Virtual reality stimulation for neuroprotection and neuroenhancement of vision in optic neuropathy patients: a prospective clinical trial.

BACKGROUND: This study aimed to assess the safety and efficacy of virtual reality (VR) visual stimulation on neuroprotection and neuroenhancement in optic neuropathy patients. METHODS: Open-label, prospective, phase I clinical trial, enrolling 21 participants with glaucoma, plus 1 patient with ischemic optic neuropathy after hydrocephalus. Participants received commercially available VR headsets (HTC Vive or Oculus Rift) for home use loaded with a study-specific visual stimulus program. The VR visual stimulation protocol consisted of up to 1-hour daily sessions, 5 days per week, for 12-week cy

Homeostatic regulation of perisynaptic matrix metalloproteinase 9 (MMP9) activity in the amblyopic visual cortex.

Dark exposure (DE) followed by light reintroduction (LRx) reactivates robust synaptic plasticity in adult mouse primary visual cortex (V1), which allows subsequent recovery from amblyopia. Previously we showed that perisynaptic proteolysis by MMP9 mediates the enhancement of plasticity by LRx in binocular adult mice (Murase et al., 2017). However, it was unknown if a visual system compromised by amblyopia could engage this pathway. Here we show that LRx to adult amblyopic mice induces perisynaptic MMP2/9 activity and extracellular matrix (ECM) degradation in deprived and non-deprived V1. Indee

A brain-wide functional map of the serotonergic responses to acute stress and fluoxetine.

Central serotonin (5-HT) orchestrates myriad cognitive processes and lies at the core of many stress-related psychiatric illnesses. However, the basic relationship between its brain-wide axonal projections and functional dynamics is not known. Here we combine optogenetics and fMRI to produce a brain-wide 5-HT evoked functional map. We find that DRN photostimulation leads to an increase in the hemodynamic response in the DRN itself, while projection areas predominately exhibit a reduction of cerebral blood volume mirrored by suppression of cortical delta oscillations. We find that the regional

Increased Prefrontal Activity with Aging Reflects Nonspecific Neural Responses Rather than Compensation.

Elevated prefrontal cortex activity is often observed in healthy older adults despite declines in their memory and other cognitive functions. According to one view, this activity reflects a compensatory functional posterior-to-anterior shift, which contributes to maintenance of cognitive performance when posterior cortical function is impaired. Alternatively, the increased prefrontal activity may be less efficient or less specific because of structural and neurochemical changes accompanying aging. These accounts are difficult to distinguish on the basis of average activity levels within brain

Neural correlates of preferred activities: development of an interest-specific go/nogo task.

The activities we choose to spend our leisure time with are intrinsically motivating and vary across individuals. Yet it is unknown how impulse control or neural activity changes when processing a preferred stimulus related to a hobby or interest. Developing a task that assesses the response to preferred interests is of importance as it would be relevant to a range of psychiatric disorders that have hyper- or hypo-arousal to such cues. During functional Magnetic Resonance Imaging (fMRI), 39 healthy adults completed a novel task to test approach behavior and cognitive control to cues that were

Targeted Reactivation during Sleep Differentially Affects Negative Memories in Socially Anxious and Healthy Children and Adolescents.

Cognitive models propose a negative memory bias as one key factor contributing to the emergence and maintenance of social anxiety disorder (SAD). The long-term consolidation of memories relies on memory reactivations during sleep. We investigated in SAD patients and healthy controls the role of memory reactivations during sleep in the long-term consolidation of positive and negative information. Socially anxious and healthy children and adolescents learnt associations between pictures showing ambiguous situations and positive or negative words defining the situations' outcome. Half of the word

Respiratory and hemodynamic contributions to emotion-related pre-syncopal vasovagal symptoms.

Vasovagal reactions are conventionally understood as resulting from systemic changes in cardiovascular activity; however, there exists a complementary perspective focused on specific changes in cerebral vasoconstriction associated with hyperventilation-induced hypocapnia. The present study investigated the role of cardiovascular and respiratory activity in self-reported pre-syncopal vasovagal reactions to a surgery video in a sample of 49 healthy women. Participants who indicated more previous real-life episodes of dizziness reported experiencing significantly more symptoms in the laboratory c

Processing of bilateral versus unilateral conditions: evidence for the functional contribution of the ventral attention network.

Processing of multiple or bilateral conditions presented simultaneously in both hemifields reflects the natural mode of perception in our multi-target environment, but is not yet completely understood. While region-of-interest based studies in healthy subjects reported single cortical areas as the right inferior parietal lobe (IPL) or temporoparietal junction (TPJ) to process bilateral conditions, studies in extinction patients with reduced ability in this regard suggested the right superior temporal cortex to hold a key role. The present fMRI study on healthy subjects aimed at resolving these

Altered amygdala-prefrontal response to facial emotion in offspring of parents with bipolar disorder.

This study aimed to identify neuroimaging measures associated with risk for, or protection against, bipolar disorder by comparing youth offspring of parents with bipolar disorder versus youth offspring of non-bipolar parents versus offspring of healthy parents in (i) the magnitude of activation within emotional face processing circuitry; and (ii) functional connectivity between this circuitry and frontal emotion regulation regions. The study was conducted at the University of Pittsburgh Medical Centre. Participants included 29 offspring of parents with bipolar disorder (mean age = 13.8 years;

The role of the amygdala during emotional processing in Huntington's disease: from pre-manifest to late stage disease.

BACKGROUND: Deficits in emotional processing can be detected in the pre-manifest stage of Huntington's disease and negative emotion recognition has been identified as a predictor of clinical diagnosis. The underlying neuropathological correlates of such deficits are typically established using correlative structural MRI studies. This approach does not take into consideration the impact of disruption to the complex interactions between multiple brain circuits on emotional processing. Therefore, exploration of the neural substrates of emotional processing in pre-manifest HD using fMRI connectivi

Retinal ganglion cells projecting to the accessory optic system in optokinetic blind albinotic rats are direction-selective.

The optokinetic deficits in albinotic rats and ferrets are caused by the loss of direction selectivity in the accessory optic system (AOS). However, the underlying mechanisms for this loss are still not clear. Here we tested the hypothesis that, in albino rats, the retinal input to the AOS lacks direction selectivity and, as a consequence, neurons in the AOS are direction non-selective. We investigated ON-center direction-selective retinal ganglion cells, the major input to the AOS, in pigmented Long Evans and albino Wistar rats using extracellular in vitro patch-clamp techniques. To visualise

Detection of third and sixth cranial nerve palsies with a novel method for eye tracking while watching a short film clip.

OBJECT: Automated eye movement tracking may provide clues to nervous system function at many levels. Spatial calibration of the eye tracking device requires the subject to have relatively intact ocular motility that implies function of cranial nerves (CNs) III (oculomotor), IV (trochlear), and VI (abducent) and their associated nuclei, along with the multiple regions of the brain imparting cognition and volition. The authors have developed a technique for eye tracking that uses temporal rather than spatial calibration, enabling detection of impaired ability to move the pupil relative to normal

Visual salience improves spatial working memory via enhanced parieto-temporal functional connectivity.

In everyday life, the brain is bombarded with a multitude of concurrent and competing stimuli. Only some of these enter consciousness and memory. Attention selects relevant signals for in-depth processing depending on current goals, but also on the intrinsic properties of stimuli. We combined behavior, computational modeling, and functional imaging to investigate mechanisms supporting access to memory based on intrinsic sensory properties. During fMRI scanning, human subjects were presented with pictures of naturalistic scenes that entailed high levels of competition between possible target ob

Functional connectivity in a frontoparietal network involving the dorsal anterior cingulate cortex underlies decisions to accept a hypothesis.

Optimally interpreting our situations and experiences frequently requires comparing the evidence supporting conflicting hypotheses and deciding which to accept. This decision is comparable to an "Aha!" moment reached during insightful problem solving. We used a probabilistic reasoning task to investigate the neural activity underlying these processes. Participants rated the probability that a given focal hypothesis, rather than its alternative, was true. Decisions to accept the focal hypothesis elicited a stronger signal than decisions to reject it in a network involving the dorsal anterior ci

Linear association between social anxiety symptoms and neural activations to angry faces: from subclinical to clinical levels.

Social anxiety disorder (SAD), which is characterized by the fear of being rejected and negatively evaluated, involves altered brain activation during the processing of negative emotions in a social context. Although associated temperament traits, such as shyness or behavioral inhibition, have been studied, there is still insufficient knowledge to support the dimensional approach, which assumes a continuum from subclinical to clinical levels of social anxiety symptoms. This study used functional magnetic resonance imaging (fMRI) to examine the neural bases of individual differences in social a

Forebrain deletion of the vesicular acetylcholine transporter results in deficits in executive function, metabolic, and RNA splicing abnormalities in the prefrontal cortex.

One of the key brain regions in cognitive processing and executive function is the prefrontal cortex (PFC), which receives cholinergic input from basal forebrain cholinergic neurons. We evaluated the contribution of synaptically released acetylcholine (ACh) to executive function by genetically targeting the vesicular acetylcholine transporter (VAChT) in the mouse forebrain. Executive function was assessed using a pairwise visual discrimination paradigm and the 5-choice serial reaction time task (5-CSRT). In the pairwise test, VAChT-deficient mice were able to learn, but were impaired in revers

Single-subject anxiety treatment outcome prediction using functional neuroimaging.

The possibility of individualized treatment prediction has profound implications for the development of personalized interventions for patients with anxiety disorders. Here we utilize random forest classification and pre-treatment functional magnetic resonance imaging (fMRI) data from individuals with generalized anxiety disorder (GAD) and panic disorder (PD) to generate individual subject treatment outcome predictions. Before cognitive behavioral therapy (CBT), 48 adults (25 GAD and 23 PD) reduced (via cognitive reappraisal) or maintained their emotional responses to negative images during fM

Biased attention to threat in paediatric anxiety disorders (generalized anxiety disorder, social phobia, specific phobia, separation anxiety disorder) as a function of 'distress' versus 'fear' diagnostic categorization.

BACKGROUND: Structural models of emotional disorders propose that anxiety disorders can be classified into fear and distress disorders. Sources of evidence for this distinction come from genetic, self-report and neurophysiological data from adults. The present study examined whether this distinction relates to cognitive processes, indexed by attention bias towards threat, which is thought to cause and maintain anxiety disorders. METHOD: Diagnostic and attention bias data were analysed from 435 children between 5 and 13 years of age; 158 had principal fear disorder (specific phobia, social pho
