, and aggregation behavior. Partial least squares regression (PLSR) and hierarchical cluster analysis (HCA) were combined to relate structural transitions and aggregation. SDS-PAGE showed pronounced d

Vaccarin Improves Myocardial Ischemia-Reperfusion Injury by Attenuating Oxidative Stress and Ferroptosis Through Reducing NOX4-Modulated JAK2/STAT3 Pathway Activation.

Vaccarin is a naturally occurring flavonoid glycoside with cardioprotective properties; however, its involvement in myocardial ischemia-reperfusion (MI/R) injury remains poorly defined. We herein investigated whether vaccarin protects cardiomyocytes from MI/R-induced dysfunction in vivo and in vitro and examined the associated molecular mechanisms. To explore the protective effects of vaccarin, we established an MI/R injury mouse model and in vitro hypoxia/reoxygenation (H/R) injury model using H9c2 cardiomyocytes. In vivo results showed that vaccarin alleviated MI/R injury. In H9c2 cells, vac

Water-induced supramolecular self-assembly of porphyrin integrated with multivalent metal-organic framework for enhanced electrochemiluminescence and ultrasensitive aptasensing of 4-fluoromethamphetamine.

Aggregation-caused quenching (ACQ) and poor aqueous solubility severely restrict electrochemiluminescence (ECL) applications of polycyclic aromatic hydrocarbons (PAHs). Herein, a water-induced supramolecular self-assembly strategy is proposed to address these drawbacks. Via a simple reprecipitation route, representative PAH meso-tetrakis(4-carboxyphenyl)porphyrin (TCPP) forms ordered supramolecular aggregates driven by noncovalent interactions, which suppress π-π stacking and ACQ and achieve a low triggering potential of - 1.0 V. Iron-based MOF supported molybdenum disulfide (MIL-101@MoS2) is

Engineered eMutS based magnetic separation system for high-fidelity DNA synthesis.

DNA synthesis fidelity remains a critical bottleneck in large-scale gene assembly. Although conventional post-synthetic purification methods (e.g., HPLC, PAGE) and enzymatic mismatch-cleavage strategies offer partial solutions, they often suffer from limited throughput, high cost, and incompatibility with automated workflows. To overcome these limitations, we developed an integrated, sensitive and high-throughput error-correction platform that combines a rationally engineered Escherichia coli MutS protein (eMutS) with a biotin-streptavidin magnetic-bead separation system (eMBS). Specifically,

Molecular dynamics simulations elucidate the misfolding mechanisms of secretion-defective pancreatic lipase variants.

Protein misfolding of pancreatic secretory enzymes is increasingly recognized as a contributor to chronic pancreatitis. Recently, missense pancreatic lipase (PNLIP) mutations that impair lipase secretion and cause intracellular aggregation and endoplasmic reticulum (ER) stress have been linked to the disease. Among these, A174P, G233E, and C254R are located in the N-terminal domain, whereas V454F is situated in the C-terminal domain of PNLIP. To elucidate how these mutations disrupt PNLIP structure, we performed detailed molecular dynamics simulations and structural modeling of human PNLIP var

RGR-Mediated Photopic Visual Cycle and Oxidative Stress: Potential Mechanisms for Cone Vision Impairment and Retinal Degeneration in Retinitis Pigmentosa Linked to D1080N-IRBP.

Interphotoreceptor retinoid-binding protein (IRBP), an extracellular glycoprotein, supports cone-mediated vision through unclear molecular mechanisms. Over 30 different mutations in the IRBP gene have been found in patients with retinitis pigmentosa (RP), childhood-onset retinal dystrophy with high myopia, and cone-rod dystrophy, yet their pathogenicity and underlying pathogenic mechanisms have not been studied in animal models. Here, we found that extracellular IRBP significantly increased the quantities of the extracellular, but not intracellular, 11-cis-retinol and 11-cis-retinal synthesize

Follicular fluid oxidative stress and antioxidant defense in relation to maternal age, ovarian reserve, and in vitro fertilization outcomes.

Oxidative stress within the follicular microenvironment plays a critical role in oocyte quality and reproductive outcomes. This prospective study examines the association between follicular fluid redox status, maternal age, ovarian reserve, and clinical outcomes in women undergoing in vitro fertilization. Follicular fluid samples were obtained from women undergoing fresh embryo transfer cycles and analyzed for oxidative damage markers (malondialdehyde (MDA), nitric oxide, ischemia-modified albumin, and sialic acid) and antioxidant parameters (superoxide dismutase (SOD), catalase (CAT), glutath

Deamidation Promotes AGE-Modifications in Human Lens γS-Crystallin.

Deamidation and advanced glycation end products (AGEs) are among the major post-translational modifications (PTMs) in eye lens proteins. Due to deamidation and AGE formation, proteins may aggregate and scatter light, contributing to lens aging and cataract formation. So far, the relation between the two PTMs remains poorly understood. γS-crystallin (γSC), a major subtype of γ-crystallin, undergoes significant modifications through deamidation, especially at the surface-exposed asparagine residues, N14, N76, and N143. In this study, deamidation of γSC was mimicked by mutating asparagine residue

Modulation of apolipoprotein E receptor-2 by ApoE4, amyloid β-peptide, reelin, and secreted amyloid precursor protein: a common point of impact in Alzheimer's disease pathogenesis.

INTRODUCTION: Apolipoprotein E (ApoE), reelin, and several other proteins bind ApoE-receptor 2 (apoER2), distinguished from other members of its receptor family by signal transduction which enhances the activity of N-methyl D-aspartate (NMDA) receptors. Evidence indicates that this signal transduction depends upon apoER2 forming dimers or other high-order clusters. It seems noteworthy therefore that protein products of major APOE gene variants differ in their numbers of cysteines capable of forming disulfide dimers, with the allele (ε4) associated with highest rates of Alzheimer's disease (AD)

Aberrant Protein S-Nitrosylation Mimics the Effect of Rare Genetic Mutations in Neurodegenerative Diseases.

Neurodegenerative disorders such as Alzheimer's disease (AD), Parkinson's disease/Lewy body dementia (PD/LBD), and amyotrophic lateral sclerosis/frontotemporal dementia (ALS/FTD) are driven by complex interactions of genetic and environmental factors. While genome wide association studies (GWAS) have uncovered a number of risk gene variants (e.g., APOE, SNCA [encoding α-synuclein], and protein disulfide isomerase [PDI]), these genetic factors alone cannot fully explain disease onset or progression. Emerging evidence suggests that post-translational modifications of proteins, particularly S-nit

The C117D oxidation mimic reveals the monomeric structure of SARS-CoV-2 main protease.

The SARS-CoV-2 main protease (Mpro) is essential for viral replication and functions as a homodimer, with dimerization being critical for catalytic activity. Mpro contains an unusually high number of cysteine residues. Among these, C117 and the catalytic nucleophile C145 can form a reversible disulfide bond under oxidative conditions. To investigate the structural and functional consequences of irreversible oxidation of these residues, we generated oxidation mimics by substituting these residues with aspartate (C117D and C145D), to mimic the sulfinic acid oxidation state. Kinetic assays reveal

Polyoxometalate-derived WSSe nanoparticles confined in carbon nanofibers for fast and durable lithium storage.

Tungsten disulfide is a promising anode material for lithium-ion batteries (LIBs) owing to its high theoretical capacity, natural abundance, and environmental benignity, but its practical implementation is constrained by sluggish charge-transfer kinetics, large cycling-induced volume variation, and rapid capacity fading. Herein, we report a synergistic heteroanion substitution and carbon confinement strategy to stabilize the electrode architecture by encapsulating selenium-substituted WSSe nanoparticles in nitrogen/phosphorus-codoped porous carbon nanofiber (WSSe@NP-CNF) via a polyoxometalate-

Protein Phase Transition Engineering of Nanochannels: A Stepwise Functionalization Strategy for Sequential Ultrasensitive Metal Ion Detection.

Water-soluble heavy metal ions pose persistent threats to ecosystems and human health, necessitating highly sensitive and selective detection strategies.
