The identification of phosducin as a novel candidate gene for hypertension and its role in sympathetic activation.

PURPOSE OF REVIEW: The primary objective of this review is to familiarize readers with the recent identification of phosducin (Pdc) as a novel candidate gene for stress-induced hypertension using comparative genetics and the elucidation of its role in sympathetic activation. RECENT FINDINGS: Phosducin was previously identified as a G-protein regulator expressed in the retina and pineal gland. Knowledge of its physiological role as a G-protein regulator was limited. A recent study by Beetz et al. based on comparative genetics of mice and humans establishes Pdc as a novel candidate gene for str

Amoxicillin-bearing microparticles: potential in the treatment of Listeria monocytogenes infection in Swiss albino mice.

The present study was aimed at evaluating the effectiveness of amoxicillin-bearing HSA (human serum albumin) and PLGA [poly(lactic-co-glycolic acid)] microparticles in combating Listeria monocytogenes infection in Swiss albino mice. Amoxicillin-bearing HSA microspheres were prepared by chemical cross-linking of a drug/albumin mixture with glutaraldehyde, and PLGA microspheres were prepared by the W/O/W (water-in-oil-in-water) emulsion technique. The microspheres were characterized for their size, ζ potential and entrapment efficiency using SEM (scanning electron microscopy) and a Zetasizer. Re

Confrontation with blood and disgust stimuli precipitates respiratory dysregulation in blood-injection-injury phobia.

Blood-injection-injury (BII) phobia patients sometimes faint during exposure to relevant stimuli. However, mechanisms and timing of physiological adjustments in BII phobia remain poorly understood. In a larger sample of 60 patients and 20 controls, we sought to replicate findings of a prior study demonstrating the role of hyperventilation in the phobic response. We also investigated the timing of respiratory adjustment across an extended exposure recovery period. In addition, because intense disgust is commonly reported by patients, responses to surgery films were compared to a pure disgust fi

Comparison of buprenorphine and meloxicam for postsurgical analgesia in rats: effects on body weight, locomotor activity, and hemodynamic parameters.

Buprenorphine is administered to humans and animals for postoperative pain management, although its use is associated with complications. Alternative analgesics, including the nonsteroidal antiinflammatory meloxicam, are available, but information on their postoperative effects is limited. The objective of the present study was to compare buprenorphine (0.03 mg/kg SC twice daily for 3 d) with meloxicam (2 mg/kg SC initial dose followed by 1 mg/kg SC once daily for 2 d) by assessing parameters relating to postsurgical recovery in rats that underwent surgical implantation of radiotelemetric tran

The effects of driving situation on sleepiness indicators after sleep loss: a driving simulator study.

Almost all studies of sleepy driving are carried out in driving simulators and with monotonous road conditions (no interaction with other cars). The present study investigated indicators of sleepy driving in a more challenging scenario after a night awake. 17 participants drove a high fidelity moving base driving simulator experiment while sleepiness was monitored physiologically and behaviourally. Short periods of situations of free driving (no other vehicles) alternated with short periods of following another vehicle (car following) with and without the possibility to overtake. The result sh

Effects of race and ethnicity on the incidence of venous thromboembolism.

Although there is strong evidence that the prevalence of venous thrombo-embolism (VTE) varies significantly among different ethnic/racial groups, the genetic, physiologic and/or clinical basis for these differences remain largely undefined. African-American patients have a significantly higher rate of incident VTE, particularly following exposure to a provoking risk factor such as surgery, medical illness, trauma, etc. In addition, African-Americans are more likely to be diagnosed with pulmonary embolism (PE) than deep-vein thrombosis (DVT) compared to Caucasian and other racial groups. Variations in the accuracy of the ECG based detection of obstructive sleep apnoea (OSA) for different numbers of ECG leads and categories of OSA events.

This study investigates the finding that there is a more pronounced change to ECG physiological predictors during apnoea events compared to hypopnoea events and therefore accurate detection of hypopnoea events is likely to be more challenging than detection of apnoea events. The relevant statistical analysis was conducted by generating logistic regression models from the two data sets: the first one containing only the apnoea events and controls and the second data set containing only the hypopnoea events and controls. The discriminating ability of the model from the apnoea data set (AUC = 0.9

Identification of differentially expressed genes in mandibular condylar and tibial growth cartilages using laser microdissection and fluorescent differential display: chondromodulin-I (ChM-1) and tenomodulin (TeM) are differentially expressed in mandibular condylar and other growth cartilages.

Mandibular condylar cartilage can be distinguished from articular and growth cartilages of long bones based on several differences in morphology, physiology, and function between these structures. However, there is almost no information available on the types of genes that contribute to these differences. In this study, genes that were differentially expressed in mandibular condylar and growth cartilages in 1-week-old rats were investigated using fluorescent differential display (FDD) and laser microdissection (LMD). A number of genes were identified by FDD including chondromodulin-1 (ChM-1),

[Fiberoptic endoscopic evaluation of swallowing--the Tel Aviv Voice and Swallowing Disorders Clinic].

Fiberoptic endoscopic evaluation of swallowing (FEES) involves passing a fiberoptic laryngoscope transnasally to visualize the hypopharynx, larynx, and proximal trachea in order to assess swallowing disorders. FEES has been compared with the modified barium swallow (MBS) (the presumed "gold standard"). To date, reports have demonstrated that FEES is as sensitive as, or even more sensitive, for use as a tool in swallowing assessment compared with the MBS. FEES provides the clinician with a safe, portable, effective, and valid means of evaluating individuals with swallowing disturbances. FEES al

Gonadal hormone regulation of glial fibrillary acidic protein immunoreactivity in the medial amygdala subnuclei across the estrous cycle and in castrated and treated female rats.

The medial amygdala (MeA) is a sexually dimorphic area that modulates neuroendocrine and behavioral activities and where gonadal hormones play an important role in neuron-glial and synaptic plasticity. Immunohistochemistry was used to identify the astrocytic marker glial fibrillary acidic protein (GFAP) in the different MeA subnuclei--anterodorsal (MeAD), posterodorsal (MePD) and posteroventral (MePV)--of intact female rats in the different phases of the estrous cycle and in ovariectomized females treated with hormonal substitutive therapy. Data semi-quantified by optical densitometry showed t

Applications of dosimetry modeling to assessment of neurotoxic risk.

Risk assessment procedures can be improved through better understanding and use of tissue dose information and linking tissue dose level to adverse outcomes. For volatile organic compounds, such as toluene and trichloroethylene (TCE), blood and brain concentrations can be estimated with physiologically based pharmacokinetic (PBPK) models. Acute changes in the function of the nervous system can be linked to the concentration of test compounds in the blood or brain at the time of neurological assessment. This set of information enables application to a number of risk assessment situations. For e

Influence of training on biomechanics of wheelchair propulsion.

A quasi-experimental longitudinal design was used to compare pre- and posttraining biomechanical and physiological characteristics of wheelchair propulsion in manual wheelchair users (MWCU) across fresh and fatigue states. An instrumented wheelchair ergometer, 3D motion analysis, and computerized open-circuit spirometry were used to collect joint kinetics and kinematics, handrim kinetics, propulsion temporal characteristics, and oxygen uptake pre- and posttraining during a submaximal exercise test to exhaustion. Each subject (n = 19) participated in a specific intervention program of supervise

Psychobiological markers of stress in pregnancy: 6-sulfatoxymelatonin--a longitudinal study.

Maternal stress, physical and psychological, has been associated with adverse pregnancy outcome. The pineal gland is a physiological transducer that reflects adrenergic input. In a recent pilot study, we found urinary 6-sulfatoxymelatonin, the melatonin metabolite, to be elevated after a women spent a day at work compared to levels after a day off work, a leisure day. To evaluate the value of melatonin as a marker of stress, we evaluate melatonin metabolite levels in 121 women, along with perceived anxiety levels and urinary cortisol. Urinary cortisol and maternal anxiety levels each were sign

Vascular protective effects of potassium.

Potential mechanisms responsible for the cardiovascular protection associated with high levels of dietary potassium were investigated in in vitro and in vivo studies. Elevation of extracellular potassium concentration within the physiological range inhibited free radical formation from macrophages and endothelial cells, inhibited proliferation and thymidine incorporation of vascular smooth muscle cells, and reduced platelet sensitivity to thrombin and other agonists. The effects of potassium on several well-characterized animal models of vascular pathology were also studied. Acute elevation of

EEG frequency analysis in obsessive-compulsive disorder.

The cerebral basis of obsessions and compulsions has attracted increasing attention in neuropsychology and neuropsychiatry. Clinical and imaging studies have suggested frontal lobe dysfunction in some cases of obsessive-compulsive disorder and Tourette syndrome with obsessional symptoms. We compared EEG spectral measures in 20 such patients not taking medications and 12 neurologically intact unmedicated controls. EEG was recorded from O1-A1+A2, O2-A1+A2, Fz-A1+A2, F7-C3, F8-C4, T5-O1 and T6-O2. One-minute epochs of artifact-free EEG were used for compressed spectral array and calculation of ti

Radiographic assessment of the second metatarsal: measure of first ray hypermobility.

Hypermobility of the first metatarsal cuneiform joint has been implicated as a cause of the hallux valgus deformity. The objective definition of hypermobility at this joint, however, has not been clearly defined. We used a modified Coleman block test to accentuate motion at the first metatarsal cuneiform joint in order to measure physiologic limits of motion in vivo. This motion was compared with radiographic analysis of the feet, which included the hallux valgus angle, intermetatarsal angle, and medial cortical thickening at the midshaft of the second metatarsal. This assessment was performed

Influences of gender, gonadectomy, and estrous cycle on GABA/BZ receptors and benzodiazepine responses in rats.

Benzodiazepines (BZ) and steroid hormone derivatives can potentiate the inhibitory actions of GABA through interactions with the GABAA/BZ/chloride ionophore complex. The present study examines whether the in vivo hormone milieu of rats modulates GABA/BZ receptors and/or benzodiazepine responses. The influences of gender, estrous cycle, and the diminution of steroid levels on GABA/BZ receptors and BZ anticonvulsant responses were tested by comparing these parameters in groups of intact male, intact female, orchidectomized, and ovariectomized rats. The hormonal milieu appears to modulate the GAB

Amelioration of nephrotoxicity associated with synthetic oxygen transport media.

Use of synthetic oxygen transport media offers the potential advantages of reducing requirements for coadministration of blood products and oxygen at the scene of mass casualty situations.
