The genetic program at the root of the biological stock exchange.

Beneath the gardens, farmlands and forest floors that surround us, a hidden world blooms in careful cooperation and intense competition. The mutualistic symbiosis of the thread-like hyphae of fungi and plant roots (collectively termed mycorrhizae from the Greek mýkēs - meaning 'fungus', and rhiza - for 'root') is present in the vast majority of plant species. As with most intimate relationships, this symbiosis functions on a principle of 'give and take'. As an autotroph, the plant is able to synthesize all the sugars it requires through photosynthesis; however, its immobility hinders its capac

Proteomic diversity in a prevalent human-infective Giardia duodenalis sub-species.

Giardia duodenalis a species complex of gastrointestinal protists, with assemblages A and B infective to humans. To date, post-genomic proteomics are largely derived from Assemblage A, biasing understanding of parasite biology. To address this gap, we quantitatively analysed the proteomes of trophozoites from the genome reference and two clinical Assemblage B isolates, revealing lower spectrum-to-peptide matches in non-reference isolates, resulting in significant losses in peptide and protein identifications, and indicating significant intra-assemblage variation. We also explored differential

A complete analysis of Relative Synonymous Codon Usage in HVRs1-4 region in adenovirus genome.

Recent outbreaks of adenovirus (FAdV) infections in poultry flocks have been determined in many countries in Europe, Asia and Australia connected with economic consequences, and loses in poultry production. To better understand the evolution and transmission of FAdV viruses, de- tailed codon usage analysis was performed for 137 recently obtained FAdV strains. A high effec- tive number of codons, and an indication the presence of low codon usage were determined. The presence of mutations, and their influence on codon usage was confirmed by a correlation be- tween nucleotide compositions at the

Morphological, histological and molecular investigations on canine uterine tissue after ovariectomy.

In the present study, we investigated the course of atrophy in canine uterine tissue and the expression of estrogen receptors (ER) and progesterone receptors (PR) within 6 months after ovariectomy (OE). In nine primipar bitches of different breeds, bilateral OE and removal of one horn was performed. Six months after surgery, the remaining uterine tissue was removed. The tissue was examined for signs of inflammation and proliferation, and for expression of ER, PR and Ki67 by means of immunohistochemistry (IHC); furthermore transcription of vascular endothelial growth factor (VEGF), transforming

How a single gene twists a snail.

The gastropod Lymnaea has unique features, that is, chirality, sinistrality, or dextrality, is displayed externally as well as internally, and is hereditary, being determined by a single-locus that functions maternally at the very early embryonic stage. Both sinistral and dextral snails exist in nature with the dextral one being dominant. Thus, the genus Lymnaea is an ideal target for studying chiromorphogenesis. This article gives a brief overview of the current state of research on chiromorphogenesis of Lymnaea (L.) stagnalis, mainly focusing on our own studies. Breeding experiments were per

Influence of stress-induced intermediates on gonadotropin gene expression in gonadotrope cells.

Despite extensive investigation, a comprehensive understanding of the mechanisms whereby stress impacts fertility remains elusive. Since the 1930s, when Hans Selye popularized studying adaptations to stress (Selye, 1937), we have learned that compensatory mechanisms involve a complex interplay of neural and hormonal processes that allow various body functions to adjust to stress, in a coordinated manner. In terms of reproduction, the adjustment to a stressor interferes with integrated functioning at multiple levels of regulation--the hypothalamus, anterior pituitary gland, gonads, and neural c

Steroid metabolism and excretion in anorexia nervosa.

Few organ systems are spared the progressive deterioration seen in critically ill patients with anorexia nervosa. Most of the endocrine disturbances discussed are due to starvation and represent appropriate metabolic adaptation of the body to food restriction and negative energy balance. In a starving patient, a lowered metabolic rate, increased cortisol and growth hormone level and reduced fertility are all appropriate adaptations to an abnormal and highly stressful state. Most metabolic abnormalities can be reversed with a well-planned refeeding program and nutritional recovery.

Babesia bovis contains an abundant parasite-specific protein-free glycerophosphatidylinositol and the genes predicted for its assembly.

Autonomous glycosylphosphatidylinositol (GPI) molecules (also protein-free GPIs or free GPIs) have been reported to be particularly abundant in some parasitic protozoa and mediate strong immunomodulatory effects on the host immune system. In the work at hand we have investigated the existence of free GPIs in Babesia bovis. Comparative thin layer chromatographic analysis of the protein-free glycolipid fraction of in vitro cultured B. bovis merozoites and erythrocyte membranes demonstrated the presence of an abundant parasite-specific band. Its chemical analysis revealed a GPI species containing

Ferritin in autoimmune diseases.

Iron, an essential element for many important cellular functions in all living organisms, can catalyze the formation of potentially toxic free radicals. Excessive iron is sequestered by ferritin in a nontoxic and readily available form in a cell. Ferritin is composed of 24 subunits of different proportions of two functionally distinct subunits: ferritin H and L. The expression of ferritin is under delicate control and is regulated at both the transcriptional and post-transcriptional levels by iron, cytokines, hormones, and oxidative stress. Mutations in the ferritin gene cause the hereditary h

Differential expression of actin modulated by temperature in mycelial and yeast cells of Paracoccidioides brasiliensis.

We used yeast and mycelial forms of Paracoccidioides brasiliensis to evaluate the effect of heat shock stress on actin expression. P. brasiliensis yeasts harvested during the exponential growth phase showed more expression of the actin mRNA when incubated at 40 degrees C than when incubated at 37 degrees C, the usual temperature at which these yeasts grow. In contrast, expression of actin mRNA was lower in yeasts incubated at 25 degrees C than in yeasts incubated at 37 degrees C. Mycelium harvested at 25 degrees C, an approximation of its normal growth temperature, and then exposed to 37 degre

Hemodynamics and the focal origin of atherosclerosis: a spatial approach to endothelial structure, gene expression, and function.

Atherosclerosis originates at predictable focal and regional sites that are associated with complex flow disturbances and flow separations in large arteries. The spatial relationships associated with hemodynamic shear stress forces acting on the endothelial monolayer are considered in experiments that model regions susceptible to atherosclerosis (flow disturbance) and resistant to atherosclerosis (undisturbed flow). Flow disturbance in vitro induced differential expression at the single gene level as illustrated for the intercellular communication gene and protein, connexin 43. Transcription p

Factors involved in leukaemogenesis and haemopoiesis.

This review describes the chromosomal abnormalities in T-cell acute lymphoblastic leukaemia (ALL) which result in the over-expression of the gene SCL, which encodes a helix-loop-helix transcription factor. Also described are how gene targeting studies have revealed a key role for SCL in normal haemopoiesis. Next, the BCR-ABL fusion protein, seen in chronic myeloid leukaemia (CML) and in some patients with ALL, is discussed. Finally, the involvement of members of the core-binding factor (CBF) gene family in leukaemogenesis are described. Members of this gene family are involved in the generatio

[The molecular basis of the virulence of Yersinia].

Pathogenic bacteria of the genus Yersinia (Y. enterocolitica, Y. pseudotuberculosis and Y. pestis) are pathogens of the invasive type. Although they provoke diseases as different as plague and enterocolitis, the molecular bases of their virulence appear to be surprisingly similar: a 70 kb plasmid confers a degree of resistance to the primary immune response and the presence or absence of chromosome encoded functions modulates virulence. The knowledge of the virulence functions that now emerges suggests the conversion of Y. enterocolitica as a life carrier.

[Ectopic calcification in Klotho mice].

We recently developed a unique short lifespan mouse strain in which a single gene mutation caused multiple aging-related disorders and identified the responsible gene as klotho. The most characteristic phenotypes seem to be caused by abnormalities in calcium metabolism. Furthermore the klotho gene is expressed principally in the important tissues for calcium homeostasis such as distal tubule cells of the kidney, choroid plexus in the brain, and the main cells of the parathyroid gland. Klotho plays a critical role for the regulation of calcium and phosphorus homeostasis by negatively regulating

Differential gene expression profiles of gastric cancer cells established from primary tumour and malignant ascites.

Advanced gastric cancer is often accompanied by metastasis to the peritoneum, resulting in a high mortality rate. Mechanisms involved in gastric cancer metastasis have not been fully clarified because metastasis involves multiple steps and requires a combination of altered expressions of many different genes. Thus, independent analysis of any single gene would be insufficient to understand all of the aspects of gastric cancer peritoneal dissemination. In this study, we performed a global analysis of the differential gene expression of a gastric cancer cell line established from a primary main

The common stress responsive transcription factor ATF3 binds genomic sites enriched with p300 and H3K27ac for transcriptional regulation.

BACKGROUND: Dysregulation of the common stress responsive transcription factor ATF3 has been causally linked to many important human diseases such as cancer, atherosclerosis, infections, and hypospadias. Although it is believed that the ATF3 transcription activity is central to its cellular functions, how ATF3 regulates gene expression remains largely unknown. Here, we employed ATF3 wild-type and knockout isogenic cell lines to carry out the first comprehensive analysis of global ATF3-binding profiles in the human genome under basal and stressed (DNA damage) conditions. RESULTS: Although expr

Transcriptional effects of E3 ligase atrogin-1/MAFbx on apoptosis, hypertrophy and inflammation in neonatal rat cardiomyocytes.

Atrogin-1/MAFbx is an ubiquitin E3 ligase that regulates myocardial structure and function through the ubiquitin-dependent protein modification. However, little is known about the effect of atrogin-1 activation on the gene expression changes in cardiomyocytes. Neonatal rat cardiomyocytes were infected with adenovirus atrogin-1 (Ad-atrogin-1) or GFP control (Ad-GFP) for 24 hours. The gene expression profiles were compared with microarray analysis. 314 genes were identified as differentially expressed by overexpression of atrogin-1, of which 222 were up-regulated and 92 were down-regulated. Joint immobilization prevents murine osteoarthritis and reveals the highly mechanosensitive nature of protease expression in vivo.

OBJECTIVE: Mechanical joint loading is critical for the development of osteoarthritis (OA). Although once regarded as a disease of cartilage attrition, OA is now known to be controlled by the expression and activity of key proteases, such as ADAMTS-5, that drive matrix degradation. This study was undertaken to investigate the link between protease expression and mechanical joint loading in vivo. METHODS: We performed a microarray analysis of genes expressed in the whole joint following surgical induction of murine OA (by cutting the medial meniscotibial ligament). Gene expression changes were
