EPHA4 signaling dysregulation links abnormal locomotion and the development of idiopathic scoliosis.

Idiopathic scoliosis (IS) is the most common form of spinal deformity with unclear pathogenesis. In this study, we first reanalyzed the loci associated with IS, drawing upon previous studies. Subsequently, we mapped these loci to candidate genes using either location-based or function-based strategies. To further substantiate our findings, we verified the enrichment of variants within these candidate genes across several large IS cohorts encompassing Chinese, East Asian, and European populations. Consequently, we identified variants in the EPHA4 gene as compelling candidates for IS. To confirm

Emerging evidence of context-dependent synapse elimination by phagocytes in the CNS.

Precise synapse elimination is essential for the establishment of a fully developed neural circuit during brain development and higher function in adult brain. Beyond immune and nutrition support, recent groundbreaking studies have revealed that phagocytic microglia and astrocytes can actively and selectively eliminate synapses in normal and diseased brains, thereby mediating synapse loss and maintaining circuit homeostasis. Multiple lines of evidence indicate that the mechanisms of synapse elimination by phagocytic glia are not universal but rather depend on specific contexts and detailed neu

Schistosoma mansoni Fibroblast Growth Factor Receptor A Orchestrates Multiple Functions in Schistosome Biology and in the Host-Parasite Interplay.

Stem cells play significant roles in driving the complex life cycle of Schistosoma mansoni. Fibroblast growth factor (FGF) receptor A (SmFGFRA) is essential for maintaining the integrity of schistosome stem cells. Using immunolocalization, we demonstrated that SmFGFRA was distributed abundantly in germinal/stem cells of different S. mansoni life stages including eggs, miracidia, cercariae, schistosomula and adult worms. Indeed, SmFGFRA was also localized amply in embryonic cells and in the perinuclear region of immature eggs; von Lichtenberg's layer and the neural mass of mature eggs; the cili

Estrogen and progesterone priming induces lordosis in female rats by reversing the inhibitory influence of the infralimbic cortex on neuronal activity of the lateral septal nucleus.

The lateral septal nucleus (LSN) exerts inhibitory control over lordosis in female rats, but the influence of forebrain structures, such as prelimbic (PL) and infralimbic (IL) regions of the medial prefrontal cortex (mPFC), on LSN activity during sexual receptivity is unknown. We hypothesized that the neural responsivity of these connections may differ depending on sexual receptivity. Gonadally intact female Wistar rats received sequential priming injections of estradiol and progesterone (E2-P4). The presence of lordosis was then confirmed by exposing the female rats to a sexually experienced

Characterization of bipolar disorder patient-specific induced pluripotent stem cells from a family reveals neurodevelopmental and mRNA expression abnormalities.

Bipolar disorder (BD) is a common neuropsychiatric disorder characterized by chronic recurrent episodes of depression and mania. Despite evidence for high heritability of BD, little is known about its underlying pathophysiology. To develop new tools for investigating the molecular and cellular basis of BD, we applied a family-based paradigm to derive and characterize a set of 12 induced pluripotent stem cell (iPSC) lines from a quartet consisting of two BD-affected brothers and their two unaffected parents. Initially, no significant phenotypic differences were observed between iPSCs derived fr

The temporal dynamics of coherent motion processing in autism spectrum disorder: evidence for a deficit in the dorsal pathway.

Individuals with autism spectrum disorder (ASD) show impairments in processing coherent motion which have been proposed to be linked to a general deficit in the dorsal visual pathway. However, few studies have investigated the neural mechanisms underlying coherent motion processing in ASD. Thus, the aim of this study was to further test the hypothesis of a dorsal pathway deficit in ASD using visual evoked potentials (VEPs). 16 children and adolescents with ASD and 12 typically developing controls were examined with VEPs elicited by a random dot kinematogram. After an initial experimental seque

Integrative control of the lower urinary tract: preclinical perspective.

Storage and periodic expulsion of urine is regulated by a neural control system in the brain and spinal cord that coordinates the reciprocal activity of two functional units in the lower urinary tract (LUT): (a) a reservoir (the urinary bladder) and (b) an outlet (bladder neck, urethra and striated muscles of the urethral sphincter). Control of the bladder and urethral outlet is dependent on three sets of peripheral nerves: parasympathetic, sympathetic and somatic nerves that contain afferent as well as efferent pathways. Afferent neurons innervating the bladder have A-delta or C-fibre axons.

Surgery for idiopathic retroperitoneal fibrosis by ureterolysis and ureteric protection with a posterior pre-peritoneal fat flap.

Tinnitus is hypothesized to be an auditory phantom phenomenon resulting from spontaneous neuronal activity somewhere along the auditory pathway. The neural abnormalities underlying tinnitus are largely unknown. We evaluated the functional characteristics and the auditory system synchronization using Auditory Brainstem Response (ABR) in normal hearing tinnitus patients. In this observational comparative cross-sectional study, patients with chief complaints of Tinnitus and equal number of age and sex matched controls without hearing loss and tinnitus were enrolled. All patients underwent a full

Simultaneous intrastriatal and intranigral grafting (double grafts) in the rat model of Parkinson's disease.

Experimental and clinical studies of neural transplantation in Parkinson's disease have focused on the placement of fetal dopaminergic grafts not in their ontogenic site (substantia nigra) but in the main nigral target area (striatum). The reason for this is the apparent inability of intranigral nigral grafts to extend axons for long distances reinnervating the ipsilateral striatum. This review presents previous work by our laboratory [I. Mendez, M. Hong, Reconstruction of the striato-nigro-striatal circuitry by simultaneous double dopaminergic grafts: a tracer study using fluorogold and horse

Cholinergic control mechanisms for immunoreactive motilin release and motility in the canine duodenum.

The relationship of immunoreactive (IR) motilin release from the duodenum to duodenal motility changes was investigated in anaesthetized dogs. Stimulation of one or both vagi at 5 or 15 Hz or field stimulation of intrinsic duodenal nerves produced significant increases in duodenal vein IR motilin concentrations and accompanying increases in duodenal motility. However, only stimulation of both vagi at 15 Hz produced significant changes in peripheral venous concentrations of IR motilin. These occurred after a delay at a time when both the duodenum and the antrum were quiescent. Either hexamethon

A corticohypothalamic circuit that mediates maternal awakening to pup cries.

Maternal caregiving requires sensory vigilance to ensure offspring survival, particularly during sleep. Infant cries are salient sensory cues that reliably trigger maternal arousal and caregiving responses in both humans and dams, but the underlying neural circuitry remains poorly defined. Here we identify a corticohypothalamic circuit in mice that mediates maternal awakening in response to pup calls. We show that layer 6 (L6) neurons in the temporal association cortex and ectorhinal cortex (TeA/Ect) projecting to the preoptic area (POA) exhibit enhanced responsiveness to pup calls after the t

Paraventricular nucleus CRH neurons regulate acute lung injury via sympathetic nerve-neutrophil axis.

Acute lung injury (ALI) and acute respiratory distress syndrome (ARDS) are severe conditions with high morbidity and mortality with limited effective therapies. Neuroimmune interactions play a critical role in lung homeostasis, but it remains unclear if specific brain regions regulate lung inflammation. Here, we perform anatomical tracing, chemogenetic modulation, and pharmacological interventions in male mice and identify a neural circuit from corticotropin-releasing hormone neurons in the paraventricular nucleus of the hypothalamus (CRHPVN neurons) to the lung. The activation of these neuron

A review of the neural basis underlying the acoustic startle response with a focus on recent developments in mammals.

The startle response consists of whole-body muscle contractions, eye-blink, accelerated heart rate, and freezing in response to a strong, sudden stimulus. It is evolutionarily preserved and can be observed in any animal that can perceive sensory signals, indicating the important protective function of startle. Startle response measurements and its alterations have become a valuable tool for exploring sensorimotor processes and sensory gating, especially in the context of pathologies of psychiatric disorders. The last reviews on the neural substrates underlying acoustic startle were published a

NAMPT as a Therapeutic Target against Stroke.

Nicotinamide phosphoribosyltransferase (NAMPT), also an adipokine known as visfatin, acts via enzymatic activity to synthesize nicotinamide mononucleotide (NMN) and then to maintain homeostasis of nicotinamide adenine dinucleotide (NAD), which plays a dual role in energy metabolism and biological signaling. Of note, the NAMPT metabolic pathway connects NAD-dependent sirtuin (SIRT) signaling, constituting a strong intrinsic defense system against various stresses. Most recently, studies have demonstrated several mechanisms by which NAMPT might serve as a therapeutic target against ischemic stro

Stem cell-like dog placenta cells afford neuroprotection against ischemic stroke model via heat shock protein upregulation.

In this study, we investigated the dog placenta as a viable source of stem cells for stroke therapy. Immunocytochemical evaluation of phenotypic markers of dog placenta cells (DPCs) cultured in proliferation and differentiation medium revealed that DPCs expressed both stem cell and neural cell markers, respectively. Co-culture with DPCs afforded neuroprotection of rat primary neural cells in a dose-dependent manner against oxygen-glucose deprivation. Subsequent in vivo experiments showed that transplantation of DPCs, in particular intravenous and intracerebral cell delivery, produced significa

The DNA glycosylases OGG1 and NEIL3 influence differentiation potential, proliferation, and senescence-associated signs in neural stem cells.

Embryonic neural stem cells (NSCs) exhibit self-renewal and multipotency as intrinsic characteristics that are key parameters for proper brain development. When cells are challenged by oxidative stress agents the resulting DNA lesions are repaired by DNA glycosylases through the base excision repair (BER) pathway as a means to maintain the fidelity of the genome, and thus, proper cellular characteristics. The functional roles for DNA glycosylases in NSCs have however remained largely unexplored. Here we demonstrate that RNA knockdown of the DNA glycosylases OGG1 and NEIL3 decreased NSC differe

Zebrafish con/disp1 reveals multiple spatiotemporal requirements for Hedgehog-signaling in craniofacial development.

BACKGROUND: The vertebrate head skeleton is derived largely from cranial neural crest cells (CNCC). Genetic studies in zebrafish and mice have established that the Hedgehog (Hh)-signaling pathway plays a critical role in craniofacial development, partly due to the pathway's role in CNCC development. Disruption of the Hh-signaling pathway in humans can lead to the spectral disorder of Holoprosencephaly (HPE), which is often characterized by a variety of craniofacial defects including midline facial clefting and cyclopia 12. Previous work has uncovered a role for Hh-signaling in zebrafish dorsal

Brain-Inspired Multi-Pathway Motion Decision-Making for Obstacle Avoidance of Humanoid Arms.

Achieving rapid and accurate obstacle avoidance in complex and dynamic environments remains a significant challenge for robots. To enhance the adaptability and flexibility of humanoid arms for obstacle avoidance, a brain-inspired multi-pathway motion decision-making method is proposed to modulate rational planning and habitual actions of humanoid arms. Firstly, a novel framework integrating both a slow and a fast pathway is designed for motion decision-making tasks. Imitating the rational planning function of the prefrontal cortex, the slow pathway employs an improved planning approach based o

Linking systemic metabolic state to long-term motor memory: Insights from respiratory exchange ratio and glucose manipulation.

Respiration is a crucial metabolic process that converts macronutrients, carbohydrates and fats, and oxygen into energy and carbon dioxide to support motor actions. Moreover, the brain is a significant energy consumer, accounting for large portions of the body's total energy expenditure and relying primarily on carbohydrates for neural activity and plasticity. However, it is not known whether gas composition in breathing can serve as an indicator of neural activity and plasticity as they can for movement intensity. In human reaching movement tasks, we evaluated time-constants of sensorimotor l
