Insights into the genetic aetiology of diabetes in Africa.

Despite the high public health burden of diabetes in Africa, research into its genetic aetiology has been slow, limiting the continent's ability to benefit from an emerging era of diabetes precision medicine. Some progress is evident. In monogenic diabetes, where a molecular diagnosis enables tailored treatment, two cases from Africa successfully illustrate this approach despite the absence of routine affordable genetic testing, and control data from diverse African populations. Although limited, genome-wide association studies from African populations have discovered novel African-specific ty

Genetic links between female reproductive milestones and major depressive disorder: evidence from a common genetic factor model.

BACKGROUND: Major depressive disorder (MDD) is more prevalent in women and has been linked to female reproductive timing. However, the genetic mechanisms underlying these associations remain unclear. Reproductive milestones are temporally structured and biologically interrelated, suggesting a shared genetic architecture that may influence MDD susceptibility. METHODS: Using large-scale genome-wide association study (GWAS)data, we analyzed five key female reproductive milestones and their genetic relationships with MDD. We applied linkage disequilibrium score regression (LDSC), Genomic Structur…

Genetic mechanisms of primary biliary cholangitis and its association with immune cells using mendelian randomization and biological annotation.

The incidence of primary biliary cholangitis (PBC) is on the rise, attributed to autoimmune-mediated bile duct damage in genetically susceptible individuals triggered by environmental factors. The close association between immune cell infiltration around the intrahepatic bile ducts and bile duct damage, and the causal and potential relationships of immune cells in the pathogenesis of PBC, remain unclear. Utilizing summary statistics from published genome-wide association studies, we evaluated the association between 731 immune cell traits and PBC through 2-sample Mendelian randomization (MR) a

Perfluorooctane Sulfonate Exposure and Peripheral Artery Disease: Mendelian Randomization and Integrative Multi-Omics Systems Toxicology With Machine Learning.

Epidemiological and experimental studies have linked legacy and emerging PFAS to adverse cardiovascular outcomes, yet most evidence remains observational and is susceptible to confounding, measurement error, and reverse causation. This study used European-ancestry GWAS data and two-sample Mendelian randomization to assess potential causal associations between PFOA or PFOS exposure and five cardiovascular diseases. For significant MR findings, network toxicology was integrated with transcriptomic data to identify candidate targets and pathways, followed by machine learning-based prioritization.

Revealing the Shared Genetic Architecture of Metabolic Dysfunction-Associated Steatotic Liver Disease-Related Traits Through Genomic Structural Equation Modeling.

Although individual traits related to metabolic dysfunction-associated steatotic liver disease (MASLD) have been investigated through large-scale genome-wide association studies (GWASs), the shared genetic susceptibility across these traits remains unclear. We therefore conducted a multivariate GWAS of key MASLD-related traits to elucidate their common genetic architecture. We applied genomic structural equation modeling to model a latent genetic factor (MASLD-F) underlying genetically correlated MASLD-related traits, leveraging their GWAS-derived genetic correlations. We then performed functi

Genome-wide investigation highlights global and local pleiotropy linking neurodevelopmental disorders to acquired hearing problems.

BACKGROUND: Neurodevelopmental disorders have been associated with hearing problems (HP) later in life, but there is limited information regarding their shared biology. METHODS: We leveraged large-scale genome-wide datasets to estimate genetic correlation (global and local), polygenic overlap, and locus-specific pleiotropy among HP, autism spectrum disorder (ASD), attention-deficit/hyperactivity disorder (ADHD), and Tourette syndrome (TS). Then, we investigated shared molecular functions, biological processes, and cellular components, and performed a drug-repurposing analysis to identify comp

Integrated Genome-wide Association and Single-cell Transcriptomic Analysis Identifies OAT as Therapeutic Targets for Periodontitis.

Periodontitis is a chronic inflammatory disease affecting tooth-supporting tissues. Beyond microbial dysbiosis, host metabolic regulation plays a critical role. This study identified ornithine aminotransferase (OAT), a mitochondrial enzyme in amino acid metabolism, as associated with altered fibroblast phenotypes and metabolic profiles in periodontitis. Integrative genetic analysis showed a putative causal relationship between increased OAT expression and disease risk. Single-cell RNA-Seq revealed OAT enrichment in fibroblasts, especially in subsets with inflammatory and matrix-remodeling char

Evaluating the Relationship between Cathepsins and Papillary Thyroid Carcinoma: A Mendelian Randomization Study.

BACKGROUND: Papillary Thyroid Carcinoma (PTC) is the most common thyroid cancer, with an etiology and progression that are not fully understood. Research suggests a link between cathepsins and PTC, but the causal nature of this link is unclear. This study uses Mendelian Randomization (MR) to investigate if cathepsins causally influence PTC risk. METHODS: We applied univariable and multivariable MR analyses using genetic variants as proxies for cathepsin levels. Genetic data for cathepsins were sourced from the INTERVAL study, while PTC data came from the Finnish Genome-Wide Association Study …

Context-specific eQTLs provide deeper insight into causal genes underlying shared genetic architecture of COVID-19 and idiopathic pulmonary fibrosis.

Most genetic variants identified through genome-wide association studies (GWASs) are suspected to be regulatory in nature, but only a small fraction colocalize with expression quantitative trait loci (eQTLs, variants associated with expression of a gene). Therefore, it is hypothesized but largely untested that integration of disease GWAS with context-specific eQTLs will reveal the underlying genes driving disease associations. We used colocalization and transcriptomic analyses to identify shared genetic variants and likely causal genes associated with critically ill COVID-19 and idiopathic pul

Linkage disequilibrium score regression identifies genetic correlations between hepatocellular carcinoma and clinically relevant traits.

Hepatocellular carcinoma (HCC) mortality is increasing globally, partly due to the growing prevalence of nonviral liver diseases. Genome-wide association studies (GWAS) have identified genetic variants associated with HCC development. Leveraging GWAS summary statistics and linkage disequilibrium score regression (LDSR), we investigated disease co-development with hepatitis C virus-negative (HCV-negative) HCC to provide unique insights into HCC etiology and prioritize relationships for further causal inquiry. We utilized the LDSR statistical framework to estimate the genetic correlation and her

The genetics of neurodegenerative diseases is the genetics of age-related damage clearance failure.

In this perspective we draw together the data from the genome wide association studies for Alzheimer's disease, Parkinson's disease and the tauopathies and reach the conclusion that in each case, most of the risk loci are involved in the clearance of the deposited proteins: in Alzheimer's disease, the microglial removal of Aβ, in the synucleinopathies, the lysosomal clearance of synuclein and in the tauopathies, the removal of tau protein by the ubiquitin proteasome. We make the point that most loci identified through genome wide association studies are not strictly pathogenic but rather relat
