Platelet activation and bacterial interactions: linking immunothrombosis to infectious pathophysiology.

Platelets are anucleate blood cells traditionally recognized for their central role in hemostasis but are now appreciated as active sentinels of innate immunity. During bacterial infection, platelets sense pathogen-associated molecular patterns and immune complexes through toll-like receptors, FcγRIIA, complement receptors, and adhesive integrins. This recognition enables platelets to interact with leukocytes and endothelial cells and to release proinflammatory and procoagulant mediators. While these responses contribute to pathogen containment, they also promote thromboinflammation and microv

Organic acids - a dual-action mode influencing both host and pathogen integrity to reduce Salmonella infection.

In this study, we aimed to describe the mechanism by which a blend of organic acids (AuraShield - As) prevents Salmonella Typhimurium SE10/72 infection of primary chicken caecal epithelial cells (PECC) and caecal biopsies. First, our results show a MIC of 0.50% and an MBC of 1% against Salmonella Typhimurium SE10/72, with a sub-inhibitory concentration of 0.25% selected for further experiments. At this level, in vitro and ex vivo, As significantly decreased the ability of S. Typhimurium to attach to and invade PECC cells and caecal biopsies alongside bacterial motility and biofilm formation. T

Host prenylation promotes membrane association of the Salmonella effector GtgE during infection.

The Rab32 antimicrobial pathway restricts Salmonella Typhi replication in murine macrophages. The broad-host pathogen Salmonella Typhimurium has evolved a solution to disrupt this pathway via effector proteins, including GtgE. The presence of a C-terminal CaaX motif in GtgE prompted the hypothesis that the effector was prenylated by the host. Here, we show that the CaaX motif enhances GtgE membrane association and intracellular targeting. Substitution of the C-terminal cysteine (C225S) caused diffused cytosolic distribution and loss of membrane enrichment. Inhibition of host isoprenoid biosynt

Targeting USP22 to promote K63-linked ubiquitination and degradation of SARS-CoV-2 nucleocapsid protein.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) generally hijacks the cellular machinery of host cells for survival. However, how SARS-CoV-2 employs the host's deubiquitinase to facilitate virus replication remains largely unknown. In this study, we identified the host deubiquitinase USP22 as a crucial regulator of the expression of SARS-CoV-2 nucleocapsid protein (SARS-CoV-2 NP), which is essential for SARS-CoV-2 replication. We demonstrated that SARS-CoV-2 NP proteins undergo ubiquitination-dependent degradation in host cells, while USP22 interacts with SARS-CoV-2 NP and downreg

Spatial Proteomics Using BiFCPL Identifies Regulators of DMV Formation Involved in Coronavirus Replication.

β-Coronaviruses hijack host factors to remodel host endo-membranes to form double membrane vesicles (DMVs), which act as central hubs for the replication of viral genomes. Understanding the molecular mechanism underlying DMV formation is critical for developing effective antiviral strategies and has garnered significant attention. However, the host factors involved in DMV formation remain scanty. Here, we employed a bimolecular fluorescence complementation-based proximity labeling (BiFCPL) strategy to investigate the proteome of DMVs generated by co-expression of SARS-CoV-2 NSP3 and NSP4. Our

Engineering virus-like particles for safe and versatile modeling of SARS-CoV-2 host interaction and immune escape.

The continuous evolution of SARS-CoV-2 poses global health challenges. A safe, rapid, and versatile method for assessing functions of Spike protein mutations in ACE2 receptor binding and immune evasion would be highly valuable. To address this, we engineered a transcription- and replication-competent virus-like particle (trVLP) derived from the Sindbis virus, pseudotyped with the SARS-CoV-2 receptor binding domain (RBD). This trVLP exclusively propagates in BHK-21 cells engineered to express both RNA replicase and human ACE2, providing a controllable, safe model of SARS-CoV-2 RBD-ACE2 interact

IFIT3 inhibits transmissible gastroenteritis virus (TGEV) infection by promoting the phosphorylation of TBK1 and STAT1, which enhances the innate immune response.

TGEV mainly infects pig small intestinal epithelium, resulting in vomiting, diarrhea, dehydration, and death. IFIT3 is involved in resisting viral infection and is an innate immune regulator. In this study, we determined that TGEV infection could induce IFIT3 expression. The overexpression of IFIT3 inhibited TGEV infection, promoted the phosphorylation of TBK1 and STAT1, and upregulated the transcription of IFN-β and interferon-stimulated genes (ISGs). Conversely, knockdown of IFIT3 decreased the activation of the interferon immune response. Blocking the JAK-STAT1 pathway inhibited the transmi

Porphyromonas gingivalis hijacks mitophagy and lysosomal function to persist in endothelial cells.

INTRODUCTION: The direct infection of endothelial cells by Porphyromonas gingivalis (P. gingivalis), a keystone periodontal pathogen, has been implicated in the development of atherosclerosis. While non-selective autophagy facilitates its intracellular persistence in endothelial cells, the role of selective autophagy in this process remains unclear. This study investigated whether P. gingivalis hijacks mitophagy and lysosomes to persist in endothelial cells. METHODS: Human aortic endothelial cells (HAECs) were infected with P. gingivalis for 24 h. Mitophagy was detected by Western Blotting (W

Pivotal role of voltage-dependent anion channel 2 in pyroptosis induced by spring viremia of carp virus in fish cells.

Spring viremia of carp virus (SVCV) represents a significant threat to cyprinids, particularly common carp (Cyprinus carpio). The disease caused by this virus is characterized by tissue necrosis and petechial hemorrhages. However, the pathogenesis of SVCV infection remains poorly understood. Pyroptosis, a recently identified form of programmed cell death, plays a crucial role in host-pathogen interactions and provides a novel approach for studying inflammation-related diseases. This study demonstrates that SVCV induces gasdermin Eb-dependent pyroptosis through activation of NLRP3 and initiatio…

Transcriptome profiling reveals that the host BRD4 protein facilitates African swine fever virus infection and suppresses inflammatory cytokine expression by downregulating transcriptional regulatory signaling pathways.

The African swine fever virus (ASFV), a complex DNA virus belonging to the Asfarviridae family, is a significant threat to the global swine industry because of its high mortality rates and impact on international trade. The establishment of a stable and efficient cell culture model of ASFV in vitro is helpful for the development of effective vaccines. Several passaged cell lines supporting ASFV replication have been reported to meet the scientific purpose of serial passage of ASFV to a certain extent, but it remains to be determined whether gene expression is lost or whether immunogenicity cha

Ubiquitination: A Double-Edged Mechanism in Coronavirus Infections.

The emergence of SARS-CoV-2 in late 2019 had a profound impact on public health, leading to the global COVID-19 pandemic. This viral outbreak has significantly heightened interest in coronaviruses, accelerating research into their pathogenesis. Ubiquitination, a common Posttranslational protein modification, plays a crucial role in processes such as protein localization, metabolism, and degradation. During coronavirus invasion and disease progression, complex interactions involving ubiquitination are at play. On one hand, the host utilizes ubiquitination to activate innate immune signaling pat

Ubiquitin-like modifier-activating enzyme 1 interacts with Zika virus NS5 and promotes viral replication in the infected cell.

Translation errors, impaired folding or environmental stressors (e.g. infection) can all lead to an increase in the presence of misfolded proteins. These activate cellular responses to their removal, including intracellular protein degradation activities. Protein ubiquitylation is involved in two major degradation pathways, the ubiquitin-proteasome system and selective autophagy. In humans, the ubiquitin-like modifier-activating enzyme 1 (UBA1) is the primary E1 enzyme in the ubiquitin conjugation cascade. Viruses have evolved to exploit protein degradation pathways to complete their infection

Infectious bronchitis-virus-like QX strain transmission, pathogenesis, replication, and host miRNA biogenesis pathway hijacking mechanism.

The infectious bronchitis virus (IBV) is an acute, highly contagious, single-stranded RNA (ssRNA) gammacoronavirus mainly transmitted to chickens through the intranasal route. Positive-sense ssRNA viruses primarily act on mRNA and enhance the replication of viral copies. We identified the nasal entry site of the IBV-QX strain and provided insights into the minimal viral replication in systemic organs. Here is an overview of its entry mechanisms and tropism in systemic organ tissues. It enters the host cells via spike proteins, which bind to highly expressed receptors in respiratory, renal, and

From the Gut to the Brain: Transcriptomic Insights into Neonatal Meningitis Escherichia coli Across Diverse Host Niches.

Neonatal Meningitis-causing Escherichia coli (NMEC) is the leading cause of neonatal meningitis and exhibits remarkable adaptability to diverse host environments. Understanding its transcriptional responses across different host niches is crucial for deciphering pathogenesis and identifying potential therapeutic targets. We performed a comparative transcriptomic analysis of NMEC RS218, the prototype strain of NMEC, under four distinct host-mimicking conditions: colonic fluid (CF), serum (S), human brain endothelial cells (HBECs), and cerebrospinal fluid (CSF). Differential gene expression anal

ZYG11B suppresses multiple enteroviruses by triggering viral VP1 degradation.

UNLABELLED: Enterovirus 71 (EV71) is a major cause of hand, foot, and mouth disease, particularly affecting pediatric populations worldwide. The role of ZYG11B, a CUL2-complex-associated E3 ubiquitin ligase from the Zyg-11 family, in antiviral defense against EV71 remains unclear. To our knowledge, this study is the first to reveal that ZYG11B targets EV71 VP1 for proteasomal degradation via the ubiquitin-proteasome pathway, with CRL2ZYG11B complex activity specifically driving K33-linked ubiquitination. Mass spectrometry and immunoprecipitation analyses confirmed the interaction between ZYG11

GCN2 enhances host survival and drives eIF2α phosphorylation during mouse adenovirus type 1 infection.

The integrated stress response (ISR) is a cellular signaling pathway that reduces protein synthesis in the face of cellular stress, including viral infection. Two eukaryotic initiation factor 2α (eIF2α) kinases, protein kinase R (PKR) and general control nonderepressible 2 (GCN2), are commonly activated during viral infections. Mouse adenovirus type 1 (MAV-1) infection leads to a steep reduction of PKR levels by proteasomal degradation. We assayed whether GCN2, a sensor of amino acid starvation and UV damage, plays a role in the ISR to MAV-1 infection. There was more phosphorylated GCN2 in MAV

Environmental cues in different host niches shape the survival fitness of Staphylococcus aureus.

The ability of Staphylococcus aureus to adapt and thrive in diverse host niches adds to the challenge in combating this ubiquitous pathogen. While extensive research has been pursued on the adaptive mechanisms of methicillin-resistant S. aureus (MRSA) in various infection models, a comprehensive analysis of its fitness across different host niches is lacking. In this study, we employ transposon sequencing to analyze the adaptive strategies of MRSA in various infection niches. Our analysis encompasses a cell model that mimics an intracellular niche, human blood, which represents a major extrace

Heart failure-induced microbial dysbiosis contributes to colonic tumour formation in mice.

AIMS: Heart failure (HF) and cancer are the leading causes of death worldwide. Epidemiological studies revealed that HF patients are prone to develop cancer. Preclinical studies provided some insights into this connection, but the exact mechanisms remain elusive. In colorectal cancer (CRC), gut microbial dysbiosis is linked to cancer progression and recent studies have shown that HF patients display microbial dysbiosis. This current study focussed on the effects of HF-induced microbial dysbiosis on colonic tumour formation. METHODS AND RESULTS: C57BL/6J mice were subjected to myocardial infar

SARS-CoV-2, periodontal pathogens, and host factors: The trinity of oral post-acute sequelae of COVID-19.

COVID-19 as a pan-epidemic is waning but there it is imperative to understand virus interaction with oral tissues and oral inflammatory diseases. We review periodontal disease (PD), a common inflammatory oral disease, as a driver of COVID-19 and oral post-acute-sequelae conditions (PASC). Oral PASC identifies with PD, loss of teeth, dysgeusia, xerostomia, sialolitis-sialolith, and mucositis. We contend that PD-associated oral microbial dysbiosis involving higher burden of periodontopathic bacteria provide an optimal microenvironment for severe acute respiratory syndrome coronavirus 2 (SARS-CoV
